Studies on the Maintenance of Bioluminescence from Stored Photobacterium phosphoreum (Photobacterium phosphoreum의 생체발광 유지도에 관한 연구)

  • The object of this work is to improve the maintenance of bioluminescence from stored Photobacterium phosphoreum in a view of developing continuous monitoring system for pollutants. The long-term experiments were performed to determine the effect of storage temperature and immobilization on the maintenance of bioluminescence and viability of P. phosphoreum. A naturally luminescent bacterium, P. phosphoreum was starved in 2.5% Nael solution at $20^{\circ}C$, $4^{\circ}C$, -$20^{\circ}C$ and -$70^{\circ}C$ for 30 days. In vivo luminescence was measured by luminometry, and total cell concentrations and concentrations of culturable and viable cells were determined by acridine orange staining, dilution plate counting, and direct viable counting, respectively. The bioluminescence emission from cells stored at 4De was maintained up to 10 days while those with starved cells at other temperature ranges decreased to background level within 3 days. In terms of viability of cells, concentrations of cells stored at $20^{\circ}C$ were rapidly decreased as a result of cell lysis, leading to a drop in culturable and viable counts while cells stored at $4^{\circ}C$ was shown viable but nonculturable state during starvation. With immobilized cells on strontium alginate, the bioluminescence showed higher maintenance than free cells and decreased with count number of nonculturable cells.

The Growth of Vibrio vulnificus in Meat Homogenates of Fish and Shellfish (어패육에서의 Vibrio vulnificus의 증식에 관한 연구)

  • The change of cell counts of Vibrio vulnificus in meat homogenates of fish and shellfish by the storage time and temperature was examined to get basic information for precautionary steps against septicemia from slices of raw fish (sashimi). Therefore, we inoculated raw and cooked meat homogenates of fish and shellfish with Vibrio vulificus M-8 (isolated from shellfish ) and stored them at $-20^{\circ}C,\;4^{\circ}C\;and\;30^{\circ}C$ for 72 hours. Vibrio vulnificus M-8 was not detected in 32 hours when it was frozen and stored at $-20^{\circ}C$ after inoculating them into phosphate buffer solution at concentration of $10^5\;cell/ml$, while the existance of Vibrio vulnificus was identified after 72 hours of storage at the same temperature in case of inoculation into the meat homogenate of yellow tail. The cell count of Vibrio vulnificus was decreased as about $20\%$ of initial count after 2 hours storage at $4^{\circ}C$ in phosphate buffer solution with fish and shellfish homgenates. From the experimental results it was recognized that Vibrio vulnificus was labile to the cold stress. In comparison to the growth of growth of Vibrio M-8 at $30^{\circ}C$ in the raw and cooked meat of the yellow tail(Seriola guingueradita), snapper(Chrysophrys major), ark shell(Anadra brouhgtonii), and oyster(Crassostrea gigas), the raw meat homogenates were more excellent than the cooked ones though all fish and shellfish meat homogenates were proves to be good for the growth of the microbe.

Effect of Clove (Eugenia caryophyllata Thumb) on the Survival of Listeria monocytogenes and Salmonella typhimurium during Cold Storage (저온저장중 Clove(Eugenia caryophyllata Thumb)가 Listeria monocytogenes와 Salmonella typhimurium의 생존에 미치는 영향)

  • The antibacterial activity of low concentrations of clove (Eugenia caryophyllata Thumb) in culture broth against Listeria monocytogenes and Salmonella typhimurium was tested at 35, 5, and -20$^{\circ}C$. Tryptic soy broth (TSB) containing 0∼0.5% (w/v) of clove was inoculated with 10$\^$5/∼10$\^$7/ cell/ml of L. monocytogenes and S. typhimurium and incubated at each temperature. The growth of L. monocytogenes occured only after a prolonged lag period at 0.1% clove at 35$^{\circ}C$, while viabilities of the cells decreased by 1.4 and 3.3 log cycles at 0.3 and 0.5% clove, respectively. Growth of S. typhimurium occured at the presence of 0∼0.5% clove after a longer lag period with increasing concentration of clove at 35$^{\circ}C$. During refrigerated storage at 5$^{\circ}C$, the growth of L. monocytogenes occured after 6 days of lag period at 0.1% clove while viability of the cells were decreased during 24 days of storage. During frozen storage at -20$^{\circ}C$, the viability of L. monocytogenes and S. typhimurium decreased about 4 log cycles during 3 days of early period of storage at 0.1% clove. There were no major changes in the population of L. monocytogenes and S. typhimurium in TSB with different concentrations of clove during frozen storage.

Assessment of Immune Quality and Pathogen Contamination of Colostrums Collected from Colostrum Banks in Korea (초유은행에서 수거한 초유의 병원체 오염과 면역수준의 평가)

  • Because colostrum is considered to be the sole source of passively acquired maternal antibodies for calves, newborn calves must consume colostrum to gain disease resistance during their early years of life. Storage of surplus colostrum from dairy cows right after calving and feeding newborn calves in deficiency of colostrum to assure adequate uptake of IgG for protection of the calf has been a common practice in the bovine production. In the current study, 35 colostrums were randomly collected from 3 colostrum banks located in different regions of Korea and monitored for general bacterial contamination and major bovine pathogens. Immunoglobulin concentrations and BVDV-specific antibodies were also determined to evaluate the immune quality of the colostrums. Moderate to severe bacterial contamination (up to 72,000,000 CFU/ml) was observed in most of the colostrums collected from colostrum banks. General immune quality of the colostrums was under the satisfactory level since most of the colostrums contained less than 50 g/L of IgG, which is the minimum concentration for good quality colostrums. Therefore, colostrum for colostrum bank should be collected at the first 2-3 post-partum milkings according to proper harvesting and handling procedures to guarantee the safety and quality of colostrum. In addition, it was recommended that colostrum should be heat-treated before frozen and stored in the bank because pasteurization at $63^{\circ}C$ for 30 min was very effective reducing the risk of disease transmission without causing significant degradation of immunoglobulins.

Establishment of the Successive Rearing System of Brush-footed Butterflies (Lepidoptera: Nymphalidae) (네발나비과 나비류의 계대사육법 체계확립)

  • In order to establish the successive rearing system brush-footed butterflies (Lepidoptera : Nymphalidae) were reared in a room. Artificial diets were developed for a year-round rearing. Bu-diet was best to rear these butterflies among 3 kinds of diet used. The freeze-dried host plant leaf powder in diet was better than heat-dried one $(60^{\circ}C)$ in the growth of larvae. The rearing results were best in the diet C/N ratio was 1:1. The 24-hrs old eggs could be stored for 5 days at $15^{\circ}C$ or for 3 days at $5^{\circ}C$ and showed 75% of hatchability. On the other hand, pupae could be stored for maximum 15 days at $15^{\circ}C$ because the emergence of abnormal adults appeared much more as the cold storage period got longer. And the adult was able to be stored until 60 days at refrigerator without relation of nectar-sucking period before cold-storage and storage temperature. Also a simple artificial ovipositing kit was devised by ${\Phi}9$ cm of petri-dish and a female oviposited $278{\pm}27$ of eggs with adding the ether extract of host plant to this kit. The systematic successive rearing method of brush-footed butterflies in a room was completed.

Pretreatment and Storage Condition of Abnormal Fermented Oriental Melon for Fermentation Use (참외주 제조를 위한 이상발효 참외의 원료전처리 및 저장조건의 확립)

  • Since abnormal fermentation and short storage duration of oriental melon are the main problems causing loss in commercial value, it is necessary to develop a food processing method using uncommercial melon. In this study, we suggested the effective pretreatment and storage conditions of melon as the material for alcoholic beverage production. Abnormally fermented melon had smaller carbohydrate and larger moisture content than normal one, indicating that carbohydrate in normal melon was probably converted to fermented products during fermentation. The sugar content of oriental melon was increased after fruiting and the highest value $(12.4^{\circ}Brix)$ was found at 5 weeks of storage. The maximum storage duration of normal and fermented oriental melons were 25 and 7 days at $4^{\circ}C$, and 8 and 4 days at room temperature, respectively. The oriental melon for fermentation-use could be conserved after slicing for 30 days at $4^{\circ}C$ with the addition of 1.5% citric acid and for one year at $-20^{\circ}C$ with the plastic film sealing, respectively.

Microbiological and Genotoxicological Safety of Gamma-Irradiated Chicken (감마선조사 닭고기의 미생물학적 및 유전독성학적 안전성 평가)

  • Gamma irradiation (1-10 kGy) was applied to chicken for the evaluation of their microbiological safety and possible genotoxicity. In 3 kGy-irradiated sample, the growth of psychrophile was inhibited about 1.5 log cycles and no cells were recovered in total microbial counts. All kinds of contaminated microorganism were sterilized by 7 kGy-irradiation. Also, irradiation followed by freeze-storage at the same time was very effective in inhibiting bacterial growth. The genotoxicity of 10 kGy-irradiated chicken was evaluated by Salmonella Typhimurium reversion assay and in vivo micronucleus assay using mouse bone marrow cells. The results were negative in the bacterial reversion assay with S. Typhimurium TA98, TA100, TA1535, and TA1539. Clastogenic effects were not shown in vivo mouse micronucleus assay at 10 kGy-dose tested.

진공펌프 물 배기속도 측정 설비 구축

  • 현민지브이티(Genesis)는 중소기업청 중소기업개발지원사업의 일환으로 진행된 2014년도 구매조건부 신제품 개발사업에 선정되어 '극저온 G-M냉동기를 이용한 대용량 Cold Trap개발'과제를 수행하면서 32인치 급으로 수분에 대해서 30,000[L/s] 이상의 배기속도를 가지는 대형 CWP를 개발하고 있다. 1차년도(2015년) 목표는 80K에서 200W급 단단 G-M극저온 냉동기를 개발하는 것이고, 2차년도(2016년) 목표는 이를 장착하여 30,000[L/s]의 물 배기속도 능력을 갖춘 32인치(800mm)급 직부형(appendage) CWP를 개발하는 것이다. 여기에서 가장 큰 문제점은 CWP 시스템의 물 배기속도를 실제로 측정하는 것이다. 왜냐하면 지금까지는 물(H2O)이 가진 독특한 물리적 특성으로 인해 배기속도 측정에 많은 어려움이 있어 이론적으로 계산한 값을 사용해 왔다. (심지어 크라이오 펌프 제조사 조차도 실험하지 않고 이론적인 계산 값을 일반적으로 사용한다.) 그러나 최근 본 과제 외에 물 배기속도 측정에 관한 요구사례와 일부 크라이오 펌프 제조사에서 수행하고 있다는 보고가 있는 바, 실제 물배기속도 시스템을 구축하여 이론과 실제 사이의 차이와 측정의 어려움 등에 관해 규명하고자 하였다. 물 배기속도 측정 방법은 크게 2가지로 나눌 수 있다. 첫째, 시스템으로 흘리는 물의 양을 Liquid MFC를 이용하여 먼저 측정한 후 Vaporizer로 보내어 기화 시키며 배기속도를 측정하는 방법. 둘째, 물을 Vaporizer로 먼저 기화시킨 후에 High Temp. MFM으로 기체 유량을 측정하며 배기속도를 측정하는 방법이 그것이다. 이에 국내 최초로 두 가지 방법 모두를 사용하여 표준화 된 물 배기속도 측정 설비를 구축하였고, 20인치(500mm) 크라이오 펌프와 인라인(inline)형 CWP 모델에 대한 물 배기속도 측정을 성공적으로 완료할 수 있었다. 향후 본 시험 방법과 결과를 토대로 32인치(800mm) 직부형 CWP 모델에 대한 물 배기속도 측정시험을 수행하고자 한다.

  • The groundwater heat pump system(GWHP) is one of the most efficient ground source heat pump system(GSHP) which uses low grade and shallow geothermal energy for cooling and heating purpose. The GWHP system shall be designed properly based on peak block load performance and optimum pumping rate of groundwater comparable to ground coupled heat pump system(GCHP). The optimum pumping rate depends on groundwater temperature at a specific site, size of plate heat exchanger, and total head loss occurred by whole system comprising pumps and pipings. The required optimum flow rates of the system per RT are ranged from 3.8 to 9.8lpm being less than the typical building loop flow of 9.5 to 11.4lpm.

A Study on the N2O Separation Process from Crude N2O (Crude N2O로부터 정제된 N2O 분리공정에 관한 연구)

  • Liquid phase nitrous oxide ($N_2O$) contains air, carbon monoxide, water, carbon dioxide and NOx as main impurities. It is known to be very dangerous to obtain a very pure $N_2O$ product by using solidification at low temperature. In this study a new method to obtain a high purity of $N_2O$ product based on a continuous distillation process was introduced. For the modeling of the continuous distillation process to obtain a product having a purity over 99.999% of $N_2O$ stream, Intalox wire gauze packing- No. SCH-80S gauze packing column was used. Peng-Robinson equation of state was used for the modeling of the continuous distillation process and refrigeration system. Computational results performed in this work showed a good agreement with Aspen Plus simulation results.
