Selection of a Triploid Poplar by Flow Cytometric Analysis and Growth Characteristics of its in vitro Grown Plants (유세포 분석을 통한 현사시나무 3배체 선발 및 계통별 기내생장 특성)

  • Triploids are a useful tool for biomass production and molecular breeding of trees with a long life span. Triploids of the poplar 'Hyunsasi' (Populus alba ${\times}$ P. glandulosa) have been developed by crossing between female diploids and a male tetraploid. The tetraploid was developed around the 1970s at Korea Forest Research Institute by colchicine-induced chromosome doubling. Seedlings of the $F_1$ generation were analyzed using flow cytometry to verify their ploidy status. The mean relative fluorescence index of 3 F1 poplars, labeled as Line- 1, Line-17, Line-18, were approximately 1.5 times higher than those of diploid poplars, and the results clearly indicated that they were triploids. The phenotype of the F1 poplars included larger leaves and thicker stem than diploids, and abnormal leaf morphology, especially in the triploid 'Line-18'. Three triploid lines developed roots more slowly and had less roots than diploid. However, 3 poplar cytotypes (2x, Line-1, Line-17) rooted within 10 days on MS medium. In contrast, compared with the 3 cytotypes, the Line-18 showed about 80% and 70% in the rooting rate and the number of roots. The triploid poplars could be directly utilized for biomass production and with their sterility, they could serve as basic material for genetic transformation. In addition, flow cytometric analysis proved to be an effective and reliable method for screening forest trees for their ploidy level.

A Study on the Production Structure and Biomass Productivity of Quercus variabilis Natural Forest (굴참나무천연림(天然林)의 생산구조(生産構造) 및 물질생산력(物質生産力)에 관(關)한 연구(硏究))

  • Growth and biomass production of natural stands of Quercus variabilis in relation to tree density were studied to obtain basic guide lines for future tending operation. Two natural stands of Quercus variabilis located at 900m (A stand: 6,600trees/ha, $15.84m^2/ha$, $\frac{19}{17-20}$) and 800m (B stand: 4,300trees/ha, $16.65m^2/ha$, $\frac{20}{17-21}$) elevation in Sancheong, Kyongnam Province were selected for the comparative study and following results were obtained through a sample plot method. After diameter of individual trees in the sample plots was measured, twelve average trees from each diameter class were cut felled to measure dry weight of $W_S$, $W_B$, $W_L$, $W_{Ba}$, and standing biomass and biomass production rates by a allometrior regressions related to $D^2H$. Vertical distribution of leaves along the stems indicated that photosynthesis was carried out 2.2m above the ground in Stand A and 1.2m in Stand B. Maximum photosynthesis was located 4.2m and 6.2m above the ground in Stand A and B, respectively. Leaf area index was 4.25ha/ha for Stand A, and 3.89ha/ha for Stand B. Above-ground standing biomass was 49.51 ton/ha for Stand A and 59.20 ton/ha and net annual production was 6.75 ton/ha/yr. for Stand A and 8.99 ton/ha/yr. for Stand B. The ratio of net annual production to standing biomass was 17.5% for Stand A and 16.7% for Stand B. Net assimilation rate was 2.75kg/kg/yr. for Stand A and 3.58kg/kg/yr. for Stand B. Stem wood production rate was 1.46kg/kg/yr. for Stand A and 2.09kg/kg/yr. for Stand B. Bark production rate was 0.60 kg/kg/yr. for Stand A and 0.34kg/kg/yr. for Stand B. Above data indicated that Stand B utilized growing spaces and sites more efficiently than Stand A. It is concluded chat productivity of natural stands of Quercus variabilis can be enhanced through optimization of basal areas and number of tree per hectare and that sound management of natural oak stands should be based on systematic sampling of the area for periodic productivity estimation.

Effects of Tree Density Control on Carbon Dynamics in Young Pinus densiflora stands (소나무 유령림의 임목밀도 조절이 탄소 동태에 미치는 영향)

  • The objective of this study was to examine carbon dynamics with biomass, soil $CO_2$ efflux, litter and root decomposition after tree density control in young Pinus densiflora stands. The stands were established with 50% thinning, clear-cut, and control stands with three pseudo-replicated plots and a bare soil plot in 8-year-old Pinus densiflora nursery field. Monthly measurements were conducted from March 2012 to February 2014 and aboveground biomass and coarse-roots were estimated by derived allometric equations. Average diameter growth at root collar in control and thinned was 0.89 cm and 1.48 cm per year, respectively, and the diameter growth of control stand was significantly higher than that of thinned stands (p<0.05). Total biomass was estimated to 5.17, $4.85kg\;C\;m^{-2}$ per year in control and thinned, respectively. Annual soil $CO_2$ efflux in control, thinned, clear cut, and bare soil was 3.71, 3.90, 4.17, $4.56kg\;CO_2\;m^{-2}\;yr^{-1}$, respectively and removing trees significantly increased soil $CO_2$ efflux (p<0.05). Net Ecosystem Production (NEP) was 1.57, 1.36, -0.67, $-1.25kg\;C\;m^{-2}\;yr^{-1}$ in control, thinned, clear cut and bare soil in the young Pinus densiflora stands. NEP was significantly decreased by removing trees. Thinning increased diameter at root collar and carbon of individual tree and recovered 86% of carbon removed by thinning after one-year. In addition, soil $CO_2$ efflux increased and NEP increased by thinning. Results of this study, tree density control such as thinning increased the carbon storage and growth of the young Pinus densiflora stands.

Characteristics of Suillus bovinus fairy rings and genets associated with thinning intensity in Pinus densiflora forests (소나무림에서 간벌강도에 따른 황소비단그물버섯(Suillus bovinus)의 균환과 genet 특성)

  • To study the fairy ring and genet characteristics of Suillus bovinus based on thinning intensity in Pinus densiflora forests, a simple sequence repeat (SSR) analysis was performed on the fruiting bodies of the plant. In pine wood production forests, the thinning strengths applied were 34%, 45%, and 60%. As a result, the number of fruiting bodies in the 34% treatment area was 104, which was higher than that in the other treatment areas. In the 34% treatment area, fruiting bodies occurred in a circular shape, within a diameter of approximately 5 meters (m) of the trees. In the 45% treatment area, the fruiting bodies were randomly distributed between 6 to 7 m from the trees, while in the 60% treatment, fruiting bodies occurred in a narrow oval shape, 6 m from the trees. In the control area, two fruiting bodies were present around the root collar. Hybridity was confirmed in the SSR markers of Sb-CA1 and Sb-CA3. The fruiting bodies in the 34% treatment area had a He / Ho value lower than that in the 60% treatment area. In fruiting bodies of the 34% treatment area, a total of 20 genets were identified, with an average size of 14±11 ㎡; 60% of genets were formed by a single fruiting body. In fruiting bodies of the 45% treatment area, a total of 6 genets were identified and the average size was 11±12 ㎡; 50% of genets were formed by a single fruiting body. In fruiting bodies of the 60% treatment area, a total of 10 genets were identified, with an average size of 1.1±0.8 ㎡; 70% of genets were formed by a single fruiting body. Thus, the formation ratio of a new genet increases when the thinning intensity is increased.

Potassium Supply Characteristics in Different Forest Soils (지역별(地域別) 산림토양(山林土壤)의 K(Potassium) 공급(供給) 특성(特性))

  • Generally potassium in the soil can be remained as water soluble, exchangeable, and available types, respectively. Theses types of potassiums are also known to keep their kinetic relationships to each other. The Purpose on this research was to investigate types and relationships of potassium in the soil, and the characteristics of potassium supply in different forest soils. The results could be summarized as follows; 1. The amounts of available potassium in A and B horizon soils by continuous leaching with 0.01N-HCl were as follows : for A Horizon soils, pohang(Gray brown forest soils), Changsung (Brown forest soils), and Youngwol(Dark red forest soils) were ranged from 0.2me/l00g to 0.8me/100g, Taean(Red and Yellow forest soils) was 0.1-0.6me/100g, Kapyung(Brown forest soils) was 0.2-0.4me/100g. For the B layer, Youngwol was 0.1-0.5me/100g, Pohang, Taean, Kapyung were 0.1-0.4me/100g, Changsung was 0.1-0.3me/100g, respectivly. 2. Of ten times-pulse leaching with 0.01N-HCl for A layer soil, more 80% of total available potassium leaching were recovered by the second pulse as leachate for Pohang, Changsung, Youngwol, while similar amounts of potassium in the leachate were obtained with the third pulse leaching for Kapyung, Taean, respectively, On the other hand, the 80 % release of available potassium from the B layer soil was obtained by the second pulse leaching for all areas investigated. 3. For the relationships between soluble potassium and exchangeable potassium, the exchangeable potassium was increased while the soluble potassium was not changed significantly in B layer. And both soluble potassium and exchangeable potassium in A layer were similar. For the relationship between exchangeable potassium and available potassium in both A and B layers, the amounts of available potassium increased by 1.2 to 1.5 times as the exchangeable potassium increased. 4. For distribution of all types of potassium throughout locations investigated, the types of potassium were in the decreasing orders of available, exchangeable, and soluble. 5. The simplified method for an analysis of all types of potassium by sequential leaching with 0.01N-HCl should be developed not only with respect to time-saving and efficiency but also verification of the relationship between available potassium and tree growth.
