Search for Medicinal Plants on Improvable Effect of Intestinal Microflora (장내 미생물 개선효과가 있는 약용식물소재 탐색)

  • Extracts of 13 medicinal plants were tested to investigate the improvable effects on microflora growth in the intestines of human. Water extracts of Artemisia capillaris, Taraxacum platycarpum and Ethanol extracts of Allium tuberosum, Artemisia capillaris, Artemisia vulgaris, Plantago asiatica, Schizandra chinensis, Sedum sarmentosum, Taraxacum platycarpum, Zingiber officinale inhibited the growth of C. perfringens. Water and ethanol extracts of Sedum sarmentosum, Schizandra chinensis inhibited the growth of E. coli. While 13 medicinal plants were not remarkably inhibited the growth of B. longum. Therefore, ethanol extracts of Schizandra chinensis, Sedum sarmentosum can be utilized as functional food materials to improve in the intestines.

Effect of Extract of Fermented Dropwort on Intestinal Bacteria and Enzymes In Vitro (미나리발효액이 장내 유해세균 및 유익균의 In Vitro 생육 및 효소활성에 미치는 영향)

  • Effect of extract of fermented dropwort (Oenanthe stolonifera) on growth of intestinal harmful/useful bacteria and enzyme activity were investigated in vitro. The extract showed strong inhibition on harmful microbes including Vibrio and Salmonella, but mild inhibition on Bifidobacterium longum in both agar plate and liquid cultivation. Minimum inhibitory concentration (MIC) value of B. longum was the highest among tested microbes. Inhibition effect of fermented extract on harmful microbes increased according to fermentation period. Extract of fermented dropwort showed inhibitory effects on activity of microbial ${\beta}$-glucuronidase and tryptophanase. The inhibitory effects were also proportional to fermentation period. As consequence, it is assumed that the uptake of fermented dropwort might be useful for human intestinal health.

Effect of Maintained Microorganisms against to The Phytoncide on Pr. intermedia (피톤치드 처리 후의 잔존 구강 세균이 Pr. intermedia에 미치는 영향)

  • The present study was performed to observe the effect of phytoncide on oral normal microflora and the inhibitory effect of the surviving resident oral bacteria on Pr. intermedia. In this study, saliva from each of 20 healthy subjects was treated with 1% phytoncide from Japanese Hinoki (Chamaecyparis obtusa Sieb. et Zucc.). Surviving salivary bacteria were isolated on blood agar plates and identified by 16S rDNA sequencing. In order to select inhibitory isolates against Pr. intermedia, the isolates from the phytoncide-treated saliva were cultured with Pr. intermedia. The results were as follows: 1. Among the 200 surviving resident oral bacterium, 148(74.0%) bacterium inhibit the growth of Pr. intermedia on blood agar plates. 2. The 200 surviving resident oral bacterium were 109 Streptococcus salivarius(54.5%), 25 Streptococcus sanguinis(12.5%), 15 Streptococcus mitis(7.5%). 3. Among the 148 bacteria which inhibit Pr. intermedia, Streptococcus salivarius was 85.3%(93/109), Streptococcus sanguinis was 64.0%.(16/25), Streptococcus mitis was 54.3%(8/15), Streptococcus parasanguinis was 66.7%(6/9), and Streptococcus Alactolyticus was 100%(8/8). Taken together, among the surviving resident oral bacterium, Streptococcus salivarius, Streptococcus sanguinis, Streptococcus mitis were mainly observed to inhibit Pr. intermedia. and they may exert an additional inhibitory activity against the periodontopathic bacterium. Therefore, phytoncide can be used for preventing and ceasing the progress of periodontal disease and halitosis, and thus is expect to promote oral health.

Characterization of Pyrenophora avenae Isolated from Discolored Black Oat Seeds in Korea (귀리 흑변 종자에서 분리된 Pyrenophora avenae의 특성)

  • In January 2017, discolored black oat seeds were found in the storage depot of a farmhouse in Jeongeup. Pyrenophora sp. was detected in 45% of the oat seeds surveyed. All Pyrenophora isolates obtained from the seeds were identified as Pyrenophora avenae based on the sequences of internal transcribed spacer (ITS) rDNA regions and glyceraldehyde 3-phosphate dehydrogenase (GPDH) gene and validated by morphological and cultural characterization. A phylogenetic tree constructed using the ITS and GPDH sequences showed that the Korean isolates of P. avenae comprise of four genetically distinct groups. Pathogenicity test validated that the fungus is an infectious agent responsible for discolored black seeds and leaf blotch in oat plants. This is the first study report that P. avenae causes leaf blotch disease of oat in Korea.

음란사이트 접속방지 기능을 갖는 웹브라우즈 개발

  • 청소년 유해 사이트의 대명사인 음란 사이트의 증가는 인터넷의 역효과를 대신하는 대표적인 사례라 할 수 있다. 본 연구는 음란 사이트의 접속을 차단하기 위해 웹 브라우즈가 접속한 모든 사이트를 데이터베이스에 기록할 수 있도록 하여, 보호자에 의해 유해 사이트로 판정 받은 사이트는 접속 할 수 없도록 하는 기능을 갖는 웹 브라우즈를 개발하였다. 본 연구에서 개발한 웹 브라우즈는 한번 접속한 모든 사이트를 데이터베이스를 통해 관리할 수 있으며, 유해 사이트에 대한 정보는 패스워드에 의해 데이터베이스의 접근이 가능한 보호자에 의해 보호될 수 있도록 하였다. 특히 아직 한번도 접속하지 않았던 사이트들에 대해서도 보호자에 의해 유해성이 인정되는 특정 단어를 등록함으로서 특정단어가 들어 있는 유해 사이트에 대한 접속을 예방 할 수 있도록 하였다.

Effect of Wildlife Access on Microbial Safety of Irrigation Water Used in the Cultivation of Chinese Cabbage in Goesan (야생동물 출입이 괴산 지역 배추 재배 농업용수의 미생물 안전성에 미치는 영향)

  • Water is an important component in the production of fresh produce. It is mainly used for irrigation and application of pesticides and fertilizers. Several outbreaks cases related to fresh produce have been reported and water has been identified as the most likely source. On the other hand, wildlife has been identified as a possible source of the waterborne pathogens. The purpose of this study was to investigate the effect of wildlife access on irrigation water used in the cultivation of Chinese cabbage in Goesan. The frequency of wild animals access to upstream water source and the contamination level of bacteria such as Escherichia coli and Enterococci of irrigation water used in Chinese cabbage farm was examined. A total of 37 wildlife including the wild bear and water deer were observed in upstream of water source during the cultivation of Chinese cabbage. The result indicated the presence of hygienic indicator bacteria from the upstream where there is no human access. The contamination range of coliforms, E. coli, and Enterococcus spp. Detected in the irrigation water were 2.13~4.32 log MPN / 100 mL, 0.26~2.03 log MPN / 100 mL, and 1.43~3.49 log MPN / 100 mL, respectively. Due to low water temperatures, the contamination levels of coliform bacteria and E. coli in the irrigation water during harvesting time was lower compared to those recorded during transplanting of Chinese cabbage. However, no significant difference was detected in the number of Enterococci during the cultivation of Chinese cabbage. The results indicated the need to manage the microbial risk in irrigation water to enhance safety in cultivation of Chinese cabbage.

Acaricidal and antimicrobial toxicities of Cyanachum paniculatum root oils and these components against Haemaphysalis longicornis and human intestinal bacteria (산해박 뿌리에서 추출한 정유 및 구성성분의 인간 장내미생물에 대한 항균활성 및 작은소피참진드기에 대한 살비활성)

  • Anaerobic growth-inhibiting and acaricidal activities of 2'-hydroxy-5'-methoxyacetophenone derived from Cyanachum paniculatum oil and its derivatives against five intestinal bacteria (Bifidobacterium bifidum, B. longum, Clostridium pefringens, Escherichia coli and Lactobacillus casei) and Haemaphysalis longicornis were examined. In the packet test against the larvae of H. longicornis, none of the C. paniculatum oil exhibited acaricidal activity, while the C. paniculatum oil showed only antimicrobial activity against five intestinal bacteria in the disc diffusion method. Based on the inhibition zones and MIC values, 2',4'-dimethoxyacetophenone, 2',5'-dimethoxyacetophenone, 2'-hydroxy-4'-methoxyacetophenone, 2'-hydroxy-5'-methoxyacetophenone, 2'-methoxyacetophenone, and 4'-methoxyacetophenone, containing a methyl group on the acetophenone skeleton, possessed growthinhibiting activities against C. perfringens and E. coli. However, acetophenone, 2'-hydroxyacetophenone, 4'-hydroxyacetophenone, 2',4'-hydroxyacetophenone and 2',5'-hydroxyacetophenone, which contained a hydroxyl group on the acetophenone skeleton, had no growth-inhibiting activity against intestinal bacteria. These results indicated that 2'-hydroxy-5'-methoxyacetophenone and its derivatives could potentially be developed as natural antimicrobial agents to specific control of C. perfringens and E. coli.

동치미 유래 생균제로서 가능성이 있는 Lactobacillus sp. FF-3의 분리와 특성

  • 생균제로서 가능성 있는 유산균의 선별을 위해 발효식품인 동치미로부터 분리된 균주들의 내산성, 내담즙성, 내췌장액성 및 유해균 억제능을 조사하였다. 특히 분리된 균종 중에서 FF-3가 인공위액과 담즙산에 대한 높은 내성을 나타내었을 뿐만 아니라, Salmonella sp. 과 Escherichia coli에 대하여 높은 항균 활성을 나타내었다. FF-3유래 박테리오신은 그람양성 및 음성 세균에서 넓은 항균 범위를 나타내었다. 본 연구에서 최종선별된 FF-3 균주를 16s rDNA sequence 동정한 결과 Lactobacillus sp.으로 확인되어 FF-3으로 명명하였다.

Effect of Kimchi ingredients on the growth of pathogenic and lactic acid bacteria (김치부재료의 유산균 증식 및 유해균 억제효과)

  • When ingredients of Kimchi were mixed and stored in $18^{\circ}C$, lactic acid bacteria, such as Leuconostoc mesenteroides and Lactobacillus plantarum, were selectively grown up. Herefore, to understand why lactic acid bacteria were selectively cultured in Kimchi, antibacterial activities of Kimchi ingredients against some pathogens and Kinlchi lactic acid bacteria were investigated. Kimchi mixed with all ingredients significantly inhibited the growth of all tested pathogens: S. typhimurium, S. sonnei, and E. coli. Kimchi without green onion, garlic or ginger inhibited the growth of S. typhimurium, but did not E. coli and S. sonnei. However, Kimchi without red pepper powder did not inhibit the growth of all tested pathogens. All ingredients of Kimchi did not inhibit the growth of L. plantarum and L. mesenteroides. These results suggest that Kimchi ingredients can synergistically inhibit the growth of pathogens and Kimchi may be a selective medium for lactic acid bacteria.
