Study on Incineration Behavior of Heavy Oil Fly Ash for Valuable Metal Recovery (유가금속(有價金屬) 회수(回收)를 위한 중유회(重油灰)의 연소거동(燃燒擧動)에 관한 연구(硏究))

  • To design and construct a moving bed stoker incinerator for incineration treatment of the domestic oil fly ash, operating condition and moving bed area of incinerator were determined by performing incinerate experiment of the oil fly ash in the muffle furnace which simulates moving bed stoker incinerator in all conditions. Incineration process of the oil fly ash could be divided into 3 stages, every stage needs the appropriate operating condition for effective incineration. The optimum content of water in the heavy oil fly ash was found to be 20 wt% to prevent the ash from flying and reduce the volume. Science combustion rate of oil fly ash depends on the oxygen content, the incinerator must have a equipment to control the oxygen content in the combustion air. The optimum temperature was 750800C in order to prevent adhesion to the stocker and evaporation of metal compounds of low melting point. Uniform combustion reaction and acceleration of combustion rate required agitation during the combustion of oil fly ash. The incineration rate was 12.53kg/m2hr and the working area of moving bed incinerator was found to be 60m2 to incinerate 18 tons of oil fly ash per day.

Protective effect of Thymus quinquecostatus extracts UVB-induced matrix metalloproteinase-1 via suppressing MAPKs phosphorylation in human keratinocyte (사람의 각질세포에서 UVB 유도에 따른 MMP-1의 발현 조절과 MAPKs 인산화에 타임 추출물이 미치는 효과)

  • Ultraviolet rays are electromagnetic waves with a shorter wavelength than visible light, and ultraviolet rays that pass through the ozone layer are the main cause of skin aging. Chronic exposure of skin tissue to ultraviolet light activates the Mitogen-activated Protein Kinases (MAPKs) signaling pathways in human keratinocytes, resulting in increased production of matrix metalloproteinases (MMPs). In this study, we investigated the herbal extracts from Jeju Island on the anti-aging effect in human keratinocytes (HaCaTs) by ultraviolet stimulation. We examined that herb extract from Jeju Island were decreased in anti-aging activity on measuring the level of MMP-1 gene and protein expression in ultraviolet-induced keratinocytes. As a result, it was confirmed that Thymus quinquecostatus extract (TQE) significantly reduced the expression of MMP-1 in a dose-dependent manner by UV irradiated HaCaTs. According to our data, TQE significantly attenuated UV-induced phosphorylation of the MAPKs signaling elements ERK1/2, JNK1/2 and p38 proteins. These results suggest that the MAPKs pathway may contribute to the inhibitory effect of TQE on UV-induced MMP-1 production in human keratinocytes. Our results suggest that TQE may be a protective agent against skin aging by preventing UV-induced MMP-1 production.

Mineral Processing Characteristics of Titanium Ore Mineral from Myeon-San Layer in Domestic Taebaek Area (국내 태백지역 면산층 타이타늄 광석의 기초 선광 연구)

  • Titanium's importance as a mineral resource is increasing, but the Korean industry depends on imports. Ilmenite is the principal titanium ore. However, research and development from raw materials have not been investigated yet in detail. Hence, measures to secure a stable titanium supply chain are urgently needed. Accordingly, through beneficiation technology, we evaluated the possibility of technological application for the efficient recovery of valuable minerals. As a result of the experiments, we confirmed that mineral particles existed as fine particles due to weathering, making recovery through classification difficult. Consequently, applying beneficiation technologies, i.e., specific gravity separation, magnetic separation, and flotation, makes it possible to recover valuable minerals such as hematite and rutile. However, there are limitations in increasing the quality and yield of TiO2 due to the mineralogical characteristic of the hematite and rutile contained in titanium ore. Hametite is combined with rutile even at fine particles. Therefore, it is essential to develop mineral processing routes, to recover iron, vanadium, and rare earth elements as resources. On that account, we used grinding technology that improves group separation between constituent minerals and magnetic separation technology that utilizes the difference in magnetic sensitivity between fine mineral particles. The development of beneficiation technology that can secure the economic feasibility of valuable materials after reforming iron oxide and titanium oxide components is necessary.

EFFECT OF CURCUMIN AND RESVERATROL ON THE CELL CYCLE REGULATION, APOPTOSIS AND INHIBITION OF METASTASIS RELATED PROTEINS IN HN-4 CELLS (Curcumin과 resveratrol에 의한 두경부암 유래의 HN-4 세포의 세포주기, 세포사 및 전이관련 단백질의 발현 조절)

  • Nontraditional or alternative medicine is becoming an increasingly attractive approach for the treatment of various inflammatory disorders and cancers. Curcumin is the major constitute of turmoric powder extracted from the rhizomes of the plant Curcuma longa. Resveratrol is a phytoalexin present in grapes and a variety of medicinal plants. In this report, We investigated the effect of curcumin and resveratrol on regulatory protein of cell cycle, induction of apoptosis and MMP activity. Treatment with 75 M curcumin for 24 hrs produced morphological changing in HN-4 cells. Curcumin and resveratrol inhibited the cellular growth in HN-4 cells. Inhibition of cell growth was associated with down-regulation of cell cycle regulatory proteins. Curcumin-induced caspase-3 activation and Bax degradation were dose-dependent with a maximal effect at a concentration of 100 M. The elevated caspase-3 activity in curcumin treated HN-4 cells are correlated with down-regulation of survivin and cIAP1, but not cIAP2. Curcumin induced a dose-dependent increase of cytochrome c in the cytosol. Curcumin induced-apoptosis was mediated through the release of cytochrome c. In addition, curcumin-induced apoptosis was caused by the generation of reactive oxygen species, which was prevented by antioxidant N-acetyl-cysteine (NAC). Cotreatment with NAC markedly prevented cytochrome c release, Bax cleavage and cell death. Also resveratrol-induced apoptosis was preceded by down-regulation of the anti-apoptotic Bcl-2, cIAP1, and caspase-3 activity. However, resveratrol-induced apoptosis was not prevented by antioxidant NAC. In addition, HN-4 cells release basal levels of MMP2 when cultured in serum-free medium. Treatment of the cells with various concentrations of PMA for 24 hr induced the expression and secretion of latent MMP9 as determined by gelatin zymography. HN-4 cells were treated with various concentrations of curcumin and resveratrol in the presence of 75 nM PMA, and MMP2 and 9 activities were inhibited by curcumin and resveratrol. These findings have implications for developing curcumin-based anticancer and anti-inflammation therapies.

Cognitive-enhancing Effects of a Fermented Milk Product, LHFM on Scopolamine-induced Amnesia (발효유 산물인 LHFM의 인지기능 개선 효과)

  • Probiotics and their products, such as yogurt and cheese have been widely consumed in many countries with proven health benefits including anti-microbial activity and anti-diarrheal activity. LHFM (Lactobacillus helveticus - fermented milk) is a processed skim milk powder, fermented by a probiotics, L. helveticus IDCC3801. In the present study, we aimed to investigate the neuroprotective effects and the cognitive improvements of LHFM. LHFM itself did not show any cytotoxicity to the human neuroblastoma cell line, SH-SY5Y; however, it dose-dependently protected against glutamate-induced neuronal cell death. LHFM also attenuated scopolamine-induced memory deficit in Y-maze and Morris-water maze. In the analysis of hippocampus after a behavior test, LHFM significantly increased the acetylcholine level and also inhibited acetylcholine esterase activity. Therefore, the raised acetylcholine release partially contributes to the improvement of learning and memory by a treatment with LHFM. These results suggest that LHFM is an effective material for prevention or improvement of cognitive impairments caused by neuronal cell damage and central cholinergic dysfunction.

The Study of anti-cancer mechanism with Cobrotoxin on Human prostatic cancer cell line(PC-3) (전립선 암세포에 대한 Cobrotoxin의 항암(抗癌) 기전(機轉) 연구(硏究))

  • Objective : The purpose of this study was to investigate the anti-caner effect of cobrotoxin on the prostatic cancer cell line (PC-3).The goal of study is to ascertain whether cobrotoxin inhibits tile cell growth and cell cycle of PC-3, or the expression of relative genes and whether the regression of PC-3 cell growth is due to cell death or the expression of gene related to apoptosis. Methods : After the treatment of Pc-3 cells with cobrotoxin, we performed 형광현미경, MTT assay, Western blotting, Flow cytometry, PAGE electrophoresis and Surface plasmon resonance analysis to identify the cell viability, cell death, apoptosis, the changes of cell cycle and the related protein, Adk, MAP kinase. Results : 1. Compared with normal cell, the inhibition of cell growth reduced in proportion with the dose of cobrotoxin(0-16nM) in PC-3. 2. Cell viabilities of 0.1, 1, 4nM cobrotoxin treatment were decreased and those of 8, 16nM were decreased significantly. 3. S phase of cell cycle was decreased at the group of 1, 2, 4, 8, 16nM cobrotoxin, but M phase was increased at 0.1, 1, 2, 4, 8, 16nM cobrotoxin. 4. Cox-2 expression after cobrotoxin was peaked at 12hours and was decreased significantly after 6, 12, 24 hours. 5. The expression of Cdk4 was decreased dose-dependently at 1, 2, 4, 8nM cobrotoxin and was decreased siginificantly at 4, 8nM Cyclin D1 was decreased at 1, 2, 4, 8nM and Cycline E was not changed. Cycline B was decreased at 1, 2, 4, 8nM dose-dependently and was decreased siginificanlty at 2, 4, 8nM. 6. The expression of Akt was decreased at 1, 2, 4, 8nM dose-dependently and was decreased significantly at 2, 4, 8nM. 7. ERK was increased at 1, 2nM and decreased at 4, 8nM, p-ERK was increased at 1, 2, 4 nM, but decreased at 8nM. JNK and p-JNK were increased at 1, 4, 8 nM. p38 was increased at 2nM p-p38 was increased at lnM but decreased significantly at 2, 4, 8nM. 8. The nucli of normal cells were stained round and homogenous in DAPI staining, but those of PC-3 were stained condense and splitted. Apoptosis was increased dose-dependently at 2, 4, 8, 16nM and increased significantly at 2, 4, 8, 16nM. 9. Bax wasntchandat1,2,4,8nMandBcl-2wasdecreasedsignificantlyat1,2,4,8nM.Caspase3and9werent changed at 1, 2, 4nM but were decreased significantly at 8nM. Conclusions : These results indicate that cobrotoxin inhibits the growth of prostate Cancer cells, has anti-cancer effects by inducing apoptosis.

Induction of Apoptosis and Inhibition of Growth in Human Gastric Cancer by Piperine (Piperine에 의한 위암세포 AGS 증식 억제와 Apoptosis 유도)

  • Piperine [(E,E)-5-(3,4-methtylenedioxyphenyl)-2,4-pentadienolypiperidide] is a principal of Piperaceae, including Piper nigrum L. and Piper longum Linn., which has been used as a spice and traditional medicine. In this study, we investigated whether or not piperine has anti-cancer effects on AGS human gastric cancer cells. The results demonstrated that piperine not only inhibited proliferation using MTT assay but also induced apoptotic bodies using DAPI assay in a dose-dependent manner in response to piperine. Expression levels of p53, Bax (pro-apoptotic), cleaved caspase-9, and cleaved-PARP increased, whereas expression levels of Bcl-2, XIAP (anti-apoptotic), and Akt decreased in a dose-dependent manner compared with the control by western blotting analysis. To identify the connection between phospo-Akt and Bcl-2 family in response to piperine, LY249002 (Akt inhibitor) was treated with piperine (150μM). The results were shown that expression of phospo-Akt was reduced whereas expression of Bax and cleaved-PARP increased in a dose-dependent manner. These results indicate that piperine induced apoptosis in AGS cells and may serve as a chemopreventive or therapeutic agent for human gastric cancer.

Chemical Characteristics and Immuno-Stimulatory Activity of Polysaccharides from Fermented Vinegars Manufactured with Different Raw Materials (상이한 재료로 제조된 발효식초 유래 다당의 화학적 특성과 면역증진 활성)

  • To elucidate the immuno-stimulatory activity of traditional fermented vinegar, six kinds of crude polysaccharides were isolated from traditional fermented vinegars manufactured with different raw materials in domestic or foreign countries, after which their chemical properties and immuno-stimulatory activities were evaluated. Of the six samples, three kinds of crude polysaccharides prepared from Korean brown rice vinegar (KBV-0), Japanese brown rice vinegar (JBV-0), and Korean persimmon vinegar (KPV-0) showed higher yields and interleukin (IL)-6 production by macrophages and were thus selected for further study. Anti-complementary activities of KBV-0, KPV-0, and JBV-0 increased dose-dependently. KBV-0 and KPV-0 showed higher anti-complementary activities (ITCH50 62 and 65%) than JBV-0 at 1,000μg/mL. KBV-0, KPV-0, and JBV-0 did not affect growth of peritoneal macrophages at a dose of 1.6 to 1,000μg/mL, where as they significantly augmented production of IL-6, IL-12, and TNF-α in a dose-dependent manner. However, immuno-stimulatory activity of KPV-0 was the most potent among the tested polysaccharides. These results suggest that Korean fermented vinegars contain selected polysaccharides that confer immuno-stimulatory activities beneficial to human health.

A Study on Environmental Design Treatments of Subway Stations in Terms of Visual Cues for Place Indication - Focused on the Cases of Seoul, Tokyo and Taipei - (장소지시단서로서의 지하철역 환경디자인 기법에 관한 연구 -서울, 동경, 타이페이 사례를 중심으로-)

  • Currently, finding one's way in the underground spaces associated with subway stations in large cities is becoming a serious problem, and an environment that facilitates wayfinding is becoming more important. This study investigated interior treatments that function as visual cues in the subway stations of Seoul, Tokyo, and Taipei in order to develop design guidelines that would improve the place-cognition environment. A theoretical study has shown that there are three categories of visual cue for place cognition: marked trails, regional differentiation, and landmarks. The case study showed that marked trails involve treatments like vertical markers, confrontational positioning, and subway color codes. Regional differentiation involves patterns on the floors and walls, and accent colors on the walls and columns. Landmarks include treatments such as super graphics, symbolic features, artistic decoration and special lights. In the cases of Seoul, marked trails and regional differentiation were predominant. In addition, the Olympic symbol 'Hodori' and the ancient Korean character "Hunminjungeum" were used as landmarks. In the cases of Tokyo, regional differentiation predominated over marked trails because of the complexity of the spatial structure and subway line connection system. In the cases of Taipei, simple, unified images predominated, and there was minimal dependence on marked trails and landmarks. The need for regional differentiation was weak, because the stations studied have open structures that guarantee an open field of view. The results of this research could be used as design guidelines, after further experimental verification, that would help to improve the public environment in terms of wayfinding.

Analysis of the Spatial Distribution and Characteristics of Talented Labor Attraction in Korea - Focused on Talent Magnet Potential Index(TMPI) - (인재유인력의 지역분포와 특성분석 - 인재유인잠재력지수(TMPI) 산출을 중심으로 -)

  • The purpose of this study is to analyze regional spatial distribution and characteristics with the Talent Magnet Potential Index(TMPI), which is calculated through the extra-regional talent attracting environment(Magnet factors) and intra-regional talent cultivating environment(Incubator factors) of each region. As a result, Seoul, Daejeon, Gyeonggi, Ulsan etc. ranked highest, and regions developed in knowledge-based services, R&D capability, and manufacturing turned out to have a higher TMPI. The regions were categorized into 4 types, and the regional type analysis showed that the cumulative effects of Type I(Talent comparative advantage type) and Type III(Talent absolute shortage type) contribute to widening the economic gap among these regions. In addition, although the metropolitan based Type IV(intra-regional talent dependant type) had excellent talent training capability, there seemed to be an outflow of talent into Type I or Type II(extra-regional talent dependant type). This paper has done a correlation analysis to test the validity of the TMPI. As a result, the correlation between talent by type and TMPI turned out to be very high. The correlation coefficient with 2T turned out to have the highest correlation, showing that tolerance contributes a much greater deal to talent attracting and training than expected. Finally, based on the main analytical results, implications for talent attraction & training policy projects and 7 improvement plans have been proposed.
