Carotenoid Production of Transformed Carrot Hairy Root and Its Culture in Bioreactor (형질전환된 당근 모상근의 Carotenoid 생산과 생물반응기에서의 배양)

  • 다양한 배양조건을 이용하여 당근 모상근의 생장과 carotenoid 함량을 조사하였으며, 이러한 배양조건을 airlift형생물 반응기에 적용하여 모상근의 다량 배양을 시도하였다. 고형배지 배양에서의 carotenoid 함량은 광조건에서 배양된 모상근이 암조건에서 배양된 모상근보다 10배 정도 증가하였으며, 모상근의 생장은 광조건이 암조건보다 낮은 수준이었다. 공기 유출기 배양에서는 모상근이 광조건과 암조건 모두에서 매우 빠르게 생장하여 생줄량의 차이가 없었으며, 모상근의 생장과 carotenoid 함량에 있어서 빛의 조사는 모상근의 전체적인 생중량을 증가시키기 보다는 배양 후기 생장단계에서 carotenoid 함량을 증가시켜서 암조건에 비하여 10배 정도의 carotenoid(11 $\mu\textrm{g}$/g)를 생산하였다. 모상근을 생물 반응기에서 배양하였을 때, 모상근이 지수함수적으로 빠르게 생장하여 배양 25일 동안에 처음 무게의 11배를 나타내었다. 그리고 pH는 처음 5.5에서 5일후 4.9로 떨어지다가 그 이후에 일정한 수준(pH 4.8)을 보여주었으며, 광주기와 광도을 조절하지 않은 상태에서의 carotenoid 함량은 2$\mu\textrm{g}$/g으로 나타났다.

  • The effect of low dose ${\gamma}$-radiation on the callus growth of Lithospermum erythrorhizon S. cultured on different medium and lighting condition was investigated. The 8 Gy irradiation stimulated callus growth on LS medium supplemented with BA 2 mg/L and NAA 2 mg/L, however, the growth of callus was more effective on LS medium supplemented with BA 1 mg/L and NAA 1 mg/L under 16 hrs day light. And on the LS medium containing IAA 0.2 mg/L, 16 Gy irradiation increased the callus growth by supplement with kinetin 2 mg/1 and the effect of kinetin was higher than BA at same concentration. The growth of callus was more vigorous on LS medium than MS medium in general. On M-9 medium, the growth of callus was poor regardless lighting conditions, however, by ${\gamma}$-ray irradiation of 16 Gy or 30 Gy, callus growth rates were increased by 30% or more than 30%, especially, under 16 hrs day light condition.

Comparative Evaluation of Colon Cancer Stemness and Chemoresistance in Optimally Constituted HCT-8 cell-based Spheroids (적정 구성 배양 HCT-8 기반 대장암 스페로이드의 암 줄기세포능 및 항암제 내성 평가의 비교 평가 연구)

  • Cancer is a complex disease heterogeneously composed of various types of cells including cancer stem-like cells responsible for relapse and chemoresistance in the tumor microenvironment. The conventional two-dimensional cell culture-based platform has critical limitations for representing the heterogeneity of cancer cells in the three-dimensional tumor niche in vivo. To overcome this insufficiency, three-dimensional cell culture methods in a scaffold-dependent or -free physical environment have been developed. In this study, we improved and simplified the HCT-8 colon cancer cell-based spheroid culture protocol and evaluated the relationship between cancer stemness and responses of chemosensitivity to 5- Fluorouracil (5-FU), a representative anticancer agent against colon cancer. Supplementation with defined growth factors in the medium and the culture dish of the regular surface with low attachment were required for the formation of constant-sized spheroids containing $CD44^+$ and $CD133^+$ colon cancer stem cells. The chemo-sensitivities of $CD44^+$ cancer stem cells in the spheroids were much lower than those of $CD44^-$ non-stem-like cancer cells, indicating that the chemoresistance to 5-FU is due to the stemness of colon cancer cells. Taken together, the inflammation and oncogenic gut environment-sensitive HCT-8 cell-based colon cancer spheroid culture and comparative evaluation using the simplified model would be an efficient and applicable way to estimate colon cancer stemness and pharmaceutical response to anticancer drugs in the realistic tumor niche.

  • An effective micropropagation system for Liriodendron tulipifera L via somatic embryogenesis was established using immature seeds. Immature seeds from five individual trees were bisected longitudinally and cultured on two basal media (MS and B5) containing different combinations of 2,4-D and TDZ to induce callus and embryogenic tissue under light ($40{\mu}mol\;m^{-2}s^{-1}$, 16 hr/day) or complete darkness at $25{\pm}2^{\circ}C$. There was no distinctive difference on callus and embryogenic tissue induction between the two basal media with PGRs. Optimum culture medium appeared to be MS medium supplemented with 1.0mg/L 2,4-D and 0.01mg/L TDZ plus 3% sucrose. Nonembryogenic callus induction rate was not significantly different among the genotypes. However, However, the embryogenic callus induction frequency differed greatly by the genotypes ranging from 55% to 72% when cultured in the dark. Generally, the cultures maintained in the dark tended to show normal somatic embryo development as well as embryogenic tissue formation and this was confirmed by histological examination. Above results suggest that a proper selection of mother tree and dark culture condition are necessary to optimize somatic embryogenesis system of Liriodendron tulipifera.

Mass production of potato microtubers by bioreactor culture (바이오리액터 배양기에 의한 감자소괴경 대량 생산)

  • Highest increase of biomass was observed when tissue-cultured potato (Solanum tuberosum L. cv. Chubaek) shoots were cultured in a liquid medium containing 1/3 MS solution in a 18 L bioreactor, as compared to 1/4 and 1/2 MS solution. The medium containing 1/4 MS solution showed higher increase of shoot biomass than one containing 1/2 MS solution. Potato microtubers were formed when the medium was exchanged with the medium for microtuber formation and incubated under dark condition. The microtubers were observed first at some axillary buds one week after incubation under dark condition and then at most of the axillary buds by the end of 3 weeks. The 1.5 MS liquid medium and $20^{\circ}C$ were optimal conditions. By the end of 6 weeks, more 1,000 microtubers were formed in the 18 L bioreactor. Then, greened microtubers were harvested after one week culture under light condition.

  • The experiment was carried out to investigate the influence of light condition and polarity of explant on adventitious shoot formation from cotyledon of apple in vitro The treatment of light culture after 10days dark treatment showed effective result than other treatments on the rate of adventitious shoot formation on light intensity treatments and also the treatment of blue light treatment after 4days dark treatment showed more effective result than other treatments in light quality treatments. The polarity of explant influence on adventitious shoot formation. Adventitious multiple shoot formation occured at the proximal end of an excised cotyledon. In other words. shoot organogenesis occured at the proximal cut surfaces of both proximal and distal explants rather than at the distal cut surface of proximal explants.

  • Effects of salicylic acid (SA) on anthocyanin synthesis in cell suspension cultures of grapes (Vitis vinifera L.) were investigated. tow concentrations (0.1 to 1$\mu$M) of SA did not affect the cell growth and anthocyanin accumulation whereas high concentrations (5 to 10$\mu$M ) of SA inhibited cell growth with increasement of anthocyanin synthesis. Five micromoles of SA promoted anthocyanin accumulation 4 folds compared to control cells. When SA was treated on the different culture times (0 to 7day), the highest pigment accumulation was obtained at the cells of second day. A low productivity of anthocyanin under continuous dark incubation was also recovered by adding SA which mimicked light irradiation effect These results suggest that SA is one of essential agents in anthocyanin biosynthesis.

Chemopreventive Effects of the Extracts from Soybean Fermented with Basidiomycetes (들송이(Agrocybe cylindracea)와 말똥진흙버섯(Phellinus igniarius)을 배양한 대두추출물의 암예방 효과)

  • The inductions of phase II enzymes, such as NAD(P)H : quinone reductase (QR), glutathione S-transferase (GST), glutathione (GSH) level and the inhibition of polyamine metabolism were tested for the chemopreventive potentials of the extracts from the soybean fermented with Agrocybe cylindracea (AC) or Phellinus igniarius (PI). The soybean fermented with AC or PI was potent inducer of QR activity in murine hepatoma Hepa1c1c7 cells. GST activities of the extracts from soybean fermented with AC or PI were higher than that of the extract from soybean not fermented with basidiomycetes. In addition, GSH levels of the extracts from soybean fermented with AC or PI were increased about 1.2 fold or 1.4 fold, respectively. In addition, proliferation of Acanthamoeba castellanii in a broth medium was inhibited by the extracts from soybean fermented with AC or PI at the both concentration of 20 and 40 mg/3 ml. These results suggest that soybean fermented with AC or PI may have chemopreventive potentials by inducing QR activity, increasing GSH and GST levels and inhibiting polyamine metabolism.
