Inhibitory Effect of PG-Platycodin D on the Development of Atopic Dermatitis-Like Skin Lesions in ICR Mice (Platycodin D를 포함하는 도라지 추출물이 DNCB 유도 알레르기성 아토피 피부염에 미치는 영향)

  • Atopic dermatitis is characterized by chronically pruritic and inflammatory dermatitis. In this study, we investigated the effect of Platycodon grandiflorum including platycodin D (PG-Platycodin D) in an atopic dermatitis-like mouse model. An atopic dermatitis-like skin lesion was induced by repeated treatment of 2,4-dinitrochlorobenzene (DNCB) on the dorsal skin of ICR mice. The efficacy of PG-Platycodin D was tested by observing scratching behavior, the skin severity score, and histopathologic analysis. PG-Platycodin D reduced the DNCB-induced increase in scratching behavior and the skin severity score. In addition, histopathologic analysis revealed a reduction in the thickening of the epidermis in the PG-Platycodin D group. These results may contribute to the development of a therapeutic drug for the treatment of atopic dermatitis.

  • With the advent of 'The Forth Industrial Revolution' and the growing demand for quality of life due to economic growth, needs for the quality of medical services are increasing. Artificial intelligence has been introduced in the medical field, but it is rarely used in chronic skin diseases that directly affect the quality of life. Also, atopic dermatitis, a representative disease among chronic skin diseases, has a disadvantage in that it is difficult to make an objective diagnosis of the severity of lesions. The aim of this study is to establish an intelligent severity recognition model of atopic dermatitis for improving the quality of patient's life. For this, the following steps were performed. First, image data of patients with atopic dermatitis were collected from the Catholic University of Korea Seoul Saint Mary's Hospital. Refinement and labeling were performed on the collected image data to obtain training and verification data that suitable for the objective intelligent atopic dermatitis severity recognition model. Second, learning and verification of various CNN algorithms are performed to select an image recognition algorithm that suitable for the objective intelligent atopic dermatitis severity recognition model. Experimental results showed that 'ResNet V1 101' and 'ResNet V2 50' were measured the highest performance with Erythema and Excoriation over 90% accuracy, and 'VGG-NET' was measured 89% accuracy lower than the two lesions due to lack of training data. The proposed methodology demonstrates that the image recognition algorithm has high performance not only in the field of object recognition but also in the medical field requiring expert knowledge. In addition, this study is expected to be highly applicable in the field of atopic dermatitis due to it uses image data of actual atopic dermatitis patients.

  • Objective : It is well known that patients with atopic dermatitis are prone to cutaneous infections. Especially Staphylococcus aureus has been regarded as having an influence on atopic dermatitis. We observed and treated a case of an adult atopic dermatitis patient with sepsis by cooperation of east-west medicine. We present this case because there were no former reports of atopic dermatitis with sepsis treated by cooperation of east-west medicine. Method : We used herbal medication, acupuncture, herbal wet dressing combined with antibiotics and analgesics for 19 days. Atopic dermatitis grade was determined using SCORAD index. Result : The SCORAD index was lowered 32.3 at discharge than before of which the SCORAD index was 62.5 at admission. Other systemic inflammatory response such as fever, pus was also resolved. Conclusion : From this result, we suggest that herbal therapy associated with antibiotic therapy were shown to treat atopic dermatitis with secondary infection effectively.

  • A canine patient exhibited partial anorexia and sudden respiratory distress. Diagnostic imaging and cytology of tracheal-lavage fluids revealed fungal pneumonia. Molecular identification and phylogenetic analysis detected Candida catenulata. Treatment with oral itraconazole for 3 weeks was effective. This is the first report of C. catenulata infection in a dog.

  • SNS 의 발달로 이를 활용한 제품의 광고가 활발하게 이루어지고 있다. 다양한 제품군 중에서도 사용자의 피부 및 건강의 개선 효과가 나타나는 화장품, 건강보조제 등은 후기 글을 보고 실제 효과를 판단하기에 어려움이 있다. 이는 많은 양의 광고에 가려진 실질적 후기를 찾는 것이 어렵고, 포스팅의 전문을 읽는 것은 비효율적이라는 점에서 기인한다고 할 수 있다. 본 논문에서는 소셜 미디어를 바탕으로 아토피 치료법의 효과를 분석할 수 있는 효과 분석 모델을 개발하고 그 결과를 제시하였다. 먼저 많은 후기가 존재하는 키워드를 기반으로 최대 1000 개의 블로그 포스팅을 수집하였고, 광고성 글을 제외하는 자동 처리 알고리즘을 실시하였다. 다음으로 각각의 후기 글에 나타난 효과를 한눈에 알아볼 수 있도록 점수화하는 효과 분석 알고리즘을 제안하고 실험하였다. 실험결과 감마리놀렌산, 플라즈마, 락토바실러스 등이 긍정적 효과가 있는 치료법으로 나타났다. 본 논문에서 제시한 알고리즘은 제품의 효과를 점수화할 수 있으므로 아토피 치료법에 한정되지 않고, 해당 제품군인 화장품 및 건강보조제 등에 다양하게 적용될 수 있을 것으로 보인다.

Anti-Allergic Activities of Ultra-fine Powder from Persimmon (감 초미세 분말의 추출물을 이용한 항산화 및 항아토피 활성)

  • Ultra-fine powder (FA) extracts from persimmon were investigated as potential food materials in industrial processes because such extracts have various useful biological activities. We examined the antioxidant and anti-allergic activity of FA extracts using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and ferric reducing activity of plasma (FRAP) models, and by measuring interleukin 4 (IL-4) expression in spleen cells and in an atopy animal model. FA extracts were prepared using solvents including distilled water and 50% (v/v) acetone. The results showed that FA extracts had potent DPPH and FRAP activities, decreased IL-4 expression in spleen cells, and inhibited ear thickness and inflammation in a mouse animal model. The data indicate that a FA extract could be used as an antioxidant and/or anti-inflammation agent, and in industrial food preparation processes.

  • Recently, since atopic dermatitis is sensitive to skin irritation, it has been suggested that the development of a patch that can effectively exhibit adhesion and absorption to a specific local area while minimizing skin irritation, and capable of appropriate drug release should be given priority. In this study, we tried to develop a hydrogel patch that minimizes skin irritation, adheres effectively to a specific area, and promotes absorption. The atopic patch was formulated into a super-absorbent hydrogel sheet using a freeze drying method. Cell viability assay was carried out using keratinocytes (HaCaT cell) and fibroblasts (L929 cells). In order to investigate the physical properties, FT-IR, FE-SEM, porosity analysis and swelling behavior were investigated. As a result, the newly prepared HA-Dex hydrogel patch was verified by biocompatibility and physical evaluation. In addition, the manufactured hydrogel patch has sufficient moisture absorption capacity and can relieve itching of atopic skin, and is expected to be applied to various drug delivery products for the treatment of atopic dermatitis in the future.

Anti-inflammatory and Anti-allergic Effects of Herbal Extracts on Atopic Dermatitis ( Part II ) (약용식물 추출물의 아토피성 피부염에 대한 항염증 및 항알레르기 효과 (제 2 보))

  • Atopic dermatitis is a chronic, relapsing inflammatory skin disease associated with dysfunction of skin barrier and cutaneous hyper-reactivity to environmental triggers. In the previous study, cytotoxicity, antioxidant, anti-inflammatory and anti-allergic activities were investigated for various herbal extracts such as Aloe vera L. (AV), Viola mandshurica W. Becker (VM), Punica granatum L. (PG), and Dendrobium nobile L. (DN) in order to develop effective therapeutic herbal extracts for atopic dermatitis, In this study, anti-inflammatory activities of these herb extracts in lipopolysaccharide (LPS)-induced macrophage RAW264.7 cells were further examined to find the underlying molecular mechanisms. The RT-PCR (reverse transcription polymerase chain reaction) analysis showed that PG, DN and AV inhibited effectively the gene expression of pro-inflammatory cytokines IL-6 and IL-$1{\beta}$ in LPS-stimulated macrophages, while VM did not. The transfection and luciferase analysis exhibited that all herbal extracts hindered the activation of transcription nuclear factor kappa B (NF-${\kappa}B$). The western blot analysis indicated that AV blocked the activation of only JNK MAP (c-Jun N-terminal kinase mitogen-activated protein) kinase not p38 MAP kinase, while VM, PG and DN did not show the activation of both JNK and p38 MAP kinases. These results suggest that AV, VM, PG, and DN have anti-inflammatory activities and thus have the potential to reduce and alleviate the symptoms of atopic dermatitis.

Effect of Anti-atopic Allergic Reaction in Response to Oriental Herb Extracts (생약재 추출물의 아토피 완화효과)

  • The in vivo and in vitro effects of oriental herb extracts of Cassia obtusifolia, Taraxacum platycarpum and Ulmusmacrocarpa on anti-atopic allergic reaction were evaluated in this study. A mixture of these extracts exhibited more potent anti-allergic activities in human mast cells than those from individual extracts. The herbal mixture significantly inhibited the release of compound 48/80-induced $\beta$-hexosaminidase release in the human mast cell line, HMC-1. The mixture also suppressed the production of PMA and A23187-induced inflammatory cytokines in HMC-1 cells. To further investigate the in vivo effects of the herbal mixture, a Dermatophagoides farinae (DF)-induced atopic dermatitis mouse model was utilized. Oral administration of the herbal mixture significantly decreased the ear thickness and swelling in DF treated NC/Nga mice in a dose dependent manner. Furthermore, serum levels of IgE and interleukin-4 (IL-4) were significantly decreased, whereas interferon-gamma (IFN-$\gamma$) levels were increased in the mixture administrated groups when compared to the control. Taken together, our data indicate the possibility of using a mixture of the oriental herb extract to relieve symptoms of atopic dermatitis.

Inhibitory Effect of Angelica keiskei Extract in an Atopic Dermatitis Animal Model (신선초 추출물의 아토피 억제활성)

  • The purpose of this study was to evaluate the anti-atopic-dermatitis effect of Angelica keiskei extract using a DNFB-induced animal model of atopic-dermatitis symptoms. A. keiskei was prepared via extraction with DW, 50% ethanol, and 100% ethanol in addition to fresh juice. All the extracts reduced the thickness levels of the ears and ear epidermis against swelling by DNFB inducement, especially in the aqueous-extract-treated group. The mRNA expressions of matrix metalloproteinases (MMPs), however, were not observed, which may indicate that Angelica keiskei extract alleviates the atopic-dermatitis symptoms in an MMP-independent manner. Finally, the level of the inflammatory cytokine IL-4 was inhibited only in the juice-treated group, although the IL-13 level was inhibited in the juice-, 50%-ethanol-extract-, and 100%-ethanol-extract-treated groups in a dose-dependent manner, and was not inhibited in the aqueous-extract-treated group. Taken together, these results suggest that Angelica keiskei extract has an inhibitory effect on atopic dermatitis, and may be a useful biomaterial for the development of cosmeceuticals.
