Effect of medicinal plant extract for hangover relief (약용식물 추출물의 숙취 해소 효과에 관한 연구)

  • The present study was performed to evaluate the effect of medicinal plant extract on relieving hangovers in mice administered alcohol. The animals were divided into three groups. Each group was treated with fermented plant extract, non-fermented plant extract, or water 30 min after consuming ethanol (2 mL/kg). A locomotor activity test showed that all groups had decreased motor activity until 40 min after plant extract administration. The mice treated with water had lower motor activity until 100 min post-administration. However, the group treated with non-fermented plant extract showed increased motor activity 40 min post-administration, and the higher activity level was maintained until 120 min post-administration. The animals treated with fermented plant extract had a level of motor activity between those of the groups treated with water or non-fermented plant extract. Blood was collected from each mouse 120 min post-administration and aldehyde concentration was measured. The group treated with non-fermented plant extract had a significantly higher (p < 0.05) aldehyde concentration than the other groups. These results demonstrate that the non-fermented medicinal plant extract helped alleviate hangovers 40 min after administration by reducing aldehyde concentrations in the blood.

  • Reactive oxygen species (ROS) are continuously produced at a high rate as a by-product of aerobic metabolism. Several lines of evidence provided that ROS appears to cause to develop aging and various diseases. High level of hyaluronic acid with decreased molecular weight has been detected in patients with inflammatory diseases including rheumatoid arthritis. In this study, we have conducted to investigate the antioxidant and hyaluronidase inhibitory activities of Prunus persica Batsch var. davidiana Maximowicz in order to screen the bioactive substances which can be developed as possible anti-inflammatory agents. Among the extracts of Prunus persica Batsch var. davidiana Maximowicz, EtOAc extract exhibited the strongest effect on antioxidant experiment such as DPPH, Ferric- Thiocyanate and Rancimat. Also, EtOAc extract showed a potent hyaluronidase inhibitory activity.

  • This experiment was conducted to study the effects of medicinal plant extract on performance in broiler chickens. A total of eight hundred forty, 1-d-old broiler chicks(Ross) were randomly allocated to 7 treatments with 4 replicates of 30 birds per replicate. The treatments were negative control (NC, diets without antibiotics), positive control (PC, diets with-containing antibiotics), Schisandrae fructus (T1), Scutellariae radix (T2), Camellia sinensis (T3), Artemisia capillaris (T4) and mixed medicinal plants (T5) extracts added to drinking water. The final body weight, body weight gain and feed conversion rate were significantly improved in all treatments compared to NC (P<0.05). No significant differences were observed in the relative weights of liver, spleen, pancreas, bursa of Fabricius and intestinal tract among the groups. The relative lengths of duodenum, jejunum and ileum were significantly decreased in PC compared with other groups (P<0.05). No significant differences were observed in the cecal microflora. The contents of triacylglycerol (TG), blood urea nitrogen (BUN), glutamic-oxaloacetic transaminase (GOT) and glutamic-pyruvic transaminase (GPT) in blood serum were not significantly influenced. The concentration of cholesterol in blood serum was significantly decreased in all treated groups as compared to those of the control (P<0.05). There was no difference in the components of leukocytes and erythrocytes among the groups.

Screening of Some Plant Extracts for Inhibitory Effects on HIV-1 and Its Essential Enzymes (Human Immunodeficiency Virus Type I에 대한 수종 식물 추출물의 억제활성 검색)

  • In order to elucidate the relationship between anti-HIV-1 enzyme activity and inhibition of HIV-1 replication by natural sources, extracts from some plants using the foods and oriental medicines were tested for inhibitory effects on the viral replication, reverse transcriptase (RT), protease and ${\alpha}-glucosidase$. In the anti-RT test, water extracts of Ficus carica (leaf), Houttuynia cordata (aerial part) and Ixeris tamagawaensis (aerial part) showed more than 79% inhitibion at a concentration of $100\;{\mu}g/ml$. The protease and ${\alpha}-glucosidase-inhibiting$ samples in the screening were water extract of Syringa dilatata (leaf) and methanol extract of Hibiscus syriacus (leaf and stem), which showed more than 40% inhibition at a concentration of $100\;{\mu}g/ml$. In the primary anti-HIV-1 test, water extracts of Equisetum arvense (aerial part), Hibiscus syriacus (leaf), Ixeris tamagawaensis (aerial part) and Pueraira thunbergiana (leaf) showed the potent inhibition against HIV-1 induced cytopathic effects.

  • The antioxidative activity of marine microalgae, Phaeodactylum tricornutum (P. tricornutum) of Bacillariophyceae, was determined by measuring radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. The chloroform fraction of P. tricornutum showed strong antioxidative activity, The potential antioxidative activity of factions extracted with mixture solution of organic solvents was detected in dichloromethane : methanol (2 : 1) fraction, This fraction was further purified by preparative thin layer chromatography (PTLC) and repeated reverse-phase HPLC. On the basis of chemical and spectoscopic evidence from results obtained by UV, FT-IR, EIMS and NMR, the compound purified from P. tricornutum was identified as zeaxanthin.

Screening of Chinese Herbal Medicines with Inhibitory Effect on Aldose Reductase (III) (중국 약용식물 추출물의 알도즈 환원 효소 억제 효능 검색 (III))

  • Aldose reductase (AR) is a critical enzyme in the development of the diabetic complications. AR, the first enzyme in the polyol pathway, catalyzes the reduction of the aldehyde form of glucose to sorbitol with concomitant conversion of NADPH to $NADP^+$. None of aldose reductase inhibitor (ARI) has achieved worldwide use because of limited efficacy or undesirable side effects. Therefore, evaluating natural sources for ARI potential may lead to the development of safer and more effective agents against diabetic complications. Forty eight Chinese herbal medicines have been investigated for inhibitory activities on AR. Among them, seven herbal medicines, Buddleja officinalis (whole plant), Lonicera japonica (leaf and flower), Polygonum aviculare (aerial part), Polygonum aviculare (whole plant), Salvia miltiorrhiza (root), Schisandra chinensis (stem), and Zanthoxylum armatum (leaf and stem) exhibited a significant inhibitory activity against AR. Particularly, L. japonica and P. aviculare showed two times more potent inhibitory activity than the positive control, 3,3-tetramethyleneglutaric acid (TMG).

  • Aldose reductase (AR), the key enzyme in the polyol pathway, plays an important role in the pathogenesis of diabetic complications such as cataract formation. AR inhibitors (ARI) can prevent or reverse early abnormalities in diabetic complications. However, none of ARI has achieved worldwide use because of limited efficacy or undesirable side effects. Therefore, evaluating natural sources for ARI potential may lead to the development of safer and more effective agents against diabetic complications. 49 Vietnam herbal medicines have been investigated with an in vitro evaluation system using aldose reductase inhibitory activities. Of these, 7 herbal medicines, Artemisia annua (leaf, stem), Duranta repens (leaf, stem), Excoecaria cochinchinensis (aerial part), Flueggea virosa (leaf, stem), Khaya senegalensis (leaf), Polygonum orientale (aerial part), Toxicodendron succedanea (leaf, stem) exhibited a significant inhibitory activity against aldose reductase. Particularly, Duranta repens (leaf, stem) and Toxicodendron succedanea (leaf, stem) showed two times more potent inhibitory activity than the positive control, 3.3-tetramethyleneglutaric acid (TMG).

  • 피마자유(Castor oil)는 합성수지, 그리스, 유압용 오일, 윤활기유 등의 다양한 용도로 쓰이는 오일로서 점도가 높고 무색에서 황갈색을 띈다. 피마자유 추출은 압착 착유 또는 용매 추출로 얻게 되며 본 실험에 사용된 오일은 압착 착유한 것으로 매우 진한 갈색을 띄는 정제전 오일을 이용하였다. 실험에 사용된 피마자유의 초기 산가는 1mgKOH/g 이하로 낮은 유리지방산 함량을 보였으며 수분은 0.3%로 전이에스테르화 반응을 위해서는 수분 증발이 필요했다. 피마자유의 바이오디젤 생산을 위해 진행된 물성 분석 항목은 산가, 수분, 인함량, 황함량, 점도, 고형물이며 원료유와 그 바이오디젤에 대해 각각 물성 분석을 실시하였다. 피마자유의 가장 큰 특징은 다른 식물성 오일과는 다르게 오일이 알코올에 녹는 특성이 있으며 이런 이유로 전이에스테르화 반응 후 바이오디젤과 글리세롤이 분리되지 않는 문제점을 갖고 있다. 피마자유를 이용해 제조한 바이오디젤, 즉 지방산메틸에스터의 함량을 분석한 결과 약 90%의 메틸에스터화 반응 전환율을 나타내었으나 국내외 품질규격상의 탄소수 C14~C24:0의 지방산 에스터(fatty acid methyl esters)로 검출되는 바이오디젤의 함량은 10% 미만으로 나타났으며 나머지 90%는 라이신올레익산메틸에스터(ricinoleic acid methyl ester)로 분석되었다. 따라서, 기존의 대두유, 유채유, 팜유, 폐식용유로부터 제조한 바이오디젤과 물성이 매우 상이하고 특히 끊는점(boiling point)과 점도가 높아 경유 대체연료로는 활용이 불가능할 것으로 판단된다. 하지만 기존의 다양한 용도의 오일로 사용하기 위해 정제하는 과정에서 전체 착유 오일중의 약 10%만을 선택적으로 분리하여 바이오디젤 원료로 활용하는 방안은 가능할 것으로 판단된다.

  • Aldo-keto reductase family 1 B10 (AKR1B10) has been considered as a potential cancer therapeutic target. Ethanol extracts prepared from 82 Korean local plants were examined for their antioxidant activity and inhibitory effects on recombinant human AKR1B10 (rhAKR1B10) in vitro. 21 extracts showed more than 80% of ABTS radical scavenging activity at $100\;{\mu}g/mL$ and 11 extracts inhibited more than 50% of rhAKR1B10 activity at $10\;{\mu}g/mL$. Especially, 9 extracts showed potent inhibition on rhAKR1B10 activity compared with positive control tetramethylene glutaric acid.

  • To induce the phytoalexins in plant cell culture systems, we surveyed the antimicrobial activity following the feeding of five naphthoquinones in cell cultures of petunia. Among naphthoquinones treated, 2,5,7-trihydroxy-3-(5'-hydroxyhexyl)-1,4-naphthoquinone (3-OH NQ ) was most efficiently absorbed into the cells within 48 hr. The crude extracts of cells treated with 3-OH NQ showed a strong inhibition activity on spore germination of Aspergillus candidus $(MIC:\;32\;{\mu}g/ml)$, whereas the untreated cells showed no activity. The two active compounds, 4,2',4',${\beta}$-tetrahydroxychalcone and 4',7-dihydroxyflavone, were isolated from petunia cells treated with 3-OH NQ. The major phytoalexin, 4,2',4',${\beta}$-tetrahydroxychalcone, inhibited strongly the spore germination of A. candidus $(MIC:\;16\;{\mu}g/ml)$.
