Isolation and Identification of a Purple, Non-Sulfur Bacterium from Korea Coast

  • A species of facultative photo-organotrophic, purple, non-sulfur bacterium was isolated from the west coast and the south coast 47 area of Korea at 2001 September. Separated 13 samples of changes with red color under $28{\sim}32\;^{\circ}C$, 3000 lux, anaerobe conditions for 7 days cultivated in Basal medium. For a pure isolation from 13 samples it used agar-shake tube method (0.4 % agar) and it separated 5 strains through 13-repetition test. The RAPD(Random Amplified Polymorphic DNA)-PCR result of strains (EGH-9, EGH-13, EGH-23, EGH-24, EGH-30) that EGH-24 and EGH-30 was same strain. For wastewater biodegradation test that 4 isolation strains cultivated in synthesis wastewater in 7 days. EGH-24 was high 63000 mg/L (CODcr) to 43400 mg/L (CODcr). EGH-24 was selected with efficient wast water treated strain. Based on the results obtained from morphology, nutrient requirements, major bacteriochlorophyll content, 165-rDNA phylogenetic analysis, this strain may be identified as a new strain of the genus Rhodobacter and named Rhodobacter sp. EGH-24.

A Study on the Neo-Classicism Expressed in Contemporary Fashion Design (현대복식에 표현된 신고전주의 특성에 관한 연구)

  • The purpose of this thesis is to study the social and cultural background and characteristic of neo-classicism which appered in the modern fashion of the pluralistic society of the latter half of 20th century to understand that the product of societh such as fashion mode reflects the situation of society and culture. For this purpose, documentary studies about the concept and background of neo-classicism, were preceded, and analyzed the occurrence background and characteristic of neo-classicism of the modern fashion after the 1980's, which showed up in post-modernism fashion. The characteristics of neo-classicism expressed in modern fashion is as follows; First, they are expressed in modern fashion in forms of simplicity by means of minimizing process of sewing or ornamenting and deletion of dart. Second, retro-style in terms of concerning of the past, is featured in various styles of Greco-roman drapery and expresed in forms of mixing clothing elements of modern and classic in modern times. Third, the trend of ecology in terms of interest of the nature and the thought that the spiritual world is more important than the material world. It is expressed by using natural elements adn natural materials, which wanted to get the nature and human into one and search for the losed nature of modern men. Fourth, the pursuit of the beauty of the human body, is expressed in forms of body-prioity style through using see-through or elastic materials, which is knit, lycra, spandex, etc. In conclusion, we can recognize that the social product reflects social and cultural situation. And the characteristic of neo-classicism has the meaning of harmonizing the human and nature and the returing the humanity.

Antioxidative Enzyme Activities and O-J-I-P Transients of Crinum asiaticum var. japonicum $B_{AK}$. Leaves under Winter Stress (겨울철 저온스트레스에 의한 문주란 잎의 항산화효소 활성과 O-J-I-P 곡선 패턴)

  • The diurnal and seasonal variations of antioxidative enzyme activity and the O-J-I-P transients were investigated from the leaves of Crinum asiaticum var. japonicum under winter stress in natural habitat, in order to diagnose quantitatively physiological states of plants under stresses. The activities of superoxide dismutase and peroxidase increased slightly in winter. Especially, peroxidase acitivity was higher at dawn and night in winter and some isoforms were detected only in early winter. In the O-J-I-P transients, the fluorescence intensity of J, I, P steps decreased significantly in winter season, contrary to its high value in summer season. Of the chlorophyll fluorescence parameters derived from the O-J-I-P transients, Fm and $\Phi_{po}$ decreased with the increase of ABS/RC depending on temperature drop in winter.

The Analysis of Actual State of Working Environment and Working Uniforms for Burning Waste Workers (생활폐기물 소각장 작업자의 작업환경과 작업복 현황 분석)

  • This study analyzes the actual state of the working uniforms worn by burning waste workers to protect their body from the hazardous environment. Directions for improvement of the clothing fabrics is also suggested. We visited and interviewed those persons who worked in the burning waste plant in Seoul and in Gyunggi Province. 245 workers were surveyed by questionnaires, and 211 workers out of 245 were statistically analyzed. Frequencies and percentages were calculated and $\chi$$^2$ test, ANOVA and Scheffe Post hoc comparison methods were utilized to test some hypothesis. The working uniforms were classified as four general types: separate type, disposable-coverall. overall, and winter clothes. At present, the working uniforms are not suitable for the conditions which they are used. There is a lack of ventilation and an inability to absorb perspiration limited elasticity, and the uniforms are prone to static shock. The degree of satisfaction varied significantly, based on the worker's educational background and main work. However, only a small percentage of workers interviewed were satisfied with the appearance and the fit of the present working uniforms. The working duration had a negative impact on the satisfaction of the worker. This results suggest that materials, colors, design and size of working clothes must be improved. The suggestions are as follows: Firstly. to use disposable non-woven fabrics finished to increase moisture regain. Secondly. to use the bad air permeability fabrics to protect the skin from absorbtion of harmful gases, flying dust, heavy metals, etc.

Study on the Resazurin test as a Rapid Method for the Estimation of Maturity of Kimchi (김치숙성도 판정기준을 위한 신속 검사법 Resazurin-test 에 관한 연구)

  • An attempt was made to establish a simple and rapid method for the evaluation of Kimchi quality. Four kinds of Kimchi included one home made traditional Kimchi were stored at $4^{\circ}C$ and$25^{\circ}C$ and biochemical changes, including pH, total acidity, volatile acidity, volatile basic nitrogen, vitamin C, NaCl, glucose, nitrite and resazurin value were determined and tested for the relationship with sensory evaluation. The change in pH, total acidity and resazurin value were highly correlated with the sensory maturity of Kimchi-Resazurin value was strongly influenced by the sample dilution and fermentation temperatures. It was concluded that resazurin test with sample solution of $2{\sim}4%$ could be used as a rapid method for the estimation of the maturity of Kimchi.

Effect of Na-Acetate, Na-Malate and K-Sorbate on the pH, Acidity and Sourness during Kimchi Fermentation (Na-Acetate 및 Na-Malate와 K-Sorbate가 김치발효중 pH, 산도 및 산미에 미치는 효과)

  • This paper was carried out to investigate the relation of pH, aciidity and sourness during Kimchi fermentation and preservatives on Kimchi fermentation. Na-acetate, Na-malate, K-sorbate and K-sorbate+acetic acid were added to Kimchi samples. These Kimchi samples were fomented for 7 days at $37^{\circ}C$ and $20^{\circ}C$. In the experiment about the sourness and buffer action by organic salts which showed that the intensity of sourness was differented by the difference of pH in the same acidity. Na-acetate (0.3%) and Na-malate (0.3%) acted as good buffer, whereas K-sorbate (0.1%) and K-sorbate (0.1%)+acetic acid (0.05%) acted as lactobacilli growth enhancer in the fermentation.

  • This study was carried out to investigate effects of the mindmap note-taking for the formation of the mathematical concepts structure and the matjematical creativity. Two classes were randomly chosen for this study from the third grade students of a middle school located in a medium size city. Thirty one lecture hours of the mindmap note-taking on the quadratic equation and functions were administered to the experimental class of 41 students, while same lecture hours of the ordinary instruction on the same contents were administered to the control class of 40 students. It was shown from this experiment that there ware significant evidences of improvement both in the formation of students' mathematical concepts structure and mathematical creativity through the mindmap note-taking lecture. Hence, the mindmap note-taking lecture is suggested for the improvement in the formation of student's mathematical concepts structure and mathematical creativity.

Comparison of Electricity Generation and Microbial Community Structure in MFCs Fed with Different Substrates (미생물연료전지에서 공급기질에 따른 전기발생량 및 미생물 군집구조 비교)

  • Electricity generation of microbial fuel cells (MFC) is greatly affected by the kind of feed substrates because substrates would change microbial community of electrochemically active bacteria (EAB) able to transfer electrons to electrode. The effect of different substrates on electricity generation and microbial community of MFC was investigated. Two-chamber MFCs fed with acetate (A-MFC), butyrate (B-MFC), propionate (P-MFC), glucose (G-MFC) and a mixture (M-MFC) of the 4 substrates (acetate : butyrate : propionate : glucose = 1 : 1 : 1 : 1 as $COD_{Cr}$ base) were operated under continuous mode. The maximum power density was found from the M-MFC ($190W/m^3$) which showed the lowest internal resistance ($89{\Omega}$). The maximum power densities of the pure substrates feed MFCs were in order of A-MFC ($25W/m^3$), P-MFC ($21W/m^3$), B-MFC ($20W/m^3$) and G-MFC ($9W/m^3$). In DGGE analysis, the microbial community structure in suspension was quite different from each others depending on feed substrates, while the community structure in the biofilm was relatively similar regardless of the substrates. This result suggests that the feed substrates would affect the microbial community of suspended growth bacteria than attached growth bacteria resulting in difference of electricity generation in MFCs.

Grain Boundary Trap Levels in ZnO-based Varistor (ZnO계 바리스터의 입계포획준위)

  • The trap levels of ZnO-based varistor are obtained by Isothermal Capacitance Transient Spectroscopy method. Here ICTS measuring system consists of YHP 4192A Impedance Analyzer and a personal computer for the data acquisition. Between $-40^{\circ}C$ and $60^{\circ}C$, the grain boundary trap levels of 0.48 and 0.94eV were detected for $ZnO-Bi_2O_3-MnO$ system. The hole omission spectra are observed in the case of the addition of CoO into the $ZnO-Bi_2O_3$ system, while the electron emission spectra are detected in the case of the addition of MnO. The nonlinear resistance coefficient $\alpha$ increases with the decrease of the dormer concentration. Finally, the trap level density of $ZnO-Bi_2O_3-MnO$ system is found to decrease with the amount of CoO, while $\alpha$ is found to increase with the amount of CoO.

Preparation of $Pb(Zr,\;Ti)O_3$ thin films by MOCVD using ultrasonic spraying (초음파분무를 이용한 MOCVD법에 의한 $Pb(Zr,\;Ti)O_3$박막의 제조)

  • Thin films of )$Pb(Zr, \;TiO_3$ are fabricated by MOCVD using ultrasonic spraying. The films having perovskite structure are made at low deposition temperature, $300-450^{\circ}C$. The phase and composition of the films vary with the composition of the starting solution and the deposition temperature. Dielectric constant of the films is 187 at 1MHz. Ferroelectric hysterysis loop measurements indicate a remanant polarization of $5.5{\mu}C/cm^2$, and coercive field of 65kV/cm. Resistivity of thin films is about $10^{11}{\Omega}cm$ and the breakdown electric field abort 35kV/cm.
