  • This study evaluated disease resistance responses of pure lines of tomato plants at various temperature conditions against Ralstonia solanacearum strains isolated from Korea. Evaluation of six tomato lines with various strains of R. solanacearum showed that many strains can infect the resistant lines of tomato plants previously known as highly tolerant to bacterial wilt. One of the most virulent strains, SL341 (race 1 and biovar 4) caused severe infection on all six tomato lines, irrespective of temperature. In contrast, a moderately virulent strain SL1944 (race 1, biovar 4) showed the remarkable difference in disease progress on some resistant lines dependent on temperature. Moneymaker and Bonny Best were susceptible to SL1944 at all tested conditions with different temperature. However, tomato lines, such as Hawaii 7998, Hawaii 7996, Bblocking which were previously known as highly tolerant lines, were severely infected by SL1944 at relatively higher temperature ($35^{\circ}C$ for 14 hr light and $28^{\circ}C$ for 10 hr dark cycle). The disease progress at high temperature was much faster than those at low temprature on the same tomato line and those on Moneymaker and Bonny Best at the same high temprature. This result suggested that R. solanacearum strains isolated in Korea were highly virulent to bacterial wilt resistant tomato lines and some strains may cause severe infection on those plants at higher temperature.

  • To estimate the genetic characteristics and cumulative power of discrimination (CPD) within Korean native commercial chicken, we used a total of 395 genomic DNAs from six breeds population (Korean Native Red chicken: R, Korean Native Yellow chicken: Y, Korean native Commercial Chicken: C, Ogal chicken: S, Hy-Line Brown: H, White Leghorn: W). Genetic diversity indices including mean allele number among loci, unbiased heterozygosity ($h_i$) within locus, effective number of alleles ($N_e$) and polymorphism information content (PIC) as well as the unbiased average heterozygosity (H) among loci in the populations were calculated using the generated allele frequencies by each marker. Frequencies of microsatellites markers were used to estimate heterozygosities and genetic distances. The nearest distance (0.119) was observed between the C and Y strains. The generated unbiased average heterozygosity among loci in each population was integrated to the global formula of CPD and the result demonstrated that the CPD within the six chicken populations was 99.461%.

  • Hypocotyl explants from 7 days old seedlings of one $F_1$ hybrid cultivar and two pure lines of cucumber formed embryogenic calli at frequencies of up to 8% when cultured on Murashige and Skoog medium (MS) supplemented with 1 mg/L 2,4-D for 3 weeks. Embryogenic calli gave rise to somatic embryos. When slices of somatic embryos were cultured on the same medium for 4 weeks, they formed embryogenic calli. Embryogenic cell suspension cultures were established with embryogenic calli in MS liquid medium with 1 mg/L 2,4-D. Embryogenic potential of cell suspension cultures was maintained by subculturing every seven days. When the level of 2,4-D in the medium was lowered to 0.2 mg/L by diluting with liquid MS basal medium, embryogenic cell suspension cultures underwent development into numerous somatic embryos. When plated onto MS basal medium, over 95% of somatic embryos developed into plantlets. Plantlets were transplanted to potting soil and grown to maturity.

  • Fragaria ${\times}$ ananassa (Duch.) cultivars, 'Wongyo 3 115' a nd 'Wongyo 3 116', we re bre d as a n inte rme diate parent l ine using heterosis of $F_1$ hybrids in octoploid strawberry. These two new cultivars were developed as inbred lines ($S_9$ generation) propagated from both seed and runners/stolons. 'Wongyo 3115', an inbred line derived from the 'Benihoppe' cultivar, has a semi-upright plant shape showing a vigorless growth habit, weaker than the original 'Benihoppe' cultivar; it has early flower bud differentiation and produces 10 flowers per cluster when grown from healthy seed. 'Wongyo 3115' has low yields of conical type fruit with pink colored skin; average fruit weight is approximately 11 g. The important characteristic of 'Wongyo 3115' is its excellent fruit firmness, firmer than the original variety. 'Wongyo 3116', an inbred line derived from the 'Doyonoka' cultivar, has an upright plant shape, vigorless growth habit, weaker than that of the original 'Doyonoka' cultivar; it has early flower bud differentiation and produces 8 flowers per cluster when grown from healthy seed. 'Wongyo 3 116' has oblate type fruits with red skin and an ave rage weight of approximately 12 g. The important characteristics of 'Wongyo 3116' are excellent fruit shape and higher yield than other inbred lines, although it produces small fruits compared to the original cultivar. Both cultivars have excellent taste, high sugar/acid ratio, and good texture. In regards to disease and pest resistance, 'Wongyo 3115' and 'Wongyo 3116' have a tendency to be sensitive to powdery mildew, anthracnose and the two-spotted spider mite.

  • This study aimed to identify the growth performance of Korean indigenous chicken pure-line by sex and twelve strains conserved in Poultry Research Institute, National Institute of Animal Science, Rural Development Administration. The effect of sex and strain on body weight was significantly different in every period, with males being heavier in all periods than females. In the case of biweekly weight gain, the tendency to increase rapidly from birth to six weeks old, and to decrease in the period from twelve to fourteen weeks old was common across all sex and strains. Depending on sex and strain, there were significant differences in age and the number of peaks. Regardless of sex and strain, the determination coefficient and adjusted determination coefficient showed high goodness of fit (99.1~99.9%) to growth functions. However, for each model, the goodness-of-fit had variations by sex and strains. von Betalanffy function had the best fit to growth curves in all the female strains except strain D. On the other hand, Gompertz function had the best fit for all the male strains except strain C. Logistic function showed the lowest goodness-of-fit in all sex and strains. Mature weights were in the order of von bertalanffy, Gompertz, and Logistic models, while growth ratio and maturing rate followed the order of logistic, gompertz, and von bertalanffy functions. This information could be useful for Korean indigenous chicken management and designing crossbreeding tests and breeding programs.

  • This experiment was conducted to evaluate the diversity and purity of the Korean ginseng (Panax gjnseng) lines developed by the pure line selection using RAPD markers. Four primer (OPA 19, OPM 11, URP 3 and UBC 98) out of the 48 primer tested produced band which showed within-line polymorphisms at least in one line. Within-line polymorphisms were detected in six lines by OPA 19, in four lines by URP 03, in five lines by OPM 11, and in one line by UBC 98 respectively. Five plants obtained from the commercial cultivar 'Cheonpung' were differentiated using the primers OPA 19 and OPM 11. Five plants obtained from the 'Yeonpung were differentiated using the primer OPM 11. Detection of within-line RAPD polymorphisms might be attributed to the fact that cross pollination appear in P. gjnseng and a long period of three to four years required to reach the reproductive stage thereby delay the process to homozygosity.

  • To obtain pure lines for breeding $F_1$ varieties of Primula malacoides, 10 lines that were considered uniform in growth and flowering characteristics were selected from self-crossings of the sixth generation ($S_6$). Plant height of the selected lines ranged from 20.8 to 33.2 cm, and number of leaves ranged from 33.6 to 54.2. The selected lines flowered at 181.4 to 207.8 days after sowing. Three lines, including 'PR-03-81', flowered in less than 190 days from sowing, while six lines, including 'PR-03-228', only flowered after more than 200 days from sowing. In terms of petal color, six lines, including 'PR-03-81', were pink, three, including 'PR-03-236', were purple, and line 'PR-03-275' was white. Flower diameter ranged from 2.4 to 2.9 cm, and number of pedicels per plant was between 15.4 and 26.4. Number of flowers per pedicel ranged from 20.2 to 30.2. Germination was more than 70%, and number of seeds per capsule (pod) ranged from 110.0 to 162.8. In summary, plant height was low and flower diameter was small and plants had many flowers and readily formed seeds. The selected lines are not only valuable as ornamental plants but could also be used as breeding materials for $F_1$ hybrids of Primula polyantha.

  • In this study, a total of 25 lines consisting of five native ginseng collections from Poonggi area, five lines from Geumsam area and 15 varieties were analyzed and clustered for the selection of Poonggi native ginseng in Korea using DNA markers. The results indicated that the long cluster distance were observed between the collections of 331002, 331004, 331005, 331007 and 331026 from Poonggi area, and the collections of 332009, 332021, 332046, 332050 and 332066 from Geumsan area because of the sensible differences on the number of leaves per stem, stem color and petiole color. Thus, the collections from Poonggi area with specific characters consisting of one stem per plant, five leaves per stem and broad elliptic leaflet shape were finely classified using nine primers including OPD05, OPD20, OPG17, OPH05 and so on. In this study, the collection of 331007 from Poonggi area was considered as the respective collection with above characters. Thus, the nine primers such as OPD05 and so on, will be used to select the Poonggi ginseng in the future studies.
