In this study, in order to evaluate the anti-obesity effect of sargassum horneri extract, the effects of the extract on lipase activity and preadipocyte differentiation in 3T3-L1 cells were investigated. S. horneri extract between 0.0 and 1.0 mg/mL showed no cytotoxicity and inhibited lipase activity by 68.1%. When S. horneri extract was utilized at levels of 0.25, 0.5, and 1.0 mg/mL in 3T3-L1 cells, preadipocytes differentiation decreased by 11.4, 19.7, and 25.6%, respectively, showing anti-obesity effects. In addition, after treatment with 1.0 mg/mL S. horneri extract, the mRNA expression levels of sterol regulatory element binding proteins-1c (SREBP-1c), peroxisome proliferator activated receptor-γ (PPAR-γ), CCAAT enhancer binding protein-α (CEBP-α), fatty acid synthase (FAS), and stearoyl-CoA desaturase1 (SCD1) in 3T3-L1 cells were significantly decreased (p < 0.05) by 65.2, 54.9, 50.0, 33.8, and 33.8% respectively. These results showed that S. horneri extract suppresses lipase activity and prophylactic preadipocyte differentiation in 3T3-L1, and thus can be used as an anti-obesity agent in functional foods and medicines.
Objectives The purpose of this study is to confirm the regulate effect of T helper (Th) 2 differentiation that Sabaek-san extract may produce to improves skin lipid barrier damage. Methods Four-weeks-old NC/Nga mice were divided into four groups: control group (Ctrl, n=10), lipid barrier eliminated group (LBE, n=10), Dexamethasone treatment after lipid barrier elimination group (DXT, n=10), and Sabaek-san extract treatment group after lipid barrier elimination (SBT, n=10). Sabaek-san extract was administered for 3 d after removal of the skin fat barrier in SBT group. Then, we identified changes in external symptoms of the skin, factors affecting skin barrier such as potential of hydrogen (pH), filaggrin, transepidermal water loss (TEWL) and Th2 differentiation factors like Interleukin (IL)-4, Kallikrein Related Peptidase 7 (KLK7), and protease activated receptor 2 (PAR-2) through our immunohistochemistry. Results After lipid barrier elimination, the reduction of morphological skin inflammations was less in SBT group than in LBE and DXT group. Also, pH and TEWL were significantly decreased with SBT group. However, filaggrin was significantly increased in SBT group compared to LBE, DXT, and Ctrl group. All kinds of Th2 differentiation factors (IL-4, KLK7 and PAR-2) were also decreased in SBT compared to the LBE and DXT. Conclusions As a result of this study, SBT administration decreased pH, TEWL, and increased filaggrin, thus participating in recovering damaged skin barrier. Since Th2 differentiation factors were decreased as well, SBT's regulatory effect in sequential immune reactions may be a possible explanation of how it enhances recovery of the damaged lipid barrier.
Objectives : Decrease in muscle mass and loss of muscle function due to aging are associated with various diseases. As interest in healthy aging increases, efforts to prevent and treat muscle hypoxia as an illness are increasing. Considering the physical limitations, a pharmacologic approach to the treatment of myopenia is needed. Methods : Terminalia chebula Rets has a wide range of pharmacological effects and is used as a medicinal product in traditional medicine. However, the drug effect on the treatment of muscle disorders has not been revealed. The purpose of this study was to evaluate the value of water extract of Terminalia chebula (WETC) as a therapeutic agent to relieve symptoms of muscle hypoxia. Results : WETC showed strong radical scavenging ability. In addition, WETC increased cell activity of myoblast, and we observed that WETC induces myoblast differentiation by immunoblot analysis using differentiation protein markers as well as cell morphology of myoblast. Based on these results, we examined the effect of chebulic acid, chebulagic acid, gallic acid, geraniin, and punicalagin on cell activity and differentiation of myoblasts. Gallic acid significantly increased cell activity of myoblast, and it was found to be an effective substance which not only induces myoblast differentiation but also promotes proliferation. Conclusions : We suggest that the WETC with antioxidant effect and its indicator gallic acid on cell activity, proliferation and differentiation of myoblast can be studied and developed as a food and medicine for prevention and treatment of various muscle diseases.
Objectives : Sarcopenia is a disease that leads to a decrease in skeletal muscle, and the importance of prevention and treatment thereof is increasing in an aging society. However, there is a definite limitation of exercise therapy for sarcopenia, and thus, there is an urgent need for a pharmacologic research to the treatment of sarcopenia. Methods : 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) assay was performed to confirm the antioxidant efficacy of water extract of Lacca Sinica Exsiccata (WELSE). To determine the effect of WELSE on myoblast activity, 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay was performed. To confirm the effect of WELSE on the differentiation of myoblast into myotubes, protein expression levels of myosin heavy chain 3 (Myh3) and paired box 3/7 (pax3/7) were confirmed by immunoblot analysis. In addition, immunofluorescence microscopy was performed to confirm the effect on myotube formation of WELSE. Results : It was confirmed that WELSE had high antioxidant activity and showed no cytotoxicity to myoblast up to 200 ㎍/㎖ concentration. Myoblast was treated with WELSE at a concentration of 100 ㎍/㎖ and differentiated for 5 days. The expression of Myh3, which forms myotubes, was promoted and the morphology of myotubes was changed and Increasing the thickness. Conclusions : In this paper, we confirmed the excellent antioxidant efficacy of WELSE and positive effects on muscle differentiation and myotube formation. These results suggest valuable as a material for pharmaceutical research on the prevention and treatment of sarcopenia.
Objectives : At present, aging-related degenerative muscle diseases are considered a serious problem. However, the effects on muscles regarding the efficacy of blueberry have not been studied. In this study, we tried to find out the correlation between blueberry and muscle. Methods : 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) assay was performed to confirm the antioxidant efficacy of blueberry hydrothermal extract. To determine the effect of blueberry hydrothermal extracts (BHE) on myoblast activity, 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay was performed. Results : Antioxidative efficacy and low toxicity were confirmed through ABTS assay and MTS assay of blueberry extract for myoblasts. As a result of immunoblot analysis and immunofluorescence analysis, the decrease in myogenic marker Pax3/7 was not confirmed, but myotubes The specific expression inhibitory activity of the forming protein Myh3 was confirmed. Through this, it was confirmed that the blueberry extract has a negative activity against myoblast differentiation. Conclusion : This experiment confirmed that blueberry hydrothermal extract has excellent antioxidant efficacy and negative results in inhibiting the differentiation and proliferation of myoblast. This requires deep study of certain ingredients and requires reassessment of the dietary intake of blueberries.
Obesity is a disease in which an abnormally large amount of fat accumulates in the body. Various diseases such as type 2 diabetes, dyslipidemia, high blood pressure, fatty liver, gallbladder disease, and coronary artery disease are induced. In this study, we investigated the effect of betaone, a type of barley, on obesity suppression. After the betaone extract was treated with 3T3 L1 adipocytes, the effect on adipocyte formation was investigated through Oil Red O staining. It was observed that differentiation was inhibited without affecting the viability of 3T3 L1 adipocytes. The effect of betaone extract on obesity inhibition in a mouse model was investigated. As a result of administering betaone extract after a high-fat diet, it was confirmed that the level of blood sugar and body weight was decreased, and glucose uptake ability was improved in a glucose tolerance test. The formation of mouse adipose tissue was suppressed, and the expression of genes involved in the formation and degradation of obesity in liver tissue was improved. These results suggest that betaone extract is a useful substance for improving obesity and is an excellent material for health functional food.
Objectives : This study aimed to investigate the synergistic effect of combining Cynanchi Wilfordii Radix extract with Humuli Lupuli Flos extract on estrogenic and anti-osteoclastogenic activity. Methods : Estrogenic effect of a mixture of Cynanchi Wilfordii Radix extract and Humuli Lupuli Flos extract (CWHL), Cynanchi Wilfordii Radix extract, Humuli Lupuli Flos extract, caudatin (an active ingredient of Cynanchi wilfordii Radix extract) and 8-prenylnaringenin (an active ingredient of Humuli Lupuli Flos extract) were examined by proliferation E-screen assay and expression of estrogen inducible gene, pS2 via Real Time-PCR (RT-PCR) in MCF-7 estrogen responsive cells. And their estrogenic activities were investigated how to modulate Estrogen receptor 𝛽 by binding affinity assay. Inhibitory effect of CWHL, Cynanchi Wilfordii Radix extract, Humuli Lupuli Flos extract, caudatin and 8-prenylnaringenin on RANKL-induced osteoclast differentiation were tested by TRAP (Tartrate-resistant acid phosphatase) staining in osteoclastogenic RAW 264.7 cells. Results : CWHL, Humuli Lupuli Flos extract and 8-prenylnaringenin accelerated the proliferation of MCF-7 and the expression of pS2 in MCF-7. CWHL, Cynanchi Wilfordii Radix extract, Humuli Lupuli Flos extract, caudatin and 8-prenylnaringenin bind to estrogen receptor 𝛽. CWHL, Cynanchi Wilfordii Radix extract, Humuli Lupuli Flos extract, caudatin and 8-prenylnaringenin inhibited RANKL-induced osteoclastogenesis in osteoclastogenic RAW 264.7. CWHL is more effective for all markers than Cynanchi Wilfordii Radix extract or Humuli Lupuli Flos extract alone. Conclusions : CWHL may a potential therapeutic agent for menopause and osteoporosis as a natural food resource. CWHL as a natural food source has therapeutic potential in cases of menopause and osteoporosis.
Purpose: We evaluated whether Spatholobus suberectus extract (SSE) can be used as a means of preventing and treating osteoporosis by measuring its effect on osteoclast differentiation, gene expression, and bone resorption. Methods: SSE was used to examine the effect on RAW 264.7 cells stimulated with RANKL to induce bone resorption. The inhibitory effect of TRAP formation and the expression of the bone resorption factors TRAP, cathepsin K, and MMP-9 during differentiation were measured. The effects on the differentiation-related factors NFATc and TRAIL and on the expression of OC-STAMP, DC-STAMP, ATP6v0d2, MITF, c-Fos, and inflammation-related factors were also evaluated. The effect on bone resorption was evaluated by culturing RANKL-treated osteoclasts on artificial bone fragments and observing the resulting resorption traces. The effect on bone damage in experimental animals was also measured. Results: SSE inhibited the differentiation of RANKL-stimulated osteoclasts into osteoclasts and suppressed the expression of cathepsin K, TRAP, MMP-9, NFATc1, TRAIL, MITF, OC-STAMP, DC-STAMP, ATP6v0d2, and c-Fos genes. Bone pore formation due to osteoclast action was also inhibited, and LPS-induced bone loss was suppressed in animal experiments. Conclusions: SSE could be useful for the prevention or treatment of osteoporosis by inhibiting osteoclast differentiation and bone resorption and suppressing bone loss induced in experimental animals. However, studies of larger populations are required.
In order to maintain bone homeostasis, it is necessary to balance bone resorption and remodeling through the differentiation of osteoclasts that absorb old bone and osteoblasts that form new bone. However, bone resorption due to excessive osteoclast differentiation is a major cause of osteoporosis and controlling excessive osteoclast differentiation has been known as a treatment strategy for osteoporosis. Therefore, in this study, the effect of an ethanol extract of Sphaerotylus antarcticus Kirkpatrick, 1907 (SAE), polar-derived sponge with unknown biological activity, on the osteoclast differentiation process of RANKL-induced RAW264.7 cells and the generated ROS was evaluated. In the study results, SAE down-regulated the formation and function of RANKL-induced osteoclasts and osteoclast differentiation specific proteins, genes in a concentration-dependent manner. In addition, it was possible to confirm the result of restoring the lost antioxidant enzyme along with down-regulation of ROS generated by RANKL. Therefore, in this study, we propose the possibility of SAE as a potential regulator of osteoporosis due to excessive osteoclast differentiation and report the biological value of the diversity of marine-derived natural products by identifying the first biological activity against SAE that is not yet known.
일본 우주항공연구개발기구(Japan Aerospace Exploration Agency, JAXA)는 2007년부터 2017년까지 달 탐사선 셀레네(Selenological and Engineering Explorer, SelEnE)가 관측한 데이터를 수집하고, 연구했다. CIPS는 실제 같은 영상을 고해상도로 합성한다. 합성할 데이터의 최적인 가중치 초기화 및 파라미터 갱신 방법, 활성 함수 조합은 실험을 통해 확인한다. 필요에 따라 앙상블 학습을 할 수도 있다. 성능평가는 FID(Frechet Inception Distance), 정밀도, 재현율을 사용한다. 제안한 방법은 진행 중인 연구의 시간과 비용을 절약하고, 인과관계를 더욱 명확히 밝히는 데 도움 될 수 있다고 사료된다.
