Metabolic profiling and method validation of marker compounds from Saposhnikoviae Radix and Peucedani Japonici Radix (방풍, 식방풍의 대사체 프로파일링을 통한 지표성분 선정 및 분석법검증)

  • Saposhnikoviae Radix (SR) and Peucedani Japonici Radix (PR) have been used as the main traditional herbal medicines in Korea, China and Japan. In this study, ultra-performance liquid chromatography coupled to quadrupole time of flight mass spectrometry (UPLC-QTOF/MS)-based metabolomics was applied to evaluate the quality of SR and PR using the marker compounds. In the S-plot of SR and PR, 5-O-methylvisammioside and peucedanol were selected as a marker compound for SR and PR, respectively. Also, an UPLC method was established and well validated for marker compounds of SR and PR. These results suggested that the established analysis method could be used one of the good methods for the classification and quality assessment of SR and PR.

Agronomic Characteristics of "Sikbangpoong 1" a New High Variety of Peucedanum japonicum Thunberg (방풍 신품종 "식방풍 1호" 의 생육특성 및 수량성)

  • A new Sikbangpoong(Peucedanum japonicum Thunberg) variety, 'Sikbangpoog 1', was developed through a pure line selection at the Kyougpuk Provincial RDA during the period of 1990 to 1995. The variety was characterized to have higher stem height, leaf number and seed production, and larger root diameter compared with the check variety of Bonghwa Sikbangpoong but plant height of 'Siknagpoong 1' was similar to that of the check variety and it was also more tolerant to water and heat stress. Root color of 'Sikbangpoong 1' was yellowish brown and root shape of the variety was straight and conical. The root yields of Sikbangpoong 1 in yield trial, regional adaptation trial and farmer's field trial were always 10-28% higher than those of the check variety.

Development and Application of PCR-based Markers for the Discrimination of Bang-Poong and Related Species (방풍류의 감별을 위한 분자마커의 탐색과 활용)

  • Bang-Poong and related species are an important herbal medicine. However, it is difficult to determine the commercial dry material through anatomical and chemotaxonomical characteristics. Here, we used a PCR-based technique for an accurate discrimination of Bang-Poong and related species. With the RAPD primers, 215 RAPDSs(random amplified polymorphic DNAs) were obtained, and 98% of them showed polymorphic patterns. RAPDs from the four primers were appropriate for the discrimination of S. divaricata $(T_{URCZ{\cdot}})\;S_{CHISKIN}$, those from the six primers for P. japonicum $T_{HUNBERG}$, those from the four primers for P. terebinthaceum $F_{ISHER}$, and those from the six primers for G. littoralis Fr. $S_{CHMIDT}$. The specific bands from the primer 425 were obtained and used to develop SCAR (sequence characterized amplified region) markers, based on the sequence information of the RAPD markers. The SCAR primers generated a 215 bp fragment specific to Peucedanum terebinthaceum $F_{ISHER}$, and a 177 bp and a 300 bp fragment specific to G. littoralis Fr. $S_{CHMIDT}$. As a result, the three SCAR markers were able to discriminate from two Bang-Poong related species.

Wood Properties of Japanese Cedar (Cryptomeria japonica) Planted as a Windbreak Forest of Mandarin Orange Field in Jeju Island

  • The anatomical characteristics, green moisture content, inorganic element concentration, and black heartwood percentage were compared between the Japanese cedar (Cryptomeria japonica) planted in a general mountain forest and in a windbreak forest for a mandarin orange field in Jeju island. The results obtained were as follows: 1. Black heartwood percentage was found to be much higher in the trees of windbreak forest than in those of general mountain forest. 2. Green moisture content of wood appeared to increase with the color change of heartwood from reddish brown to black. 3. Inorganic elements of $K_2O$ and CaO in the ray cells of black heartwood were thought to be one of the important factors for black heartwood formation. 4. Wood specific gravity and strength properties were comparatively lower in the trees of Jeju island than in those of southern part of the Korean peninsula. These properties, however, were found to be not significantly different between the trees of windbreak forest and general mountain forest in Jeju island.

The Effect of the Attached Glazing and Windbreak on the Thermal Performance and Air Tightness of Sliding window (덧유리 및 방풍재 적용을 통한 슬라이딩 창의 단열 및 기밀성능 개선효과 분석)

  • Purpose: Thermal performance and air tightness of window are improved for the building energy efficiency. As the deteriorated houses are increased, the improve measures with low cost and easy installation are developed in the energy performance of window. Attached glazing and windbreak can be easily applied to the window with low cost. In this paper, the effect of the attached glazing and windbreak on the thermal performance and air tightness of window is analyzed as the measure to improve performance of window. Method: Thermal transmittance of glazing is evaluated through WINDOW simulation according to thickness of attached glazing and air cavity. Based on the simulation results, thermal transmittance, air tightness and condensation resistance performance of four cases are tested according to Korea standards. One type of PVC sliding double window is chosen as the specimen. For the analysis on low performance of window, the outside of window is excluded in the PVC sliding double window. Result: This study shows that thermal performance of glazing can be increased by the application of attached glazing. Furthermore, lower thermal performance of glazing can obtain the higher effect of attached glazing. The application of attached glazing and windbreak can effect on increasing thermal performance and air tightness of window.

Vegetation Structure and Conservation of the Jeongdori Windbreak Forests on Wando Island in Dadohaehaesang National Park (다도해해상국립공원 완도 정도리 방풍림의 식생구조와 보전)

  • The Jeongdori windbreak forests of Wando island, located between the sea and the land, are very important as ecological succession areas that protect cultivated lands and fishing villages. Among the Jeongdori windbreak forests, the rocky seashore areas are occupied by Sageretia theezans mantle community, and their inner sections by sub-tree layers such as Ligustrum japonicum and Ligustrum obtusifolium. More specifically, the inner sections of the rocky seashore areas were found to be the habitat of Ligustrum japonicum-Quercus variabilis community, Ligustrum jarponicum-Viburnum erosum community, Ligustrum japonicum-Cinnamomum japonicum community, Cinnamomum japonicum-Carpinus tschonoskii community, and Pinus densiflora-Viburnum erosum community. The average height of tree layers of the Jeongdori windbreak forests is 10.3m, and the average diameter at breath height are 30.3cm. Though the windbreak forests was artificially made, it is now virtually natural forest and deserves protection for ecological reasons. Designated as a scenic spot 3, it is always in the danger of being damaged by frequent tourist visits. So the Jeongdori windbreak forest needs to be continuous maintenance and management by the National Park Service. Considering of the precious cultural treasures of the famous pebble stones covering Gugyedeung and the naturality of the Jeongdori windbreak forests, the Jeongdori windbreak forests as a scenic spot have to designated as the national monument in future and will preserve by the Cultural Properties Administration.

Discrimination of Geographical Origin for Herbal Medicine by Mineral Content Analysis with Energy Dispersive X-Ray Fluorescence Spectrometer (에너지분산형 X-선 형광분석기를 이용한 한약재의 무기질 분석 및 이에 의한 원산지 판별)

  • In this study, the macromineral content ratios of four herbal medicine samples(Saposhnikoviae Radix, Bupleuri Radix, Cnidii Rhizoma, and Astragali Radix) were analyzed to discriminate their geographical origins using an energydispersive x-ray fluorescence (EDXRF) technique. EDXRF is a rapid, non-destructive, and multi-elemental analysis technique. Initially, samples of both domestic and imported herbal medicines were pulverized, and then their macromineral contents, including P, S, K, and Ca, were analyzed using EDXRF. For the discrimination of their geographical origins, canonical discriminant analysis was carried out based on the estimated macromineral relative content ratios of the samples. According to the results, the discrimination accuracies were as follows: 93.3% for Saposhnikoviae Radix, 95.7% for Bupleuri Radix, 98.8% for Cnidii Rhizoma, and 87.5% for Astragali Radix. Overall, the results imply that this technique could be used as a standard method, to discriminate their geographical origins between domestic and imported herbal medicines.

Comparative Effect of Plant Growth Regulators on Callus Induction and Plant Regeneration in Glehnia littoralis Schmidt et Miquiel (갯방풍(Glehnia littoralis Schmidt et Miquiel)에서 캘러스 유도 및 식물체 재분화에 미치는 생장조절제의 효과)

  • Glehnia littoralis is known as an edible and medicinal plant using green loaves and mature roots of plant. In the present paper, the influence of plant growth regulators on callus induction and plant regeneration was investigated. Callus induction and regeneration occurred from leaf and petiole explants in Glehnia littoralis. Optimal condition of plant growth regulators for callus induction from leaf and petiole explants was MS basal medium supplemented with 2mg/L 2,4-D and 2mg/L BA. The frequency of callus induction was higher in petiole explant than leaf. When the callus was cultured on MS basal medium supplemented with 0∼1 mg/L IAA, 0∼1mg/L NAA and 0∼2mg/L BA for about 65 days, the most effective plant growth regulators on plant regeneration from callus were 1mg/L NAA and 2mg/L BA. The plantlets acclimatized successfully and grown in vermiculite matrix.

Cultivation Limit of Vitex rotundifolia, Tetragonia tetragonoides and Glehnia littoralis at Coastal Area and Physiological Vitality of RAW 264.7 cell and HL-60 cell (해안지대 순비기나무, 번행초, 갯방풍의 재배한계 및 RAW 264.7 세포와 HL-60세포의 생리활성)

  • The cultivation boundary areas and biological activator efficacies of Vitex rotundifolia, Tetragonia tetragonoides and Glehnia littoralis were investigated. Investigation of coastal areas revealed the East and West cultivation boundary areas of Vitex rotundifolia and Gelhnia littoralis to be the Taean Peninsula, Taean-gun and Gangwon-do, Donghae-City. The East and West coastal cultivation boundary areas of Tetragonia tetrragonoides are Jeollanam-do and Kyeongsangbuk-do, Ulleung-gun. The natural habitat is less than 200 m from the coastal area. Most of the Vitex rotundifolia, Tetragonia tetragonoides and Glehnia littoralis grow naturally in a region of mixed sand, clay and loam, in direct sunlight. No dominant vegetation was located. To analyze anti-inflammatory efficacy, RAW 264.7 cells were used. Vitex rotundifolia, Tetragonia tetragonoides and Glehnia littoralis inhibited inflammation by 37%, 12% and 10% in $100{\mu}g/m{\ell}$, respectively. However, the anti-inflammatory efficacy of Glehnia littoralis was invalid. Analysis of anti-cancer activity was accomplished using HL-60 cells. Glehnia littoralis revealed 60% and 72% inhibition activity in $100{\mu}g/m{\ell},\;and\;200{\mu}g/m{\ell}$ respectively. No cytotoxicity inhibition activity was investigated in Vitex rotundifolia and Tetragonia tetragonoides.

Effect of Vinyl Mulching on Growth and Yield of Peucedanum japonicum $T_{HUNB}$ (비닐피복이 식방풍의 생육과 수량에 미치는 영향)

  • This study was carried out to know the effect of mulching on growth and yield of Peucedanum japonicum. Time from sowing to germination was 8day at raising seedling(RS) and 40day at direct sowing(DS). The order of goodness in top part growth as follows : RS+white vinyl mulching (WVM), RS+black vinyl mulching (BVM), RS+no mulching(NM) and DS+NM. Tap root length at RS + NM was significantly shorter than those at the others. Tap root at the others was about same length. Tap root diameter was significantly larger at vinyl mulching than at no mulching and the thinnest at DS+NM. The number of supporting root at RS was more than DS as much as $4.7{\sim}7.1$. Rate of dried root weight to fresh weight was highest(30.5%) at DS+ NM and low at vinyl mulching. Dried root yield per 10a was 542kg at RS+WVM, 506kg at RS+BVM, 419kg at RS+NM and 357kg at DS. These results suggested that raising seedling and white vinyl mulching was good for culture of Peucedanum japonicum.
