Optimization of Synthesis Condition and Determination of Residue for Polyamine Type Flocculant (폴리아민계 고분자 응집제의 합성조건 최적화 및 잔류물분석)

  • Aluminium based inorganic flocculants are extensively used in this country in the removal of fine particles present in the raw water for the production of drinking water. These inorganic flocculants, however, have potential hazard of high residual aluminium ions in the treated waters, which is known to be one of the reasons of alzheimer's disease. Inorganic flocculants alone are sometimes incapable of treating water when there are excessive turbidity in the raw water sources due to flooding. A polyamine type polymeric flocculant has long been used to treat raw water in the drinking water production in the European countries and United State of America. The optimum reaction conditions such as mole ratio of epichlorohydrin(EPI) to dimethylamine(DMA), reaction temperature and time for each stage for the pilot scale preparation of polyamine from EPI-DMA was studied in this work. The variation of intrinsic viscosity and flocculating efficiency in the water treatment of the synthesized polyamines were evaluated. The residual materials after polymerization reaction were analyzed by gas chromatography to study the effect of variation of reaction conditions.

Isolation and Optimization of Cultivating Conditions of Alkalophilic Strains for Biodegradation of Azo Dye (Azo 염료의 분해를 위한 호알카리성 균주의 분리 및 배양조건의 최적화)

  • In order to treat of alkaline dye-processing wastewater, alkalophilic strains biodegrading azo dye, Acid red 1, is isolated from natural system, and optimal culture conditions are examined using response surface analysis, statistical analysis system program. 15 different species which grow in alkaline culture media are isolated from the effluent and river soil discharged from wastewater treatment plant in dye industrial complex. One strain which has the best decolorization efficiency is chosen, and named as AR-1. The result of the examination of carbon, nitrogen and phosphorus sources which have influence on growth and decolorization reveals that optimum carbon, nitrogen and phosphorus sources are 1.0% fructose, 1.0% polypeptone, 1.0% yeast extract and 0.5% $K_2HPO_4$, respectively. In order to optimize of biodegradation conditions of dye by response surface analysis, the characteristics of decolorization and cell growth according to culture temperature and time are monitered. The result shows that the one is optimum 34.77$^{\circ}C$ for 12.97 hours; the other at 34.73$^{\circ}C$ for 12.96 hours. While, optimal conditions of culture that satisfy both cell growth and decolorization are the temperatures from 32.86$^{\circ}C$ to 36.36$^{\circ}C$ and the period of 10.96 to 15.75 hours, respectively.

Optimization of Extraction Conditions for Cabbage (홍월적 양배추의 추출조건 최적화)

  • This study was conducted to monitor the extraction yields and functional properties from cabbage by a response surface methodology. The extract yield was maximized as 44.47$\%$ under the temperature of 79.86$^{\circ}C$, ethanol concentration of 56.84$\%$ and solvent to sample ratio 25.58 mL/g . The maximum value of electron donating ability was 85.46$\%$ at 46.38$^{\circ}C$,57.06$\%$ of ethanol concentration and 27.71 mL/g of solvent to sample ratio. The maximum value of tyrosinase inhibitory effect was 69.37$\%$ at 37.5$^{\circ}C$,47.71$\%$ of ethanol concentration and 16.03 mL/g of solvent to sample ratio. The maximum value of SOD-like activity was 48.36$\%$ in 66.12$^{\circ}C$, 70.35$\%$ of ethanol concentration and 29.13 mL/g of solvent to sample ratio. Estimated conditions for the maximized extraction including yield, electron donating ability and SOD-like activity were 20 $\∼$ 30 mL/g in ratio of solvent to sample, 25$\∼$85$\%$ in ethanol concentration, and 40$\∼$90$^{\circ}C$ in extraction temperature.

Optimization of the re-extraction process of ethanol from catechin components from Korean green tea extract (한국산 녹차추출물 catechin 성분의 ethanol 재추출 공정 최적화)

  • The purpose of this study was to determine the optimum re-extraction conditions of ethanol from catechin in Korean green tea (Camellia sinensis L.) using response surface methodology (RSM). The experiments were carried out according to a five level and two variable central composite design (CCD). The two independent variables were solvent ratio to sample content (1, 4, 7, 10, 13 mL/g) and extraction temperature (-20, -10, 0, 10, $20^{\circ}C$) on the dependent variables including yield, epigallocatechin (EGC), epicathchin (EC), epigallocatechingallate (EGCG), epicatechingallate (ECG), total catechin and caffeine. ANOVA results showed that Coefficients of determination ($R^2$) of estimated models for dependent variables were ranged from 0.9054~0.9778, while $R^2$ of caffeine were estimated 0.8770. The optimum ranges for the maximized extraction including yield, EGC, EC, EGCG, ECG, caffeine and total catechin were 4.5~7.5 mL/g in ratio of ethanol to sample and $-8{\sim}8^{\circ}C$ in extraction temperature. The actual values of yield, EGC, EC, EGCG, ECG, caffeine and total catechin, respectively, at the optimized conditions were 35.02%, 13.31%, 3.978%, 19.11%, 4.29%, 5.30% and 40.68%.

Optimization of Ethanol Extraction Conditions for Antioxidants from Zizyphus jujuba Mill. Leaves Using Response Surface Methodology (반응표면분석법을 이용한 대추잎 항산화물질의 에탄올추출조건 최적화)

  • The leaves of Zizyphus jujuba have been used for various purposes including medicine and nutrition. In this study, the conditions for the ethanol extraction of antioxidant from Zizyphus jujuba were optimized using response surface methodology (RSM). A Box-Behnken design containing 15 experimental runs with three replicates was employed to study the effects of solvent extraction conditions such as extraction temperature ($^{\circ}C$, $X_1$), extraction time (min, $X_2$), and ethanol concentration (%, $X_3$) on the extraction yield of antioxidants from Zizyphus jujuba. The yields of total polyphenols and total flavonoid, and electron donating activity (EDA) were considered as response variables. The second-order polynomial model gave a satisfactory description of the experimental results showing different patterns of extraction conditions with variation in the linear, quadratic, and interaction effects of the independent variables. Based on four-dimensional RSM, one of the optimized sets of conditions was 45% ethanol, $45^{\circ}C$, and an extraction time of 15 min. Under the optimal conditions, the predicted values were 177.64 mg/g dry basis, 35.99 mg/g dry basis, and 86.14% Vit.C equivalents for total polyphenols, total flavonoids, and EDA, respectively. The experimental values showed good agreements with the predicted values.

Preparation of Electrochemically Stable and SERS Active Silica@Gold Microshell (전기화학 반응용 표면증강라만산란 활성 실리카@금 마이크로쉘의 제작)

  • In order to monitor in situ electrochemical reaction we prepared the gold microshells on silica microspheres of $2{\mu}m$ in diameter which were able to not only work as electrodes but also surface enhanced Raman scattering (SERS) active substrates. Previously reported gold microshell using polystyrene as core material have a few serious problems, mostly coming from solubility in organic solvent, nonuniform distribution in size and toxicity of the polystyrene. Here we prepared silica core-gold microshell to obtain a strong SERS active platform benefitting from the physicochemical stability, uniformity and non-toxicity of silica. Varying the concentration of 3-aminopropyl triethoxysilane (APTES), the surfaces of silica beads were modified and the optimal condition was determined to be 1% APTES that made the SERS activity of gold microshell strongest. The gold microshells as made were characterized by homemade Micro-Raman system spectrometer, Field-Emission Scanning Electron Microscope.

Synthesis of a Nitrogen-rich Insensitive Energetic Material, DNAM(Dinitroammeline) (고질소 둔감에너지 물질인 DNAM(Dinitroammeline) 합성공정 개발)

  • We studied the Lab-scale process for the development of DNAM in aspect of safety and overall efficiency. Melamine is used for starting material of process, DNAM was synthesized by oxidation and nitration reaction. In order to optimize the process parameters with the product in higher yield and purity, a systematic study with variation of different parameters like molar ratio of nitrating mixture, conditioning time and order of reactant was carried out. The optimized reaction conditions for the synthesis of DNAM were : conditioning time of $3{\pm}3.5$ hour and nitrating mixture in 1:1.4 molar ratio. In order to proceed a stable reaction, melamine was added with enough time to relieve reaction heat. From these reaction condition, DNAM could be obtained in a yield of more than 60%.

Numerical Calculations for the Optimal Performance of Regenerative Catalytic Oxidation(RCO) (축열식 촉매 산화(RCO) 반응의 성능 최적화를 위한 전산 해석)

  • The computational fluid dynamics was analyzed for the pressure distribution, stream velocity distribution, stream line field, retention time and temperature distribution which are applied to the catalyst layer in the RCO reactor to derive the optimum operating condition of the heat condensing type catalytic oxidation (RCO) reactor. The results from the computational analysis revealed that the pressure loss due to the ceramic honeycomb in the catalytic bed of the reactor which is operating currently is not significant and the stream velocity (1.8~2.7 m/s) after the ceramic filter is working in stability without big channeling. To improve the stream velocity distribution of the air stream, it is necessary to extension of the connecting range between the plenum and catalytic bed inside the facility. However, the method of attaching the air stream guide vane or the perforated plate inside the reactor was not so effective.

Preparation of Branched-chain Amino Acid (BCAA)-enriched Hydrolysates from Corn Gluten (고 분지아미노산 함유한 옥수수 단백가수물의 제조조건 탐색)

  • The process of the preparation of branched-chain amino acid (BCAA)-enriched hydrolysates from corn gluten was optimized through the parameters of pre-treatment (heating and cellulosic hydrolysis), hydrolysis method (acid, protease, and microbe plus protease), concentration, and spray drying condition. The protein yield of corn gluten was increased by heating and cellulase treatments. Among three different hydrolysis methods, the combined use of microbes and protease was the most effective in terms of free amino acid (FAA) and BCAA content of the corn gluten hydrolysates. In addition, the FAA and BCAA content in the hydrolysates prepared by microbial and enzymatic combined treatment were improved by a concentration process. Spray drying conditions for the preparation of the powder from the hydrolyzed reactant were an inlet temperature of $185^{\circ}C$, outlet temperature of $80^{\circ}C$, and the use of maltodextrin as an anticaking agent. Thus, this study established an economical process for preparation of value-added hydrolysates of excellent productivity and quality, in terms of high BCAA content and product stability.

Optimization of Production Yield for Neohesperidin by Response Surface Methodology (반응표면 분석법을 이용한 neohesperidin 생산 수율의 최적화)

  • Neohesperidin is a natural new nutrition sweetener, widely existing in plants of dry citrus peel, which can be derived from extraction. Since the sweetness is 1,300-1,500 times greater than that of sugar, neohesperidin are widely used in fruit juices, wines, beverages, bakeries and pharmaceutical formulations, and are particularly suitable for consumption by diabetic patients. However, the yield of extraction from citrus peel waste is very low. In this study optimal yield conditions were determinedusing response surface methodology (RSM) in order to increase the neohesperidin extraction yield. The critical factors for maximum extraction yield were selected extraction pressure ($x_1$), extraction time ($x_2$), and concentration of ethanol ($x_3$). As a result, the extraction yield was improved when the extracting pressure increased. The extraction yield also increased in a time-dependent manner. When adding ethanol as an assistance solvent to the supercritical carbon dioxide, extraction yield was increased as more ethanol concentration was added. Finally, the extraction yield of neohesperidin was improved to about 162.22% compared to ethanol extraction as a conventional method.
