Effect of Toluene Application to Skin on the Enhancement of Liver Injury in $CCl_4$-Pretreated Rats

  • To evaluate an effect of toluene application to skin on the enhancement of liver damage in $CCl_4$-pretreated rats, toluene (35 mg/$cm^2$) was sequentially applied for 5 days to the skin of liver damaged rats with $CCl_4$ (6 times every other day: 0.1 ml/100 g body weight-50% $CCl_4$ in olive oil) On the basis of the functional and morphological findings in rat liver, appling toluene to the skin in liver damaged animals led to the more enhanced liver damage. In addition, by applying toluene to skin of liver damaged rats, the hepatic cytochrome P450 content was somewhat more increased, but the hepatic benzylalcohol dehydrogenase activity was significantly decreased (P<0.001), whereas benzaldehyde dehydrogenase activity was not statistically changed. In conclusion, the toluene application to skin in liver-damaged rat led to enhancement of liver injury that may be due to the accumulation of toluene metabolite in liver.

The Beneficial Effects of Vitamin E on the Irradiation-induced Testis Damage

  • Radiation exposure may cause tissue damage by ionized or excited atoms. The molecules are transformed from water or they directly interact with cells. Antioxidants such as vitamin E, vitamin C are used to reduce the interference of radiation. Especially, vitamin E inhibits the formation and accumulation of lipid and oxidation and the activity of drug-metabolizing enzymes. After irradiation of 3 Gy to the testis, the entire shape of testis was partly depressed and an irregular vacuole was observed to the edge. The 3 Gy group after treatment with vitamin E showed the low destruction of spermatogonia and partial tissue damage as compared to the 3 Gy group. Vitamin E inhibited the production of vacuoles from the cell death and generated the block to prevent the condensation of the seminiferous tubules.

Characteristics of Oncolytic Adenovirus Replication and Gene Expression in Hypoxic Condition

  • Adenovirus type 5 (Ad5) vectors have been used for gene transfer to a wide variety of cell types in vivo and in vitro. The advantages of adenovirus vectors include the high titer of virus readily obtained in large scale preparations, their ability to transduce dividing and non dividing cells, and the high level of transgene expression. Since adenovirus vectors do not integrate in host cell DNA, there is a lack of insertional mutagenesis. However, many human tumor cells lack expression of the adenovirus 5 receptors and contain areas of hypoxia. In order to identify the pattern of replication and gene expression of oncolytic adenovirus in hypoxic condition, multiple different fiber modified Ads (Ad5F/S11, Ad5F/S35, Ad5F/K7, Ad5F/K21, and Ad5F/RGD) was compared. The replication of all fiber modified adenovirus was inhibited in hypoxic condition in HEK 293 cells, but gene expression has variety on different tumor cell lines and the level of coxackievirus and adenovirus receptor (CAR) expression. These data suggest that CAR expression pattern and hypoxic condition of tumor are considered for optimal oncolytic adenovirus application.

Role of Cannabinoid on Neuronal Differentiation of P19 Cells

  • P19 cells are pluripotent embryonal carcinoma cells and can be differentiated into neuronal cell type by treatment with retinoic acid (RA) and aggregation culture. Cannabinoids are the active components of Cannabis sativa and they have diverse pharmacologic activities, such as pain control, anti-inflammatory effects, neuro-protection effects and tumor regression. Cannabinoids also involved in neuronal proliferation, migration, differentiation and survival in developing brain. Here, we studied the role of cannabinoids on neuronal differentiation of P19 cells. Treatment with cannabinoids increased the neuronal differentiation induced by RA and also promoted transcriptional activity of neurogenin 1, key transcription factor for neuronal differentiation of P19 cells. These results suggest that the cannabinoids can accelerate neuronal differentiation of P19 cells.

The Development and Characterization of a pH Dependent Matrix Tablet Containing Probiotics

  • The objective of this study was to develop a pH dependent oral matrix tablet containing probiotics. In this study, hydroxyl-propyl-methyl-cellulose (HPMC) and polyvinyl pyrrolidone K30 (PVP K-30) was utilized as a binder, sodium starch glycolate (SSG) was used as a disintegrant material for the tablet formulation. The disintegration test, hardness test, angle of response were performed to examine the characteristics of prepared tablet. Lactobacillus vitality test was applied to analyze the total Lactobacillus viable count. The results demonstrated that the pH dependent matrix tablet was prepared successfully and can thus be industrialized instead of the current methodologies used for preparation of conventional probiotics.

Association of Anti-apoptotic Mechanism Due to House Dust Mite in Neutrophils with Protein Synthesis and Bad

  • House dust mite is an essential allergen in the pathogenesis of allergic diseases. Abnormal regulation of neutrophil apoptosis is an important pathogenic process in allergic diseases. In the present study, we investigated the effects of house dust mites on spontaneous apoptosis of neutrophils and its associated mechanisms. Extract of Dermatophagoides pteronissinus (DP) inhibited neutrophil apoptosis in a time-dependent manner. Cycloheximide (CHX), an inhibitor of translation, increased apoptosis of DP-treated neutrophils as well as control cells. The pro-apoptotic effect of CHX was blocked by DP in neutrophils. In addition, DP increased the phosphorylation of Bad in a time-dependent manner, indicating that it exerted an inhibitory effect on the function of Bad. These results suggest that DP has anti-apoptotic effects of neutrophils and may regulate protein synthesis and activation of Bad. Moreover, these findings may shed light on elucidation of allergy pathogenesis due to house dust mites.

Beneficial Effects of Oat (Avena sativa L.) Bran Extract in Loperamide-induced Constipated SD Rats

  • The efficacy of extracts of oat (Avena sativa L.) bran in loperamide-induced constipation in SD rats was evaluated. The rats were divided into six groups of five rats each. The animals in Group 1 (control) and Group 2 (constipated control) were administered with distilled water orally. Groups 3, 4 and 5 comprised of constipated rats administered 100, 200 and 300 mg/kg body weight per day of extract of oat (Avena sativa L.) bran respectively while Group 6 were constipated rats administered bisacodyl (0.25 mg/kg body weight). Constipation was induced by oral administration of loperamide. The feeding characteristics, body weight, fecal properties were monitored. The results show that oats (Avena sativa L.) bran possesses laxative effects in loperamide-induced constipated rats.

Anti-obesity Effect of Black Vinegar Fermented with Herbal Extracts

  • Vinegar has been widely produced for a variety of industrial and domestic use as well as medicinal use. For sale of the commercial vinegar with herbal extracts, we produced an experimental black vinegar through sequential fermentation of alcohol, followed by acetic acid according to the manufacturer's procedure. To investigate the effect of anti-obesity of black vinegar on biochemical values, we evaluated enzyme activities via acetyl-CoA carboxylase (ACC), which plays a critical role in the lipid metabolism. We found that increased phosphorylated adenosine monophosphate (AMP) activated protein kinase (AMPK) and ACC in L6 mouse muscle cells treated with the manufactured vinegar. Based on the results, supplementation of experimental herbal black vinegar inactivates ACC, enhancing the phosphorylation of AMPK. Thus, the lipid oxidation and inhibitory effect of fatty acid synthesis by the black vinegar expects to facilitate the anti-obesity activity.

Cherry Tomato Supplementation Reduces Cardiovascular Risk

  • Tomatoes are rich in nutrients and have many beneficial advantages on human health. Four cherry tomato supplementation diets (CTSDs) were prepared from the juice and cake of fresh and processed (heat-treated) cherry tomatoes. Rats were fed CTSDs for 28 days and the changes in health indices in the serum were analyzed. CTSDs significantly decreased (P < 0.05) food efficiency ratio compared with the control. CTSD feeding significantly increased (P < 0.05) the high-density lipoprotein cholesterol level compared with the control, which resulted in a significant (P < 0.05) decrease in coronary artery risk index and atherogenic index. Furthermore, CTSD feeding increased serum serotonin level. These results indicate that CTSD shows antihyperlipidemic effect.

The Ameliorative Effect of β-sitosterol on DNCB-induced Atopic Dermatitis in Mice

  • ${\beta}$-sitosterol, one of phytosterols, exhibited numerous pharmacological effect including anti-inflammatory, anti-cancer and immune-modulating properties. This study attempted to determine the pharmacological effects of ${\beta}$-sitosterol on atopic dermatitis (AD). We investigated to ascertain the pharmacological effects of ${\beta}$-sitosterol on 2, 4-dinitrochlrobenzene (DNCB)-induced AD symptom and histamine-induced scratching behaviors in mice. Additionally, we evaluated the effects of ${\beta}$-sitosterol on the interleukin (IL)-6 levels in HaCaT cells and skin tissue of AD. The findings of this study demonstrated that ${\beta}$-sitosterol reduced AD clinical symptoms such as eczematous, erythema and dryness and serum histamine and IgE levels in DNCB-induced AD model and histamine-induced scratching behaviors in mice. Additionally, ${\beta}$-sitosterol inhibited the IL-6 expression in AD-like skin lesion and HaCaT cells. Collectively, these findings provide that ${\beta}$-sitosterol could be a therapeutic agent for skin inflammation including AD.
