근육 및 지방세포를 분화 관련 유전자의 DNA Marker가 한우의 도체특성 및 육질에 미치는 영향

  • 본 연구는 근육 및 지방세포 분화에 관여하는 leptin, MYF5 및 H-FABP의 3개 기능성 후보 유전자가 한우의 도체특성 및 육질에 미치는 영향을 분석하기 위하여 이들 유전자의 PCR-RFLP marker와 도체형질과의 관련성을 분석하였다. Leptin, MYF5 및 H-FABP 유전자에서 AA, AB 및 BB 3종류의 RFLP 유전자형이 각각 검출되었고 A와 B 대립유전자 빈도는 각각 0.57과 0.43, 0.61과 0.39 그리고 0.90과 0.10으로 추정되었다. 육질 등급에 따라 고급육과 저급육으로 분리 선발한 두 그룹간의 대립유전자 출현빈도를 비교한 결과 leptin과 MYF5 유전자에서 각각 통계적 유의차(P< .05)가 인정되었다. 또한 각 후보유전자의 RFLP marker 유전자형이 도체형질에 미치는 효과를 분석한 결과 leptin 유전자는 등지방 두께 그리고 MYF5유전자는 배장근 단면적에 각각 유의적인 영향(P< .05)을 미치는 것으로 분석되었다. 그러나 H-FABP 유전자는 도체형질들과 유의성이 인정되지 않았다. 따라서, leptin과 MYF5 유전자는 한우의 도체특성 및 육질 개선을 위한 DNA marker로 이용 가능할 것으로 사료된다.

Thymic Stromal Lymphopoietin (TSLP) Gene Polymorphisms are not Associated with Rheumatoid Arthritis in a Korean Population (TSLP 유전자의 다형성은 한국인 류마티스관절염 발생에 영향을 미치치 않는다)

  • Thymic stromal lymphopoietin(TSLP) is a novel IL-7-like hematopoietic cytokine. Human TSLP is produced by epithelial cells, stromal cells, and mast cells. The TSLP gene is highly expressed in synovial fluid specimens derived from rheumatoid arthritis (RA) patients. We previously identified four single nucleotide polymorphisms (SNPs) and one variation site in human TSLP gene. In this study, we analyzed the genotypic and allelic frequencies of the TSLP SNPs between RA patients and healthy controls. We also investigated the relationships between SNP genotypes and the RF levels and anti-synthetic cyclic citrullinated peptide (CCP) levels in RA patients. We then calculated the haplotype frequencies defined by these SNPs for both groups. The genotype and allele frequencies of the TSLP SNPs did not differ significantly between the RA patients and the healthy controls. We also found that TSLP SNPs in the RA patients had no significant association with the levels of RF or anti-CCP. Our results suggest that TSLP SNPs are not associated with susceptibility to RA.

A Polymorphism Analysis and Visualization Tool for Specific Variation Pattern Identification in Groups of Nucleotide Sequences (특정변화패턴 식별을 위한 염기서열 집단간의 다형성 분석 및 시각화 도구)

  • A genome contains all genetic information of an organism. Within a specific species, unique traits appear for each individual, which can be identified by analyzing nucleotide sequences. Many Genome-Wide Associations Studies have been carried out to find genetic associations and cause of diseases from slightly different base among the individuals. It is important to identify occurrence of slight variations for polymorphisms of individuals. In this paper, we introduce an analysis and visualization tool for specific variation pattern identification of polymorphisms in nucleotide sequences and show the validity of the tool by applying it to analyzing nucleotide sequences of subcultured pOka strain of varicella-zoster virus. The tool is expected to help efficiently explore allele frequency variations and genetic factors within a species.

  • Collected 38 colored soybeans, which was cultivated under the conventional field conditions, were investigated about their organeloptic parameters and correlations among factors such as organeloptic score, texturometer parameters and chemical components of seeds. Most of the organeloptic parameters were affect much more by the characteristics of each cultivars than by grain weight, and then showed significant differences with the effect of interactinon between grain weight and cultivar, too. Also among contribution degrees of texturometer scores to overall acceptance, cohesiveness was higher in small, medium and super grains, however in large grains, hardness. And among sensory attributes, suitable toughness was contribute most positively to acceptance.

Association between Economic Traits and Candidate Gene Polymorphism in Korean Native Pig and Duroc (한국 재래 돼지와 듀록의 경제형질과 후보 유전자 다형성간의 연관성 분석)

  • MC4R, PRKAG3, FABP3, and ESR have reported as important candidate genes related to some economic traits in pigs. To investigate the association between these genes and economic traits, the analysis of restriction fragment length polymorphism (RFLP) was conducted on 147 individuals (96 Durocs and 86 Korea native pigs; KNP) using single nucleotide polymorphism (SNP). Different genotype frequencies of 4 candidate genes were observed in Duroc and KNP. There were significant associations between MC4R polymorphic site and average daily gain (ADG, p<0.05) and backfat thickness (BF, p<0.05) in the Duroc, ADG (p<0.05) and days to 70 kg (p<0.05) in KNP. PRXAG3 polymorphic site were significantly .elated to BF (p<0.05) in the Duroc, ADG (p<0.05) and days to 70 kg (p<0.05) in the KNP. In FABP3, association with BF (p<0.05) in the Duroc, ADG (p<0.05) and days to 70 kg (p<0.05) in the KNP were found. ESR polymorphic site was not significantly associated to any other traits.

Polygenic Association of ACE and ACTN3 Polymorphisms with Korean Power Performance (ACE와 ACTN3의 다중유전형질과 근력운동 경기력간의 관계)

  • This study aimed to examine whether the polygenic profile of ACE ID and ACTN3 R577X polymorphisms is associated with muscle power performance in Korean athletes. For this study, 106 top-class power athletes (top-class group), 158 elite power athletes (elite-class group), and 676 healthy adults (control) aged 18-39 yrs were recruited and their genotypes were analyzed. The top-class group showed higher frequencies of the II genotype and I allele in ACE, as well as higher frequencies of the RR genotype and R allele in ACTN3 (top-class vs. control: 41.4% vs. 32.1% for II genotype, 67.1% vs. 57.7% for I allele, p<0.05; 42.3% vs. 29.0% for RR genotype, 65.3% vs. 54.8% for I allele, p<0.05). In the polygenic profile, the top-class group had significantly higher frequencies of combined-II/ID+RR/RX genotype than the control group (top-class vs. control: 82.9% vs. 66.7% for II/ID+RR/RX, p<0.05), and there was even a sharp increase in total genotype score (TGS) in this group compared to the elite-class and control groups ($66{\pm}0.9$ vs. $58{\pm}1.9$ vs. $56{\pm}2.3$, p<0.05). The combined-II/ID+RR/RX genotype showed the possibility of succussion in the top-class muscle power performance with an odds ratio of 2.3 (CI:1.4-4.1, p<0.05). These results suggested that ACE and ACTN3 need to interact with each other to affect muscle-power performance in an additive form. Furthermore, the polygenic profile of ACE and ACTN3 can predict muscle performance with high success in a homogeneous dominant combined genotype (II/ID+RR/RX). A further study could identify and combine other genes into ACE and ACTN3 for muscle strength.

Effects of Variety and Acetosyringone Influencing Transformation of Rice Mediated by Agrobacterium tumefaciens (Agrobacterium tumefaciens에 의한 벼 형질전환에 미치는 품종과 Acetosyringone의 영향)

  • The cytosolic glutathione reductase (GR) gene of Brassica campestris L. was introduced into several Japonica cultivars of rice by Agrobacterium tumefaciens and a large number of transgenic plants were produced. Three-week old calli were co-cultivated with A. tumefaciens strain EHA101 carrying the plasmid pIGR1. The efficiency of transformation was differed from rice cultivars. A Japonica cultivar, 'Daeribbyeo' appeared the highest efficiency (42.5%) of transformation among the four cultivars tested. The addition of acetosyringone (50 $\mu$M) during co-cultivation was a key to successful transformation. Transgene fragments were identified by PCR amplification and further confirmed by Southern blot analysis. Mendelian inheritance of the transgenes was confirmed in T$_1$ progeny.

Analysis of Pyrethroid Resistance Allele in Malaria Vector Anopheles sinensis from Malaria High-risk Area (말라리아 위험지역에서 채집된 말라리아 매개모기 Anopheles sinensis의 피레스로이드계 저항성 대립형질 분석)

  • Malaria is mainly transmitted by Anopheles sinensis which is dominant species in malaria high-risk area, northern part of Gyeonggi province in Korea. Pyrethroid insecticide is used for malaria vector, An. sinensis in Korea and the previous investigation consistently reported insecticide resistance from the vector. This study investigated insecticide susceptible and resistant alleles from An. sinensis and the status of malaria vector control in malaria high-risk area. For the study, An. sinensis collected from Paju, Gimpo and Ganghwa were sequenced for kdr detection. In Paju, there was no homozygous susceptibility and all of tested samples had homozygous or heterozygous resistance. There were 6.7% for susceptible homozygosity and 93.3% for resistant homozygosity or heterozygosity in Gimpo. Furthermore, the percentages of homozygous susceptibility and homozygous or heterozygous resistance in Ganghwa were 5.7% and 94.3% respectively. The results showed that the frequency of the insecticide resistance from An. sinensis in malaria high-risk area were increased much more than the previous investigation. Hence, this study suggests that malaria vector control programs should have to be prepared for the management of pyrethroid insecticide resistance.

Genetic and Morphological Relationship among Korean Hylotelephium Species(Crassulaceae) (한국산 꿩의비름속(돌나물과) 식물의 유전적, 형태적 유연관계)

  • Genetic and morphological studies using morphological and isozymic data from 20 populations of 6 Korea Hylotelephium were conducted to resolve the species relationships and the origin of polyploid species. The results of morphological studies suggest that triploid H. viviparum is closely related to H. verticillatum, and the high frequency alleles of two species were very similar each other in isozyme analyses. Unique fixation of $PGI-1^b$ in two species supports that H. verticillatum should be the diploid ancestor of H. viviparum. However, a unique $ALD-1^c$ allele is present in H. viviparum, and the lack of the common alleles of H. erythrostictum and H. spectabile in this species eliminated them from the candidates of another tetraploid ancestor of H. viviparum. Based on the high morphological and genetic similarities, our results supported that the Korea endemic H. viridescens might be originated from the H. erythrostictum or share a common ancestor with it. Besides, H. ussuriense is closely related with H. erythrostictum and H. viridescens.
