The Effect of Plant Hormones and Light Quality on the Invertase Activity in Maize (Zea mays L.) and Mung Bean (Phaseolus radiatus L.) (옥수수와 녹두의 Invertase Isozymes 활성에 미치는 식물호르몬 및 광선의 효과)

  • The effects of plant hormones (NAA, $GA_3$ and BA) and light qualities (white, red, green and blue light) on the changes of reducing sugar contents and invertase isozyme activities in leaves of maize (Zea mars L.) and mung bean (Phseolw radiatus L.) seedlings were investigated. NAA accelerated the increase of reducing sugar contents and invertase isozyme activities, on the contrary, $GA_3$ had little effect in the accumulation of reducing sugar and in the increase of enzyme activities from the leaves of maize and mung bean seedlings. On the other hand, BA accelerated an increase in the activities of the invertase isozyme from the leaves of mung bean seedlings whereas it had little effect in the increase of the enzyme activities from those of maize seedlings. The accumulation of reducing sugar in leaves of both seedlings was promoted by red light irradiation compared to white light irradiation, while the activities of the enzyme were little affected by various light Qualities. In the simultaneous applications of plant hormone and light quality, NAA with white light was very effective in the increase of reducing sugar contents and the enzyme activities from the leaves of mung bean seedlings, whereas NAA application with blue light showed a prominent enhancement in the reducing sugar contents and the enzyme activities from those of maize seedlings. These results suggest that plant hormone, particularly NAA, may be a more important factor than various light Qualities in the stimulation of invertase activity.

Impact of School Violence on Psychological Well-being: Korean Students' Happiness and Suicidal Impulse (한국 청소년의 학교폭력 경험과 심리적 안녕 -주관적 행복감과 자살충동을 중심으로-)

  • This study examines the impact of school violence on South Korean students' psychological well-being. The research is designed to analyze how different types of experiences of school violence-victim, perpetrator, victim-perpetrator, and non-involvement-influence students' suicidal impulse and sense of happiness. Empirical data was gained from a national-scale survey conducted by Social Development Research Institute at Yonsei University in 2013, and was analyzed using binary logistic and multiple regression methods. Findings suggest that any types of experiences of school violence had significant effect on suicidal impulse. In particular, the likelihood of victim-perpetrator to experience suicidal impulse was 7.1 times stronger than non-involvement. However, the effect of experiences of school violence on students' sense of happiness was insignificant. While the impact of school violence on sense of happiness among victims and victim-perpetrators was significant, its impact among perpetrators proved insignificant. Rather, stress factor-in particular, stress from peers-had more significant impact on the level of students' happiness. This study contributes to understanding the multiple effect of school violence by examining both negative and positive dimensions of psychological well-being.

Development of Herbicide Resistant Plant Through Plant Tissue Culture (제초제(除草劑) Butachlor 및 Simetryne에 저항성(抵抗性)인 식물체(植物體) 선발육성(選拔育成))

  • This study was conducted to select and develop herbicide resistant plant through tissue culture. Growth response of seedlings and callis of various rice varieties with Echinochloa species was assessed under the treatment of various rates of butachlor [N-(butoxy methyl)-2-chloro-2', 6'-diethyl acetamide] and simetryne [2,4-bis(ethyl amino)-6-methyl thio-1,3,5-triazine]. Further, succinate dehydrogenase activity was determined in herbicide treated callus to characterize different response of plants to herbicide. Rice variety like Sangpung showed relative resistance in both callus and seedling states against butachlor, indicating maintenance of resistance. However, in the simetryne treatment, the similar response was not observed in callus and seedling state, although there was a great different response among plant materials against simetryne. Rice variety which exhibited resistance in callus and seedling states showed low succinate dehydrogenase inhibition index. Succinate dehydrogenase inhibition index can be used as an important marker characters to differentiate varietal response of plant to herbicide. Rice plant was differentiated from butachlor and simetryne tolerant callus treated at $2.5{\times}10^{-5}$ M is growing under the growth chamber and can be used for resistant source.

Relationship between stress and subjective oral dryness in the elderly in a rural region: a pilot study (농촌 노인의 스트레스와 주관적 구강건조감의 관련성: 예비조사)

  • Objectives: Stress is a psychological reaction to stimuli such as anxiety or threat felt by a person either physically or mentally when placed in a difficult situation. Although a relationship between stress and dry mouth has been reported, it remains understudied. The purpose of this study was to investigate the association between stress and subjective dry mouth among the elderly living in a rural region. Methods: A total of 214 people aged ${\geq}60years$ living in a rural area were recruited with an informed consent for a cross-sectional analysis. Information about stress and subjective dry mouth was obtained by an interview survey with a constructed questionnaire. Data on other potential confounding factors (including oral factors) such as socio-demographic data, health-related behaviors, the number of remaining teeth, and subjective chewing ability were also gathered at the same time. Logistic regression models were used to analyze the relationship of stress and dry mouth using SPSS. Results: The Crude odds ratio (OR) of stress awareness affecting subjective dry mouth was 2.59 (95% confidence interval: 1.43-4.68). After adjusting for sex, education, income, smoking, and alcohol intake, the adjusted OR was 2.52 (95% confidence interval: 1.30-4.87) which was statistically significant. Conclusions: Elderly people who were stressed had an approximately 2-fold increase in experiencing subjective dry mouth when compared to their stress-free counterparts.

The Influence of Pretreatment Period, 2-Hydroxynicotinic Acid and Anther Co-pretreatment on Embryo Induction in Isolated Microspore Culture of Capsicum annuum L. (고추의 나출 소포자 배양시 전처리 기간, 2-Hydroxynicotinic Acid 및 약-공동전처리가 소포자배 발생에 미치는 영향)

  • Microspores were isolated from pepper (Capsicum annuum L.) anthers by using a micro-blender and cultured in modified NLN medium at $25^{\circ}C$. The influence of pretreatment period at $32^{\circ}C$, adding the 2-hydroxynicotinic acid to a pretreatment medium, and co-pretreatment anthers with microscopes on the induction of embryo were examined. Globular and torpedo embryos were observed from 3 weeks after culture. Embryo development was not synchronized within culture. After 4 weeks in culture, in addition to globular and torpedo embryos, cotyledonary embryos were observed. Normal cotylodonary embryos developed into plantlets when transferred to a solid hormone free B5 medium containing $2\%$ sucrose. Embryo yields were significantly higher after 1- and 2-day pretreatment at $32^{\circ}C$. However the development of embryo ceased at the globular or heart stage. In contrast, embryo yields were lower after 3- to 6-day pretreatment at $32^{\circ}C$ and embryo developed at the cotyledonary stage. After adding the 2-hydroxynicotinic acid to anther pretreatment solution, embryo yields were slightly increased. However most embryos occurred were at the globular or heart stage. Co-pretreatment of microspores with anthers was deleterious for embryo induction and development. AS far as we know, this is the first report of success in obtaining high frequency of embryogenesis and plantlets formation from isolated microspores of pepper. Although the culture conditions have to be optimized further, this promising microspore culture system can be used for genetic transformation, selection for dominant and recessive traits as well as for the production of homozygous doubled haploid plants.

Landscape Configuration Reading of 'Jangseong Pilmaseowon' through the Recomposition of Landscape (경관적 재구성을 통한 '장성 필암서원' 경관짜임의 독해(讀解))

  • This study was conducted to identify landscaping elements such as location, situation and feng shui included in the spatiality of Jangseong Pilmaseowon and to interpret aesthetic features of visual-perceptual spatial composition according to its arrangement. As it is shown in 'Pilamseowon', 'Pilbongseowon', and 'Gimhaseoseowon' appearing in antique maps, the awareness considering 'Pilam' as 'Pilbong' and 'Gimhaseo' was revealed. Mountain Pilamsan[Mountain Munpilsan] which is the location of seowon and Pilam(Brush-shaped rock) is the core of establishment of location identity of Pilamseowon and the symbol of Haseo Kim In-hu, which shows that they are deeply related to Ingeoljiryeong(人傑地靈: 'a place derives reflected glory from an illustrious human') based on connection. Pilamseowon shows locational characteristics of living in stream(溪居) facing panoramic 'jeungsan field' without Ansan(案山). Based on the teachings of Neo-Confucianism, Village Maekdong which is the birth place of Haseo, Pilam, seowon geomancy considering the Danbonghamseo-type(丹鳳含書形) geographical shape, formative reflection, Pilmaseowon and structures revealed in building naming more clearly show symbolic landscaping features resulting from 'theory of 'Heaven-Man Unity'(天人合一)' representing the union of nature and haman, than other seowons. The maximization of centrality through connected yards constructed with the 'jeondang hujae(前堂後齋)' arrangement in the order of Whakyeon-lu, Chenogjeol-dang, Jindeak-jae or Sungui-jae, and Woodong-sa is a unique feature of spatial frame of Pilmaseowon. In addition, it reveals the centrality reinforced with 'the move of inner center through arrangement of Kyeongjang-kag and Kyesengbi inside 'YuSik(遊息)' space and religious space' and the landscaping arrangement of Pilmaseowon from installation and device for reinforcement of territoriality. Moreover, it was found that orders and aesthetic features based on Neo-Confucianism were logically realized in the formation of Pilmaseowon with visual and compositional landscaping arrangement such as 'reinforcement of view centrality through composition of windows and doors', 'securement of visual transparency through framing and duplication', and 'realization of hierarchy through height of jaesil toenmaru'. The meaning system and spatial or visual aesthetic features of Pilmaseowon newly arranged and interpreted through landscaping recomposition is not a coincidental but inevitable result. It is another resource basis and an element that can improve the internal exuberance of Pilamseowon. This landscaping reading study is expected to improve the understanding of landscapes of Pilmaseowon and elevate the sensibility of unrevealed cultural landscapes.

Effects of Regular Walking Exercise on Health-Related Parameters in Persons with Chronic Diseases (규칙적인 걷기운동이 질환별 노인의 건강변인에 미치는 효과)

  • The purpose of this study was to investigate the effects of regular walking exercise on health-related parameters in the elderly with chronic diseases - apoplexy, overweight, impaired fasting glucose, and overweight + impaired fasting glucose. A total of 85 subjects, 27, 17, 21 and 20 in apoplexy (A), overweight (O), impaired fasting glucose (IFG), overweight + impaired fasting glucose group (O_IFG), respectively, completed a 12-week walking exercise. The health-related parameters were measured before and at the completion of the exercise program including anthropometric measurements, functional physical fitness levels, blood pressure, fasting glucose, blood lipid profiles and chronic inflammatory markers (CRPs). Significant improvements in body weight, BMI, %body fat, blood pressure, all blood lipid measurements and all measured physical fitness items were shown in A; those in %body fat, TC, HDL-C and LDL-C in O; those in body weight, BMI, %body fat, fasting glucose, TC, TG and HDL-C in IFG; and those in body weight, HDL-C and LDL-C in O_IFG (p<0.05). The results of the present study demonstrated that a 12-week walking exercise brought positive effects on body weight, bloody lipid profiles, fasting glucose and functional physical fitness levels in the elderly with chronic diseases. In conclusion, this study suggested that walking regularly is very effective in lowering the risks of developing chronic diseases.

Studies on the Anther Culture of Prunus mume S. et Z. and the Other Three Species (Prunus mume S. et Z. 외(外) 삼종(三種)의 약배양(葯培養)에 관(關)한 연구(硏究))

  • When haploid plant would be appeared by the anther culture, the large quantity of young plant multiplied maternal inheritance and the same pure line rapidly in the short length of time, which will be effected to cut down much expences efforts and time for the production of seeds or seedlings. Therefore, the development of the technique for this would be much profited in the country industry. In the late of a few years studies were early attempted in this field, but up this time there were a few success of plants only and none of perennial plant. In this status of the country condition required earnestly for the development of the green industry, this researcher attempted to culture the anther of late uninucleate microspore or early binucleate microspore of the Prunus mume and other three psecies, economic trees estimated specially economic, on the place of Modified Murashige and Skoog's medium supliment with Kinetine, 2.4-D, and N.A.A for inducing haploid plants. The obtained results were as follows: 1. 2,000 anthers were cultured and there were shown that 2N callus in Prunus mume had 82 as 4.1%, 2N callus in Prunus tomentosa 15 as 0.8%, 2 N callus in Prunus salisina 75 as 4% 2. N callus had shown 40 as 2% from Prunus armeniaca var. ansu only, and the other trees showed all 2N callus. 3. Callus had appeared in every tree but 2N callus appeared was all filaments and there showed from only connective tissue N callus appeared was all from anther locule inside. 4. Then Prunus armeniaca var. ansu only was not callus of somatic anther tissue origin, but as there was callus origined from microspore which was changed in to swollen microspores or polynucleate microspores, it was certained to need haploid plant.

Identification of Novel Salt Stress-responsive Genes Using the Activation Tagging System in Arabidopsis (애기장대에서 activation tagging system을 이용한 새로운 고염 스트레스 반응 유전자의 동정)

  • Abiotic stresses limit the growth and productivity of plants. Cellular adaptation to abiotic stresses requires coordinated regulation in gene expression directed by complex mechanisms. This study used the activation tagging system to identify novel salt stress-responsive genes. The study selected 9 activation tagging lines that showed salt stress-tolerant phenotypes during their germination stages. Thermal asymmetric interlaced-PCR (TAIL-PCR) was used to identify the T-DNA tagging sites on the Arabidopsis genome in selected activation tagging lines, including AT7508, AT7512, AT7527, AT7544, AT7548, and AT7556. RT-PCR analysis showed that ClpC2/HSP93-III (At3g48870), plant thionin family (At2g20605), anti-muellerian hormone type-2 receptor (At3g50685), vacuolar iron transporter family protein (At4g27870), and microtubule-associated protein (At5g16730) were activated in AT7508, AT7512, AT7527, AT7544, and AT7556, respectively. Interestingly, in AT7548, both the genes adjacent to the T-DNA insertion site were activated: Arabinogalactan protein 13 (AGP13) (At4g26320) and F-box/RNI-like/FBD-like domains-containing protein (At4g26340). All of the seven genes were newly identified as salt stress-responsive genes from this study. Among them, the expression of ClpC2/HSP93-III, AGP13, F-box/RNI-like/FBD-like domains-containing protein gene, and microtubule-associated protein gene were increased under salt-stress condition. In addition, AT7508, AT7527, and AT7544 were more tolerant to salt stress than wild type at seedling development stage, functionally validating the screening results of the activation tagging lines. Taken together, our results demonstrate that the activation tagging system is useful for identifying novel stress-responsive genes.

Comparisons between a Forest Road with a Coniferous Plantation and Distributed Vegetation on the Edge of a Forest, and Reclaimed Soil Seed Bank (식재 침엽수 숲길과 숲 가장자리 분포 식생 및 매토종자 비교)

  • The purpose of this study is to compare the differences in aboveground flora and underground flora between a forest road and a forest edge and to clarify each characteristic through ecological approach to a forest road. The study site was the forest planted with Pinus koraiensis and Abies holophylla, and located at an altitude of 45m($36^{\circ}36'23''N127^{\circ}21'45''E$). The width of the forest road is 3.2m. This research set the forest edge within the areas 5m away from the forest road and also conducted a survey on vegetation 5 times from september 2006 to August 2007. In addition, it installed thirty six quadrats to make an analysis of reclaimed soil seed bank. Soil amounting to 600$cm^3$ was collected from each quadrat using soil samplers(100$cm^3$),which was preserved in low temperature refrigeration for a month. Soil was thinly strewed evenly on trays and watered every four or five days; then, this research did experiment for six months until no more germination took place. Through this process, this research identified species and counted the number of germinating individuals by using emerging seedlings. The research result showed that on the whole, the similarity index between aboveground flora and underground flora was low. The correlation coefficient between the aboveground flora vegetations both on the forest road and on its edge was found to be 0.36, showing a correlation with each other(p<0.05). On the other hand, the correlation coefficient between underground flora vegetations through the analysis of reclaimed soil seed bank was 0.20, showing no correlation with each other(p>0.05). As the survey result of naturalized plants, there existed 7 species of naturalized plants on the forest road in case woody plants were included, showing 11.11% naturalization rate and 2.61% urbanization index(UI). On the other hand in case woody plants were not included among the naturalized plants, the naturalization rate on the forest road was 12.50% while the naturalization rate on the edge of the forest was 9.09%.
