This study was undertaken in order to evaluate effects of methanol extracts of Stachys sieboldii MIQ and its related enzyme activities in brain tissues of rats. Sprague-Dawley(SD) male rats were fed within a control group, which is a basic diet group. The experimental diet group was given 100 and 200 mg/kg to supervise 100 and 200 mg/kg body weight per day for 20 days. Lipid peroxide levels and acetylcholine esterase activity in brain tissues were slightly decreased at a dose dependent manner, in vitro. Lipid peroxide levels were also decreased at a dose dependent manner; methanolic extracts of Stachys sieboldii MIQ demonstrated significant inhibitory effects, in vivo. Monoamine oxidase and xanthine oxidase activities were significantly inhibited in the brain tissues of experimental group compared to control group and the ratio of type conversion of xanthine oxidase were decreased.

In this study we wanted to investigate the effect of taurine supplement on the lipid peroxide formation and the activities of glutathione(GSH) dependent enzyme in diabetic model mice. We induce type I diabetes mellitus with alloxan injeciton in ICR mice and type II with high calorie diet in genetically hyperglycemic KK mice. Taurine was given in drinking water at the level of 5%(w/v) for seven days. In type I diabetic model, the malondialdehyde(MDA) of liver and islet significantly increased compared to control group and they significantly decreased by taurine supplement. In type II diabetic model, the concentration of MDA was not changed by taurine supplement. The activities of GSH-peroxidase(GPX) of liver and islet increased in type I diabetic group while decreased in type II. GPX activities were not changed by taurine supplement in the liver of both types but increased in the islet of type II. Taurine supplement has no effect on the activities of GSH S-transferase(GST) in both types. From these results, we suggest that taurine supplement protect against lipid peroxide formation in diabetic model of type I.
Glycoprotein (60 kDa) isolated from Ficus Carica Linnoeus (FCL glycoprotein) was examined by evaluating its hypolipidemic effects on plasma cholesterol levels and hepatic detoxicant enzyme activities in ICR mice. FCL glycoprotein $(100{\mu}g/mL)$ had strong scavenging activities (38%) against lipid peroxyl radicals. When mice were treated with Triton WR-1339 (400 mg/kg), levels of total cholesterol (TC) and low-density lipoprotein (LDL)-cholesterol in plasma significantly increased by 53.9 and 47.5 mg/dL, respectively, compared to the control, whereas, when pretreated with FCL glycoprotein $(100{\mu}g/mL)$, decreased remarkably by 55.4, and 47,0 mg/dL, compared to Triton WR-1339 treatment alone. Interestingly, high-density lipoprotein (HDL)-cholesterol level did not change. Body and liver weights did not change significantly after Triton WR-1339 treatment in presence of FCL glycoprotein. FCL glycoprotein $(100{\mu}g/mL)$ stimulated activities of antioxidative detoxicant enzymes such as superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx), whereas GPx activity significantly increased compared to the control. These results suggest FCL glycoprotein has abilities to scavenge lipid peroxyl radicals, lower plasma lipid levels, and stimulate detoxicant enzyme activity in mouse liver.

The purpose of this study was to identify the inhibitory effect of hepatic toxicity and liver lipid metabolism after administration of Artemisia capillaris and Paecilomyces japonica. Along with the control, SD rats were divided into ethanol treated group with subgroups of 6% Artemisia capillaries (6A), 4% Artemisia capillaris+2% Paecilomyces japonica (4A2P), 3% Artemisia capillaris+3% Paecilomyces japonica (3A3P), 2% Artemisia capillaris+4% Paecilomyces japonica (2A4P) and 6% Paecilomyces japonica (6P). In this study we also intended to verify the optimum ratio of Artemisia capillaris and Paecilomyces japonica which can reduce hepatotoxicity. Artemisia capillaris and Paecilomyces japonica reduced cholesterol and triglyceride levels which were increased by ethanol. Total-cholesterol level was decreased the most in the groups of 4A2P and 3A3P. On the other hand, activity of superoxide dismutase (SOD) was enhanced significantly (p<0.05). Malondialdehyde (MDA) activity was decreased significantly (p<0.05) in the subgroup of 6A and 4A2P. When the ratio of Artemisia capillaris and Paecilomyces japonica was 2:1, the improvement of the rat serum and liver lipid metabolism and the alleviation of hepatic damage by ethanol were the most effective in this study. Therefore, it can be considered that the symptoms of severe chemically induced hepatotoxicity could be reduced by Artemisia capillaris and Paecilomyces japonica administration.
The effect of antioxidants on the myocardial cellular damage which occurs during reoxygenation of hypoxic myocardium was examined in isolated rat hearts. The roles of oxygen free radical and lipid peroxidation in reoxygenation injury of myocardium were also investigated. In Langenorff preparation of isolated rat heart, which was made hypoxic by perfusion with the substrate free, hypoxic cardioplegic solution ($37^{\circ}C$, 90 min), the release of cytosolic enzymes (creatine phosphokinase, lactic dehydrogenase) and a lipid peroxidation product, malondialdehyde into the coronary effluent were abruptly increased by reoxygenation. The release of enzymes was closely parallel to that of MDA. These increases of enzymes and lipid peroxidation product were suppressed to various degrees in the presence of scavengers of superoxide anion (superoxide dismutase, 10,000 U), hydrogen peroxide (catalase, 25,000 U) and hydroxyl radical (dimethyl sulfoxide, 10%). A natural antioxidant, ${\alpha}-tocopherol$(4.5 uM) and a synthetic one, butylated hydroxytoluene (2 uM) suppressed the release of cytosolic enzymes with the concomittent reduction of lipid peroxidation as measured by malondialdehyde release into the coronary effluent. These effects of antioxidants were dose dependent, and were more pronounced when the antioxidants were administered throughout hypoxic and reoxygenation periods than given during reoxygenation period only. These results suggest that cytotoxic oxygen free radicals produced in the myocardium during reoxygenation may be responsible fur the myocardial cellular injury by enhancing the lipid peroxidation of cellular membranes. Furthermore, the antioxidants may exert protective effect against reoxygenation damage of hypoxic myocardium through the inhibition of lipid peroxidation reaction.
The purpose or this study was to investigate the effect or H $_2$O fraction or Dioscorea japonica Thunb(DJT) and selenium(Se) on the lipid peroxidation in streptozotocin (STZ)-induced diabetic rats. Male Sprague-Dawley rats weighing 180∼220g were divided into 5 groups: One normal rat group and 4 diabetic rat groups(the STZ-Control group, the DJT group, the DJT-Se group and the Se group). Diabetes was induced in the male rats by injection of STZ into tail vein at a dose of 45 mg/kg. The H$_2$O-fraction of DJT(500 mg/kg) was administered orally for 14 days. The supplementation was achieved with the AIN-76 recommendation diet by adding 2 mg/kg diet of selenium as Na$_2$SeO$_3$ which was prepared freshly everyday. The levels of glycogen in liver and muscle and protein in kidney, liver and muscle were measured. The liver concentrations of cholesterol and triglyceride were analyzed. Also, the malondialdehyde(MDA) levels in kidney, liver and lung were determined. The glycogen levels of liver and muscle in diabetic groups were not significantly different from the normal group. The protein concentrations of kidney, liver and muscle were not significantly different either, but the level of muscle protein was higher than STZ-Control group. The levels of liver cholesterol were significantly different between normal and STZ-Control groups and decreased in all diabetic experimental groups fed on H$_2$O-fraction of DJT and Se supplementation compared with the STZ-Control group. The levels of liver triglyceride were higher in the DJT-Se group than the STZ-Control group. The concentrations of MDA in lung decreased greatly by the administration of Se among all and the concentration of liver MDA was significantly reduced and that of DJT-Se group was the lowest. In conclusion, the results indicated that the administration of H$_2$O-fraction of DJT with selenium supplementation has a synergistic antioxidative effect by influencing on lipid metabolites and peroxidation especially in liver.
본 연구는 retinyl acetate와 $\beta$-carotene보충식이가 에탄올에 의한 산화적 손상에 미치는 영향을 조사하고자 흰쥐에게 비타민 A와 $\beta$-carotene을 보충한 식이를 5주간 급여한 후 에탄올을 급성으로 투여하여 혈청과 간 조직에서 생성되는 지질과 지질과산화물 함량 그리고 glucose-6-phosphatase 활성도를 측정하였다. 본 실험에서 얻은 결과를 요약하면 체중 증가량은 전 실험군에서 유의적인 차이없이 실험 기간 동안 증가되었으며 사료섭취량과 식이효율을 그리고 간 중량 역시 각 군 모두 유의적인 차이를 나타내지 않았다. 혈청 중성지질 함량은 대조군과 retinyl acetate 보충군에서 높은 함량을 보였으나 통계적인 유의성은 없었다. 총 콜레스테를 함량은 대조군과 $\beta$-carotene 보충군은 비슷한 수준을 나타낸 반면 retinyl acetate 보충군에서는 유의적으로 감소되었다. HDL-콜레스테롤 함량은 대조군에서 가장 높았으며 LDL-콜레스테롤 함량은 유의적이지는 않았으나 retinyl acetate 보충군에서 가장 낮은 함량을 보였다. 동맥경화지수는 전 실험군에서 유의적인 차이는 없었으나 retinyl acetate 보충군에서 가장 낮았다. 간 마이크로솜 내에서 지질과산화물 함량은 재도군에 비하여 $\beta$-carotene 보충군에서 유의적으로 감소되었다. Retinyl acetate와 $\beta$-carotene 보충군 사이에서는 유의적인 차이가 없었으나 급성적인 에탄올 투여시에 $\beta$-carotene의 보충 급여가 지질과산화반응에 방어효과가 있는 것으로 나타났다. 간 조직중의 glucose-6-phosphatase 활성도는 비타민을 정상적으로 공급시킨 대조군이 retinyl acetate나 B-carotene 보충군에 비하여 유의적으로 높았으며 retinyl acetat와 B-carotene 보충군 사이에는 유의적인 차이가 없었다.

The purpose of this study was to investigate the effects of vitamin C supplementation and yoga practice on total antioxidant status (TEAC), lipid peroxidation, and blood lipid profiles of middle-aged women. Thirty two women ($53.34{\pm}5.08$ years old) in B city volunteered and the signed consents were received before the study. Experimental groups were normal (n=8), vitamin C intake group (n=8), yoga group (n=8), and vitamin C intake with yoga group (n=8). Total experimental period was 12 weeks. Yoga group practiced yoga 3 times a week for 12 weeks with intensity of RPE $13{\sim}15$. Each practice was 60 minutes. Vt. C group was instructed to take 1 g of Vt. C every day after dinner meal. Changes in TEAC after 12 weeks were 28.23% for the normal group, 30% for Vt. C group, 26.58% for yoga group, and 43.66% for Vt. C with yoga group; the increases in TEAC among groups were not significantly different. Serum malondialehyde (MDA) concentration of the normal group increased by 10% during 12 weeks of experiment while that for the Vt. C, yoga, and Vt. C with yoga group were significantly decreased by 25%, 32.14%, and 33.87%, respectively (p<0.05) compared with the normal group. Neither yoga, Vt. C supplementation nor combined program for 12 weeks were effective enough to change the serum lipid profiles compared with the normal group. In conclusion, Vitamin C supplementation or regular yoga practice seems to have health promoting effects of retarding the oxidative stress by decreasing lipid peroxidation in middle-aged woman.
This study was performed to investigate the antioxidative effect of Ligusticum chuanxiong Hort extracts (LCE) against the hyperlipidemia of high-fat diet-fed obese rats. The rats were divided into the three groups (normal group, control group and sample group) to perform the experimental research. 1.5 ml/kg of LCE was intraperitoneally administered into the sample group for 21 days. The equal dose of 0.9% saline was intraperitoneally administered into the normal group and the control group. On day 22, they were anesthetized with ether and dissected. The levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT) were examined in serum of rats. Superoxide dismutase (SOD) was measured in mitochondrial fraction. Malondialdehyde (MDA), catalase (CAT), and glutamate peroxidase (GPx) were determined in liver homogenate. High-fat diet markedly increased the levels of AST, ALT and MDA, significantly decreasing those of SOD, CAT and GPx. But Ligusticum chuanxiong Hort-pretreatment decreased the levels of AST, ALT, and MDA. increasing those of SOD, CAT and GPx. These results demonstrated the antioxidative effects, suggesting that LCE could be the candidate for the functional material.

The effect of Thuja biotae water extract on lipid composition, blood glucose, cholesterol, triglyceride and lipid hydroperoxide was studied in rabbits. In the animalfed Thuja biotae water extract $1m{\ell}/kg\;or\;2m{\ell}/kg$, blood glucose, cholesterol, triglyceride and lipid hydroperoxide was lower values than the control group. The lipid composition of plasma was not great differencies, but palmitic acid $C_{16:0}$ was very elevated with higher than 50%, n-3/n-6 ratio was higher than 0.3 and p/s ratio was higher than 4.0. It was concluded that Thuja biotae water extract may be one of the blood glucose, cholesterol and triglyceride level lowering factor.
