Nerve-Agent Selective Chemiresistors Fabricated by Oxime Decorated Polypyrrole Layer on Cellulose Paper (셀룰로오스 종이 상에 Oxime 도입된 polypyrrole 층을 제조한 신경작용제 선택적 화학저항 센서)

  • In continuous research of detecting highly toxic chemical warfare agents to ensure preparedness for the future battlefield, flexible and wearable sensor platforms with high sensitivity are still demanding. Herein we demonstrate a facile fabrication of polypyrrole-based chemiresistors on cellulose paper for the detection of nerve gas simulants. In order to optimize electrical properties of sensor platform, conducting polymer made of polypyrrole were first synthesized on flexible cellulose paper and interdigitated electrodes were formed thereon. Following confirmation of polypyrrole and/or oxime moiety through FT-IR analyses, electrical characteristics were measured in the various ratio of monomers between simple pyrrole and oxime-modified one. Typically for the optimized chemiresistor(2:8 molar ratio of simple pyrrole and oxime-modified one), eleven species of chemical warfare agents were examined and enhanced conductivity(104~105 order) was observed for three simulants(diethyl cyanophosphonate, diisopropyl fluorophosphonate and diethyl chlorophosphonate), which was mainly attributed to intermolecular hydrogen bonding, while no significant responses was recorded against sixteen common volatile organic chemicals.

Agrobacterium-mediated Transformation of PAT into Platycodon grandiflorum A. De. candolle (Agrobacterium을 이용한 Phosphinothricin Acetyl Transferase의 도라지로의 형질전환)

  • This study was conducted to introduce phosphinothricin acetyl transferase (PAT) gene, resistant to basta which was non-selective herbicide, into balloon flower (Platycodon grandiflorum A. De. candolle). Seeds were germinated on MS medium, and 10-day-old immature cotyledon explants and 30-day-old leaf explants were cocultured with Agrobacterium tumefaciens strain MP 90 (pBinSyn) on 1/10 MS medium for 48 hours in the dark at $25^{\circ}C$. The cultures were transferred for selection of kanamycin-resistant shoots to the MS medium supplemented with 0.2 $mg/{\ell}$ NAA, 1.0 $mg/{\ell}$ BA, 3% sucrose, 100 $mg/{\ell}$ kanamycin, 500 $mg/{\ell}$ carbenicillin. Shoots were obtained from 10-day-old immature cotyledon explants after 4 weeks of culture. The shoots were subcultured twice every 4 weeks on the same medium for growth of transgenic shoots. Successful transformation was confirmed by histochemical GUS assay, PCR analysis, RT-PCR analysis, 10 $mg/{\ell}$ phosphinothricin treatment and 0.3% basta spray. The basta-resistant transgenic plants flowered normally.

Selection of Herbicide Resistant Potatoes Transformed with Phosphinothricin Acetyltransferase Gene (Phosphinothricin Acetyltransferase 유전자 도입에 의한 제초제 저항성 감자의 선발)

  • This experiment was conducted to introduce PAT (phosphinothricin acetyltransferase, non-selective herbicide bialaphos resistant gene) gene into potato (Solanum tuberosum. cv. Desiree). Optimal shoot regeneration from leaf discs and stem segments was obtained in MS medium supplemented with 0.1 mg/L IBA and 0.5 mg/L BA, and the frequency of shoot regeneration was 54% in left discs and 46% in stem segments. In this condition, leaf discs and stem segments of potato were co-cultivated with A. tumefaciens MP90 which contained binary vector with GUS: :NPTII gene and PAT gene. Transgenic shoots were regenerated from leaf and stem-derived calli on selection medium with 100mg/L kanamycin. The 100${\mu}M$ acetosyringone treatment during the co-cultivation highly enhanced(4 times than the control) the shoot regeneration on selection medium. When the putative transgenic plants were transferred to medium with 10mg/L basta, all of them were survived. After PCR. GUS test, and Southern blot analysis of the survived plant, we confirmed that the gene was stably integrated into the potato genome and expressed. After the transgenic plants were transplanted in soil, and the transgenic plants were sprayed with the herbicide basta (300ml/10a), the transgenic plants remained green but control plants were died.

  • 국내의 내진설계 기준은 1988년에 처음 도입되었으며, 최근 점차 강화되고 있는 실정이다. 공동주택에 주로 적용되는 전단벽식 구조시스템에서 증가된 지진력에 저항하기 위해서는 벽량과 철근이 증가하게 되어 공사비가 상승하게 된다. 이러한 단점을 보완하기 위한 제진설계의 필요성이 대두되고 있는 실정인데, 기존의 제진장치는 주로 가새형 또는 벽체형을 대부분이라 평면계획에 제약이 있다. 따라서 전단벽식 구조의 공동주택의 제진설계 시에는 우리나라와 같은 중 약진 지역에 적합하고 저렴한 비용으로 충분한 내진성능과 평면의 가변성을 확보할 수 있는 댐퍼의 선택이 필요하다. 본 연구의 목적은 기존의 가새형 및 벽체형 제진장치의 국내 공동주택 적용시의 문제점인 평면의 가변성 확보에 유리하고, 수동형 제진장치의 장점을 추구할 수 있는 마찰댐퍼를 삽입한 커플링보 제진시스템의 내진성능을 조사하는 것이다. 내진성능을 평가하기 위해서 실대형 커플링보 실험체를 계획하고 제작하였다. 실험체는 2개로 구성되어 있으며, 하나는 기존의 철근배근 상세를 갖는 철근콘크리트 커플링보 실험체와 커플링보에 마찰댐퍼가 삽입된 실험체이다. 횡하중에 대한 성능을 평가하기 위해서 유사정적 반복가력실험을 실시하였다. 엑츄에이터로부터 실험체 상보의 가력지그를 통해 하중이 전달되도록 하였으며, 가력은 최초 0.25%의 층간변형각부터 변위제어를 통해 목표 층간변형각인 1.5% 이상까지 진행되도록 하였다. 실험결과, 두 실험체의 이력곡선과 에너지 흡수능력을 평가하였다. RC 실험체는 핀칭현상이 관찰되었고, 가력이 진행됨에 따라 커플링보와 벽체에서의 균열이 확산되어 종국적으로 취성적인 커플링보의 전단파괴가 발생하였다. 마찰댐퍼를 삽입한 실험체는 계획된 마찰거동이 잘 발휘되어 목표 층간변형각인 1.5%까지 이선형거동이 잘 나타났다. 최대 내력은 RC 실험체가 3배 이상 크지만, 누적층간변형각에 따른 에너지 흡수능력은 마찰댐퍼 실험체가 2배 이상 우수한 결과를 보였으며, 커플링보 및 벽체에서의 균열이 매우 저감되었다.

Earthquake Simulation Tests on a 1:5 Scale 10-Story R.C. Residential Building Model (1:5 축소 10층 내력벽식 R.C. 공동주택의 지진모의실험)

  • This paper presents the results of shaking table tests on a 1:5 scale 10-story R.C. wall-type residential building model. The following conclusions are drawn based on the test results. (1) The model responded linear elastically under the excitations simulating an earthquake with a return period of 50 years, and showed a nonlinear response under the excitations simulating the design earthquake of Korea. (2) The model showed a significant strength drop under the maximum considered earthquake, with a return period of 2400 years. (3) The major portion of the resistance to lateral inertia forces came from the walls used for the elevator and stair case. (4) Finally, the damage and failure modes appear to be due to the flexural behavior of walls and slabs. A significant deterioration of stiffness and an elongation of the fundamental periods were observed under increased earthquake excitations.

  • Natural cavitied were found at shallow depth during construction of a huge bridge in Cambro-Ordovician Limestone Basin in the central part or Korea. The distribution patterns of cavities in this area were investigated carefully with a supplementary field job such as a structural geological survey, a geophysical survey, and a rock mechanical test in laboratory or field. A structural geological mapping produced a detail geological map focusing the route of the Proposed highway. It suggested that there were three faults in this wet and these faults had an influence on the mechanism of natural cavities. Among many kinds of geophysical surveys, an electrical resistivity prospecting was applied first on the specific area that was selected by results from the geological survey. Many evidences far cavities were disclosed from this geophysical data. Therefore, a seismic tomography was tested on the target wet which was focused by results from the electrical resistivity Prospecting and was believed to have several large cavities. A distinct element numerical simulation using the UDEC was followed on the target area after completing all of field surveys. Data from field tests were directly dumped or extrapolated to numerical simulations as input data. It was verified from numerical analysis that several natural cavities underneath the foundation of the bridge should be reinforced Based on the project result, finally, most of fecundations far the bridge were re-examined and the cement grouting reinforcement was constructed on several foundations among them.

Studies on the Resistance of 'Tong-il' Variety (IR-667) to Brown Planthopper, Nilaparvata lugens STAL (벼멸구에 대한 통일벼(IR-667)의 내충성에 관한 연구)

  • This study has been carried out to investigate the varietal resistance of Tong-il rice variety (IR-667) at the seedling stage to the brown planthopper. Nilaparvata lugens STAL, and further to know the biological effects of the varieties to the insects. The results obtained were as followings; 1. Tong-il variety tested in this experiment was susceptible to the brown planthopper but it seemed to be a little more tolerable than the Japonica varieties which had been recommended in Korea. 2. Tong-il variety (Suwon 213, Suwon 213-1, Suwon 214, and Suwon 215) was highly prefered by brown planthopper for feeding to the Korean recommended varieties Paltal, Jinheung, Akibare, and Shin #2. 3, The resistant varieties Mudgo, Kayama MGL-2, and Karsamba ASD-7 which showed low feeding preference exhibited higher ovipositional preference than the susceptible varieties. Relatively greater number of eggs was recorded on the Tong-il variety (Suwon 213, Suwon 213-1, and Suwon 215) in comparison with the Korean recommended varieties, 4, When the female brown planthopper adults were caged on Tong-il variety (Suwon 214) and the resistant variety Karsamba ASD-7 separately, greater number of eggs were observed on Suwon 214 and less on Karsamba ASD-7, while when two varieties Suwon 214 and Karsamba ASD-7 were given together in the same cage, Karsamba ASD-7 was much more prefered for oviposition to Suwon 214. 5. There was no difference in e99 Periods and their hatchal\bility between the resistant and susceptible varieties, but nymphal periods, rate of adult emergence, longevity of the adult, and its fecundity were significantly different according to the varieties.

  • National Academy of Agricultural Science and eight province Agricultural Research & Extension Services investigated the occurring area of herbicide resistant weeds in paddy field of Korea. In order to estimate the occurring areas of herbicide resistant weeds, we collected paddy soils randomly from 3,200 sites in 2011 and 2012, and treated 30 kg $ha^{-1}$ of pyrazosulfuron-ethyl+pyriminobac-methyl GR to the paddy soil. 176,870 ha, approximately 22.1% of cultivated area excluding organic and eco-friendly cultivated area, was estimated to be infested by SUherbicide resistant paddy field weeds. Several species of resistant weeds were occurred at Jeonranam-do with 44.3%, followed by 30.5% at Chungchungnam-do, 27.8% at Chungchungbuk-do, and 24.5% at Jeonrabuk-do, respectively. Monochoria vaginalis showed the highest distribution with 57,018 ha, 32.2% followed by Scirpus juncoides, and Lindernia procumbens, respectively.

  • The groundwater survey was carried out by electrical resistivity and resistivity tomography using several kinds of electrode array to delineate the depth of the saturated zone and basement complex, the aquifer system of fractured rock, and the 2-D resistivity structure was obtained by inversion technique. And the hole-to-surface and crosshole tomography were applied for two boreholes and the inverted resistivity tomogram are obtained. The comparison of those data with core logging data was performed and those results were relatively well correlated. And it was possible to find out the configuration of basement and the fracture zone and the aquifer system from the 2-D resistivity structure and resistivity tomogram.

Development of Molecular Markers for Alternative Oxidase Synthesis Genes in Brassica rapa L. (배추 alternative oxidase 합성 유전자와 연관된 분자마커 개발)

  • The low and high temperature stress might affect the yield and quality of many crop species. Alternative oxidase (AOX) gene is known as factors related to stress resistance in plants. In order to develop molecular markers related to stress resistance in Chinese cabbage, fifteen ESTs sharing sequence similarity to arabidopsis AOX genes were found using Brassica rapa EST database from NCBI. The polymorphic DNA sequences using the ESTs were then screened between Chinese cabbage, 'Chiifu' and 'Kenshin'. We found four ESTs that have either insertion or deletion between the two cultivars. These polymorphic sites were then targeted for development of the four PCR based molecular markers. These molecular markers developed in this study could be useful for a test of their relationship with abiotic stress resistance in Chinese cabbage.
