Effect on active transport of cell membrane model which irradiated by radiation (방사선이 조사된 세포막 모델이 물질의 능동수송에 미치는 영향)

  • The effect on active transport of $K^+$ and $Na^+$ of cell membrane model which irradiated by radiation was investigated. The cell membrane model used in this experiment was a $Na^+$ type sulfonated copolymerized membrane of styrene and divinylbenezene. The initial flux of the ion was increased with increase of both $H^+$ ion concentration. In this experiment range(pH $0.5^{-3}$), the initial flux of $K^+$ which was not irradiated by radiation was found to be from $7.9{\times}10^{-4}$ to $7.49{\times}10^{-3}mole/cm^2{\cdot}h$ and that of Na+ from $10.6{\times}10^{-4}$ to $7.68{\times}10^{-3}mole/cm^2{\cdot}h$. The initial flux of $K^+$ which was irradiated by radiation was found to be from $35.0{\times}10^{-4}$ to $42.4{\times}10^{-3}mole/cm^2{\cdot}h$ and that of $Na^+$ from $52.0{\times}10^{-4}$ to $43.3{\times}10^{-3}mole/cm^2{\cdot}h$. The membrane was selective for $K^+$ and the ratio $K^+/Na^+$ was about 1.10. And the driving force of pH of irradiated membrane was significantly increased about 4-5 times than membrane which was not irradiated. As active transport of $K^+$ and $Na^+$ of cell membrane model were abnormal, cell damages were appeared at cell.

  • This study examined the possibility on the dietary value and cryopreservation of the marine microalga, Isochrysis galbana. Four cryoprotectants, dimethyl sulfoxide (DMSO), ethylene glycol (EG), glycerol (Gly) and 1.2-propanedial (PD) were tested at the concentrations of 1.0, 1.5, 2.0, 2.5 and 3.0 M respectively. The highest survival rates were obtained with 1.5 and 2.0 M of four cryoprotectants yielding a survival rate of 80%. Cell concentration of $30\;{\times}\;10^4\;cell/ml$ at the initial point of the experiment was increased to $365\;{\times}\;10^4\;cell/ml$ in 1.5 M of dimethyl sulfoxide, $298\;{\times}\;10^4\;cell/ml$ in 1.5 M of ethylene glycol, $512\;{\times}\;10^4\;cell/ml$ in 1.5 M of glycerol, and $385\;{\times}\;10^4\;cell/ml$ in 1.5 M of 1.2-propanedial after five days, respectively. In dietary value experiment, survival rate and growth were not significantly different.

  • Methods to determine L-dopa(L-3,4-dihydroxyphenylalanine) in aqueous solution by spectrofluorimetry based upon the ligand sensitized luminescence of Tb(III) ion L-dopa complex have been studied. Tb(III) ion and Lu(III) ion were used as ligand sensitized fluorescencer and co-fluorescence enhancer, respectively. The effects of excitation wavelength, pH, concentration of Tb(III) ion, concentration of Lu(III) ion and emission wavelength on the fluorescence intensity were investigated. The fluorescence intensity of the Tb(III) ion L-dopa complex was further increased with addition of Lu(III) ion. The calibration curve for L-dopa was linear over the range from 5.0 ${\times}$ $10^{-7}$ M to 1.0${\times}$ $10^{-4}$ M and the detection limit was 4.0 ${\times}$ $10^{-8}$ M under the optimal experimental conditions of 300 nm, 8.0, 1.0 ${\times}$ $10^{-4}$ M and 545 nm for excitation wavelength, pH, concentration of Tb(III) ion and emission wavelength, respectively. When Lu(III) ion was added to Tb(Ⅲ) ion L-dopa complex, the concentration range of linear response and detection limit were 1.0${\times}$$10^{-8}$ M to 2.0 ${\times}$ $10^{-4}$ M and 1.0 ${\times}$ $10^{-9}$ M, respectively under the optimal experimental conditions of 300 nm, 8.5, 1.0 ${\times}$ $10^{-5}$ M, 1.0 ${\times}$ $10^{-5}$ M, 545 nm for excitation wavelength, pH, concentration of Tb(III) ion, concentration of Lu(III) ion and emission wavelength, respectively.

Effect of Ozone Treatment for Safety Improvement of Fresh Vegetable Juice (녹즙의 위생화를 위한 오존처리의 효과)

  • Fresh vegetable juice is a non-heat treated product and the only step to reduce microbial growth is washing. Therefore, the materials for fresh vegetable juice including Angelica keiskei, Brassica loeracea var. acephala, and Daucus carota L. were treated by ozone after the first washing process and investigated for microbial and chemical changes. The number of the total aerobic bacteria in materials after selection step were $8.2{\times}10^5{\sim}5.0{\times}10^6\;CFU/g$, which was a higher contamination level than the limit of Korea food code ($10^5\;CFU/g$). However, after the 1st washing process and ozone treatment, the total aerobic bacterial number was reduced to $4.7{\times}10^4{\sim}6.7{\times}10^4\;CFU/g$, which showed 2 log microbial reduction. After the 2nd washing step followed by ozone treatment, there was no difference in microbial number. The number of colifroms in the materials of fresh vegetable juice were $8.0{\times}10^3{\sim}3.5{\times}10^3\;CFU/g$ initially but showed $1.5{\times}10^2{\sim}3.0{\times}10^2\;CFU/g$ after the ozone treatment (1 log reduction). On the other hand, there was no changes in the contents of ascorbic acid, flavonoids, polyphenols, minerals (cadmium and lead) during all processes. In addition, no color changes were observed during washing process. Therefore, ozone treatment in the materials of fresh vegetable juice decreased the microbial numbers. Also, chemical characteristics of ozone treated sample were not different when compared with control.

Evaluation of Factors Related to Productivity and Yield Estimation Based on Growth Characteristics and Growing Degree Days in Highland Kimchi Cabbage (고랭지배추 생산성 관련요인 평가 및 생육량과 생육도일에 의한 수량예측)

  • This study was carried out to evaluate growth characteristics of Kimchi cabbage cultivated in various highland areas, and to create a predicting model for the production of highland Kimchi cabbage based on the growth parameters and climatic elements. Regression model for the estimation of head weight was designed with non-destructive measured growth variables (NDGV) such as leaf length (LL), leaf width (LW), head height (HH), head width (HW), and growing degree days (GDD), which was $y=6897.5-3.57{\times}GDD-136{\times}LW+116{\times}PH+155{\times}HH-423{\times}HW+0.28{\times}HH{\times}HW{\times}HW$, ($r^2=0.989$), and was improved by using compensation terms such as the ratio (LW estimated with GDD/measured LW ), leaf growth rate by soil moisture, and relative growth rate of leaf during drought period. In addition, we proposed Excel spreadsheet model for simulation of yield prediction of highland Kimchi cabbage. This Excel spreadsheet was composed four different sheets; growth data sheet measured at famer's field, daily average temperature data sheet for calculating GDD, soil moisture content data sheet for evaluating the soil water effect on leaf growth, and equation sheet for simulating the estimation of production. This Excel spreadsheet model can be practically used for predicting the production of highland Kimchi cabbage, which was calculated by (acreage of cultivation) ${\times}$ (number of plants) ${\times}$ (head weight estimated with growth variables and GDD) ${\times}$ (compensation terms derived relationship of GDD and growth by soil moisture) ${\times}$ (marketable head rate).

  • This paper is a study on the method that it is bruting a peace of a rough diamond with keeping the maximum volume of it. When a diamond will be bruting, how to keep the maximum diameter of one is very important because of its own characteristics that include shapes, mineral characteristics, and so on. In the middle of bruting. concentrating on the work is also required because girdle will be break down. We tried to bruting with gaining maximum diameters of rough diamond. As the result. an octahedron is made into the round shape with size $6.75\times 6.75\times 3.83$ mm , a dodecahedron the cushion shape with size $6.86\times 6.86\times 3.95$mm , a fancy the marquise with size $6.92\times 3.78\times 3.46$mm . Their rates of loss are round brilliant 24.5%, cushion barion 15.5 %, and marquise 18.7 %, respectively.

  • Nine kinds of amphoteric surfactants were derived from 1-(2-hydroxyethyl)-2-undecyl-2-imidazoline. Their surface activities including surface tension and isoelectric points were measured in aqueous solution and critical micelle concentration(cmc) was also evaluated by the measurement of surface tension. From the measurement of surface tension, carboxylated amides revealed to be 26~40dyne/cm at $4.0{\times}10^{-4}{\sim}1.0{\times}10^{-3}mol/{\ell}$, sulfonated or sulfated imidazolines, 30~35 dyne/cm at $1.5{\times}10^{-3}{\sim}2.5{\times}10^{-3}mol/{\ell}$ and sulfonated amides, 25~33 dyne/cm at $5.8{\times}10^{-4}{\sim}8.0{\times}10^{-4}mol/{\ell}$ concentration range. It was found that isoelectric points of carboxylated amides were pH 7.2~7.9 and those of sulfonated or sulfated imidazolines and sulfonated amides were pH 4.5~5.5.

The Analysis of Old Person's Total Antioxidant Capacity and Free Oxygen Radical According to Exercise Frequency of Combination Exercise (빈도에 따른 복합운동프로그램이 노인들의 체내 총항산능 및 활성산소 유리기에 미치는 영향)

  • Purpose:The purpose of this study was analyze the effect combination exercise program on plasma total antioxidant capacity and free oxygen radical according to exercise frequency of elderly. Age range of subject is 65~70 years. Methods:Subjects divided into three groups: control group(0-time), week-3 times group(3-times), week-5 times group(5-times). Each group were performed combination exercise during 8 weeks. Results:The results of each group by ANOVA free oxygen radical have a change(p<.05). We performed comparison of total antioxidant capacity and free oxygen radical pre-test and post-test on each groups. Total antioxidant capacity was increased in the 3-times and 5-times groups but there have not significant. Total antioxidant capacity is improve better than to 5-times. Free oxygen radical is significant increased in the 0-time (p<.05). But 5-times but 3-times is no significant decrease. Free oxygen radical is improve better than to 3-times. Conclusion:In the old people, exercise frequency week 3 times doing appeared effectively in improvement of the free oxygen radical and 5 times doing appeared effectively in improvement of the total antioxidant capacity. So rightly must apply a exercise frequency well in characteristic of old person each person.

Fabrication of semiconductor optical switch module using laser welding technique (반도체 광스위치 모듈의 제작 및 특성연구)

  • Semiconductor optical switch modules of 1$\times$2, 1$\times$4, and 4$\times$4 types for 1550 nm optical communication systems were fabricated by using laser welding technique, embodying in 30-pin butterfly package. For better coupling efficiency between switch chip and optical fiber, tapered fibers of 10~15mm lens radii were used, which provided up to 60% optical coupling efficiency. With the help of new laser hammering process, we could recover the lost optical power almost completely up to average 82% of initially obtained power. The fabricated optical switch modules showed good thermal stability of less than 5% degradation even after 200 times thermal cycling test. The 2.5 Gbps optical transmission characteristics of the 4$\times$4 switch module showed low sensitivities of less than -30dB for all possible switching paths. The transmission penalties of 1$\times$2 switch module at $10^{-10}$ BER were 0.6dB and 0.7dB for 50Xm and 90 Km optical fibers, respectively.

Microspore-derived Embryo Formation in Response to Cold Pretreatment, Washing Medium, and Medium Composition of Radish (Raphanus sativus L.)

  • Cold pretreatment, washing medium and composition of nutrient media may have marked effects on microspore embryogenesis. When microspores isolated from radish (Raphanus sativus L. cv. Gwanhun) flower buds were washed with Nitsch & Nitsch (NLN) medium liquid medium containing $130g{\cdot}L^{-1}$ sucrose (NLN-13), yields of microspore-derived embryos were greater than when using B5 liquid medium containing $130g{\cdot}L^{-1}$ sucrose. Microspore viability is known to decrease rapidly with storage; however, in this experiment, microspore viability was maintained for 24 h at $4^{\circ}C$ without media. Among the various medium concentrations used ($0.25{\times}$, $0.5{\times}$, $1.0{\times}$, $2.0{\times}$, and $4.0{\times}$ NLN liquid medium), $0.5{\times}$ NLN liquid medium induced the most efficient formation of microspore-derived embryos. In addition, microspore-derived embryos yields were greater when microspores were cultured in $0.5{\times}$ NLN liquid medium supplemented with $0.25{\times}$, $0.5{\times}$, and $1.0{\times}$ NLN microelements, compared to medium not supplemented with microelements. In this study, the highest yield of microspore-derived embryos was observed when the microspores derived from flower buds were washed using NLN-13 liquid medium and then cultured on $0.5{\times}$ NLN liquid medium supplemented with $0.25{\times}$ NLN microelements, followed by incubation at $25^{\circ}C$ for 30 days.
