In Vitro Antineoplastic Effects of Chitosan Hydrolysates on Various Tumor Cell Lines (키토산 가수분해물의 In Vitro 항종양성)

  • In this study, the antineoplastic effects of chitosan hydrolysates were assessed. The chitosan hydrolysates showed no cytotoxicity in in vitro trials using the normal cell line, Vero E6(Africa green monkey kidney cells). The $IC_{50}$ value of the chitosan hydrolysates on Vero E6 was 1,107.95 ${\mu}g/m{\ell}$. The hydrolysates exhibited in vitro antineoplastic activity in five human tumor (lung carcinoma, bladder carcinoma, colon carcinoma, stomach carcinoma, breast carcinoma) cell lines. The $IC_{50}$ values of the hydrolysates on A549, J82, SNU-C4, SNU-1, and ZR75-1 cells were 421.06, 417.99, 445.54, 380.65 and 460.49 ${\mu}g/m{\ell}$, respectively.

  • 본 연구는 콜레스테롤을 제거한 우유에 미세캡슐 키토올리고당을 첨가한 랫트 사료의 섭취량에 따른 체중 증가량, 총 콜레스테롤의 함량과 중성지질, HDL-cholesterol의 함량 변화를 연구하였다. 이 미세캡슐 키토올리고당을 SD 랫트에 매일 섭취시킨 결과HDL-cholesterol의 증가가 높게 나타났다. 본 실험에서, 혈중콜레스테롤 저하 기능이 있는HDL-cholesterol 수치가 증가하는 결과를 나타내므로 혈중 콜레스테롤 저하 기능성 우유로서의 기능이 있다고 사료된다.

A Study on the Deposition of Chitosan for Enhancing the Adsorption Ability of Activated Carbon (활성탄의 흡착력 향상을 위한 키토산 첨착에 관한 연구)

  • The preparative methods of a chitosan-deposited activated carbon and its characteristics were studied by using three kinds of chitosan with different degree of deacetylation and average molecular weight. The procedure was consisted of the dissolution of chitosan into acid solution, impregnation of activated carbon, agitation, evaporation, and drying. When the chitosan-dissolved acid and its concentration, amounts of chitosan deposited, and agitation conditions were changed, the specific surface area, deposition state on surface, and stability were investigated, and amounts of Cr(VI) adsorbed was measured. In the preparation process, it was proper to agitate the chitosan-dissolved acetic acid solution at room temperature for 1hr. In the deposition of chitosan with low molecular weight, the specific surface area of activated carbon was greatly decreased even at low chitosan loading, but in the case of high molecular weight it was not nearly changed to 10wt% loading. It was known that chitosan was uniformly and physically deposited on activated carbon. The chitosan-deposited activated carbon was stable into the solution over about pH 6. The removal of Cr(VI) was remarkably enhanced by adding the adsorption function of chitosan to the surface of activated carbon with about 5wt% chitosan. It may be therefore used as an adsorbent for removing the pollutants in air and wastewater.

Application Effects of Chitosan Fertilizer on the Growth of Cabbage and GABA Contents in the Cabbage (배추의 생장 및 배추 중의 ${\gamma}-aminobutyric$ acid 함량에 미치는 키토산비료의 시비효과)

  • To investigate the effects of chitosan on growth and quality improvement of vegetables, we utilized cabbage as a model plant system and SL-chitosan as a chitosan fertilizer. The chitosan fertilizer treatment increased the leaf lengths of cabbage seedlings compared with those of control groups. In addition, the content of ${\gamma}-aminobutyric$ acid (GABA) in the fertilizer-treated cabbage seedlings was higher than that in the control group. Peripheral lengths and head weights of cabbages along with their GABA contents were also measured during the growth of cabbages in field. The fertilizer treatment, without changing the physico-chemical properties of main field soil after the cultivation of cabbage, significantly increased the peripheral length, average weight and GABA content compared with control treatment. These results may suggest that the quality and quantity of cabbage can be improved by chitosan treatments.

  • In this study, the optimal conditions for hair dyeing using sappan wood (Caesalpinia sappan L.) powder were determined, and then the effect of chitosan treatment on the hair dyeing properties of sappan wood powder was investigated. Treatment of sappan wood powder and water with the ratio of 1:3 (w/w) at 40℃ for 60 minutes was found to be the most effective to hair dyeing. In addition, the pretreatment of low molecular weight (~30 kDa) chitosan at a concentration of 1% (w/w) prior to treatment with sappan wood powder effectively improved and maintained the hair dyeing properties of sappan wood. In conclusion, the low molecular weight chitosan is considered to be of great help as a mordant in improving the dyeing efficiency and retention of sappan wood powder in hair dyeing.

Effect of Polymer, Calcium, Perlite and Chitosan in Organic Amendment on Growth in Kentucky Bluegrass (유기질개량재에서 폴리머, 칼슘, 펄라이트 및 키토산 성분이 켄터키 블루그래스의 생장에 미치는 효과)

  • Research was initiated in greenhouse to investigate effects of polymer, calcium, perlite and chitosan on the growth of Kentucky bluegrass (KB). A total of 24 treatment combinations were used in the study. Treatments were made of water-swelling polymer (WSP), calcium, perlite, and chitosan in soil organic amendment (SOA). Significant differences were observed in germination rate, turfgrass coverage, turfgrass density and top growth among treatments. Germination rate, density and plant height varied with time after seeding. A proper mix of WSP is considered to be lower than 3% for turfgrass coverage and density. Regarding survival capability and top growth, however, it was good under 6%. Overall KB growth was more influenced by calcium and perlite than chitosan. Calcium and perlite were the most effective elements for early survival capacity and turfgrass density, respectively. But no effect was found by chitosan. Top growth increased with three elements, being perlite > calcium > chitosan. The chitosan was effective in early germination, but there was no effect on top growth until 3 weeks, when compared with others. A further study is needed for investigating the effect of these materials on the growth characteristics in mixtures of sand and SOA before a field application.

  • In order to obtain microbial endochitosanase for enzymatic production of chitooligosaccharides from chitosan, we screened four microbes from soil and selected. Bacillus sp. HSB-21 which showed highest activity. Chitosanase, produced from isolating microbe, was endo-type and molecular mass of the enzyme was estimated as 21,000 by active staining. Its optimum pH and temperature were 5.5 and $50^{\circ}C$, respectively. It was stable in the pH range of 3.0 to 8.0 and up to $40^{\circ}C$. It did not produce chitomonosaccharide and produced chitooligosaccharide ranging from chitobiose to chitooctaose as major end-products from chitosan. The chitosanase from Bacillus sp. HSB-21 can be applicable to enzymatic production of chitooligosaccharide which has high degree of polymerization .

Stepwise Addition Technology for the High Yield Production of Chitosanoligosaccharide (키토산올리고당 생산을 위한 다단계 첨가방법)

  • Optimization of the chitosanoligosaccharide production was studied with chitosanase. The optimum conditions for the enzymic reaction have been determined. Enzyme stability was maintained above 90% after 6 days at pH 5.0. The optimum initial reaction rate was appeared in 1.0% of chitosan solution. The production yield of chitosanoligosaccharides was over at 0.4%~2.0% of chitosan. At 4.0% of chitosan solution, however, the production yield was decreased to 64.6%. To increase the yield, stepwise addition of substrate into the reactor was investigated. In this case, the yield was increased from 64.6% to 83.2% and the final concentrations of chitosanoligosaccharide was 12.26 mg/mL. By TLC analysis, most of the chitosanoligosaccharides produced were 3-5 mers.

  • In an effort to develop a potent system for the production of various dp (degree of polymerization) chitooligosaccharides, 32 enzymes or microbial systems were screened for chitosanolytic acitivity using chitosan as a substrate. The efficiency of each enzyme system was evaluated by the changes of turbidity and viscosity of chitosan solution, the amount of precipitate and the reducing sugar-producing activity in the enzymatic reaction mixture. Based on these assay methods for the chitosanase activity, Bacillus sp. P2l out of 32 screened systems showed highly potent endochitosanase, which was comparable with a commercially available enzyme (E7). Chitooligosaccharides of dp 3-7 were separated by TLC as major enzymatic reaction products, suggesting that the chitosanase from Bacillus sp. P2l be endo-splitting type.

Effect of Inclusion of Chitosan-Oligosaccahariede in Drinking Water on the Blood Component Profile, Immunity and Antioxidative Enzyme in Broiler Chickens (키토산 올리고당의 음수내 급여가 육계의 혈액성상, 면역력 및 항산화 효소계에 미치는 영향)

  • The study was conducted to the effect of inclusion of chitosan-oligosaccharide in drinking water on the blood component profile, immunity and antioxidative enzyme of broiler chickens. A total of 28,000 broiler chickens (Arbor Acre) at 1 days of age were fed the commercial diet until 35 days of age, the treatment divided into two treatments, normal control group and chitosan-oligosaccharide in drinking water group. In concentration of glucose, treatment was significantly decreased than control (P<0.05), but not statistically different on the Triglyceride, Total cholesterol, HDL-cholesterol and LDL-cholesterol in the Blood component profile of broiler chicks. The concentration of glutamic oxalacetic transaminase (GOT) was statistically different but the concentration of glutamic pyruvate transaminase (GPT) was significantly decreased in treatment fed chitosan-oligosaccharide than control (P<0.05). Immunoglobullins in the blood, concentration of IgG was not significantly different among control and treatment but concentration of IgM was significantly increased in fed chitosan-oligosaccharide than control (P<0.05). Antioxidant and super oxide dismutase (SOD) was not different among control and treatment and concentration of catalase and glutathione peroxidase were significantly increased in fed chitosan-oligosaccharide than control (P<0.05).
