Development of Small-scale Hybrid Rocket Motor using $PE-N_2O$ Propellants ($PE-N_2O$ 추진제를 이용한 소형 하이브리드 로켓 모터 개발)

  • In this study, a hybrid rocket motor with separable and detachable oxidizer tank from combustion chamber is developed. Initially, the measured thrust of the motor showed about 30% of the design thrust since the oxidizer supply was not enough. In order to solve this problem, application is made to expand the orifice diameter of oxidizer injector empirically, so that the mass flow rate of oxidizer was improved. The improved performance was about 60% of design thrust, 18kgf, and thrust-to-weight ratio was reasonable, compared with other sounding rockets.

Determination of Boron by Ion Pair Liquid Chromatography with Chromotropic Acid (Chromotropic Acid를 착화제로 이용한 이온쌍 액체 크로마토그래피에 의한 붕소의 분리와 정량)

  • The separation and determination of boron with chromotropic acid (1,8-Dihydroxynaphthalene-3,6-disulfonic acid) as a complex agent has been studied using ion pair liquid chromatography. The use of tetrabutylammonium bromide added as an ion pair reagent to mobile phase (MeOH 61%, phosphate buffer 39% pH=8.5) allowed good separation of boron-chromotrophic acid complex anion and chromotrophic acid on poly(styrene-divinylbenzene) based reversed phase column (PRP-1, 15 $cm{\times}4.6$ mm i.d.). The complex formation between boric acid and chromotrophic acid was enhanced in the presence of 0.1 M tetrabutylammonium bromide, resulting in high sensitivity. The linear calibration was achieved over the boron concentration range of 0.5∼1000 ${\mu}g/L.$ The detection limit was 0.5 ${\mu}g/L$ (S/N=2). The proposed method was applied to the determination of boron in commercially available chemicals, $Na_2SO_4$, NaOH, KCl.

Isolation and Expression of Aluminum Induced Protein(ClAIP) Gene from Codonopsis lanceolata (더덕에서 Aluminum Induced Protein (ClAIP) 유전자의 분리 및 발현분석)

  • A cDNA clone (GenBank accession no. CF924621) homologous to aluminum induced protein gene was isolated and characterized from Codonopsis lanceolata (ClAIP). The ClAIP is 906 nucleotides long and has an open reading frame of 711 bp with a deduced amino acid sequence of 236 residues. The ClAIP shows high homology to A. marina (84%), G. hirsutum(83%), V. radiata (83%), A. thaliana (80%), B. nap us (78%) and T. aestivum (68%). The deduced amino acid sequence of ClAIP also has homology to the N-terminal end of plant Asn synthetase. This region does not contain the active sites of the enzyme and the significance of this conservation is currently not clear. To investigate the expression of ClAIP against several heavy metal stresses, we treated the sliced tap root of C. lanceolata with various heavy metals. The expression of ClAIP was increased by 25 uM $Al_2$(SO$_3$)$_4$ in proportion to incubation time and also increased by 50 uM CdCl$_2$.

  • Bifidobacteria have been known as beneficial inhabitant of human intestine. Therefore, bifidobacteria began to be noticed as a starter in the manufacture of fermented dairy products. Perhaps the key for effective use of bifidobacteria in commercial dairy products is the maintenance of viability of bifidobacteria during large scale preparation of starter culture and distribution of products. So we tried to obtain the bifidobacteria having suitable characteristics for using as a starter in the manufacture of fermented dairy products. Among bifidobacteria isolated from Korean, E-4 strain showed the highest resistance to oxygen. To know whether the selected strain will be fit for manufacture of fermented dairy products, we also investigated resistance of the selected strain to HCI. The selected strain, E-4, was more resistant to environmental stresses such as oxygen, H2O2 and HCI than Bifidobacterium longum known as resistant strain to environmental stresses. According to carbohydrate fermentation patterns and morphological characteristics, E-4 strain was identified as B. bifidum. In conclusion, the selected strain, E-4, was thought to be fit for manufacture of fermented dairy products.

Studies on the Separation and Preconcentration of Metal Ions by XAD-16-[4-(2-thiazolylazo)] orcinol Chelating Resin (XAD-16-[4-(2-thiazolylazo)]orcinol 킬레이트 수지에 의한 금속이온의 분리 및 농축에 관한 연구)

  • The sorption and desorption properties of U(VI), Th(IV), Zr(IV), Cu(II), Pb(II), Ni(II), Zn(II), Cd(II) and Mn(II) ions on XAD-16-[4-(2-thiazolylazo)orcinol] (TAO) chelating resin were studied by elution method. The effect was examined with respect to overall capacity of each metal ion, separation of mixed metal ions, flow rate and concentration of buffer solution for optimum condition of sorption. The overall capacities of some metal ions on this chelating resin were 0.35nmol U(VI)/g resin, 0.49nmol Th(IV)/g resin, 0.41nmol Cu(II)/g resin, and 0.31nmol Zr(IV)/g resin, respectively. The elution order of metal ions obtained from breakthrough capacity and overall capacity at pH 5.0 was Th(IV)>Cu(II)>U(VI)>Zr(IV)>Pb(II)>Ni(II)>Zn(II)>Mn(II)>Cd(II). The group separation of mixed metal ions was possible by increasing pH in pH range 2~5 at a flow rate of 0.28mL/min. Characteristics of desorption were investigated with desorption agents such as $HNO_3$, HCl, $HClO_4$, $H_2SO_4$, and $Na_2CO_3$. It was found that 2M $HNO_3$ showed high desorption efficiency to most of metal ions except Zr(IV) ion. Also, desorption and recovery of Zr(IV) ion were successfully performed with 1M $H_2SO_4$. Recovery of trace amount of U(VI) ion from artificial sea water was over 94%. The chelating resin, XAD-16-TAO was successfully applied to group separation of rare earth metal ions from U(VI) by using 2M $HNO_3$ as an eluent.

Flavonoids from the arial parts of Artemisia agryi and their antioxidant capacity through GSH recovery effect (황해쑥(Artemisia agryi)으로부터 flavonoid 화합물들의 분리 동정과 세포 내 GSH 회복능을 통한 항산화 활성 평가)

  • The arial parts of Artemisia argyi were extracted with methanol : water (70:30), and the concentrates was partitioned into EtOAc (ethyl acetate), n-BuOH (normal butanol), and H2O (water) fractions. The repeated silica gel and ODS (octadecyl silica gel) column chromatographies for EtOAc and n-BuOH fractions led to isolation of four flavonoids without any ambiguity based on intensive interpretation of several spectroscopic data including nuclear magnetic resonance, and mass spectrometry. The chemical structure of the isolated compounds revealed to (2S)-naringenin (1), 3-methylkaempferol (2), 3,3'-dimethylquercetin (3), and 3,3',4'-trimethylquercetin (4). These four compounds were first isolated from A. argyi through this study. In this study, four compounds isolated from A. argyi showed an increase in glutathione mean and a decrease in glutathione heterogeneity so that the compounds uniformly raised the intracellular glutathione (GSH) level. Based on these results, it is considered that it can be used as a functional pharmacological material.

A Study on the Oxy-Combustion of the Coal in Drop Tube Furnace (Drop Tube Furnace에서 석탄의 순산소 연소 특성)

  • The oxy-combustion system is one of the carbon recovery and storage technologies (CCS: Carbon capture & storage) that performs coal combustion using pure oxygen and recirculated flue gas. This is a technology that facilitates storage of carbon dioxide by generating an exhaust gas consisting of only carbon dioxide without a process of separating carbon dioxide and nitrogen when coal is burned using pure oxygen and recirculated flue gas mixture instead of a conventional air combustion system that produces carbon dioxide and nitrogen mixed exhaust gas. In this study, the characteristics of generated NO and SO2 as atmospheric pollutants during oxy-combustion were examined using O2/CO2 mixed simulation gas. The reaction temperature was varied from 900 ℃ to 1200 ℃ and oxygen partial pressure was varied from 30% to 50%. The results showed that NO and SO2 concentrations in flue gas increased as the oxygen concentration and the reaction temperature in the furnace increased. The partial pressure of CO2 in flue gas also increased as the oxygen concentration and the reaction temperature in the furnace increased. As a results of comparing NO production of 30% O2/CO2 oxy-combustion with air combustion, NO in flue gas increased with reaction temperature in both experiments and NO of oxy-combustion was 40 ~ 80 ppm lower than that of air combustion.

  • Marine microorganisms to produce functional biopolymers were isolated from the seashore of the Kyungsang province. Microorganisms to hydrolyze carboxy-methyl cellulose(CMC) were cultured in marin broth and the other liquid medium that contained 2.0% (w/v) glucose, 0.25% yeast extract, 0.5% $K_2HPO_4$, 1% NaCl, 0.02% $MgSO_4{\cdot}7H_2O$ and 0.06% $(NH_4)_2SO_4$ to investigate the ability to produce carboxymethyl cellululase (CMCase) under aerobic conditions. Twelve microorganisms among them showed higher activities of CMCase than B. amyloliquefaciens DL-3, which was known as a cellulase-producing strain. The microorganism showing highest activity of CMCase in this study was identified as Bacillus subtilis subsp. subtilis with 16S rDNA partial sequencing and gyrase A partial sequencing and named as B. subtilis subsp. subtilis A-53.

  • A yeast, Torulopsis bombicola, was cultured in the media fortified with soybean oil as a additional carbon source for 7 days with reciprocal shaking. From the culture broth, sophorose-lipid was isolated and treated with alkali to afford sophorose. The sophorose contained in the medium was acetylated and isolated through silica gel column chromatography. The aceylated sophorose was hydrolyzed with 5% KOH at room temperature to give rise to sophorose. Meanwhile, the MeOH extracts obtained from the pod of Sophora japonica was solvent-fractionated with n-BuOH and $H_2O$, and butanolic layer was chromatographed on silica gel column to afford a flavonoide-glycoside. The glycoside was hydrolyzed with 0.02 N $H_2SO_4$ to yield sophorose.

Increased Production of an Alkaline Protease from Bacillus clausii I-52 by Chromosomal Integration (Bacillus clausii I-52의 Chromosomal Integration에 의한 Alkaline Protease의 생산성 향상)

  • TTo increase productivity of a strong extracellular alkaline protease (BCAP), stable strains of Bacillus clausii I-52 carrying another copy of BCAP gene in the chromosome were developed. Integrative vector, pHPS9-fuBCAP carrying BCAP promoter, ribosome binding site, signal sequence and active protease gene was constructed and transferred into B. clausii I-52, and integration of the constructed plasmid into chromosome was identified by PCR. An investigation was carried out on BCAP production by B. clausii I-52 and transformant C5 showing the highest relative activity of alkaline protease using submerged fermentation. Maximum enzyme activity was produced when cells were grown under the submerged fermentation conditions at $37^{\circ}C$ for 48 h with an aeration rate of 1 vvm and agitation rate of 650 rpm in a optimized medium (soybean meal 2%, wheat flour 1%, sodium citrate 0.5%, $K_2HPO_4$ 0.4%, $Na_2HPO_4$ 0.1%, NaCl 0.4%, $MgSO_47H_2O$ 0.01%, $FeSO_47H_2O$ 0.05%, liquid maltose 2.5%, $Na_2CO_3$ 0.6%). A protease yield of approximately 134,670U/ml was achieved using an optimized media, which show an increase of approximately 1.6-fold compared to that of non-transformant (83,960 U/ml). When the stability of transformant C5 was examined, the integrated plasmid pHPS9-fuBCAP was detected in the transformant after cultivation for 8 days, suggesting that it maintained stably in the chromosomal DNA of transformant C5.
