Research on the potential biological activity of red alga Symphyocladia spp. has been limited to Symphyocladia latiuscula, which is widely used as a food ingredient in Korea. Here, we examined the biological activity of another species, Symphyocladia linearis, which is found in Korea and was reported as a new species in 2013. The aim of this study was to evaluate the antioxidant, anti-inflammatory, and antibacterial properties of a 70% ethanol extract of S. linearis. Antioxidant activity, which was evaluated using radical scavenging assays, revealed half maximal inhibitory concentration values for 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) of 34.57 and 11.70 ㎍/mL algal extract, respectively. Anti-inflammatory activity of the S. linearis ethanolic extract was evaluated using RAW 264.7 cells by measuring the inhibition of lipopolysaccharide-induced nitric oxide (NO) and prostaglandin E2 (PGE2) production. The potential cytotoxicity of NO and PGE2 was first examined, confirming no toxicity at concentrations ranging from 10-100 ㎍/mL. NO production was inhibited 61.1% and 78.0% at 50 and 100 ㎍/mL S. linearis extract, respectively; and PGE2 production was inhibited 69.1%, 83.2%, and 94.8% at 25, 50, and 100 ㎍/mL S. linearis extract, respectively. Thus, the S. linearis extract showed very strong efficacy against PGE2 production. The cellular production of reactive oxygen species, measured using 2',7'-dichlorofluorescin diacetate fluorescence, was inhibited 48.8% by the addition of 100 ㎍/mL S. linearis extract. Antibacterial activity was evaluated using the disc diffusion method and minimum inhibitory concentration (MIC). S. linearis was effective only against gram-positive bacteria, exhibiting antibacterial activity against Staphylococcus aureus with a MIC of 256 ㎍/mL extract and against Bacillus cereus with a MIC of 1,024 ㎍/mL extract. Based on these results, we infer that a 70% ethanolic extract of S. linearis possesses strong anti-inflammatory properties, and therefore has the potential to be used in the prevention and treatment of inflammatory and immune diseases.
에탄올이 면역계의 항원전달세포이며, 동시에 감마인터페론 등의 림포카인에 의해 활성화되면 작동세포의 역활을 하는 단핵구-대식세포의 arachildonic acid 대사의 cyclooxygenasc 경로의 프로스타글라딘 생산에 미치는 영향을 연구하기 위하여 감마 인터페론으로 전처치하여 활성화시킨 단핵구와 대조군 단핵구를 사용하여 방사면역측정법으로 프로스타글라딘들을 측정하였다. 건강한 성인의 말초혈액으로 Ficoll-Hypaque용액을 이용한 농도구배법과 젤라틴 피복 플라스크를 이용하여 단핵구를 순수하게 분리한 다음 800단위의 감마 인터페론을 사용하여 3일동안 단핵구를 활성화시켰다. 감마인터페론으로 활성화 시키지 않은 $10^6$개의 단핵구에 100mM,300mM,600mM의 에탄올을 30분간 반응시킨 후 6시간후에 thromboxane B$_2$, PGEB$_2$ 와 6-keto-PGF1$\alpha$ 와 PGE$_2$ 모두 대조군에 비하여 생산이 증가하였다. 감마인터페론으로 전처치한 경우에는 알코올 처치에 관계없이 세가지 프로스타글란딘들 모두 6시간과 24시간에서 대조군보다 감소되었다. 이와같은 성적들로 미루어 볼 때 감마인터페론 처치가 에탄올로 유발한 단핵구 arachidonic acid 대사의 변화를 억제한다고 사료된다.
BACKGROUND/OBJECTIVES: The aim of this research was to study the different long term effects of consumption of dietary oil sources with varying omega-6/omega-3 (${\omega}-6/{\omega}-3$) polyunsaturated fatty acids (PUFAs) ratios on bone marrow fatty acid level, ex vivo prostaglandin $E_2$ ($PGE_2$) release, and mineral content of bone in rabbits. MATERIALS/METHODS: For this purpose, weaning and female New Zealand white rabbits were purchased and randomly divided into five groups and offered ad libitum diets containing 70 g/kg of added oil for 100 days. The dietary lipid treatments were formulated to provide the following ratios of ${\omega}-6/{\omega}-3$ fatty acids: 8.68 soy bean oil (SBO control), 21.75 sesame oil (SO), 0.39 fish oil (FO), 0.63 algae oil (DHA), and 0.68 algae oils (DHA/ARA). DHA and ARA are two types of marine microalgae of the genus Crypthecodinium cohnii. RESULTS: The dietary treatments had significant effects on the bone marrow fatty acids of rabbits. Rabbits fed the FO diet, containing the highest ${\omega}-3$ PUFA concentration, and those fed the SBO diet showed the highest ${\omega}-6$ PUFA. On the other hand, a positive correlation was observed between Ex vivo $PGE_2$ level and the ${\omega}-6/{\omega}-3$ dietary ratio. Significant effects of dietary treatment on femur Ca, P, Mg, and Zn contents were observed in both genders. CONCLUSIONS: Findings of the current study clearly demonstrated that dietary PUFA, particularly ${\omega}-6/{\omega}-3$ and ARA/EPA ratios are important factors in determining bone marrow fatty acid profile, and this in turn determines the capacity of bone for synthesis of $PGE_2$, thereby reducing bone resorption and improving bone mass during growth.
Objectives : This study investigated the anti-oxidant and anti-inflammatory activities of Changchulgeumryeontang (CCGRT) extract. Methods : The macrophage cell line RAW 264.7 cells were used and MTT assay was performed to measure the cell viabilities at the various concentrations of CCGRT ($25-200{\mu}g/m{\ell}$). Nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) were measured in LPS-induced RAW 264.7 cells. Expressions of iNOS, $NF-{\kappa}B$, $IL-1{\alpha}$, $IL-1{\beta}$ and IL-6 were also performed by real-time PCR. The anti-oxidant activities of CCGRT was measured by DPPH radical scavenging activity. Results : 1. there was no cytotoxicity in RAW264.7 cells treated with CCGRT compared to the control. 2. CCGRT treated group significantly inhibited NO and $PGE_2$ production compared to the LPS treated group. 3. CCGRT treated group significantly decreased mRNA expressions of iNOS, $NF-{\kappa}B$, $IL-1{\alpha}$, $IL-1{\beta}$ and IL-6 compared to the LPS treated group. 4. CCGTR was found to have high DPPH free radical scavenging ability. Conclusions : According to the above results, CCGRT may be a potentional choice for the treatment of inflammatory skin disease. MspTL is the major surface protein of Treponema lecithinolyticum associated with periodontitis and endodontic infections. Our recent investigation revealed that MspTL induces proinflammatory cytokines and intercellular adhesion molecule 1 in THP-1 cells and periodontal ligament cells. In this study we conducted oligonucleotide microarray analysis to investigate the global transcriptional regulation in THP-1 cells stimulated with purified recombinant MspTL. MspTL upregulated the expression of 90 genes in THP-1 cells at least four fold, and the functions of these genes were categorized into adhesion, apoptosis/antiapoptosis, cell cycle/growth/differentiation, chemotaxis, cytoskeleton organization, immune response, molecular metabolism, proteolysis, signaling, and transcription. The majority of the modified genes are known to be NF-${\kappa}B$-responsive and interferon-stimulated genes (ISGs). The expression of 12 selected genes was confirmed by real-time RT-PCR. Because prostaglandin $E_2(PGE_2)$ is an important inflammatory mediator and Cox-2 was found to be induced by MspTL in the microarray analysis, we determined the level of $PGE_2$ in the culture supernatants of MspTL-treated cells and found that MspTL significantly increased $PGE_2$. Our results provide insight into the gene regulation of host cells in response to MspTL, and may contribute to the understanding of the molecular mechanism in periodontitis.

Biotin is a water-soluble vitamin used as a skin conditioning agent and promotes the formation of intercellular lipid layers through increased lipid synthesis, which improves the skin's natural barrier function. The anti-inflammatory effects of biotin have been investigated using in vitro assay models, such as MTT assay, measurements of concentrations of nitric oxide(NO), prostaglandin E2(PGE$_2$), and inhibition rate of 5-lipoxygenase(5-LOX). In comparison with biotin, other plant extracts were tested at the same time which were kudzu vine extract, sage extract, paeonia extract, and dipotassium glycyrrhetinate. Nitric oxide is a signal molecule with functions such as neurotransmission, local vascular relaxation, and anti-inflammation in many physiological and pathological processes. NO can cause apoptosis and necrosis of target cells such as keratinocytes and is generated from L-arginine by nitric oxide synthase (NOS). Prostanoids, including prostaglandins and thromboxanes, are generated by the phospholipase $A_2$/cyclooxygenase(COX) pathway, and leukotrienes are generated by the 5-lipoxygenase pathway from arachidonic acid. Prostaglandin E2 recently have been shown to be beneficial in the resolution of tissue injury and inflammation, also has been implicated as an immunosuppressive agent and plasma levels of PGE$_2$ are elevated in patients sustaining thermal injury. Lipoxygenase metabolites from arachidonic acid have been implicated in inflammation, anti-inflammatory activity of the raw materials was evaluated in vitro by the offered inhibition of lipoxygenase.
Objectives : In this study, the effects of ethylacetate extract of Ostericum koreanum on inflammation in RAW264.7 cells were investigated. Methods : Dried roots of Ostericum koreanum was extracted with 80% methanol for 24 h, and then fractionated with n-butanol, n-hexan and ethylacetate. RAW264.7 cells, a mouse macrophage line were incubated with different concentrations of the extract for 30 min and then stimulated with LPS at indicated times. Cell toxicity was determined by MTT assay. The concentrations of nitric oxide (NO) and prostaglandin E2 ($PGE_2$) were measured by Griess assay and enzyme immunoassay (EIA), respectively. The expressions of inducible nitric oxide synthease (iNOS) and cyclooxyganase (COX) -2 mRNA and protein were determined by RT-PCR and Western blot. Results : The methanol extract of Ostericum koreanuman and its fractions were significantly inhibited the NO and PGE2 productions in LPS-stimulated RAW264.7 cells. Among the fractions of Ostericum koreanuman the ethylacetate fraction was more strongly inhibited NO and $PGE_2$ productions compared with other fractions. The ethylacetate fraction was also suppressed LPS-induced mRNA expressions of iNOS and its protein levels in RAW264.7 cells. Conclusions : This study suggests that the ethylacetate fraction of Ostericum koreanum may have an anti-inflammatory property through suppressing inflammatory mediator productions in activated macrophages, suggesting have a therapeutic potential for the treatment of various inflammatory diseases.
괴각(Fructus Sophorae)은 회화나무(Styphnolobium japonicum L.)의 열매를 건조한 것으로 전통 한의학에서 널리 사용되는 약재 중의 하나이다. 본 연구에서는 murine RAW 264.7 대식세포 모델을 이용하여 괴각 추출물 (Fructus Sophorae extracts, FSE)이 면역 조절능에 미치는 영향을 조사하였다. 이를 위한 대식세포 활성과 연관된 면역 반응 parameter로서 prostaglandin $E_2$ ($PGE_2$)와 tumor necrotic $factor-{\alpha}$ ($TNF-{\alpha}$)의 생성에 미치는 FSE의 영향을 조사하였다. 본 연구의 결과에 의하면 FSE는 대식세포의 활성을 유도하였고, $PGE_2$ 및 $TNF-{\alpha}$의 생성을 촉진하였으며, 이는 cyclooxygenase-2 (COX-2)와 $TNF-{\alpha}$ 유전자의 전사 및 번역 수준에서의 활성화와 연관성이 있었다. 또한 FSE 처리에 의하여 다양한 종류의 cytokine 발현의 증가를 cytokine array 분석을 통하여 확인하였으며, RAW 264.7 대식세포의 활성화에는 mitogen-activated protein kinases (MAPKs) 및 phosphatidylinositol-3-kinase (PI3K)/Akt 경로 활성화가 연관되어 있음을 알 수 있었다. 본 연구의 결과는 괴각 추출물이 면역 증강제로서의 개발 가능성이 매우 높음을 시사한다.
백금족 금소과 규산염을 포함하는 자동차 폐촉매 내의 백금족 금속과 주요 성분들을 ICP-AES로 동시에 분석하기 위한 두 종류의 전처리 방법이 비교되었다. HF, $HNO_3$, HCl, $HClO_4$, $H_2O_2$, 및 $H_3BO_3$ 등의 혼합산의 다양한 조합을 사용하여 열판(hot plate) 용해법과 밀폐용기를 사용하는 극초단파(microwave) 가압-가열법을 비교 검토하였다. 전통적인 열판 용해법보다 밀폐용기를 사용한 극초단파 가압-가열법이 화수율, 분석시간 및 사용하는 양 등의 측면에서 상대적으로 우수한 결과를 나타내었고, 0.25 g의 폐촉매 시료에 대하여 HF 2 mL, $HNO_3$2 mL, 그리고 HCl 6 mL를 사용하여 200 psi(13.79 bar), $180^{\circ}C$ 조건에서 1시간 동안 가열한 후, 5% (w/v) $H_3BO_3$ 16 mL를 사용하여 20 psi(1.38 bar), $100^{\circ}C$ 조건에서 10분간 가열한 다음, 왕수 10 mL로 열판에서 두 번 반복하여 처리하는 것이 백금족 금속의 회수율 측면에서 가장 좋은 결과를 가져왔고(XRF 분석 대비 85~110%), 이 방법은 2% 이내의 상대 표준 편차를 보임으로써, 자동차 폐촉매의 일상적인 분석기법으로서의 충분한 가능성을 보였다.
본 연구의 목적은 RAW 264.7 세포에 산초 종자 정유 성분의 항염 및 항산화 효과에 대하여 알아보고자 하였다. 산초 종자로부터 정유를 추출하였으며, 추출물의 항산화 및 항염 활성에 대하여 평가하였다. 추출물의 항산화 활성은 DPPH 라디칼 소거능과 SOD 유사 활성 평가법을 이용하였다. 또한 세포독성 평가를 위해 MTT를 이용하여 세포생존율을 측정하였다. LPS로 염증이 유도된 RAW 264.7 세포에 대하여 산초 정유가 NO생성과 $PGE_2$ 생성 정도에 미치는 영향을 측정하였다. 그 결과 산초 종자 정유는 농도 의존적으로 DPPH 라디칼 소거능과 SOD 유사 활성이 증가하는 것으로 나타났다. 세포생존율 평가에서는 $40{\mu}g/mL^{-1}$ 이하의 농도에서 98% 이상의 낮은 세포독성을 나타냈으며, 항염 효능 평가에서는 LPS를 단독으로 처리한 양성대조군보다 산초 정유 성분을 처리한 군에서 NO와 $PGE_2$ 생성이 현저하게 감소되는 것을 확인하였다. 이러한 결과는 산초 정유가 염증매개인자를 감소시키고 산화의 활성을 방지할 수 있는 기능성 식물소재로서의 가능성을 나타낸다.
