  • To facilitate the heterosis breeding which utilizes cytoplasmic-genetic male sterile lines in rape (Brassica napus L.), 511 improved domestic lines were tested for their fertility restoration, maintaining ability and heterosis expression in agronomic characters. About 81 % of 511 lines showed complete fertility restoration and 11 lines could be used as maintainer of male sterility. All yield components except pod length showed greater heterotic effects than that of midparent in $F_1$. These heterotic effects resulted in extraordinary yielding ability as high as 5.6 T/ha in several $F_1$`s.

Restorer Genotype for Male Sterile Cytoplasm of Genetic Resources Moderately Resistant to Phytophthora capsici in Capsicum Pepper

  • KC00256, KC00406, KC00462, KC00463, KC00820, and KC00821, the genetic resources that have previously been reported as moderately resistant to Phytophthora capsici, as well as the line KC01322, a new source of moderate resistance introduced from Laos, were tested against two strains (Pc003 and Pc005) of P. capsici. We also determined the nuclear restorer genotypes of these lines, in regards to their interaction with cytoplasmic male sterility, through crossing the resources with cytoplasmic male sterile Punggok-A (Srfrf) and determining the fertility of the $F_1$ hybrids. The studied lines exhibited a low level of resistance to both the strains of P. capsici compared to highly resistant CM334, but their response was fairly consistent for both P. capsici strains. KC00406, KC00462, KC00463, and KC01322 produced stable, male fertile $F_1$ plants indicating that they are restorers with genotype N(S)RfRf. KC00821 produced male sterile $F_1$ plants and was identified as a maintainer with genotype Nrfrf. The $F_1$ plants of the KC00820 cross, however, set a few male fertile flowers in the greenhouse at seedling stage, then became male sterile after being transplanted to the plastic greenhouse soil in May and remained so to the end of the growing season. Therefore, KC00820 is an unstable maintainer with genotype Nrfrf. The moderate resistance exhibited by these genetic resources may be integrated into breeding programs aimed at promoting higher levels resistance via recurrent selection or hybridization.

Development of Hybrid Rice Facilitated by Cytoplasmic Genetic Male Sterility II. Responses of Korean Cultivars to the Chinese Cytoplasmic Genetic Male Sterile lines (웅성불임성을 이용한 수도잡종품종개발 II. 중공의 세포형질 유전적 웅성불임계통에 대한 몇가지 한국품종의 반응)

  • Chinese CGMS (Cytoplasmic-Genetic Male Sterility) is being transfered to Korean cultivars which will be used as CGMS maintainers. These maintainer lines along with the Chinese CGMS lines and their maintainer lines were crossed with another group of Korean cultivars which will be used as the fertility restorers, and their yield heterosis were examined. The yield heterosis was calculated as high as 113.0-221.1 % in terms of heterosis (F$_1$/Midparent) with the grand mean of 150.5%, and, 86.1-179.8% in terms of heterobeltiosis (F$_1$/better parent) with the grand mean of 125.3%. Among the female parent groups, Chinese mainteiners were the highest following by Chinese MS lines and the Korean maintainers were the lowest. Heterobeltiosis of yield components were 92.7%, 120.0%, 103.0% and 112.0% for number of panicles/unit area, number of grains/panicle, fertility per cent and 1000 grain weight, respectively. Path-coefficients of yield components to the yield were calculated. The highest one was number of grains per panicle ranging 0.8073-0.8649 followed by the 1000 grain weight ranging 0.2000-0.5032. The grain yield ratios to the grand mean were calculated in terms of combining ability. It was highest with the Chinese maintainers followed by Chinese MS lines and The Korean maintainers were lowest. This facts bring cautions to the breeding efforts to transfer the Chinese CGMS in to Korean maintainers.

Development of a SCAR Marker Linked to Male Fertility Traits in 'Jinkyool' (Citrus sunki) ('진귤' (Citrus sunki) 의 웅성가임 연관 SCAR 마커 개발)

  • In Citrus, an $F_1$ segregation population of 150 plants was constructed from a cross between 'Kiyomi' (C. unshiu ${\times}$ C. sinensis) carrying the male sterility trait and 'Jinkyool' (C. sunki). Sequence-related amplification polymorphism (SRAP) combined with bulked segregant analysis was used to develop markers linked to male fertility. In the $F_1$ population, 66 out of 150 seedlings had aborted anthers and the ratio of male sterile plants to fertile plants in the progenies matched the expected Mendelian segregation ratio of 1:1 ($x^2$ =2.16 at p=0.05). From the profiling of the 197 SRAP primer sets, three SRAP primer sets (F4/R27, F39/R60, and F15/R37) that were closely linked to the target trait were identified and successfully converted into a sequence characterized amplified region (SCAR) marker for selection of male fertility in citrus. The SCAR marker, using the pMS 33U/pMS 1462L primer set specifically, produced a single 1.4-Kb fragment that was linked to male fertility. Our results suggested that this SCAR marker can be useful for marker-assisted selection of male sterile individuals in breeding $F_1$ progenies in Citrus.

  • This study was carried out to develop the efficient methods for the subsequent progeny of intersectional hybrid between Oriental and Asiatic lily hybrids. The pollen fertility and germination ability of 3 different allotetraploids (OAOA) after somatic chromosome doubling was ranged from 0 to 80 percent on artificial pollen germination medium. The number of BC$_1$ progeny using allotetraploid of F$_1$ OA-hybrid as male and female parent was different. The efficiency of BC$_1$ progeny production was increased when F$_1$OA-hybrids was used as male rather than as female parent. And in back crosses of F$_1$ OA-hybrids with the Asiatic and Oriental hybrids, Asiatic hybrids showed higher efficiency on BC$_1$ progeny production. The ploidy level between 2x or 4x Asiatic hybrid and allotetraploid F$_1$ OA-hybrid was determined and showed higher progeny production in 2x-4x crosses rather than 4x-4x.

Breeding Hybrid Rice with Good Quality and High Yield I. Breeding of Cytoplasmic-Genetic Male Sterile and Restorer Lines with Backgrounds of Korean Rice Varieties (양질 다수성 일대잡종 벼 육성 연구 I. 우리나라 품종 배경의 세포질-유전자적 웅성불임 및 임성회복 계통 육성)

  • This study was conducted to breed various cytoplasmic genetic male sterile (CGMS) and restorer lines with backgrounds of Korean japonica rice varieties. The CGMS line BT-CMS was crossed with the restorer line AR-3. The fertile $F_1 was emasculated and crossed with the Korean japonica rice varieties of the early maturing Sobaegbyeo, Odaebyeo, Gwanagbyeo and Daeseongbyeo, and of the medium maturing Hwajinbyeo, Paldal, Suwon 224 and Iri 386, and of the late maturing Nagdongbyeo, Palkweng, Hwacheongbyeo and Milyang 97. Each of the three way cross $F_{1S} was segregated into fertile and sterile individuals. The sterile individuals in each cross were discarded and the fertile individuals were emasculated and backcrossed with the Korean japonica rice varieties. The same process was applied from BC$_1$F$_1$ to BC$_4$F$_1$ generation. In the ${BC_4}{F_1}of each cross, the male sterile individual was crossed with the recurrent Korean japonica variety which was maintainer of male sterility. The male sterile lines of ${BC_6}{F_1}were named as Sobaegbyeo R, Odaebyeo R, Gwanagbyeo R, etc. The fertile individuals homozygous in pollen fertility were selected from the ${BC_4}{F_2}generation and named as Sobaegbyeo R, Odaebyeo R, Gwanagbyeo R, etc. Agronomic characteristics of the CGMS, restorer lines with backgrounds of Korean japonica, and the recurrent Korean japonica rice varieties grown in the field condition were compared. Culm length of the CGMS lines tended to be shorter than that of the recurrent parent, however no significant differences in heading date, panicle length and yield component were found among the CGMS, restorer and recurrent lines.

Development of Gene-based Markers for the Allelic Selection of the Restorer-of-fertility Gene, Rfo, in Radish (Raphanus sativus)

  • Cytoplasmic male sterility (CMS) and fertility restoration have been utilized as valuable tools for $F_1$-hybrid seed production in many crops despite laborious breeding processes. Molecular markers for the selection of CMS-related genes help reduce the expenses and breeding times. A previously reported genomic region containing the Ppr-B gene, which is responsible for restoration of fertility and corresponds to the Rfo locus, was used to develop gene-based or so-called "functional" markers for allelic selection of the restorer-of-fertility gene (Rfo) in $F_1$-hybrid breeding of radish (Raphanus sativus L.) Polymorphic sequences among Rfo alleles of diverse breeding lines of radish were examined by sequencing the Ppr-B alleles. However, presence of Ppr-B homolog, designated as Ppr-D, interferes on specific PCR amplification of Ppr-B in certain breeding lines. The organization of Ppr-D, resolved by genome walking, revealed extended homology with Ppr-B even in the promoter region. Interestingly, PCR amplification of Ppr-D was repeatedly unsuccessful in certain breeding lines implying the lack of Ppr-D in these radishes. Ppr-B could only be successfully amplified for analysis through designing primers based on the sequences unique to Ppr-B that exclude interference from Ppr-D gene. Four variants of Rfo alleles were identified from 20 breeding lines. A combination of three molecular markers was developed in order to genotype the Rfo locus based on polymorphisms among four different variants. These markers will be useful in facilitating $F_1$-hybrid cultivar development in radish.

Utilization of Male Sterility Type and Frequency of Male Sterile Genes in Carrots (당근 웅성부임성(雄性不稔性)의 형태별(形態別) 이용현황(利用現況)과 핵내(核內) 웅성부임(雄性不稔) 유전자(遺傳子)의 분포(分布))

  • When the type and ratio of male-sterile plants in $F_1$ hybrids collected from several seed companies were investigated, there were differences in the male-sterile type depending upon region, seed company and variety group. The differences were inferred due to the easiness in breeding of maintainer line among the variety groups. American seed companies mainly used petaloid type male sterility with Imperater group varieties. European companies used brown anther type male sterility mainly with Nantes group but some companies used petaloid type also in varieties that were different from Nantes group. Asian companies (Japan, Korea) used both types with Chantaney and Kuroda group varieties, but one type was mainly used depending on individual seed company. Only one type of male-sterility in one variety was observed and the results were agreed well with other's results that male-sterility type was determined by cytoplasm factor. Some breeding lines were backcrossed to both types of cytoplasm (Sa, Sp) for maintainer line selection. We could select 15 maintainer lines from 20 lines in petaloid cytoplasm (Sp) and 3 from 4 lines in brown cytoplasm (Sa). In petaloid cytoplasm, maintainer lines can be selected at considerably high frequency. But in brown anther cytoplasm, the used materials are too restricted to tell general frequency maintainer lines.

Cross Affinity of Korean Weedy Rice to the Cultivars (국내 수집 잡초성 벼의 재배 품종에 대한 잡종 친화성)

  • Weedy rice(red rice) collected in Korea were crossed with the Japonica, Javanica and Indica tester cultivars, and also crossed among those collected lines. Their seed fertility was counted and refered to their cross affinities. Some tester cultivars such as, Suweongjo, Malagkit Sinaguing and wx 817 which were regarded as belong to the Japonica, Javanica and Indica respectively, showed the typical tendencies of hybrid sterility when they were crossed each other. But, other testers such as Kwanak, Tapaiko, Belle Patna and Tarichaosun showed some differencies from those typical patterns, implying some varigations within a ecotype. The hybrids of weedy rice crossed to the Japonica testers showed higher fertility with a few semisterilities. While the crosses of weedy rice to the Javanicas or Indicas showed high sterility implying closer affinity relationships of weedy rice to the Japonicas. Even those weedy rice lines such as Samsanmyeon 8 and Sanchungaengmi B which showed semi-sterility when they were crossed to the Japonica testers showed high sterility when they were crossed to both Javanicas and Indicas. The hybrids among weedy rice lines showed high fertility raning 69% to 96%. Even those weedy rice lines which showed semi-sterility in the crosses with Japonica testers, showed high-fertility when they were crossed with those weedy rice lines. Though the amylose content, Alkali Digestive Value, Phenol reaction and grain shapes of theose weedy rice lines showed divers variations, the cross affinities of them showed closer phylogenie relationship to the Japonicas than to Japvanicas or Indicas. But the genetic influinces of Indicas to those weedy rices were not ignorable.

Cleaved Amplified Polymorphic Sequence and Amplified Fragment Length Polymorphism Markers Linked to the Fertility Restorer Gene in Chili Pepper (Capsicum annuum L.)

  • Cytoplasmic male sterility (CMS) in plants, which is due to failure to produce functional pollen, is a maternally inherited trait. Specific nuclear genes that suppress CMS, termed fertility restorer (Rf) genes, have been identified in several plants. In this study, Rfl-inked molecular markers in pepper (Capsicum annuum L.) were detected by bulked segregant analysis of eight amplified fragment length polymorphisms (AFLPs). Only AFRF8 was successfully converted to a cleaved amplified polymorphic sequence (CAPS) marker. This was named AFRF8CAPS and genotype determination using it agreed with that obtained with the original AFRF8. A linkage map with a total size of 54.1 cM was constructed with AFRF8CAPS and the seven AFLP markers using the Kosambi function. The AFRF8CAPS marker was shown to be closest to Rf with a genetic distance of 1.8 cM. These markers will be useful for fast and reliable detection of restorer lines during $F_1$ hybrid seed production and breeding programs in pepper.
