A Study on the Flora and Community Classification of Forest Vegetation in the Mt. Yumyeong (유명산의 식물상 및 산림군락분류에 관한 연구)

  • The study was carried out to investigate the flora and community classfication of forest vegetation in Mt. Yumyeong from March. 2002 to October. 2003. As a result of that. the vascular plants were 98 families, 331 genera, 570 species, 1 subspecies, 81 varieties, 4 formae, 2 hybrid, 658 taxa in all. Korean endemic plants were 10 families, 14 genera, 13 species, 2 variaties, 1 hybrid, 16 taxa in all; rare and endangered plants were 9 families, 10 genera, 11 species, 11 taxa in all: naturalized plants were 12 families, 22 genera, 28 species, 28 taxa in all. The forest vegetation was classified into 1 order, 1 alliance, 6 communities; Rhododendro - Quercetalia mongolicae (Kim, 1990) Lindero - Quercion mongolicae (Kim, 1990) A. Carex humiljs var. nana - Pinus densiflora Community B. Oplismenus undulatifoljus - Pinus koraiensis Community C. Fraxinius rhynchophylla - Quercus mongolica Community D. Cornus controversa - Quercus mongolica Community E. Quercus variabilis - Quercus mongolica Commnuty F. Lespedeza bicolor - Quercus mongolica Community.

In vitro Regeneration and Genetic Stability Analysis of the Regenerated Green Plants in Japanese Blood Grass (Imperata cylindrica 'Rubra') (홍띠 기내 재생과 재생 녹색식물체의 유전적 안정성)

  • The in vitro regeneration was established, and the genetic stability among the mother plants (control) and the micropropagated green plants was evaluated using ISSR markers in Imperata cylindrica 'Rubra', Poaceae which containing important bioenergy plants. Green shoots were multiply induced from growing point culture via callus on MS medium supplemented with 0.01 mg/L NAA and 2 mg/L BA, and the shoots were proliferated on the MS medium with rooting. Rooted plantlets were transplanted to the pot with 100% survival rate. Using ISSR markers, somaclonal variation was analyzed in eight mother plants (control), ten green-regenerant cultivated at culture room (ReR) and ten green-regenerant cultivated at field condition (ReF). All ISSRs produced a total of 97 bands, and the scorable bands varied from one to seven with an average of 4.4 bands per primer. The polymorphism rate of ReRs and ReFs was 4.1% and 3.1% respectively, showing higher rate than that of control (0%). The genetic similarity matrix (GSM) among all accessions ranged from 0.919 to 1.0 with a mean of 0.972. According to the clustering analysis, ReFs and mother plants were divided into two independent groups. The results indicate that no clear genetic diversity was detected among regenerated plants, and ISSR markers were useful tool for identification of somaclonal variation of regenerants.

Studies on Tissue Culute in Rice, Oryza sativa L. (벼 조유배양에 관한 연구)

  • In order to provide basic information for the application of tissue culture to a rice breeding program. experiments were carried out on the callus formation from seedings and anthers and organ differentiation from the callus of rice varieties and their hybrids. It appeared that in general the callus tissue was more easily induced in japonicas than in indicas with significant varietal differences. A highly significant positive correlation (r=0.896$^{**}$) was obtained between the fresh weight of callus induced in NAA medium and in 2, 4-D medium in the same variety. Callus formed on the medium with 2, 4-D was more friable than that formed on the medium with NAA. The callus formation from seedlings was better than from anthers in upland rice varieties. The easiness of the callus formation appeared to be a dominant character in crosses of rice varieties. Varietal difference was also noticed in the organ differentiation from the callus and the root differentiation seemed to be easier than the shoot differentiation. Most of the plants derived from the cal1us were albinos and the frequency of occurrence of albinos was greater in callus of seedlings than in that of anthers. Greater amount of callus tissue was formed from anthers of $F_1$ plants than anyone of their parents in remote-crosses but it was clear in close-crosses. A highly significant positive correlation (r=0.504$^{**}$) was found between the callus formation from seedlings and from anthers.

  • A leading sweet pepper cultivar, Keystone Resistant Giant #3, was crossed with a line with resistance to Xanthomonas campestris pv. vesicatoria, PI271322, for incorporation of the resistance and to study the inheritance of resistance to the disease. Seedlings of parents, $F_1$, $F_2$ and backcross populations of a cross between Keystone Resistant Giant #3 and PI271322 were inoculated with race 1 and race 3 of Xanthamonas campestris pv. vesicatoria by infiltrating bacterial suspension into abaxial side of leaves. PI271322 was carrying a gene ($Bs_3$) for hypersensitive resistance to race 1 of X. c. pv. vesicatoria and also a component of non-hypersensitive resistance to race 3 inheriting in a quantitative mode. Correlation coefficient between disease indices to race 1 and race 3 of non-hypersensitive plants in $F_2$ population was highly significant, thus indicating that a component of non-hypersensitive general resistance in PI271322 acted consistently to both races. Individual plants with resistance to both race 1 and race 3 of X. c. pv. vesicatoria were selected and a breeding program for incorporation of the resistance is continued.

Biological Control of Sesame Soil-born Disease by Antifungal Microorganisms (참깨 토양전염성병(土壤傳染性病)의 생물학적방제(生物學的防除))

  • In order to study the biological control of soil-borne disease of sesame, antagonistic isolates of Trichoderma , Bacillus sand streptomyces to Fusarium oxysporum and Rhizoctonia solani were isolated from the rhizosphere soils of sesame plants and some other habitats. Out of the isolates of microorganisms collected a strain of Trichoderma viride was selected as a biological control agent for the study and its effect on the control of damping-off and the seedling growth of sesame was investigated. The results obtained are as follows: 26 percents of Bacillus spp. isolated from the rhizosphere soil of sesame plants showed antagonism to two pathogenic fungi. Important species were B. Subtilis and B. polymyxa. Streptomyces species isolated from the rhizosphere soils of sesame lysed the cell wall of hyphae and conidia of F. oxysporum and reduced conspicuously the formation of macroconidia and chlamydospores of the fungus. 84 percents of Trichoderma spp. isolated from the rhizosphere soil of sesame plants were antagonistic to F. oxysporum and 60 percents of the isolates were antagonistic to both F. oxysporum and R. solani. Trichoderma viride TV-192 selected from antagonistic isolates of Trichoderma spp. was highly antagonistic to F. oxysporum and soil treatment with the isolate reduced notably damping-off of sesame. T. viride TV-192 showed better growth in crushed rice straw, barley straw and sawdust media than F. oxysporum. Sawdust was selective for the growth of T. viride. Supplementation of wheat bran and mixtures of wheat bran and sawdust inoculated with T. viride TV-192 in the soil reduced remarkably damping-off of sesame by F. oxysporum but high density of the fungus TV-192 caused the inhibition of seed germination and seedling growth of sesame. Inhibitory effects of Trichoderma species on seed germination and seedling growth of sesame were different according to the isolates of the fungus. Normal sesame seedlings on the bed treated with the fungus showed better growth than not treated seedlings.

  • Five $F_1$ hybrids of radish(Raphanus sativus L.) were used in the study for induction of microspore derived embryos. Anthers from the mid-uninucleate to early bicellular stage were inoculated on the modified B5 medium and modified Nitch-Nitch medium supplemented with several growth regulators. The efficiency of anther culture was dspendent on the genotype of donor plants and we obtained various culture efficiency from different genotypes. Induction of embryos from microspore was best result on Nitsch-Nitsch media supplemented with 0.1mg/l NAA and 0.05mg/l BAP. Heat treatments of anthers at $35^{\circ}C$-2days and combined with pretreatment of $4^{\circ}C$ for 2, 8, 12 and 16days . Among the treatments, $35^{\circ}C$-2 days treatment combined with $4^{\circ}C$-2days pretreatment treatment were the most effective in developing embryos from microspores.

Studies on QTLs for Bakanae Disease Resistance with Populations Derived from Crosses between Korean japonica Rice Varieties

  • Rice bakanae disease is a serious global threat in major rice-cultivating regions worldwide causing high yield loss. It is caused by the fungal pathogen Fusarium fujikuroi. Varying degree of resistance or susceptibility to bakanae disease had been reported among Korean japonica rice varieties. We developed a modified in vitro bakanae disease bioassay method and tested 31 Korean japonica rice varieties. Nampyeong and Samgwang varieties showed highest resistance while 14 varieties including Junam and Hopum were highly susceptible with 100% mortality rate. We carried out mapping QTLs for bakanae disease resistance with four F2:F3 populations derived from the crosses between Korean japonica rice varieties. The Kompetitive Allele-Specific PCR (KASP) markers developed in our laboratory based on the SNPs detected in Korean japonica rice varieties were used in genotyping F2 plants in the populations. We found four major QTLs on chromosome 1, 4, 6, and 9 with LOD scores of 21.4, 6.9, 6.0, and 60.3, respectively. In addition, we are doing map-based cloning of the QTLs on chromosome 1 and 9 which were found with Junam/Nampyeong F2:F3 population and Junam/Samgwang F2:F3 population, respectively. These QTLs will be very useful in developing bakanae disease resistant high quality rice varieties.

  • Late blight, one of the most important disease in many agricultural crops, is caused by Phytophthora infestans. Fusarium wilt is a vascular disease of many plants caused by Fusarium oxysporum. Some bacteria isolated from rhizosphere were screened for their ability to inhibit the growth of F. oxysporum and P. infestans. Productions of siderophore, $\beta-1$,3-glucanase, hydrogen cyanide and chitinase by 4 isolated strains were examined. Among them, Bacillus sp. RFO41 most effectively inhibited the growth of F. oxysporum. The highest productions of siderophore and $\beta-l$,3-glucanase were shown in the culture of Bacillus sp. RFO41. Bacillus strain PS2 was most effective against P. infestans. PS2 showed the highest production of chitinase and hydrogen cyanide. A significant relationship was shown between the antagonistic effects of isolates against F. oxysporum and P. infestans and their production level of siderophore, $\beta-1$,3-glucanase, hydrogen cyanide, and chitinase.

Putative Histone H2A Genes from a Red Alga, Griffithsia japonica

  • Histones are important proteins that interact with the DNA double helix to form nucleosome. Two putative histone genes, GjH2A-1 and GjH2A-2 were isolated from a red alga Griffithsia japonica. The putative open reading frame of GjH2A-1 and GjH2A-2 shared high similarity with the previously reported amino acid sequences of histone H2As. They have a motif consisting of seven amino acids A-G-L-Q-F-P-V, which matches the histone H2A motif [AC]-G-L-x-F-P-V. Phylogenetic trees were constructed from amino acid sequences of 38 histone H2As. The histone H2As were divided into two groups: major H2As and H2A.F/Z variants. The major histone H2A group consisted of animals, fungi, plants + green algae, and red algae H2A subgroups. The animal histone H2A subgroup was divided into vertebrates, echinoderms, nematodes, insects, and segmented worms H2As. The putative red algal histone genes, GjH2A-1 and GjH2A-2, constituted an independent lineage. This is the first report on red algal histone genes.

STRAIN RATE CHANGE FROM 0.04 TO 0.004%/S IN AN ENVIRONMENTAL FATIGUE TEST OF CF8M CAST STAINLESS STEEL

  • To define the effect of strain rate variation from 0.04% to 0.004%/s on environmental fatigue of CF8M cast stainless steel, which is used as a primary piping material in nuclear power plants, low-cycle fatigue tests were conducted at operating pressure and temperature condition of a pressurized water reactor, 15 MPa and $315^{\circ}C$, respectively. A high-pressure and high-temperature autoclave and cylindrical solid fatigue specimens were used for the strain-controlled low-cycle environmental fatigue tests. It was observed that the fatigue life of CF8M stainless steel is shortened as the strain rate decreases. Due to the effect of test temperature, the fatigue data of NUREG-6909 appears a slightly shorter than that obtained by KEPRI at the same stress amplitude of $1{\times}10^3$ MPa. The environmental fatigue correction factor $F_{en}$'s calculated with inputs of the test data increases with high strain amplitude, while the $F_{en}$'s of NUREG-6909 remain constant regardless of strain amplitude.
