Soil Microbial Community Analysis using Soil Enzyme Activities in Red Pepper Field Treated Microbial Agents (토양효소활성을 이용한 미생물제제 처리 고추경작지의 토양미생물군집 분석)

  • Increasing concerns over green farming technology, plant growth promoting rhizobacterium (PGRP) having growth promoting as well as plant disease suppressing properties was recently preferred to use for biological control of plant pathogens infecting plant. We measured the influence of the selected microbial consortium agents-a mixture of PGPR strains-, commercial bio-fungicide, and chemical pesticides on soil microbial community in red pepper field. The activities of soil enzyme such as dehydrogenase, urease, phosphatase, ${\beta}$-glucosidase, and cellulase were analyzed to investigate that of soil microbial community. We also measured plant length, main stem, stem diameter, number of branches and yields of red-pepper in order to observe the red pepper growth promotion. The results of measuring enzyme activities were dehydrogenase 3.5584 ${\mu}g$ TPF $g^{-1}h^{-1}$, urease 15.8689 ${\mu}g$ $NH_4{^-}N$ $g^{-1}h^{-1}$, phosphatase 0.5692 ${\mu}g$ PNP $g^{-1}h^{-1}$, ${\beta}$-glucosidase 2.4785 ${\mu}g$ PNP $g^{-1}h^{-1}$, and cellulase 86.1597 ${\mu}g$ glucose $g^{-1}h^{-1}$ in the soil treated with the microbial consortium agents, so it came out to be very active in the soil. Observing the growth of red-peppers, the main-stem length and the stem diameter were 6.1% and 8.1% higher in the soil treated with the selected microbial consortium agent than the chemical pesticides. After harvesting, yields were 7.3% higher in the soil treated with selected microbial consortium agents than the chemical pesticides. These results showed that microbial consortium agents contribute to increasing soil microbial diversity, growth promoting, and yield of red pepper.

  • This study was conducted to find out the effect of herbicides on soil micro-organism and soil enzyme in loam soil, and on pathogenic microorganism in continuous pepper cropping soil. The result was summarized as follows: When herbicides were treated, the number of soil microorganism generally decreased at the early stage of incubation, and gradually increased at 30 days incubation. The number of fungi was significantly decreased seven times comparison with control plot. The number of actinomycetes was increased at 20 days incubation and that of pytium was increased for all days incubation in Linuron treatment plot. Although the activity of soil enzyme tended to decrease when herbicides were treated, the activities of urease and phosphatase were higher than that of control plot at the early stage of incubation in MO, Linuron and Simetryne treatment plot. In Simetryne treatment plot, the activity of protease was increased at the early stage, and suddenly decreased after 30 days incubation. There was no significant effect of herbicides on the activities of ${\beta}$-glucosidase and polygalacturonase. The activity of cellulase was inhibited at the early stage of incubation, but that of cellulase was higher than that of control plot after 20 days incubation. High significance was showed the correlation coefficient between soil microorganisms except fungi and soil respiration as herbicides were treated, and the total microorganism and soil respiration in Linuron plot. when Dicamba and Simetryne were treated, the correlation coefficient between the total microorganism and the activities of protease and urease were considerably significant.

  • Gaucher disease (GD) is an autosomal recessive lysosomal storage disorder caused by beta-glucosidase deficiency. An 18 month-old male with hepatosplenomegaly, anemia, thrombocytopenia, and growth retardation referred to our hospital. The patient showed neurological symptoms, such as supranuclear gaze palsy and developmental delay. Bone marrow biopsy performed to rule out malignancy and the results revealed no malignant cell; however, abnormal histiocytes suggesting storage disease was noted. Based on hepatosplenomegaly, bicytopenia and unexplained neurologic manifestations, enzyme activity and genetic analysis were conducted emergently with a strong suspicion of GD. Beta-glucosidase activity in leukocyte was decreased. GBA sequencing to confirm the diagnosis revealed compound heterozygous pathogenic variants (i.e., c.754T>A, c.887G>A), both previously reported as the cause of neuronopathic GD. Under the diagnosis of type 3 GD, the patient immediately received enzyme replacement therapy (ERT). After 17 months of ERT, the size of spleen decreased, and hemoglobin and platelet count returned to normal. In addition, the activity of chitotriosidase and angiotensin converting enzyme decreased. However, myoclonic movement and generalized seizure occurred at the age of 19 months and antiepileptic drug was started. Other neurological deterioration including supranuclear gaze palsy and developmental delay also persisted. A new therapy to overcome neurologic problems should be developed for patients with type 3 GD.

Isolation and Partial Characterization of Isoflavone Transforming Lactobacillus plantarum YS712 for Potential Probiotic Use (Isoflavone 비배당화 및 항산화 활성을 지닌 Lactobacillus plantarum YS712의 선발)

  • Lactic acid bacteria (LAB) are typical probiotic microbes that are used in various industries including fermented foods, feed additives, and pharmaceuticals. The purpose of this study was to compare the ability of isoflavone biotransformation and antioxidative activity of 17 LAB. Six strains including the Lactobacillus species exhibited a 100% hydrolysis rate for daidzein during fermentation. The content of total genistein in soymilk fermented with these strains was $872-943\;{\mu}g/g$. The DPPH (1, 1-diphenyl-2-picrylhydrazyl) radical scavenging ability of the LAB was widely variable and ranged from 23-78%. A selected strain was isolated from kimchi and the strain was identified as Lactobacillus plantarum ssp. through the API carbohydrate fermentation pattern and 16S rDNA profile. The strain exhibited excellent acid tolerance in an artificial gastric solution. L. plantarum YS712 showed high $\beta$-glucosidase activity in fermentation. The concentration of genistein and daidzein in soymilk fermented with L. plantarum YS712 increased from 3.64 to $917.3\;{\mu}g/g$ and from 58.18 to $1062.17\;{\mu}g/g$, respectively. These results demonstrate the potential of L. plantarum YS712 as a probiotic culture that can be utilized in the manufacturing of fermentation foods and dietary supplements.

Isolation and Characterization of Bacillus subtilis CH-10 Secreting Cellulase from Cattle Manure (우분으로부터 Bacillus subtilis CH-10의 분리 및 균주가 분비하는 Cellulase의 특성에 관한 연구)

  • A bacterium producing the extracellular cellulase was isolated from cattle feces and screened as cellulase activity was excellent upon congo red straining method and activity measurements. Isolate was identified as Bacillus subtilis CH-10 on the basis of morphological and biochemical properties as well as cellular fatty acids composition. The enzyme which the isolate secretes had the optimum initial pH and temperature for its induction was 7.5 and 50${\circ}C$, respectively. The maximum CMCase activity in crude enzyme solution was observed at pH 7.5 and 75${\circ}C$ and was stable for pH 7.5 to 9.0 to maintain 70% activity. When the isolate was cultured in CMC media at 37${\circ}C$ for 24 hrs, CMCase and FPase activity was 1.13 U/㎖and 0.16U/㎖, respectively whereas Avicelase and ${\beta}$-glucosidase activity was not detected. When crude supernatant was used for zymogram, three major bands, cel 1, cel 2 and cel 3, were detected approximately 39, 41 and 57 KDa, respectively on CMC-SDS-PAGE.

  • Of 450 strains isolated from the soil microbes collected in various locations in Korea, a strain had a strong inhibitory activity against bacterial ${\alpha}-amylase$ and was named strain DMC-72 of the genus Streptomyces. The amylase inhibitory metabolite produced by this strain was purified by means of acetone precipitation, adsorption on Amberlite IRC-50 and SP-Sephadex C-25. The inhibitor was found to be a derivative of oligosaccharides by spectral and chemical data. The inhibitor was stable at the pH range of $1{\sim}13$ and at $100^{\circ}C$ for half an hour, also inhibited other amylases such as salivary ${\alpha}-amylase$, pancreatic ${\alpha}-amylase$, fungal ${\alpha}-amylase$ and glucoamylase. However, it showed no inhibitory activity against ${\alpha}-glucosidase$, ${\beta}-glucosidase$, dextranase, and ${\beta}-amylase$. The kinetic studies of the inhibitor showed that its inhibitory effects on starch hydrolysis by ${\alpha}-amylase$ were noncompetitive.

Evaluation of Novel Constitutive Expression Vectors Equipped with Mined Promoters from Metagenome (메타게놈에서 발굴한 프로모터를 장착한 새로운 항시발현 벡터의 가치평가)

  • The choice of expression vector is very important for industrial production of proteins. Therefore, the systematic mining of promoters over a wider range of genetic resource and/or host is required. We previously reported a novel bidirectional reporting system (pBGR) for the isolation of promoters from metagenome and screened useful promoters that functioned constitutively in E. coli under general culture conditions. Among them, three promoter sequences including each upstream region were amplified by PCR and used to construct new expression vectors. To facilitate subcloning, a multi-cloning site was incorporated into the downstream region of the revere primer sequence. At these sites, GFP, esterase and $\beta$-glucosidase were subcloned and analyzed the constitutive expression ability of new promoter in terms of protein solubility and expression level. As a result, these vectors expressed the proteins constitutively to a level of $2{\sim}3%$ of the total cell protein in soluble fraction (>80 %). This study suggested that excavation of metagenomic promoters for construction of expression vector in a certain strain could provide a way for the development of the expression systems.

The Effect of Aquatic Macrophytes on the Biogeochemistry of Wetland Sediments (습지 식물이 퇴적물의 생지화학적 반응에 미치는 영향)

  • This research investigates the influences of the presence of aquatic macrophytes on the changes of biogeochemistry in the sediments through the comparative analysis of porewater and sediments. From the in situ measurements, elevated SO$_4{^{2-}}$ concentrations were observed in the rhizosphere during the growing season, which was resulted from the oxidation of reduced sulfide in the sediments by the oxygen release from the plant roots. There was sufficient AVS in the sediments to induce observed SO$_4{^{2-}}$ concentrations. The amount of oxygen in the oxidation of AVS to produce observed SO$_4{^{2-}}$ concentrations is 0.85 g/m$^2$ day which is relevant to the results of other researches. The AVS concentrations in the vegetated sediments increased with the depth whereas there is higher mass of AVS in the surface of the non-vegetated sediments. This shows that evapotranspiration induces the transportation of SO$_4{^{2-}}$ in the surface water into the anaerobic sediments. In addition, the elevated organic content caused by the presence of plants increased $\beta$-glucosidase activities which play an important role in the carbon cycle of the sediments.

Mycelial Growth Properties of Domestically Collected Ectomycorrhizal Tricholoma Mushrooms in Various Culture Conditions (다양한 배양 환경에 따른 국내 수집 외생균근성 Tricholoma속 종의 균사생장 특성)

  • The ectomycorrhizal basidiomycete Tricholoma is one of mushroom groups that cannot be cultivated artificially. To use this mushroom as applicable resource for food production, it is necessary to obtain information about their mycelial growth properties in various environmental conditions. This study investigated the mycelial growth of four domestic isolates of Tricholoma species (T. bakamatsutake, T. fulvocastaneum, T. matsutake, T. terreum) at different physical and chemical conditions. The optimal physical conditions for their mycelia growth were found to be a temperature range of $20{\sim}25^{\circ}C$ and a pH range of 4.0~7.0 in dark condition. The growth of T. matsutake was retarded at high temperature ($30^{\circ}C$). Tests to determine the chemical factors that affected mycelial growth showed that the four Tricholoma spp. grew 1% saline. T. matsutake grew in up to 2% saline. In the presence of various heavy metals (50 ppm) and pesticides (suppliers' recommended concentration), mycelial growth was inhibited the most by cadmium and emamectin benzoate, respectively. However, all the four Tricholoma spp. grew with $Cu^+$. The growth of T. matsutake was not inhibited by abamectin, acetamiprid, and thiacloprid. Extracellular enzyme activities of amylase and ${\beta}$-glucosidase were detected only in T. bakamatsutake and T. fulvocastaneum. The results of the present study allowed us to determine suitable or harmful environmental conditions for the mycelial cultivation of the Tricholoma spp.

Influences of Rice and Barley straw Application in the Rice Rhizosphere (수도근권(水稻根圈) 환경(環境)에 미치는 볏짚과 보릿집 시용(施用)의 영향(影響))

  • This study has been made to investigate the influences of organic matter on the soil composition, nitrogen fixing organism, soil enzyme activity and nitrogen fixing activity in the paddy rice rhizosphere when rice and barley straw were applied. The results are summarized as follows: 1. The pH in the submerged soil was increased from ear formation stage to harvesting. 2. In the rhizosphere, $Fe^{{+}{+}}$ content was decreased according to the growing stage, while increased in the nonrhizosphere. 3. In the initial stage, rhizosphere was higher than nonrhizosphere but in the late stage nonrhizosphere was higher than rhizosphere on the $NH_4-N$ content. 4. In the submerged soil, rhizosphere was higher than nonrhizosphere, on the concentration of glucose and pentose. 5. Changes of the number of nitrogen fixing organism in whole soil was not high. 6. Generally, rhizosphere was higher than nonrhizosphere on the soil enzyme activity such as phosphatase, ${\beta}$-glucosidase, and protease. 7. Acetylene-reducing activity was the highest in the tillering stage, and rhizosphere, Samgang (high-yielding variety) were higher than nonrhizosphere. Dongjin (general variety) respectively. 8. In the submerged soil applied barley straw, acetylene-reducing activity was slightly higher than rice straw in the initial stage.
