Distinctive pH Dependence and Substrate Specificity of Peptide Hydrolysis by Human Stromelysin-1 (Stromelysin-1에 의한 펩타이드 가수분해에서 pH와 기질특이성 연구)

  • A kinetic profile of the catalytic domain of stromelysin-1 (SCD) using the fluorescent peptide substrate has been determined by the stopped-flow technique. The pH profile has a pH optimum of about 5.5 with an extended shoulder above pH 7. Three pKa values, 5.0, 5.7, and 9.8 are found for the free enzyme state and two pH independent Kcat/Km values of 4.1$\times$104 M-1 s-1 and 1.4$\times$104 M-1 s-1 at low and high pH, respectively. The profile is quite different in shape with other MMP family which has been reported, having broad pH optimum with two pKa values. The substrate specificity of SCD towards fluorescent heptapeptide substrates has been also examined by thin layer chromatography. The cleavage sites of the substrates have been identified using reverse-phase HPLC method.SCD cleaves Dns-PLA↓L↓WAR and Dns-PLA↓L↓FAR at two positions. However, the Dns-PLA↓LRAR, Dns-PLE↓LFAR, adn Dns-PLSar↓LFAR are cleaved exclusively at one bond. The double cleavages of Dns-PLALWAR and Dns-PLALFAR by SCD are in marked contrast to the close structurally related matrilysin. A notable feature of SCD catalysis agrees with the structural data that the S1' pocket of SCD is deeper than that of matriysin. The differences observed between SCD and matrilysin may form the basis of understanding the structural relationships and substrate specificities of the MMP family in vivo.

Potential barrier height of Metal/SiC(4H) Schottky diode (Metal/SiC(4H) 쇼트키 다이오드의 포텐셜 장벽 높이)

  • We have fabricated Sb/SiC(4H) Schottky barrier diode (SBD) of which characteristics compared with that of Ti/SiC(4H) SBD. The donor concentration of the n-type SiC(4H) obtained by capacitance-voltage (C-V) measurement was about $2.5{\times}10 ^{17}{\textrm}cm^{-3}$. The ideality factors of 1.31 was obtained from the slope of forward current-voltage (I-V) characteristics of Sb/SiC(4H) SBD at low current density. The breakdown field of Sb/SiC(4H) SBD under the reverse bias voltage was about $4.4{\times}10^2V$/cm. The built-in potential and the Schottky barrier height (SBH) of Sb/SiC(4H) SBD were 1.70V and 1.82V, respectively, which were determined by the analysis of C-V characteristics. The Sb/SiC(4H) SBH of 1.82V was higher than Ti/SiC(4H) SBH of 0.91V. However, the current density and reverse breakdown field of Sb/SiC(4H) were low as compared with those of Ti/SiC(4H). The Sb/SiC(4H), as well as the Ti/SiC(4H), can be utilized as the Shottky barrier contact for the high-power electronic device.

Effect of temperature on pharmacokinetics of nalidixic acid and piromidic acid in black rockfish Sebastes schlegeli following oral administration (조피볼락, Sebastes schlegeli에 경구투여된 nalidixic acid 및 piromidic acid의 약물동태에 미치는 수온의 영향)

  • Effects of temperature ($13{\pm}1.5^{\circ}C$, $23{\pm}1.5^{\circ}C$) on the pharmacokinetic properties of nalidixic acid (NA) and piromidic acid (PA) were studied after oral administration to cultured black rockfish, Sebastes schlegeli. Serum concentrations of NA and PA were determined using HPLC-UV detector after a single dosage of 60 mg/kg body weight. At $23{\pm}1.5^{\circ}C$, the peak serum concentrations of NA and PA, which attained at 24 h post-dose, were 5.87 and $0.43\;{\mu}g/ml$, respectively. At $13{\pm}1.5^{\circ}C$, the peak serum concentrations of NA and PA, which attained at 10 h post-dose, were 6.22 and $1.57\;{\mu}g/ml$, respectively. Better absorption of PA was noted at $13{\pm}1.5^{\circ}C$ compared to $23{\pm}1.5^{\circ}C$. However, absorption of NA was not affected significantly by temperature. The elimination of NA and PA from serum of black rockfish was considerably faster at $23{\pm}1.5^{\circ}C$ than at $13{\pm}1.5^{\circ}C$. The kinetic profile of absorption, distribution and elimination of NA and PA in serum were analyzed by fitting to a one compartment model, with WinNonlin program. The AUC, $T_{1/2}$, $T_{max}$ and $C_{max}$, respectively, were: $161.25\;{\mu}g{\cdot}h/ml$, 0.15 h, 12.29 h and $8.91\;{\mu}g/ml$ at $23{\pm}1.5^{\circ}C$, and $134.12{\mu}g{\cdot}h/ml$, 0.18 h, 8.79 h and $5.00\;{\mu}g/ml$ at $13{\pm}1.5^{\circ}C$ with NA; $41.57\;{\mu}g{\cdot}h/ml$, 0.58 h, 8.24 h and $0.21\;{\mu}g/ml$ at $23{\pm}1.5^{\circ}C$, and $40.36\;{\mu}g{\cdot}h/ml$, 0.59 h, 5.04 h and $1.20\;{\mu}g/ml$ at $13{\pm}1.5^{\circ}C$ with PA.

Histone H3K27 Modifications and Gene Transcription (히스톤 H3K27 변형과 유전자 전사)

  • Lysine residues of histone H3 and H4 are covalently modified in the chromatin of eukaryotic cells. Lysine 27 in histone H3 was acetylated (H3K27ac) or methylated at three levels; mono-, di-, and trimethylation (H3K27me1, H3K27me2, and H3K27me3). These modifications at H3K27 were related with gene transcription and/or chromatin structure in distinct patterns. Generally, H3K27ac and H3K27me1 were enriched in active chromatin, such as the locus control region or transcriptionally active genes, while transcriptionally inactive genes were highly marked by H3K27me2 and H3K27me3. These modifications appear to have been catalyzed by distinct histone-modifying enzymes. Recent studies suggest that the four kinds of modifications at H3K27 have inter-correlation in gene transcription or chromatin structure formation.

  • An H-magic labeling in a H-decomposable graph G is a bijection $f:V(G){\cup}E(G){\rightarrow}\{1,2,{\cdots},p+q\}$ such that for every copy H in the decomposition, $\sum{_{{\upsilon}{\in}V(H)}}\;f(v)+\sum{_{e{\in}E(H)}}\;f(e)$ is constant. f is said to be H-V -super magic if f(V(G))={1,2,...,p}. In this paper, we prove that complete bipartite graphs $K_{n,n}$ are H-V -super magic decomposable where $$H{\sim_=}K_{1,n}$$ with $n{\geq}1$.

Relationship of Nurses' Knowledge, Attitude and Practice in an Influenza A (H1N1) Base-Zone Hospital (일개 도시 지역거점병원 간호사의 신종인플루엔자에 대한 지식, 태도 및 수행도와의 관계)

  • Purpose: The purpose of this study was to identify the relationship of influenza A (H1N1) knowledge, attitude and practice for nurses. Methods: Data were collected by self-report questionnaires from a total of 325 nurses working in an Influenza A (H1N1) base-zone hospital in C city during September, 2009. The collected data were analyzed using of SPSS/WIN 17.0. Results: The knowledge of influenza A (H1N1) was not statistically different for gender, age, education, work unit, clinical experience, position, or previous education of Influenza A (H1N1). The attitude to influenza A (H1N1) was statistically significant according to age or clinical experience. Practice related to influenza A (H1N1) was statistically different for education, clinical experience or previous education of influenza A (H1N1). Knowledge of influenza A (H1N1) was lowest for etiology and definition compared to other subcategories. Attitude and practice were significantly different for all items. The biggest difference in items was for 'use of physical barriers (protective goggles, face masks and gowns) during procedures that may involve contact with aerosol'. There was a positive association between attitude and practice. Conclusion: An educational program focusing on strategy to change nurses's knowledge, attitude and practice can be effective for infection control in an influenza A (H1N1) base-zone hospital.

Excessive Water Tolerance Test by Waterlogging Treatments on the Indian Corn Accessions (침수처리를 통한 인도 수집 옥수수의 내습성 검정)

  • This research was carried out to excessive water tolerance test among corn accessions collected from India to breed corn cultivars targeting India market. The corn accessions were 20 Inbred lines and cultivars from India as well as Korean cultivars Gwangpyungok and Chaloksusu. Excessive water tolerance test was done in the green house by immerging the pots containing corn seedlings for two weeks. Then, the plant heights were measured to compare the control plants that were not grown in the immerging state. The results showed that seven accessions of high tolerance in flooding; H2(92.9%), H18(88.8%), CN114A(98.1%), CN351A(94.3%), Super900M(95.3%), P3394(98.8%), 31N27(96.7%) in which the percent is comparison to the control plants. Whereas nine accessions showed high damage by immerging; H1(78.9%), H8(73.4%), H10(77.1%), H19(79.0%), H26(74.1%), H31(75.7%), H34(77.5%), H36(77.4%), H40(74.6%). However, the reduction on the contents of chlorophyll a, chlorophyll b, carotenoids in leaf revealed contrast results to the flooding tolerance. Particularly, H36($7.249{\mu}g{\cdot}mg^{-1}$) and H40($7.642{\mu}g{\cdot}mg^{-1}$) showed rapid reduction in the chlorophyll a content during the flooding treatment. Whereas two Indian commercial varieties 37N27($0.630{\mu}g{\cdot}mg^{-1}$) and P3394($1.208{\mu}g{\cdot}mg^{-1}$) showed slight reduction. The reduction of chlorophyll a and carotenoid contents was positively correlated during the excessive water stress.

Structural stability of CD1 domain of human mitotic checkpoint serine/threonine-protein kinase, Bub1

  • Bub1 is one of the spindle checkpoint proteins and plays a role in recruitment of the related proteins to kinetochore. Here, we studied the structural characteristic of the evolutionarily conserved 160 amino acid region in the N-terminus (hBub1 CD1), using Circular Dichroism (CD) and NMR. Our CD results showed that hBub1 CD1 is a highly helical protein and its structure was affected by pH: as pH was elevated to basic pH, the helical propensity increased. This could be related to the surface charge of the hBub1 CD1. However, the structural change did not largely depend on the salt concentration, though the thermal stability a little increased. The previous NMR analysis revealed that the hBub1 CD1 adopts eight helices, which is consistent with the CD result. Our result would be helpful for evaluating the molecular mechanism of the hBub1 CD1 and protein-protein interactions.
