The n-butyllithium-initiated ring-opening polymerization of ethylene oxide, in a mixture of benzene and dimethylsulfoxide (DMSO), between $25-45^{\circ}C$, with potassium tert-butoxide, is a useful and powerful method to control the molecular weight as well as achieve a quantitative chain-end functionalization yield of the resulting polymeric alkoxide via a one pot synthesis. The molecular weight of the product could be controlled by adjusting the ratio of grams of monomer to moles of initiators, such as n-butyllithium ([n-BuLi]) and potassium t-butoxide ([t-BuOK]). The yields for the macromonomer and ${\omega}-brominated$ poly(ethylene oxide) (PEO) were quantitative in relation to the chain-end functionalizations of the polymeric alkoxide formed. The resulting products were characterized by a combination of $^1H-NMR$ spectroscopic and size exclusion chromatographic analyses.
본 연구에서는 순차적 음이온 및 개환중합반응을 통해 ABC 형태의 폴리스티렌-폴리에틸렌옥사이드-폴리락티드 블록공중합체를 성공적으로 합성하였다. 우선, 첫번째 블록인 폴리스티렌을 합성하기 위해서, 사이클로헥산 용매에서 스티렌/이차-부틸리튬을 각각 단량체/개시제로 사용하여 음이온중합(anionic polymerization)을 수행하였고, 이후 고분자 말단을 수산기로 전환시키기 위해, 에틸렌옥사이드를 첨가하였다. 다음 단계로 포타슘 나프탈레나이드(potassium naphthalenide)를 이용하여 폴리스티렌 말단 수산기의 수소를 제거하여, 거대개시제인 PS-$O^-K^+$를 제조하였다. 준비된 거대 개시제에 정량된 에틸렌옥사이드를 첨가하여, 음이온중합을 수행하였다. 폴리락티드 블록을 도입시키는 개환중합의 경우, THF 용매에서 트리에틸알루미늄(triethylaluminum)/피리딘(pyridine)시스템을 이용하여 PS-b-PEO-$AlEt_2$ 형태의 거대개시제를 형성한 후, $90^{\circ}C$에서 중합을 수행하였다. 합성된 블록공중합체를 수소핵자기공명법 및 겔침투크로마토그래피 방법을 통해 조사한 결과, 잘 정의된 분자량 및 낮은 분자량 분포를 나타냄을 확인할 수 있었다.
Diblock copolymers with different poly($\varepsilon$-caprolactone) (PCL) block lengths were synthesized by ringopening polymerization of $\varepsilon$-caprolactone in the presence of monomethoxy poly(ethylene glycol) (mPEG-OH, MW 2000) as initiator. The self-aggregation behaviors of the diblock copolymer nanoparticle, prepared by the diafiltration method, were investigated by using $^1H$ NMR, dynamic light scattering (DLS), and fluorescence spectroscopy. The PEG-PCL block copolymers formed the nano-sized self-aggregate in an aqueous environment by intrsa- and/or intermolecular association between hydrophobic PCL chains. The critical aggregation concentrations (cac) of the block copolymer self-aggregate became lower with increasing hydrophobic PCL block length. On the other hand, reverse trends of mean hydrodynamic diameters were measured by DLS owing to the increasing bulkiness of the hydrophobic chains and hydrophobic interaction between the PCL microdomains. The hydrodynamic diameters of the block copolymer nanoparticles, measured by DLS, were in the range of 65-270 nm. Furthermore, the size of the nanoparticles was scarcely affected by the concentration of the block copolymers in the range of 0.125-5 mg/mL owing to the negligible interparticular aggregation between the self-aggregated nanoparticles. Considered with the fairly low cac and nanoparticle stability, the PEG-PCL nanoparticles can be considered a potential candidate for biomedical applications such as drug carrier or imaging agent.
Menadione-ghitathione conjugate (MEN-SG), a metabolite of menadione, is known to be a redoxcycler in rat hepatocyte subcellular fraction. Therefore, it was assumed that MEN-SG could exert cytotoxlclty to ral platelets, another target tissue of menadione. We first synthesized MEN-SG, the identity of which was verified by mass, $^1{H}$-NMR and UV-visible spectra. In addition, the stability of MEN-SG was investigated in biological assay system. MEN-SG was degraded in a time-dependent manner in DMSO which had been used as a vehicle and thus, tris-HCl buffer was used as a vehicle of MEN-SG despite the low solubility in it. Perchloric acid as well as platelets itself did not affect the stability of MEN-SG. Our next attempt was the evaluation of cytotoxicity of MEN-SG in rat platelets. MEN-SG did not induce cytotoxicity to rat platelets measured by two different methods, LDH release and turbidity changes. The extents of oxygen consumption by MEN-SG in intact platelets were significantly lower than those by menadione, though it had been observed that oxygen consumptions by menadione and MENSG were similar in subcellular fractioas of platelets. These results suggest that MEN-SG is not toxic to rat platelets despite its redox cycling capacity and glutathione conjugation reaction of menadione could be regarded as a detoxification process.
PEO 첨가에 따른 PLA/PBAT 블렌드 필름의 물성개선 효과를 확인하였다. PEO 농도에 따른 영향을 확인하기 위하여 기계적특성, 열적특성 및 화학적 특성을 조사하였다. PEO 농도가 증가함에 따라 PLA/PBAT/PEO 블렌드 필름의 인장강도는 소폭 감소하지만 열안정성은 증가하였다. PEO 2% 첨가군은 다른 샘플들에 비해 인장강도 감소폭 대비 높은 파단연신율(46.8%) 개선 효과를 보였다. 반면, PEO 4% 수준에서 PEO 무첨가와 유사한 파단연신율과 인장강도의 저하가 발생하였다. 열특성 분석을 통해 PLA/PBAT 블렌드 내 낮은 혼화성으로 발생한 두개의 Tm 피크가 PEO 첨가로 상용성이 증가하여 피크가 하나로 합쳐지는 것을 확인하였다. 또한, 1H-NMR 분석을 통해 블렌드 내의 PEO의 농도가 증가함을 확인하였다. 향후, PEO 농도가 증가함에 따라 감소된 인장강도 유지와 파단연신율을 확보하기 위한 PEO의 분산성 향상에 대한 추가적인 연구가 필요하다.

Methanol extract of the rhizomes of turmeric, Curcuma longa L., effectively controlled the development of red pepper anthracnose caused by Colletotrichum coccodes. In addition three antifungal substances were identified from the methanol extract of C. longa rhizomes as curcumin, demethoxycurcumin, and bisdemethoxycurcumin using mass and $^{1}H-NMR$ spectral analyses. The curcuminoids in a range $0.4-100\;{\mu}g/ml$ effectively inhibited the mycelial growth of three red pepper anthracnose pathogens, C. coccodes, C. gloeosporioides, and C. acutatum. The three curcuminoids inhibited mycelial growth of C. coccodes and C. gloeosporioides to an extent similar to the synthetic fungicide dithianon did, but the synthetic agent was a little more effective against C. acutatum. The curcuminoids also effectively inhibited spore germination of C. coccodes, and bisdemethoxycurcumin was the most active. Among the three curcuminoids, only demethoxycurcumin was effective in a greenhouse test in suppressing red pepper anthracnose caused by C. coccodes.
Paljinhangahm-dan is an Oriental herbal formulation for its ability to modulate cancer cell growth and survival. This research was performed to study the anti-cancer effects of Paljinhangahm-dan water extract(PHWE) in human pro myelocytic leukemia(HL-60) cells. After HL-60 cells were routinely cultured, tetrazolium-based colorimetric(MTT) assay was performed for cytotoxicity test. To explore the mechanism of cytotoxicity. I used several measures of apoptosis to determine whether this processes was involved in PHWE-induced cell death in HL-60 cells. In addition, the experiment was practised 1 H-NMR spectroscopy to examine molecular structure of PHWE. This study suggested that PHWE control cancer cell growth through of apoptosis with less cytotoxicity in normal cells.
Skeletal muscle can be ultrastructurally damaged by eccentric exercise, and the damage causes metabolic disruption in muscle. This study aimed to determine changes in the metabolomic patterns in urine and metabolomic markers in muscle damage after eccentric exercise. Five men and 6 women aged 19~23 years performed 30 min of the bench step exercise at 70 steps per min at a determined step height of 110% of the lower leg length, and stepping frequency at 15 cycles per min. $^1H$ NMR spectral analysis was performed in urine collected from all participants before and after eccentric exercise-induced muscle damage conventionally determined using a visual analogue scale (VAS) and maximal voluntary contraction (MVC). Urinary metabolic profiles were built by multivariate analysis of principal component analysis (PCA) and orthogonal partial least square-discriminant analysis (OPLS-DA) using SIMCA-P. From the OPLS-DA, men and women were separated 2 hr after the eccentric exercise and the separated patterns were maintained or clarified until 96 hr after the eccentric exercise. Subsequently, urinary metabolic profiles showed distinct trajectory patterns between men and women. Finally, we found increased urinary metabolites (men: alanine, asparagine, citrate, creatine phosphate, ethanol, formate, glucose, glycine, histidine, and lactate; women: adenine) after the eccentric exercise. These results could contribute to understanding metabolic responses following eccentric exercise-induced muscle damage in humans.
Dried unripe fruits of Rubus coreanus Miq. were extracted with 80% aqueous MeOH and the concentrated extract was partitioned with EtOAc and $H_2O$. From the EtOAc fraction, four triterpenoids were isolated through repeated silica gel, ODS and Sephadex LH-20 column chromatographies. From the result of physico-chemical data including NMR, MS aud IR, the chemical structures of the compounds were determined as tormentic acid (1), myrianthic acid (2), hovenic acid (3) and 2${\alpha}$,3${\beta}$,19${\beta}$,23-tetrahydroxylolean-12-en-28-oic acid (4). Compounds 3 and 4 were isolated for the first time from this plant. All isolated compounds were evaluated for cytotoxic activity against human colon carcinoma cells using in vitro three-[4,5-dimethylthiazole-2-yl]-2,5-diphenyltetrazolium bromide (MTT) assay, compound 3 showed a higher cytotoxicity ($IC_{50}$ = 7.8 ${\mu}M$) than doxorubicin ($IC_{50}$= 50 ${\mu}M$).
고삼 메탄올 추출물로부터 분리한 flavonoids를 IR, NMR등의 분광학적인 방법으로Leachianone A(LA)과 Sophoraflavanone G(SFG)로 동정하였고, 카드뮴의 세포독성효과를 고삼 및 고삼추출물이 방어할 수 있는 지를 검색하기 위하여 NIH 3T3 세포에 고삼 및 LA와 SFG, LA+SFG를 처리하고 MTT assay 및 광학현미경으로 세포의 생존률을 검색하여 다음과 같은 결론은 얻었다. MTT의 흡광도는 카드뮴의 농도에 의존하여 감소 하였으며, $IC_{50}$/인 MTT50은 12.5$\mu$M이었으며 카드뮴을 $IC_{50}$/농도로 처리하고 고삼 및 LA, SFG, LA+SFG를 각 화합물의 자체 세포독성을 갖지 않는 농도로 처리한 후 MTT assay로 세포 생존률을 측정한 결과 LA, SFG, LA+SFG 처리군들은 카드뮴 처리군(MTT$_{50}$)에 비해 세포생존률이 증가되었으며 이들은 고삼 및 고삼 추출물의 농도에 의존적으로 증가되었고 각각의 단일 화합물보다 두 화합물 LA+SFG병용처리군에서 수복 효과가 더 좋은 것으로 나타났으며 광학현미경적 소견에서도 세포재생이 뚜렷하게 보였다. 이상과 같은 고삼에서 분리한 flavonoids가 카드뮴 독성에 의하여 손상된 NIH 3T3 섬유아세포의 재생효과에 영향이 있는 것으로 판단된다.다.
