$PrBa_{0.9}Sr_{0.1}Co_2O_{5+{\delta}}$ 세라믹 분리막의 제조 및 산소투과 특성 (Preparation and Oxygen Permeability of $PrBa_{0.9}Sr_{0.1}Co_2O_{5+{\delta}}$ Ceramic Membrane)

  • $PrBa_{0.9}Sr_{0.1}Co_2O_{5+{\delta}}$ 조성의 산화물을 고상반응법을 이용하여 합성하였다. 합성된 분말은 압축 성형 후 $1,250^{\circ}C$에서 소결하여 치밀한 세라믹 분리막을 제조하였다. 제조된 $PrBa_{0.9}Sr_{0.1}Co_2O_{5+{\delta}}$분리막은 XRD분석 결과 이중 페롭스카이트 (double perovskite)구조를 보였다. 밀봉재료로는 pyrex ring을 사용하여 가스누출 실험 및 산소투과 분석을 하였다. 산소투과량 분석은 $850{\sim}950^{\circ}C$범위에서 측정되었다. 산소투과실험 결과, 투과량은 온도증가에 따라 0.15에서 $0.32mL/cm^2{\cdot}min$로 증가하였다.

  • 고온균 Thermus thermophilus HJ6 유래의 N-말단 결실 Tod polymerase($\Delta$Tod polymerase)는 온도 감수성 프로모터 (lambda pR and pL)를 포함하는 pJLA503 벡터를 이용하여 대장균에서 발현하였다. N-말단 250개 아미노산이 제거된 $\Delta$Tod polymerase는 5'$\rightarrow$3' exonuclease 활성은 없어지고 DNA 중합반응의 활성은 그대로 유지되었다. $\Delta$Tod polymerase는 $MgCl_2$의 존재 하에서 매우 효율적으로 역전사 반응과 PCR 반응을 수행하였다. 또한 $\Delta$Tod polymerase는 one-step RT-PCR 반응에서 Taq polymerase 보다 높은 cDNA 증폭 효율을 나타내었다.

  • Background: Peripheral nerve injury induces up-regulation of the calcium channel alpha2delta (${\alpha}2{\delta}$) subunit and TRPM8 in the dorsal root ganglion (DRG) which might contribute to allodynia development. We investigated the expression of the ${\alpha}2{\delta}$ subunit and TRPM8 in the DRG of sympathetically maintained pain (SMP) and sympathetic independent pain (SIP) rat model. Methods: For the SMP model, the L5 and L6 spinal nerves were ligated tightly distal to the DRG. For the SIP model, the tibial and sural nerves were transected, while the common peroneal nerve was spared. After a 7 day postoperative period, tactile and cold allodynia were assessed using von Frey filaments and acetone drops, respectively. Expression of the ${\alpha}2{\delta}$ subunit and TRPM8 in the L5 and L6 DRG were subsequently examined by a Western blot. Results: There were no significant differences between the two models for the thresholds of tactile and cold allodynia. Expression of the ${\alpha}2{\delta}$ subunit in the ipsilateral DRG to the injury was increased as determined on a Western blot as compared to that in the contralateral or sham-operated DRG of the SMP model, but there was no difference in expression seen with the use of the SIP model. There was no difference in the expression of TRPM8 in the ipsilateral DRG to the injury and the contralateral or sham-operated DRG of either model. Conclusions: Up-regulation of the ${\alpha}2{\delta}$ subunit in injured DRG may play a role that contributes to tactile allodynia development in SMP, but not TRPM8 to cold allodynia after peripheral nerve injury.

  • 다공성 $La_{0.6}Sr_{0.4}Ti_{0.3}Fe_{0.7}O_{3-{\delta}}$로 코팅된 $Ba_{0.5}Sr_{0.5}Co_{0.8}Fe_{0.2}O_{3-{\delta}}$ 관형 분리막은 압출성형 및 dip coating 방법으로 제조 되었다. 코팅된 관형 분리막의 특성은 X-선 회절분석기(XRD)와 전자 주사 현미경(SEM)을 이용하여 분석하였으며, 분석결과 $2{\mu}m$의 다공성 코팅 층을 갖는 페롭스카이트 구조임을 알 수 있었다. 산소투과량 분석은 $750{\sim}950^{\circ}C$ 범위에서 공급측과 투과 측을 대기 중 공기와 진공으로 하여 수행되었다. 다공성의 $La_{0.6}Sr_{0.4}Ti_{0.3}Fe_{0.7}O_{3-{\delta}}$로 코팅된 $Ba_{0.5}Sr_{0.5}Co_{0.8}Fe_{0.2}O_{3-{\delta}}$ 관형 분리막의 산소투과량은 $950^{\circ}C$에서 $3.2mL/min{\cdot}cm^2$로 코팅되지 않은 분리막보다 높게 나타났으며, 11일 동안의 장기 안정성 실험결과 코팅 층에 의해 안정성이 증가됨을 알 수 있었다.

Effect of Increased Glutamate Availability on L-Ornithine Production in Corynebacterium glutamicum

  • Glutamate availability in the argF-argR-proB${\Delta}$ strain of Corynebacterium glutamicum was increased by addition of glutamate to the cell or inactivation of the phosphoenolpyruvate carboxykinase activity and simultaneous overexpression of the pyruvate carboxylase activity to assess its effect on L-ornithine production. When glutamate was increased in an L-ornithine-producing strain, the production of L-ornithine was not changed. This unexpected result indicated that the intracellular concentration and supply of glutamate is not a rate-limiting step for the L-ornithine production in an L-ornithine-producing strain of C. glutamicum. In contrast, overexpression of the L-ornithine biosynthesis genes (argCJBD) resulted in approximately 30% increase of L-ornithine production, from 12.73 to 16.49 mg/g (dry cell weight). These results implied that downstream reactions converting glutamate to L-ornithine, but not the availability of glutamate, is the rate-limiting step for elevating L-ornithine production in the argF-argR-proB${\Delta}$ strain of C. glutamicum.

TOPOLOGIES AND INCIDENCE STRUCTURE ON Rn-GEOMETRIES

  • An R$^{n}$ -geometry (P$^{n}$ , L) is a generalization of the Euclidean geometry on R$^{n}$ (see Def. 1.1). We can consider some topologies (see Def. 2.2) on the line set L such that the join operation V : P$^{n}$ $\times$ P$^{n}$ \ $\Delta$ longrightarrow L is continuous. It is a notable fact that in the case n = 2 the introduced topologies on L are same and the join operation V : P$^2$ $\times$ P$^2$ \ $\Delta$ longrightarrow L is continuous and open [10, 11]. It is a fundamental topological property of plane geometry, but in the cases n $\geq$ 3, it is no longer true. There are counter examples [2]. Hence, it is a fundamental problem to find suitable topologies on the line set L in an R$^{n}$ -geometry (P$^{n}$ , L) such that these topologies are compatible with the incidence structure of (P$^{n}$ , L). Therefore, we need to study the topologies of the line set L in an R$^{n}$ -geometry (P$^{n}$ , L). In this paper, the relations of such topologies on the line set L are studied.

부신수질 Chromaffin 세포의 $Ca^{2+}$ 통로유형이 카테콜아민 분비에 미치는 영향에 관한 정량적 연구 (Contribution of Different Types of $Ca^{2+}$ channels to Catecholamine Secretion in Rat Adrenal Chromaffin Cells)

  • Adrenal chromaffin cells secrete catecholamine in response to acetylcholine. The secretory response has absolute requirement for extracellular calcium, indication that $Ca^{2+}$ influx through voltage dependent $Ca^{2+}$ channel (VDCC) is the primary trigger of the secretion cascade. Although the existence of various types of $Ca^{2+}$ channels has been explored using patch clamp technique in adrenal chromaffin cells, the contribution of different types of $Ca^{2+}$ channels to catecholamine secretion remains to be established. To investigate the quantative contribution of different types of $Ca^{2+}$ channels to cate-cholamine secretion, $Ca^{2+}$ current($I_{Ca}$) and the resultant membrane capacitance increment($\Delta{C}_{m}$) were simultaneoulsy measured. Software based phasor detector technique was used to monitor $\Delta{C}_{m}$. After blockade of L type VDCC with nicardipine (1$\mu$M), $I_{ca}$ was blocked to 43.85$\pm$6.72%(mean$\pm$SEM) of control and the resultant ㅿC$_{m}$ was reduced ot 30.10$\pm$16.44% of control. In the presence of nicardipine and $\omega$-conotoxin in GVIA(l$\mu$M), an N type VDCC antagonist, $I_{ca}$ was blocked to 11.62$\pm$2.96% of control and the resultant $\Delta{C}_{m}$ was reduced to 26.13$\pm$8.25% of control. Finally, in the presence of L, N, and P type $Ca^{2\pm}$ channel antagonists(nicardipine, $\omega$-Conotoxin GVIA, and $\omega$-agatoxin IVA, respectively), $I_{ca}$ and resultant $\Delta{C}_{m}$ were almost completely blocked. From the observation of parallel effects of $Ca^{2+}$ channel antagonists on $I_{ca}$ and $\Delta{C}_{m}$, it was concluded that L, N, and also P type $Ca^{2+}$ channels served and $Ca^{2+}$ source for exocytosis and no difference was observed in their efficiency to evoke exocytosis amost L, N, and P type $Ca^{2+}$ channels.

염기성 염료 Basic Blue 3에 대한 야자계 입상활성탄의 흡착 특성 (Adsorption Characteristics of Coconut Shell-based Granular Activated Carbon on a Basic Dye Basic Blue 3)

  • 본 연구에서는 Basic Blue 3 (BB3)에 대한 야자계 입상활성탄의 흡착특성을 평가하였다. 입상활성탄의 투여량이 증가함에 따라 BB3의 제거율은 증가하는 경향을 보였고, 0.2 g 투여량에서 초기농도 50 mg/L의 BB3가 완전히 제거되었다. 흡착평형은 초기농도 25 mg/L와 50 mg/L에서 각각 270분과 420분이 소요되었으며, 실험데이터는 유사 2차 속도식으로 잘 묘사되었다. Langmuir 식에서 예측된 최대흡착량은 298, 308, 318 K에서 34.45, 46.63, 53.10 mg/g으로 온도가 증가할수록 증가하였다. 또한, Gibbs 자유에너지 변화(${\Delta}G$)는 온도 증가에 따라 -7.37, -8.19, -10.40 kJ/mol으로 변화하였고, 엔탈피 변화(${\Delta}H$) 및 엔트로피 변화(${\Delta}S$)는 34.47 kJ/mol과 0.15 J/mol K로 계산되었다. 따라서 야자계 입상활성탄에 의한 BB3 흡착은 자발적이고 흡열적이었다.

유치레진치의 전치부 색조연구 (A STUDY ON THE COLORS OF THE ANTERIOR PRIMARY RESIN TEETH)

  • 유전치의 조기상실시 가철성 또는 고정성 공간유지장치의 제작이 필요하며, 보다 심미적인 결과를 얻기 위해서는 자연치아의 색조에 부합하는 레진치의 사용이 요구된다. 현재 가장 많이 사용되는 유치레진치는 Resin Natural $Teeth^{TM}$ (Nissin dental, Japan)로, A형 1번 색조와 A형 2번 색조로 구성되어 있다. 본 연구는 유치레진치의 전치부 색조특성을 이해하고 자연치아와의 색조비교를 위한 기본 자료를 만들기 위한 목적으로, A1 색조군과 A2 색조군의 유치레진치의 상악 4전치부의 순면을 각각 17 set에 대해 치아분광측색장치(spectrophotometer)인 ShadeEye $NCC^{TM}$(Shofu, Japan)로 각 3회씩 측정하여 CIE 표색계의 $L^*,\;a^*,\;b^*$ 값을 구하고 Kruskall-Wallis Test, Mann-Whitney U Test를 이용해 95%의 신뢰도(P<0.05)로 분석하여 다음과 같은 결과를 얻었다. 1. ${\Delta}E$값 및 $L^*$값, $b^*$값의 분포로 볼 때, A1 색조군이 A2 색조군보다 치아위치별 색조차이가 작았다. 각 색조군내에서 $a^*$값의 치아위치별 색조차이는 거의 없었다(P>0.05). 2. A1 및 A2 색조군내에서의 ${\Delta}E$값이 매우 작아(최대 ${\Delta}E{\le}1.23$) 육안으로 그 차이를 구분하기 어려운 것으로 보이며, A1과 A2 색조군의 대표값 간의 ${\Delta}E$값은 3.97로 육안으로 군 간의 차이를 구분할 수 있었다. 3. A1 색조군의 측정평균값은 $L^*=73.8$, $a^*=-1.8$, $b^*=-4.7$이었으며, A2 색조군의 측정평균값은 $L^*=75.8$, $a^*=-2.7$, $b^*=-1.4$이었다. 향후 자연 유전치에 대한 색조특성을 연구하여 본 결과와의 차이점을 비교해 색조적합성이 좋은 레진치의 개발이 필요할 것으로 사료된다.

Structure-Activity Relationships of 13- and 14-Membered Cyclic Partial Retro-Inverso Pentapeptides Related to Enkephalin

  • A series of 13- and 14-membered cyclic enkephalin analogs based on the moderately $\mu$ selective prototype compound Tyr-C[D-$A_2bu$-Gly-Phe-Leu] 8a were synthesized to investigate the structure-activity relationship. The modifications of sequence were mainly focused on two positions 3 and 5, critical for the selective recognition for $\mu$ and $\delta$ opioid receptors. The substitution of hydrophobic $Leu^5$ with hydrophilic $Asp^5$ derivatives led to Tyr-C[D-$A_2bu$-Gly-Phe-Asp(N-Me)] 7 and Tyr-C[D-Glu-Phe-gPhe-rAsp(O-Me)] 5, the peptides with a large affinity losses at both $\mu$ and $\delta$ receptors. The substitution of $Phe^3$ with $Gly^3$ led to Tyr-C[D-Glu-Gly-gPhe-rLeu] 3 and Tyr-C[D-Glu-Gly-gPhe-D-rLeu] 4, the peptides with large affinity losses at $\mu$ receptors, indicating the critical role of phenyl ring of $Phe^3$ for $\mu$ receptor affinities. One atom reduction of the ring size from 14-membered analogs Tyr-C[D-Glu-Phe-gPhe-(L and D)-rLeu] 6a, 6b to 13-membered analogs Tyr-C[D-Asp-Phe-gPhe-(L and D)-rLeu] 1, 2 reduced the affinity at both $\mu$ and $\delta$ receptors, but increased the potency in the nociceptive assay, indicating the ring constrain is attributed to high nociceptive potency of the analogs. For the influence of D- or L-chirality of $Leu^5$ on the receptor selectivity, regardless of chirality and ring size, all cyclic diastereomers displayed marked $\mu$ selectivity with low potencies at the $\delta$ receptor. The retro-inverso analogs display similar or more active at $\mu$ receptor, but less active at $\delta$ receptor than the parent analogs.
