CAT (clear-air turbulence) forecasting algorithm, the Graphical Turbulence Guidance (GTG) system developed at NCAR (national center for atmospheric research), is evaluated with available observations (e.g., pilot reports; PIREPs) reported in South Korea during the recent 5 years (2003-2008, excluding 2005). The GTG system includes several steps. First, 44 CAT indices are calculated in the domain of the Regional Data Assimilation and Prediction System (RDAPS) analysis data with 30 km horizontal grid spacing provided by KMA (Korean Meteorological Administration). Second, 10 indices that performed ten best forecasting scores are selected. Finally, 10 indices are combined by measuring the score based on the probability of detection, which is calculated using PIREPs exclusively of moderate-or-greater intensity. In order to investigate the best performance of the GTG system in Korea, various statistical examinations and sensitivity tests of the GTG system are performed by yearly and seasonally classified PIREPs. Performances of the GTG system based on yearly distributed PIREPs have annual variations because the compositions of indices are different from each year. Seasonal forecasting is generally better than yearly forecasting, because selected CAT indices in each season represent meteorological condition much more properly than applying the selected CAT indices to all seasons. Wintertime forecasting is the best among the four seasonal forecastings. This is likely due to that the GTG system consists of many CAT indices related to the jet stream, and turbulence associated with the jet stream can be activated mostly in wintertime under strong jet magnitude. On the other hand, summertime forecasting skill is much less than other seasons. Compared with current operational CAT prediction system (KITFA; Korean Integrated Turbulence Forecasting System), overall performance of the GTG system is better when CAT indices are selected seasonally.
To identify and discriminate the bacterial species at the subspecific level, rep-PCR is a reliable genomic fingerprinting tool. Fourteen strains of bacteria were isolated from different food sources, identified as Leuconostoc citreum using 16S rRNA gene sequencing, and amplified using rep-primers (REP, ERIC, and $(GTG)_5$). Fingerprinting patterns generated bands in the range of 300-6,000 bp with REP, 150-6,000 bp with ERIC, and 200-1,700 bp with $(GTG)_5$ primers. In UPGMA dendrogram analysis, 14 strains were clustered into three clades (I, II, and III) with all the primers, thus differentiating them at the molecular level. The present study revealed the differentiation of L. citreum strains using rep-PCR.
Genetic relatedness of medically important Exophiala species such as E. dermatitidis, E. mansonii, and three E. jeanselmei varieties: jeanselmei, lecanii-corni, and heteromorpha was examined using PCR-RFLP(restriction fragment length polymorphism) of ribosomal DNA, M-13, $(GTG)_5$ and nucleotide sequences of ribosomal ITS(internal transcribed space) II regions. Three E. jeanselmei varieties showing distinct band patterns for each DNA markers as well as different nucleotide sequences of ribosomal ITS II regions could be considered as a separate species. E. dermatitidis and E. mansonii demonstrated the identical band patterns of RFLP of ribosomal DNA, M-13, and $(GTG)_5$ markers. However, nucleotides sequences of ribosomal ITS II region were different between these two species.
None of the numerous published canine idiograms and karyotypes has yet been generally accepted as a standard one because the dog has 76 acrocentric autosomes of similar size and shape. To establish canine banded karyotype from the 22nd chromosome to the 37th chromosome, we analyzed canine chromosomes by GTG, high resolution, and NOR-banding techniques. The GTG and high resolution banding patterns of canine chromosomes corresponded to other reports described previously except for a few chromosomes. While other researchers observed 12 bands, we observed 7 bands in the banding patterns of chromosome 24, 34 and 37. On the other hand, the banding patterns by NOR-banding technique showed that three pairs of autosomes have nucleolus organizer regions at the terminal ends of their long arm, and the Y chromosome has it in its short arm terminal. However, the X chromosome has no nucleolus organizer like other mammals.
Comparative genomic hybridization (CGH) can now be applied to detect the origin of extra or missing chromosomal material in cases with common unbalanced aberrations and in prenatal investigations. This method has been used in 13 cases of fetal samples for this study; 3 for amniocytes, 2 for cord blood and 8 for abortus tissues. These samples were previously subjected to GTG-banding. Our study showed aneuploidy in 8 cases, and partial monosomy, partial trisomy or marker chromosome in the remaining 5. The CGH disclosed further small genetic imbalances in 4 of all 13 cases: a prenatal sample showing del(20)(q13) by GTG confirmed a loss of the segment 20p13-pter by CGH; a marker chromosome manifested normal CGH profile; chromosome der(?)(?;15) found in an abortus sample by GTG turned out to be a loss of 15pter-q14 (partial monosomy) and a gain of 10pter-q22 (partial trisomy); the der(15) shown by GTG represented partial trisomy of 3q24-qter. These findings show that CGH is very useful and efficient for cytogenetic investigations of clinical cases.
Acetic acid bacteria (AAB) are versatile organisms involved in the production of variety of fermented foods, such as vinegar and kombucha, and products of biotechnological relevance, such as bacterial cellulose. In the present study, Malatya apricot, a variety with protected designation of origin (PDO), and vinegar samples produced using various fruits were used to isolate AAB. The 19 AAB isolates obtained were typed using (GTG)5 fingerprinting, and the ones selected were identified by sequencing either 16S rDNA alone or in combination with 16S-23S rRNA internal transcribed spacer region or ligA gene. While all apricot isolates (n = 10) were Gluconobacter cerinus, vinegar isolates (n = 9) were composed of Komagataeibacter saccharivorans, Acetobacter syzygii, and possible two new species of AAB, Komagataeibacter sp., and Gluconobacter sp. (GTG)5 fingerprinting showed the presence of several genotypes of G. cerinus in the apricot samples. Screening for some technologically relevant properties, including thermotolerance, ethanol tolerance, and cellulose production capability, showed that all Komagataeibacter and some Gluconobacter isolates could tolerate the temperature of 35℃, and that vinegar isolates could tolerate up to 8% ethanol. One isolate, Komagataeibacter sp. GUS3 produced bacterial cellulose (1 g/l) and has the potential to be used for cellulose production.
The soybean cyst nematode (Heterodera glycines Inchinoe; SCN) is a devastating pest of soybean and is responsible for significant losses in yield. The use of resistant cultivars is the effective method to reduce or eliminate SCN damage. The objective of this research is to identify AFLP markers linked to the SCN resistant genes. Bulked genomic DNA was made from resistant and susceptible genotypes to SCN and a total of 19 primer combinations were used. About 31 fragments were detected per primer combination. The banding patterns were readily distinguished in resistant and susceptible bulked genotypes. Polymorphic fragments were detected between resistant and susceptible bulked genotypes in the primer combination of CGT/GGC, CAG/GTG and CTC/GAG. In primer combinations of CGT/GGC and CAG/GTG, bulked resistant genotype produced a polymorphic bands. However, in primer of CTC/GAG, bulked susceptible genotype produced a polymorphic fragments. Three AFLP markers identified as a polymorphic fragments between bulked genomic DNA were mapped in 85 F2 population. Among them, only two markers, CGT/GGC and CTC/GAG, was linked and was mapped. Broad application of AFLP marker would be possible for improving resistant cultivars to SCN.

Bacillus subtilis CH3-5 isolated from cheonggukjang secretes a 28 kDa protease with a strong fibrinolytic activity. Its gene, aprE3-5, was cloned and expressed in a heterologous host (Jeong et al., 2007). In this study, the promoter of aprE3-5 was replaced with other stronger promoters (Pcry3A, P10, PSG1, PsrfA) of Bacillus spp. using PCR. The constructed chimeric genes were cloned into pHY300PLK vector, and then introduced into B. subtilis WB600. The P10 promoter conferred the highest fibrinolytic activity, i.e., 1.7-fold higher than that conferred by the original promoter. Overproduction of the 28 kDa protease was confirmed using SDS-PAGE and fibrin zymography. RT-qPCR analysis showed that aprE3-5 expression was 2.0-fold higher with the P10 promoter than with the original promoter. Change of the initiation codon from GTG to ATG further increased the fibrinolytic activity. The highest aprE3-5 expression was observed when two copies of the P10 promoter were placed in tandem upstream of the ATG initiation codon. The construct with P10 promoter and ATG and the construct with two copies of P10 promoter in tandem and ATG exhibited 117% and 148% higher fibrinolytic activity, respectively, than that exhibited by the construct containing P10 promoter and GTG. These results confirmed that significant overproduction of a fibrinolytic enzyme can be achieved by suitable promoter modification, and this approach may have applications in the industrial production of AprE3-5 and related fibrinolytic enzymes.
Diseased eel (Anguilla bicolor) displayed severe hemorrhages in the gills, and congestion and swelling in the liver. During the epizootic, the water temperature was $28^{\circ}C$ and the morality rates were about 5%. No parasites were found on the gills and skin. Bacteria were not cultured from any internal organs using TSA or SS agar at $28^{\circ}C$ for 48 hrs. Histopathologically, the gills showed epithelial hyperplasia in the base of secondary gill lamellae and hemorrhages in the capillaries. Some cells in the proliferated interlamellar epithelia exhibited marginal hyperchromatosis. And severe vacuolated changes in the parenchymal cells and congestion in the central veins were observed in the liver. The specific amplicon (396 bp) was detected from gills and opercula of affected eel PCR using Anguillid herpesvirus-1 (AngHV-1) -specific primer sets HVAPOLVPSD (5-'GTG TCG GGC TTT GTG GTG C-3') and HVAPOLOOSN (5'-CAT GCC GGG AGT CTT TTT GAT-3'). Sequencing analysis of the amplicon demonstrated that this gene was 99% homologous to the AngHV-1 sequence deposited in GenBank. This is the first report of AngHV-1 outbreak in the farmed shortfin eels (A. bicolor) in Korea. When diseased fish were maintained for 10 days at water temperatures of $32^{\circ}C$ and $35^{\circ}C$, the cumulative mortalities were 100% and 10%, respectively. Even though the AngHV-1 genome in the gills from the eel kept at $35^{\circ}C$ was detected using PCR, the structure of gill filaments was similar with that of normal fish. Increasing the water temperature to $35^{\circ}C$ was an effective way to diminish the mortality of AngHV-1 affected eel.
The task for chromosome analysis and diagnosis by experienced cytogenetists are being concerned as repetitive, time consuming job and expensive. For that reason, intelligent agent based on chromosome knowledge base using web has been developed to be able to analyze chromosomes and obtain necessary advises from the knowledge base instead of human experts. That is to say, the knowledge base of IF THEN production rule was implemented to a knowledge domain with normal and abnormal chromosomes, and then the inference results by the knowledge base could enter the inference data into the database. Experimental data were composed of normal chromosomes of 2,736 cases and abnormal chromosomes of 259 cases that have been obtained from GTG-banding metaphase peripheral blood and amniotic fluid samples. The completed intelligent agent for the chromosome knowledge base provides variously morphological information by analysis of normal or abnormal chromosomes also has the advantage of being able to consult with the user on the chromosome analysis and diagnosis.
