  • This study was carried out to investigate the elucidation of gene expression in embryo formation of the Kidney mutant, especially yolk proteins in unfertilized eggs and template activity of m-RNA extracted from them. The results were summarized as follow ; There was no recognized qualitative difference in yolk proteins of unferilized eggs between the Kidney mutant and normal. There was not any difference, between Kidney mutant and normal in the molecular species of m-RNA derived from maternal origin with the template activity in vitro.

  • Vitellins or so named yolk proteins are stored in yolk granules of oocytes. They play major role in providing energy and nutrient for developing embryo. Vitellins are very diverse group of chemically uncharacterized complexes of glyco-, lipo-, phosfoproteins and proteoglycans. Isolation of vitellins and production of antibodies specific to them would be useful for studying the physiology of yolk formation and developmental immunological techniques for investigation reproduction for commercially important marine mollusces.   • The dietary carbohydrates are mainly digested and adsorbed at small intestine. We developed a new food additive as an egg yolk antibody(1gY) against maltase, sucrase and sodium dependent g1ucose cotransporter(SGLT) for the regulation of blood glucose level and weight control. The maltase, sucrase and SGLT were purified from porcine small intestine which is very similar to that of human in physiological characteristics. The purification step contained an ultracentrifugation, ion exchange chromatography and hydrophobic chromatography. The hens were immunized by purified protein and the IgY activities against immunized antigens were determined. This antibody obtained from the immunized hen's egg yolks directly inhibited the activities of maltase and sucrase in vitro. And the IgY delayed and decreased the increment of blood g1ucose level after administration of maltose, sucrose and glucose in rat about 30 to 60%. The results of this study suggest that the IgY inhibiting the carbohydrate digestion could be used as functional food materials for weight control and regulation of blood glucose level in diabetes.

Effects of Dietary Corticosterone on Yolk Colors and Eggshell Quality in Laying Hens

  • The objective of this study was to investigate the effects of dietary corticosterone on egg quality. For 2 weeks hens received either control or experimental diet containing corticosterone at 30 mg/kg diet. Feed intake and egg production were monitored daily, and body weight measured weekly. Egg weights and egg quality were measured daily. Corticosterone treatment resulted in a remarkable increase in feed intake and sharp decrease in egg production compared with control (p<0.05) whereas body weight remained unchanged. Decreased albumen height, but no changes in egg weight, led to decreased Haugh unit (p<0.05). Corticosterone caused elevated eggshell thickness (p<0.05) without altering weight and strength, suggesting possible changes in shell structure. Yolk color and redness were increased by corticosterone (p<0.05) but lightness and yellowness were either not changed or inconsistent over the time period of measurements. Increased concentrations in plasma were also found for corticosterone, glucose, cholesterol, creatinine, uric acid, albumin, aspartate aminotransferase, creatine kinase, lactate dehydrogenase, total protein, and amylase (p<0.05), suggesting that corticosterone increased protein breakdown, renal dysfunctions and pancreatitis. Together, the current results imply that dietary corticosterone affects egg quality such as yolk colors and shell thickness, in addition to its effects on feed intake and egg production.

Effects of Calcium, Vitamin D and Egg Yolk Peptide Treatment on the Retardation of Longitudinal Bone Growth Induced by Low-Calcium Diets (저칼슘 식이로 유발한 성장 저하 흰쥐에 대한 칼슘, 비타민 D 및 난황 펩타이드의 투여가 장골 길이 성장에 미치는 영향)

  • Objectives : Egg yolk is composed of various important chemical substances for human health. A calcium shortage causes the growth retardation on the body growth. In this study, we examined the therapeutic effects of calcium, vitamin D and egg yolk peptide (EYP) treatment on the retardation of the longitudinal bone growth induced by low-calcium diet in adolescent rats. Methods : Low calcium diets were administrated for 15 days. During the last five days, calcium and/or vitamin D and/or EYP were administrated. The body weights, longitudinal bone growth rates, the heights of growth plates, and bone morphogenetic protein (BMP)-2 and insulin-like growth factor (IGF)-1 expressions were measured using histochemical analysis. Results : Low calcium diets caused the significant reduction in body weight gains and the longitudinal bone growth. The heights of growth plates and the expressions of BMP-2 and IGF-1 showed the impairment of body growth as well. Calcium and/or vitamin D administration could not significantly increase the longitudinal bone growth. However, calcium, vitamin D, and EYP administration significantly increased the bone growth, the growth plate height, and BMP-2 and IGF-1 expressions. Conclusions : These results suggest that EYP enhances the longitudinal bone growth in the calcium and/or vitamin D deficiency and it could be a promising agent for the treatment of children suffering from malnutrition.

Studies on the Nutrient Contents and Optimum Prices of Eggs in Relation to Egg Weight (계란의 란중별영양성분 및 적정가격에 관한 조사연구)

  • These studies were conducted to compare the nutritive values and optimum prices of eggs among 6 groups of different egg Weight. With the total of 100 eggs of each weight group, after the weight percentage of egg yolk, albumen and shell in the whole egg were investigated, protein and fat contents of e99 yolk and albumen were analyzed. and then protein and fat contents in the whole eggs were calculated. Finally, the optimum prices of eggs in relation to the egg weight were studied on the basis of egg weight, protein content and protein plus fat contents of eggs, respectively. The results obtained are summarized as follows; 1. As the egg weight (X, g/10 eggs) increased, egg yolk (Y$_1$, %) and shell(Y$_2$, %) percentages tended to decrease, but egg albumen(Y$_3$, %) percentage increased lineally; Y$_1$=44.34-0.02X, Y$_2$=15.358-0.006 X, and Y$_3$=40.136+0.026 X. 2. There were no significant differences in protein and fat contents of eggs among 6 different groups of egg weight. 3. Protein (Y$_1$, %), fat (Y$_2$, %) and protein plus fat (Y$_3$, %) contents in the whole eggs declined progressively as the egg weight (X, g/10 eggs) increased ; Y$_1$=11.943-0.00032X, Y$_2$=13.996-0.00614X, and Y$_3$=25.939-0.00646X. 4. Similar results were obtained whether the optimum prices of eggs were estimated on the basis of egg weight or protein content of eggs, and they were higher in the large size eggs and lower in the small size eggs than the optimum prices of eggs estimated on the basis of protein plus fat content of eggs.

Production and Characterization of Egg Yolk Antibodies (IgY) against Flagella Antigen of Salmonella sp. (살모넬라 편모 항원에 대한 난황항체(IgY)의 생산 및 특성)

  • Egg yolk antibodies(IgY) from laying hens immunized with antigens from Salmonella choleraesuis, Salmonella typhimurium and Salmonella dublin were produced. The Antigenic proteins isolated from those flagella of Salmonella sp., determined by SDS-PAGE, were pure and had a molecular mass of approximately 53.4, 51 and 54.6 kDa, respectively. The IgY titers were found at two weeks after first immunization and increased gradually to maximum of 330,000 300,000 and 440,000 respectively. According to the results of specificity test by ELISA, the IgY raised against Salmonella sp. were found highly specific activity levels. Concentration of Salmonella sp. incubated with anti-Salmonella sp. IgY were drastically reduced to the levels of 2.8∼4.0 log CFU/ml. The contents of IgY in an egg yolk was approximately 31∼33 mg/ml.

Apoptotic Killing of Breast Cancer Cells by IgYs Produced Against a Small 21 Aminoacid Epitope of the Human TRAIL-2 Receptor

  • TRAIL, tumor necrosis factor (TNF)-related apoptosis-inducing ligand belongs to one of important cytokine superfamilIES, tumor necrosis factor ($TNF{\alpha}$). TRAIL-2 receptor agonists activate several cell signaling pathways in cells in different manners and could lead to apoptosis or necrosis. Agonistic egg yolk antibodies like IgY which have been developed in a selective manner could activate TRAIL death receptors such as TRAIL-2 (DR5) and thus apoptosis signaling. We here investigated induction of apoptosis in human breast cancer cells (MCF7 cell line) by an IgY produced against an 21 aminoacid epitope of the human TRAIL-2 receptor. As the first step a small peptide of 21 aminoacids choosen from the extracellular domain of DR5 protein was produced with a peptide synthesizer. After control assays and confirmation of the correct amino acid sequence, it was injected to hens immunized to achieve high affinity IgYs. At the next step, the produced IgYs were extracted and examined for specificity against DR5 protein by ELISA assay. Subsequently, the anticancer effect of such IgYs was determined by MTT assay in the MCF7 human breast cancer cell line. The produced peptides successfully immunized hens and the produced antibodies which accumulated in egg yolk specifically recognized the DR5 protein. IgYs exerted significant toxicity and killed MCF7 cells as shown by MTT assay.

  • Receptor-mediated endocytosis of coelomic fluid proteins (CP), yolk precursor proteins, appears to be regulated by multiple GTP-binding proteins during oogenesis of a polychaete, Pseudopotamilla occelata. Transport of 125 I-CP into the oocytes of intermediate size class, at which CP is the most actively transported, is enhanced by GTP but inhibited by GTP analogues, either GTPrS or GTP$\beta$S. The effects of GTP and GTPrS on the transport were also confirmed by tracing internalization of gold-labeled CP with transmission electron microscope. Internalization of gold-labeled CP into the yolk granules was enhanced by GTP but inhibited by GTPrS.

Expression of Lily mottle virus Coat Protein and Preparation of IgY Antibody against the Recombinant Coat Protein

  • Lily symptomless virus (LSV), Lily mottle virus (LMoV), and Cucumber mosaic virus (CMV) are the most prevalent viruses infecting lilies in Korea. Leaf and bulb samples showing characteristic symptoms of virus infection were collected in 2012, and 80 field samples were analyzed by reverse transcription polymerase chain reaction (RT-PCR). The infection frequencies were 79% for LMoV, 5% for LSV, and 3% for CMV. The LMoV coat protein gene was amplified and cloned into the pET21d(+) expression vector to develop serological diagnostic tools to detect LMoV. The resulting carboxy-terminal His-tagged coat proteins were expressed in Escherichia coli strain BL21 (DE3) by induction with IPTG. The recombinant proteins were purified using Ni-NTA agarose beads and used as an antigen to produce polyclonal antibodies in laying hens. The resulting egg yolk immunoglobulin (IgY) specifically recognized LMoV from infected plant tissues in immunoblotting assays and had comparable sensitivity to that of a mammalian antibody. In addition, method of immunocapture RT-PCR using this IgY was developed for sensitive, efficient, and rapid detection of LMoV. Based on these results, large-scale bulb tests and detection of LMoV in epidemiological studies can be performed routinely using this IgY. This is the first report of production of a polyclonal IgY against a plant virus and its use for diagnosis.
