Factorial design에 의한 Acetobacter xylinum KJ1의 Bacterial cellulose 생산조건의 최적화

  • Acetobacter xylinum KJ1 efficiently producing bacterial cellulose(BC) in shaking culture was isolated from a rotten grape. The strain was used to investigate optimum operating conditions for increasing BC production and factorial design model was employed for the optimization. The results of experiments were statistically analyzed by SAS program. Reciprocal effects of each factors(carbon source concentration, shaking speeds(rpm), oxygen pressure, and CSL concentration) and culture condition of BC production were examined by getting regression equation of the dependent variable. Comparisons between experimental results and predicted results about BC concentration were done in total 24 experiments by combination of each factors using SAS program, and the correlation coefficients of BC concentration and BC yield were 0.91 and 0.81, respectively. The agitated cultures were performed in various operation conditions of factors which affected considerably to BC production in jar fermentor. The results showed that BC concentration was 11.67g/ L in 80 hours cultivation under the condition of carbon source concentration shaking speeds(rpm) : oxygen pressure: CSL concentration = 4% : 460rpm : 0.28 : 6%. On the other hand BC yield was 0.42g/g in 80 hours cultivation under the condition of carbon source concentration shaking speeds(rpm) : oxygen pressure: CSL concentration = 4% : 564rpm : 0.21 : 2%. The BC production could be enhanced up to more than 65.3% by factorial design. The result of a verifying experiment under the optimal conditions determined by the factorial design to the BC production showed that the model was appropriate by obtaining BC concentration of 11.02g/L in the optimum condition

Optimization of the High-Pressure Condition for Rice Protein Extracting Using Response Surface Methodology (RSM) (반응표면분석법을 이용한 쌀 단백질 초고압 추출조건 최적화)

  • The purpose of this study was to optimize the rice protein extracted using a response surface methodology. The experiment was designed based on a CCD (Central Composite Design), and the independent variables were the high pressure (X1, 0-400 MPa) and processing time (X2, 0-10 minutes). The results of the extraction content (Y1), residue content (Y2), and recovery yield (Y3) were fitted to a response surface methodology model (R2= 0.92, 0.92, and 0.93, respectively). Increasing the pressure and processing time has a positive effect on the extraction content (Y1), residue content (Y2), and recovery yield (Y3). Therefore, these high-pressure conditions (independent variables) can significantly affect the improvement in rice protein extraction efficiency. Thus, the optimal conditions of X1 and X2 were 400 MPa and 10 min., respectively. Under these optimal conditions, the predicted values of Y1, Y2, and Y3 were 62.93, 57.53 mg/g, and 91.76%, respectively.

Pre-establishment of Microwave-Assisted Extraction Conditions for Chinese Ginger Oleoresins (중국산 생강의 올레오레진 제조를 위한 극초단파 추출조건 설정)

  • Some fundamental conditions were investigated in microwave-assited extraction (MAE) for oleoresin from Chinese dried ginger. The fifty watts of microwave energy(2,450 MHz), 60 mesh in particle size and 1:10 (g/mL) ratio of sample to solvent were the optimum conditions. Lower yield of soluble oleoresins was obtained at higher concentrations of ethanol. Radical scavenging ability was highest in extracts by using 25% of ethanol. Total phenolics in extracts were remarkably increased at above 50% of ethanol concentration. Overall yield of oleoresin components increased in proportional to extraction time up to 7 min. Based on the above results, it is indicated that ethanol concentration is a critical parameter in MAE.

Biochemical Quantitation of PM2 Phage DNA as a Substrate for Endonuclease Assay

  • Bacteriophage PM2 has a closed circular form of double stranded DNA as a genome. This DNA from the phage is a useful source for nick-circle endonuclease assay in the fmol range. Due to difficulties in the maintenance of viral infectivity, storage conditions of the phage should be considered for the puri-fication of PM2 DNA. The proper condition for a short-term storage of less than 2 months is to keep the PM2 phage at 4$^{\circ}C$; whereas the proper condition for a long-term storage of the PM2 phage for over 2 months is to keep it under liquid nitrogen in 7.5 % glycerol. The optimal conditions for a high yield of phage progeny were also considered with the goal to achieve a successful PM2 DNA preparation. A MOI(Multiplicity Of Infection) of 0.03, in which the OD$\sub$600/ of the host bacteria was between 0.3 and 0.5, turned out to be optimal for the mass production of PM2 phage with a burst size of about 214. Considerations of PM2 genome size, and the concentrations and radiospecific activities of purified PM2 DNA, are required to measure the endonuclease activity in the fmol range. This study reports the proper quantitation of radioactivity and the yield of purified DNA based on these conditions.

Optimization of Enzymatic Hydrolysis for the Production of Antitoxic Bovine Hepatic Extract (항독성 소간추출물의 생산을 위한 효소분해의 최적화)

  • Bovine hepatic extract is recognized as possessing detoxifying activity against various liver diseases. In orderto develop a process for its mass production, various enzymatic hydrolysis conditions were tested, and bovine hepatic extracts were prepared. These extracts were then examined for composition, microorganism levels, and vitamin $B_{12}$ content. Among the enzymes tested, papain was selected based on yields for dry residue and amino nitrogen. The other enzymes tested included bromelain, ficin, pancreatin, and protease NP. The optimal hydrolysis conditions were established at 65$^{\circ}C$ for 24 hr, with an addition of 1%(w/w) papain to the beef liver. The prepared spray-dried bovine hepatic extract showed an 11% recovery yield on a raw beef liver basis, with 95% dry residue and 11.8% total nitrogen content. Microorganisms were not detected in the dried extract, and its vitamin $B_{12}$ content was 4.1 ${\mu}$g/g. In summary, the conditions established in this study could be applied for the high yield mass production of bovine hepatic extract.

Optimum Conditions for Extracting Alginic Acid from Undaria Pinnatifida and Amino Acid Composition of Its Extraction Residue (미역 알진산의 추출조건과 그 추출잔사의 아미노산 조성)

  • The optimum conditions for extracting alginic acid from the powdered Undaria pinnatifida and amino acid composition of its extraction residue were investigated. Extraction with 60 volumes of 1.0% sodium carbonate solution to the sample at $80^{\circ}C$ gave a maximum yield of alginic acid and the optimum extraction time was 3 hours when all the other extraction conditions had teen satisfied. In the process of precipitating alginic acid gel from algin solution, the highest yield was obtained at pH 2.0 and 1.0% $H_{2}SO_{4}$ was more suitable than 10% HC1 as a precipitating agent. Extraction residue remained by separating algin solution contained 51.5% (on the dry baiss) of crude protein, and its limiting amino acid and protein score were lysine and 41.5, respectively.

Adsorption Characteristics of NH4+ by Biochar Derived from Rice and Maize Residue (벼와 옥수수 부산물로 제조한 바이오차의 NH4+ 흡착 특성 평가)

  • BACKGROUND: Biochar has ability to reduce N loss, increase crop yield, and sequestrate carbon in the soil However, there is still limited study concerning the interactive effects of various biochars on NH3 loss and plant growth. This study, therefore, was conducted to investigate the NH4+ adsorption characteristics of biochar derived from rice and maize residues. METHODS AND RESULTS: By-products were pyrolyzed under oxygen-limited conditions at 300-700℃ for 1 hour and used for experiment of NH4+ adsorption in aqueous solution. The adsorption characteristics of biochar were studied using Langmuir isotherm. Biochar yield and hydrogen content decreased with increasing pyrolysis temperatures, whereas pH, EC, and total carbon content increased. The biochar pyrolyzed at lower temperatures was more efficient at NH4+ adsorption than those produced at higher temperatures. In addition, the RL values, indicating equilibrium coefficient were between 0 and 1, confirming that the result was suitable for Langmuir isotherm. CONCLUSION: The maize stalk biochar pyrolyzed at 300℃ was the most efficient to adsorb NH4+ from the aqueous solution. Furthermore, the adsorption results of this experiment were lower than those of other prior studies, which were ascribed to different experimental conditions such as ingredients, and pyrolysis conditions.

Evaluation and Identification of Promising Bivoltine Hybrids of Silk-worm, Bombyx mori L., for Tropics

  • Ten newly evolved bivoltine hybrids of silkworm (Bombyx mori L) were evaluated with control hybrid KA${\pm}$NB4D2 during three seasons of a year for their seasonal performance. Analysis of variance and other statistical methods were employed and the performance was observed in respect of 10 quantitative traits. The results showed significant genotype${\pm}$environment interaction with respect to four quantitative characters viz. fecundity, yield/10,000 larvae, filament length and raw silk (%). Environmental effects were significant for nine characters out of ten characters evaluated. A 105${\pm}$J2 and B${\pm}$NB4D2 were considered as highly adaptable hybrids to local conditions with high mean for maximum characters studied and found suitable to rear in all seasons.

  • The growing interest in the application of arachidonic acid (ARA) in various fields of health and dietary requirements has elicited much attention on the industrial production of ARA-containing oil by the cultivation of Mortierella fungi. For the industrial production of ARA, various studies, such as isolation of a high-potential strain and optimization of culture conditions, have been conducted. Studies including the investigation of morphology are important because ARA is accumulated in the mycelia, and thus cultivation with high biomass concentration is essential for obtaining a high ARA yield. Combining the results derived from various studies, a high ARA yield was attained in an industrial fermentor. These ARA production techniques are applicable to the production of other polyunsaturated fatty acids (PUFAs), and will contribute to the improvement of fermentation technology especially in the field of fungal cultivation.

  • In this paper, a finite element method for the determination of initial blank shape in sheet metal forming process will be introduced. The initial blank shape is determined by the only one step from the final to the initial blank. The used finite element inverse method adopted Henky's deformation theory, Hill's anisotropic yield criterion and simplified boundary conditions. Based on this theory. a three-dimensional membrane finite element code was developed. The developed code will be applied to several sheet metal forming examples for the demonstration of its validity.
