Release Profile of 14C-Butachlor from Controlled Release Formulation Prepared with Alginate-Kaoline Matrix (Alginate와 Kaoline을 이용(利用)한 방출조절제(放出調節劑)의 14C-Butachlor 용출특성(溶出特性))

  • The herbicide $^{14}C$-butachlot[N-(butoxymethyl)-2-chloro-2', 6'-diethylacetanilide] labelled uniformly in benzene ring was incorporated in alginate-based granules to get controlled release properties. The influence of kaoline addition on the formulation characteristics and release profiles were evaluated under a closed dark and an opened sunlight condition. Incorporation efficiency of $^{14}C$-butachlor in alginate-kaoline matrices was over 91.8%. Formulation yield was decreased with increase of kaoline concentration. The release rate from all the granules prepared with alginate was slower than that from the commercial granule impregnated in zeolite. The release rate from the granule containing kaoline was decreased as the kaoline content was increased under both conditions. Losses of butachlor from the leacheate solution of the alginate-kaoline matrices under an opened sunlight condition was diminished by increasing the kaoline content.

  • To separate and utilize the main components of hardwood (Quercus mongolica) by organosov pulping (ester pulping), chips were cooked at various conditions such as; the solvent ratio (acetic acid: ethylacetate: water, 50:25:25; 33:33:34; 25:50:25), maximum temperature (165, 170, $175^{\circ}C$), and cooking times (2, 2.5, 3 hr). The pulps were bleached by the sequences of CEDED, C/DEDED, PEDED. Lignin, sugars, and acetic acid were separated from black liquor and washing liquors. 1. The selective delignification at optimal pulp yield (43-45%) was obtained by cooking at acetic acid: ethylace tate: water ratio of 33:34:34 for 3 hr at $170^{\circ}C$. But in this case, kappa no. of the pulp was not reduced under 60 points. 2. Kappa no. of the pulp could be dropped by an acetone wash to remove reprecipitated lignin a t cooked pulp. 3. The unbleached pulps had a brightness of 45-50%, whereas the bleached pulps gave at 88-93% brightness. Tensile, burst, and tear strengths of the bleached pulps were lower than those of kraft pulp, especially in tear strength. The pulps which were bleached with CEDED sequence were higher in strengths than another bleaching sequences. 4. Lignin of 90-95%(lignin base on wood)was separated from black liquor and washing liquors, while the purified sugars and recovery of acetic acid were a low. An organic phase composed of acetic acid, ethylacetate, and water was separated to a two-phase system by proper adjustment of the solvent ratios.

  • This study presents results of an experimental investigation of the laser-arc, hybrid, butt welding process of SS400 structural steel. Welding parameters including laser power, welding current and speed were varied in order to obtain one-pass, full-penetration welds without defects. The conditions that resulted in optimal beads were identified. After welding, hardness measurements and microstructure observations were carried out in order to study weld properties. The mechanical properties of both the base material and welded specimen were compared based on the results of tensile strength measurements. The yield and tensile strengths were found to be similar.

  • This study was carried out to develop the efficient freeze concentration process of milk through controlling the recrystallisation phenomena of ice. Freeze-concentration was progressed with multi-stage freeze concentrator and there was artificial temperature control to induce recrystallisation phenomena. In each stage of freeze concentration process, the regular recrystallisation time was fixed as 1, 2, 4 and 8 hr to compare the solute increment, yield, brix and ice-crystal size among experimental conditions. Higher concentration as total solids was observed due to the elapse of recrystallisation time, and the maximum total solids in final products: 32.67% was obtained at the ripening time of 8 hr in two-stage process. This result was excessively high concentration comparing to the existing researches and presented the possibilities of milk freeze concentration in the dairy industry, The results of brix and ice-crystal size showed the direct correlation with the recrystallisation time that meant the increased processing time showed the increment of brix and ice-crystal size. Obtained results were numerically modelled to predict the progress of concentration in the industrial process and all of them had fairly high R2 of determination. Therefore, we regarded that these numerical models could be utilized for the development of efficient technology in industrial freeze concentration process.

Biocontrol Characteristics of Bacillus Species in Suppressing Stem Rot of Grafted Cactus Caused by Bipolaris cactivora

  • One of the most important limiting factors for the production of the grafted cactus in Korea is the qualitative and quantitative yield loss derived from stem rots especially caused by Bipolaris cactivora. This study is aimed to develop microbial control agents useful for the control of the bipolaris stem rot. Two bacteria (GA1-23 and GA4-4) selected out of 943 microbial isolates because of their strong antibiotic activity against B. cactivora were identified as Bacillus subtilis and B. amyloliquefaciens, respectively, by the cultural characteristics, Biolog program and 16S rRNA sequencing analyses. Both bacterial isolates significantly inhibited the conidial germination and mycelial growth of the pathogen with no significant difference between the two, of which the inhibitory efficacies varied depending on the cultural conditions such as temperature, nutritional compositions and concentrations. Light and electron microscopy of the pathogen treated with the bacterial isolates showed the inhibition of spore germination with initial malformation of germ tubes and later formation of circle-like vesicles with no hyphal growth and hyphal disruption sometimes accompanied by hyphal swellings and shrinkages adjacent to the bacteria, suggesting their antibiotic mode of antagonistic activity. Control efficacy of B. subtilis GA1-23 and B. amyloliquefaciens GA4-4 on the cactus stem rot were not as high as but comparable to that of fungicide difenoconazole when they were treated simultaneously at the time of pathogen inoculation. All of these results suggest the two bacterial isolates have a good potential to be developed as biocontrol agents for the bipolaris stem rot of the grafted cactus.

Biocontrol of Phytophthora Blight and Anthracnose in Pepper by Sequentially Selected Antagonistic Rhizobacteria against Phytophthora capsici

  • We previously developed a sequential screening procedure to select antagonistic bacterial strains against Phytophthora capsici in pepper plants. In this study, we used a modified screening procedure to select effective biocontrol strains against P. capsici; we evaluated the effect of selected strains on Phytophthora blight and anthracnose occurrence and fruit yield in pepper plants under field and plastic house conditions from 2007 to 2009. We selected four potential biocontrol strains (Pseudomonas otitidis YJR27, P. putida YJR92, Tsukamurella tyrosinosolvens YJR102, and Novosphingobium capsulatum YJR107) among 239 bacterial strains. In the 3-year field tests, all the selected strains significantly (P < 0.05) reduced Phytophthora blight without influencing rhizosphere microbial populations; they showed similar or better levels of disease suppressions than in metalaxyl treatment in the 2007 and 2009 tests, but not in the 2008 test. In the 2-year plastic house tests, all the selected strains significantly (P < 0.05) reduced anthracnose incidence in at least one of the test years, but their biocontrol activities were variable. In addition, strains YJR27, YJR92, and YJR102, in certain harvests, increased pepper fruit numbers in field tests and red fruit weights in plastic house tests. Taken together, these results indicate that the screening procedure is rapid and reliable for the selection of potential biocontrol strains against P. capsici in pepper plants. In addition, these selected strains exhibited biocontrol activities against anthracnose, and some of the strains showed plant growth-promotion activities on pepper fruit.

Identification of Novel Source of Resistance and Differential Response of Allium Genotypes to Purple Blotch Pathogen, Alternaria porri (Ellis) Ciferri

  • Purple blotch, caused by Alternaria porri (Ellis) Cifferi, is a serious disease incurring heavy yield losses in the bulb and seed crop of onion and garlic worldwide. There is an immediate need for identification of effective resistance sources for use in host resistance breeding. A total of 43 Allium genotypes were screened for purple blotch resistance under field conditions. Allium cepa accession 'CBT-Ac77' and cultivar 'Arka Kalyan' were observed to be highly resistant. In vitro inoculation of a selected set of genotypes with A. porri, revealed that 7 days after inoculation was suitable to observe the disease severity. In vitro screening of 43 genotypes for resistance to A. porri revealed two resistant lines. An additional 14 genotypes showed consistent moderate resistance in the field as well as in vitro evaluations. Among the related Allium species, A. schoenoprasum and A. roylei showed the least disease index and can be used for interspecific hybridization with cultivated onion. Differential reaction analysis of three A. porri isolates (Apo-Chiplima, Apn-Nasik, Apg-Guntur) in 43 genotypes revealed significant variation among the evaluated Allium species (P = 0.001). All together, the present study suggest that, the newly identified resistance sources can be used as potential donors for ongoing purple blotch resistance breeding program in India.

Publishing a Web Based Crop Monitoring System and Performance Test (웹 기반 농업생산환경 모니터링 시스템 시범구축 및 성능평가)

  • In developed countries such as USA and Europe, agricultural monitoring system is developed and utilized in various fields in order to predict crop yield, observe weather conditions and anomaly, categorize crop fields, and calculate areas for each crop. These system is Web Map Service(WMS) which utilizes open source and commercial softwares, and various information collected from remote sensing data are provided. This study will utilize tools such as GeoServer, ArcGIS Server, which are widely used to monitor agricultural production, to publish Map Server and Web Application Server. This enables performance test study for future agricultural production monitoring system by making use of response time and data transfer test. When tested in identical condition GeoServer showed a better result in response time and data transfer for performance test.

Onion yield estimation using spatial panel regression model (공간 패널 회귀모형을 이용한 양파 생산량 추정)

  • Onions are grown in a few specific regions of Korea that depend on the climate and the regional characteristic of the production area. Therefore, when onion yields are to be estimated, it is reasonable to use a statistical model in which both the climate and the region are considered simultaneously. In this paper, using a spatial panel regression model, we predicted onion yields with the different weather conditions of the regions. We used the spatial auto regressive (SAR) model that reflects the spatial lag, and panel data of several climate variables for 13 main onion production areas from 2006 to 2015. The spatial weight matrix was considered for the model by the threshold value method and the nearest neighbor method, respectively. Autocorrelation was detected to be significant for the best fitted model using the nearest neighbor method. The random effects model was chosen by the Hausman test, and the significant climate variables of the model were the cumulative duration time of sunshine (January), the average relative humidity (April), the average minimum temperature (June), and the cumulative precipitation (November).

Hormonal Requirements Induced Different Regeneration Pathways in Alhagi graecorum

  • Hormonal requirements inducing different regeneration pathways with particular emphasis on somatic embryo-genesis in Alhagi graecorum were studied. While combination of 0.5 $\mu{M}$ 2,4-dichlorophenoxyacetic acid (2,4-D), 2.5 $\mu{M}$ 6-benzylaminopurine (BAP) and 5 $\mu{M}$ 1-naphthaleneacetic acid (NAA) in MS medium induced callus formation and callus maintenance from internodal explants, each alone or in combination with other induced distinct regeneration pathway. Adventitious bud formation was induced on MS medium supplemented with 2.5 $\mu{M}$ BAP. It was improved when 2.5 $\mu{M}$ BAP was used in combination with 5 $\mu{M}$ NAA. MS medium containing 0.5 $\mu{M}$ 2,4-D or 5 $\mu{M}$ NAA induced the formation of abnormal direct somatic embryos. While increase of 2,4-D concentration (1.125-9) resulted in the formation of viable embryogenic mass, increase of NAA did not change its effect. NAA should be used in combination with 2,4-D even at low concentration (0.5 $\mu{M}$) to form embryogenic mass. In A. gaecorum, the role of 2,4-D as trigger of somatic embryogenesis and BAP as trigger of adventitious bud formation was deduced, but for maximum yield certain auxin-cytokinin ratio should be applied. Embryogenic masses characterized by high water content, low peroxidase activity, and low number of peroxidase and glutamate oxaloacetate transaminase bands in comparison with calli obtained under conditions stimulating adventitious bud formation. The resulted differential gene expression, which could be detected by native-PAGE patterns, could be used as marker for organogenic pathway in A. graecorum.
