Remaining Fatigue Life Evaluation of Steel Railroad Bridge (강철도교의 잔존피로수명 평가)

  • A systematic procedure to evaluate fatigue damages and to predict remaining fatigue lives is introduced for a steel railway bridge. Fatigue damages are evaluated by using the currently available fatigue damage theory. Fatigue lives with the condition of fatigue crack initiation are estimated by the probabilistic approach based on the reliability theory as well as the simplified procedure. A equivalent deterministic procedure is also suggested to assess the remaining fatigue life under various traffic conditions. Numerical simulations are used to assess dynamic stress histories with correction factors. Loading models are obtained from the passenger volume data. Train coincidences are also considered. Based on the results, the fatigue life is found to be underestimated by without considering the coincidence of trains on the bridge. The simplified method proposed in this study are found to yield approximately the same results as the systematic procedure.

The Removal Efficacy of Heavy Metals and Total Petroleum Hydrocarbons from Contaminated Soils by Integrated Bio-phytoremediation

  • In this study, the bio-phytoremediation and phytoremediation technologies were applied to the soils contaminated with total petroleum hydrocarbons (TPH) and heavy metals to evaluate the remediation efficacy from May 2012 to December 2013. Poplar (Populus bonatii Levl.) and Sun Hemp (Crotalaria juncea L.) were selected and planted in phytoremediation practice. These plants were also utilized in the bio-phytoremediation practice, with the addition of earthworm (Eisenia fetida) and petroleum-degrading bacteria (Pseudomonos sp. NKNU01). Furthermore, physiological characteristics, such as photosynthesis rate and maximal photochemical yield, of all testing plants were also measured in order to assess their health conditions and tolerance levels in adverse environment. After 20 months of remedial practice, the results showed that bio-phytoremediation practice had a higher rate of TPH removal efficacy at 30-60 cm depth soil than that of phytoremediation. However, inconsistent results were discovered while analyzing the soil at 100 cm depth. The study also showed that the removal efficiency of heavy metals was lower than that of TPH after remediation treatment. The results from test field tissue sample analysis revealed that more Zinc than Chromium was absorbed and accumulated by the tested plants. Plant height measurements of Poplar and Sun Hemp showed that there were insignificant differences of growth between the plants in remediation plots and those in the control plot. Physiological data of Poplar also suggested it has higher tolerance level toward the contaminated soils. These results indicated that the two testing plants were healthy and suitable for this remediation study.

Effect of Brevibacterium iodinum RS16 and Methylobacterium oryzae CBMB20 Inoculation on Seed Germination and Early Growth of Maize and Sorghum-sudangrass hybrid Seedling under Different Salinity Levels

  • Salinity is one of the most relevant abiotic factor limiting crop yield and its net primary productivity. In addition, salinity induces an increased stress ethylene synthesis in plants which, in turn, exacerbate the responses to the stressor. Bacterial single or co-inoculation effect was tested using previously characterized plant growth promoting (PGP) bacteria Brevibacterium iodinum RS16 and Methylobacterium oryzae CBMB20 on maize and sorghum-sudan grass hybrid under different concentrations of NaCl. Non-inoculated maize and sorghum-sudangrass hybrid showed 33.4% and 20.0% reduction in seed germination under highest NaCl (150 mM) level tested. However, under the same NaCl concentration, co-inoculation with B. iodinum RS16 and M. oryzae CBMB20 PGP strains increased the seed germination in maize (16.7%) and sorghum-sudangrass hybrid (4.4%). In Gnotobiotic growth pouch experiments conducted for maize and sorghum-sudangrass hybrid, co-inoculation of PGP B. iodinum RS16 and M. oryzae CBMB20 mitigated the salinity stress and promoted root length by 22.9% and 29.7%, respectively. Thus the results of this study could help in development of potential bioinoculants that may be suitable for crop production under saline conditions.

Efficient Production of Porcine Circovirus Type 2 Capsid Protein using Baculovirus

  • Porcine circovirus type 2 (PCV2) is a single-stranded circular DNA virus associated with Postweaning multisystemic wasting syndrome (PMWS), which is considered to be an important infectious swine viral disease. PCV2 capsid protein encoded by ORF2 is a structural protein and expected as the high immunogenicity protein. In this study, we generated recombinant baculovirus containing ORF2 of PCV2 and analyzed the optimal conditions for the production of capsid protein in insect cell. Production and status of recombinant capsid protein in insect cell were confirmed by SDS-PAGE and Western blot analysis using His tag antibody and anti-PCV2 serum. The yield of recombinant capsid protein was high like as shown visible on SDS-PAGE. Optimal multiplicity of infection (MOI) and infection time of recombinant virus were determined as 5 MOI and 4 days, respectively. ORF2 is known to have N-linked glycosylation site, but we couldn't detect the glycosylation of recombinant protein in insect cells.

Evaluation of nanoscale zero valent iron filled column for nitrate reduction (영가철 나노입자가 충진된 컬럼을 이용한 질산성 질소 환원 성능 평가)

  • In this study, we compared the MZVI (Microscale Zero-Valent Iron) and NZVI (Nanoscale Zero-Valent Iron) for reactivity and mobility in a column to reduce nitrate, which is a major pollutant in Korea, and investigated the effect of operational parameters on the NZVI filled column. For the comparison of MZVI and NZVI, samples were collected for 990 minutes using fractionator in the similar operation conditions (MZVI 10g, NZVI 2g). The nitrate reduction efficiency of NZVI was about 5 times higher than that of MZVI, which was about 7.45% and 38.75% when using MZVI and NZVI, respectively. In the mobility experiment, the MZVI descended due to gravity while NZVI moved up with water flow due to its small size. Furthermore, the optimum condition of NZVI filled column was determined by changing the flow rate and pH. The amount of Fe ions was increased as the pH of the nitrate solution was lowered, and the nitrate removal rate was similar due to the higher yield of hydroxyl groups. The removal rate of nitrate nitrogen was stable while flow rate was increased from 0.5 mL/min to 2.0 mL/min (empty bed contact time: 2.26 min to 0.57 min). NZVI has a high reduction rate of nitrate, but it also has a high mobility, so both of reactivity and mobility need to be considered when NZVI is applied for drinking water treatment.

Characteristics of Acid Hydrolysis Indigo Extracted from Indigo(Polygonum tinctorium L.) Leaves (쪽잎 추출 산가수분해 인디고의 특성)

  • Indigo (Polygonum tinctorium L.) is a typical blue dye which had been used from ancient times. This study was going to shade the complicated traditional methods extracting indigo dye by the fermentation and producing as adsorbate on calcium hydroxide, which says so called as the 'Indigo lime'. Accordingly we were going to make indigo through the hydrolysis of the hot water extractives of indigo leaves simply. During hot-water extraction, ${\beta}$-glucosidase which required hydrolysis of the linkage between indigo and glucose was not activated. To achieve this goal, indican was acid-hydrolyzed to glucose and indigo. The acetic acid, citric acid, hydrochloric acid, and sulfuric acid were used for the hydrolysis of hot water extractives. The hydrolysis conditions of extractives performed in water bath at $80^{\circ}C$ for 120 minutes and in an autoclave for 120 minutes. In the acid hydrolysis of extracted indican by hot water, the indican yields of acetic acid and hydrochloric acid hydrolysis were higher than sulfuric acid in water bath. Also, the indican yield of hydrochloric acid hydrolysis was better than sulfuric acid in autoclave. The hot water extracted indican was confirmed by HPLC analysis and its structure was confirmed by UV-Vis and FT-IR spectroscopy, compared with isolated indigo and commercial synthesized indigo. This improved extraction and hydrolysis methods can be replace the traditional indigo making method.

Estimating magnitude of suspended sediment transport in ungauged east coastal zone (미계측 동해안 유역의 토사유출 규모의 평가에 관한 연구)

  • Coastal sediment archives are used as indicators of changes on shore sediment production and fluvial sediment transport, but rivers crossing coastal plains may not be efficient conveyors of sediment to the coast. In some case there is a net loss of sediment in lower coastal plain reaches, so that sediment input from an upstream exceeds the sediment yield (SY) at the river mouth. The main source of sediment in coastal area is the load from land. In Korea, data on suspended SY are limited owing to a lack of logistic support for systematic sediment sampling activities. This paper presents an integrated approach to estimate SY for ungauged coastal basins, using a soil erosion model and a sediment delivery ratio (SDR) model. For applying the SDR model, a basin specific parameter was validated on the basis of field data. The proposed relationships may be considered useful for predicting suspended SY in ungauged basins that have geologic, climatic and hydrologic conditions similar to the study area.

Comparison in Productivity of Pleurotus ostreatus Sawdust Spawn Under Different Storage Conditions (저장기간에 따른 Pleurotus ostreatus 톱밥 종균의 생산성 비교)

  • The contamination rates of Pleurotus ostratus spawn after 120 days of storage at $5^{\circ}C\;and\;20^{\circ}C$ were 2.1% and 86.5%, respectively. Longer periods of storage resulted in longer culture periods at both temperatures. The yield of oyster mushroom produced from sawdust spawn stored at $5^{\circ}C$ was higher than at $20^{\circ}C$, and yields decreased with increasing storage periods.

Studies on the Production of Gluconic Acid by Resting Cell System of Aspergillus niger (Aspergillus niger의 휴지균체에 의한 Gluconic Acid생성에 관한 연구)

  • The production of gluconic acid from glucose by the resting cell system of Aspergillus niger was studied. It was found that the conversion products from glucose by the resting cell system were markedly influenced by the pH, temperature, substrate concentration, aeration, metal ions, cultivation time and storage conditions of the resting cells. Conversion products were identified as gluconic acid by the thin layer chromatography and infrared spectrophotometry. These conversions were greatly stimulated by addition of $Mg^{++}$, and S $n^{++}$, but showed inhibitory effects by C $u^{++}$, H $g^{++}$, C $d^{++}$, A $g^{+}$ and cyanide. For the optimum cell storage, it was effective to be kept at -$25^{\circ}C$ in 0.05M phosphate buffer solution of pH 7.0. The gluconic acid production by the resting cell system was more effective than those of the fermentation with respect to cultivation time, yield, recovery and re-use of the cell.l.l.l.l.l.l.

Production of Lactic Acid from 1,2-Propanediol by Yeast (1,2-Propanediol로부터 Lactic acid의 생성)

  • 1,2-propanediol-utilizing yeast, Y-1-4, was isolated from sludge sample by the enrichment culture technique. The product produced from 1,2-propanediol by the selected strain was identified as lactic acid by paper chromatography and infrared absorption spectrum. The strain assimilated ethanol, 1,2-propanediol, glycerine and glucose, but it produced lactic acid from 1,2-propanediol used as the sole carbon source. Under optimal conditions, the strain Y-1-4 was cultured with shaking at 3$0^{\circ}C$ for 4days in the medium containing 1,2-propanediol 20.0g, NH$_4$Cl 5.0g, KH$_2$PO$_4$ 1.0g, MgSO$_4$.7$H_2O$ 0.5g, FeSO$_4$.7$H_2O$ 0.25g, yeast extract 0.4g, CaCO$_3$ 3. 0g and tap water to one liter, and then the yield of lactic acid was about 12. 1g per liter of the culture broth.
