  • This paper employs the reproducing kernel (RK) approximation for evaluation of field theory-based incompatibility tensor in a polycrystalline plasticity simulation. The modulation patterns, which is interpreted as mimicking geometrical-type dislocation substructures, are obtained based on the proposed method. Comparisons are made using FEM and RK based approximation methods among different support sizes and other evaluation conditions of the strain gradients. It is demonstrated that the evolution of the modulation patterns needs to be accurately calculated at each time step to yield a correct physical interpretation. The effect of the higher order strain derivative processing zone on the predicted modulation patterns is also discussed.

  • To industrialize the foods using wormwood extract, the optimal extraction yield of growth-inhibitory component of wormwood (Artemisia princeps) against Clostridium perfringens in vitro was investigated at various conditions. The optimal extraction of wormwood was produced from the condition of $40^{\circ}C$, 4 hours and 100% ethanol (30 volume).

  • To develop functional food and anti-obesity drug through inhibition of dietary lipid absorption, inhibitory effects of herb extracts on pancreatic lipase were investigated. Due to high yield and simplicity of isolation, lipase inhibitor (ELI) was isolated from ethyl acetate extract of Eugenia aromaticum, which showed highest inhibitory activity, and characterized for development of novel functional material. Stability of ELI at high temperature and low pH was investigated. Results showed ELI is relatively stable under thermal and acidic conditions, reversible, and noncompetitive inhibitor of pancreatic lipase.

Ethanol Fermentation in Lactose Medium Using a Fusant Strain of Saccharomyces cerevisiae and Kluyveromyces fragilis

  • The fermentative characteristics in ethanol production from lactose, with increased ethanol tolerance, of a fusant yeast strain constructed by protoplast fusion of Saccharomyces cerevisiae and Kluyveromyces fragilis were studied. The ethanol tolerance of this strain was increased to 8.0%, compared with the parent K. fragilis. During batch ethanol fermentation the optimal cultivation conditions for this fusant yeast were an initial pH of 4.5, a culture temperature $30^\circ{C}$. stirring at 100 rpm without aeration in 10% lactose medium (supplied with 1.0% yeast extract). Using this fusant strain in whey fermentation to ethanol, maximum ethanol production reached 3.41% (w/v) (theoretical yield; 66.7%) after a 48 hour cultivation period.

Bacillus sp. YH-001에 의한 DL-Phenylglycine으로부터 Benzoylformic acid 의 생산

  • For the production of benzoylformic acid from DL-phenylglycine, 8 strains of bacteria capable of producing benzoylformic acid were isolated from soil. Among them, the strain YH-001 showed the highest activity for production of benzoylformic acid, and was partially identified as a Bacillus sp. For the production of benzoylformic acid, a reaction mixture and reaction conditions were optimized as follows: the reaction mixture contained 2 g of DL-phenylglycine and 50 g of wet cells in 1 l of 50 mM potassium phosphate buffer (pH 8.0) and the reaction was carried out at 40$circ$C with shaking. After 6 hr incubation, 1.01 g/l of benzoylformic acid was produced which coffesponded to a conversion yield of 50.5%.

  • In this study, purification of cold-adapted protease from Janthinobacterium sp. was investigated. First, using gradient precipitation, protease was confirmed to be deposited in the 30-80% range of ammonium sulfate. Next, DEAE-Sepharose column was used for the binding of the protease under various conditions. The optimal binding condition was found to be pH 8.5 and flow rate of 30 ml/h. Under the optimal condition, the protease was purified with 29% recovery yield. This result can be useful for the purification of other cold-adapted protein.

  • The peels of Carya cathayensis Sarg. (PCCS) were treated under subcritical water conditions ($130^{\circ}C$ to $280^{\circ}C$ for 0 to 120 min). The extract from PCCS included reducing sugar, proteins, and compounds with radical scavenging activity. Addressing the reducing sugar that is a resource of bioethanol, we could maximize the reducing sugar under the subcritical water ($190^{\circ}C$ for 60 min) and obtain 0.24 g/g-sample together with 9.7 units/mg-sample of radical scavenging activity. The obtained extract was estimated to correspond to 1 L of bioethanol/100 g-sample. It was therefore considered that the treatment by subcritical water could yield reducing sugar and natural compounds with radical scavenging activity.

  • Sesame oil is a simple pressed oil as unrefined oil. During manufacturing process of roasting-expression, benzo(a)pyrene[B(a)P] formed as a strong carcinogenic substance is cause a social problem. In manufacturing process of sesame oil, it had following the forming pathway of benzo(a)pyrene[B(a)P] as well as minimizing plan of B(a)P formation. Suitable roasting condition by roaster was during 15~20min at $220^{\circ}C$, B(a)P content in sesame oil was $1.35{\sim}1.57{\mu}g/kg$. Between roasting temperature and/or roastingtime and forming amount of B(a)P was showed a linear correlation. As a point of view the turbidity and yield of final product, roasting process of the more regular level was required.

  • A lactic acid bacterium was isolated from kimchi. The isolated strain was identified as the genus Lactobacillus through its morphological, cultural, and physiological characteristics and named as Lactobacillus sp. JC-7. The optimum conditions for the electrofusion between streptomycin (2.5 mg/ml) resistant mutant of Lactobacillus acidophilus 88 and kanamycin (600 $\mu$g/ml) resistant mutant of Lactobacillus sp. JC-7 were evaluated. The highest number of fusants were obtained at a capacitance value of 120 msec (1670 $\mu$F), a field strength of 100 V/cm, and a pulse controller setting of 72$\Omega$. The optimum pH of electroporation buffer was 7.5 and the concentration of divalent cation was 1 mM Mg$^{2+}. Electrofusants were efficiently obtained by addition 20% polyethylene glycol to elec- troporation buffer. The yield of fusion was better than that of using polyethylene glycol mediated chemical induction.

  • Conditions for the production and regeneration in Lactobacillus bulgaricus protoplasts were investigated. Protoplasts of L bulgaricus strains were obtained by treatment with mutanolysin and lysozyme together in a protoplast forming buffer containing 0.02 M N-2-hydroxyethylpiperazine-N'-2-ethanesulfonic acid (HEPES) (pH 7.0) and 0.5 M sucrose. High protoplast yield was obtained from cells cultured in the de Man, Rogosa and Sharpe(MRS) medium at the middle to late logarithmic growth phase. Regeneration was efficiently accomplished with a complex medium containing 1% sucrose, 20 mM $MgCl_2$, 5% gelatin, and 0.5% bovine serum albumin. The frequency of regeneration of protoplasts was 10~20% after 5 days of incubation at $30^{\circ}C$.
