Biodegradation of monochlorophenols by wood rot fungi such as Daldina concentrica, Trametes versicolor and Pleurotus ostreatus was evaluated by determining their resistance or toxic test and biodegradability. The metabolites of monochlorophenols were also analyzed. Among the three fungi, T. versicolor was the most resistant to 200 ppm of 2-, 3- and 4-chlorophenols, and did not show any inhibitory mycellium growth. But D. concentrica had a little inhibition effect at more than 100 ppm of 3- or 4-chlorophenol. Control cultures of P. ostreatus took even 14 days far the completion of mycellium growth, but the hyphal growth was improved when 2- or 3-chlorophenol were added to the culture. In biodegradation analysis, P. ostreatus showed the highest degradation of 2- or 3-chlorophenol, while T. versicolor was the most effective in 4-chlorophenol. D. concentrica and P. ostreatus slowly degraded 4-chlorophenol. However, T. versicolor had similar degradation capability in the three monochlorophenols, suggesting that the biode- gradation nude is dependent on the fungi as well as the type of monochlorophenol. Several metabolites such as 1,3,5-trihydroxyl benzene, 1-ethyl-1-hydroxyl pentane, 2-propenoicacid, methylmalonic acid and 2-methyl-4-keto-pentan-2-ol were found as products of primary oxidation of 2-, 3- and 4-chlorophenols by intact fungal cultures. fatty acids including tetradecanoic, heptadecanoic and octadecanoic acids were also detected The order of increase of mycellium weight during incubation were P. ostreatus > T. versicolor > D. concentrica. The pH in the culture was not constantly changed depending on incubation days, but the mycellium weight was slightly increased, indicating that the biodegradation of monochlorophenol might have low relationship with the mycellium growth Laccase activities of T. versicolor and P. ostreatus were continuously increased depending on the incubation days, suggesting that the ligninolytic enzyme activity play an important role in the biodegradation of monochlorophenol.
Lignin degrading enzymes from white rot fungi show broad substrate specificities, and therefore they can degrade variety of recalcitrant compounds. We have used three different protocols for the generation of immobilized laccase and manganese peroxidase crude enzymes from the genetically transformed strains of Merulius tremellosus Fr. These immobilized enzymes were used in the decolorization of Remazol Brilliant Blue R (RBBR), and they showed about 75% decolorization rates during the 48 h reactions. Although the decolorization efficiency decreased by 10-15% after a repeated use of the immobilized enzymes, these can be reused in various degrading reactions.
Laccases are multicopper-containing enzymes which catalyze the oxidation of phenolic and nonphenolic compounds with the concomitant reduction of molecular oxygen. They often occur as isoenzymes, either constitutive or inducible, that oligomerize to multilateral complexes, what allow for penetration to the woody cell wall structure. White rot basidiomycete fungi may produce a number of laccase isoenzymes, some constitutively and others after induction. Fungal laccase is commonly induced by many ions, such as $Cu^{2+}$, $Cd^{2+}$$Ca^{2+}$, $Li^+$, $Mn^{2+}$, $Ag^+$, $Hg^{2+}$, Mn and $Fe^{3+}$, phenolic compounds, some organic compounds, such as ethanol, isopropanol, cAMP, caffeine, p-anisidine, viscosinamide and paraquat, and nitrogens and even heat shock. A combination of Cu and pHB (p-hydroxybenzoic acid) made it possible to extend the inducible laccase activities over 30-fold. But the most effective inducer of laccase in the basidiomycete and other higher fungi is 2,5-xylidine, over 160-fold stimulation of laccase activity. The laccases are frequently encoded by gene families, as e.g. in Pycnoporus cinnabarinus, from which the lcc3-1 or the allelic form lac1 and lac3-2 have been cloned and sequenced. In the case of inducible forms the post-inductional laccase formation depends upon the synthesis of mRNA and the induction is due to the synthesis of a new protein.

In this study, the physical properties of the medium and changes in the wood chemical composition of the sawdust were investigated during the cultivation of oak mushroom sawdust bags, and the following results were obtained. After inoculation, the weight of the medium decreased during the incubation period. It is determined that this is not due to evaporation of moisture containing the medium or decomposition of sawdust, but to decomposition of rice bran, a low molecular substance added to the medium. It was confirmed that the moisture content of the medium was steadily increased during incubation, and it was estimated that the organic substrates such as rice brane in the medium was decomposed by mycelium, and water, one of the decomposition products of organic substrates, caused an increase in the moisture content of the medium. Along with the increase in the harvest of oak mushrooms, the proportion of organic substances such as holocellulose and lignin, the main components of the wood cell wall of sawdust, steadily decreased. In particular, the degradation characteristics of the wood cell wall component of shiitake, which is a white rot fungi, were confirmed by higher lignin reduction rate than that of holocellulose. On the other hand, ash, which is an inorganic material, increased with an increase in the number of mushroom harvests. The increase in the amount of ash in the medium may have been due to the decrease in the organic matter content such as holocellulose and lignin.
Laccase was applied to improve the drying speed of urushi lacquer to confirm a potential use of laccase as an enzyme additive. As a result of applying laccase of lacquer and white-rot fungi, drying times for both Korean and Chinese specimens were reduced. All of the specimens to which laccase was applied were dried(set to touch) within 60 minutes from the start of the drying, and the drying rate was improved as the content ratio of laccase increased. In addition, although there were differences in hardening drying time between Korean and Chinese lacquers, it was confirmed that hardening drying took place even at room with temperature of 20 ± 2 ℃ and humidity of 55-60%. As a result of lacquer layer analysis of the dried specimens, the drying speed was improved when the content ratio of laccase to urushiol was 5:1, and there were no differences in chromaticity and adhesion, confirmed that the layers were stable. It was observed that the gloss of both Korean and Chinese lacquers were reduced when laccase was applied. However, according to the analysis of FT-IR and Py-GC/MS, the changes in components were not as sufficient to affect the physical properties. Thus, its applicability as an additive was confirmed.
White rot fungi have been useful source of enzymes for the degradation of environmental pollutants including polycyclic aromatic hydrocarbons (PAHs) and synthetic dyes. PAHs are widespread organic compounds present in fossil fuels and are routinely generated by incomplete fuel combustion. PAHs are some of the major toxic pollutants of water and soil environments. Synthetic dyes are major water-pollutants, which are toxic to organisms in water environments and interfere photosynthesis of water plants. Removal of PAHs and synthetic dyes has been of interests in the environmental science especially in the environmental microbiology. Mushrooms are fungal groups that function as primary degraders of wood polyphenolic lignin. The ligninolytic enzymes produced by mushroom, including manganese peroxidase, lignin peroxidase, and laccase, mediate the oxidative degradation of lignin. The catalytic power of these enzymes in the degradation of aromatic ring compounds has been sought for the degradation of various organic compounds. In this project, we have screened 60 wild mushroom strains for their degradation activity against two representative PAHs, naphthalene and anthracene, and five aromatic dyes, including alizarin red S, crystal violet, malachite green, methylene blue, rose bengal. The degradation of PAHs was measured by GC while the decolorization of dyes was measured by both UV spectrophotometer and HPLC. As results, 9 wild mushroom strains showed high activity in degradation of PAHs and textile dyes. We also describe the secretive enzyme activities, the transcription levels, and cloning of target genes. In conjunction with this, activities of degradative enzymes, including laccase, lignin peroxidase, and Mn peroxidase, were measured in the liquid medium in the presence of PAHs and dyes. Our results showed that the laccase activity was directed correlated with the degradation, indicating that the main enzyme acts on PAHs and dyes is the laccase. The laccase activity was further simulated by the addition of $Cu^{2+}$ ion. Detailed studies of the enzyme system should be sought for future applications.
Sixty strains of Pleurotus ostreatus, white-rot fungi, were screened for production ability of their lignocellulolytic enzymes to selectively wood degradation. That results were shown that all of screened strains were produced lignocellulolytic enzymes on 2nd screening liquid culture medium. However, cellulase activity of selected six strains of P. ostreatus was low in avicel-yeast-peptone liquid culture medium. In the case of xylan degrading enzyme, No. 6 and No. 38 strains produced a xylanase(above 1.0U/ml) and a 1,4-${\beta}$-xylosidase (above 0.15 U/ml). Examination of the ligninolytic enzyme profiles of selected thirteen strains of the P. ostreatus, in the presence of Remazol Brilliant Blue R(RBBR), were observed that laccase(Lac) activity were earlier reached maximum level(0.8-2.0 U/ml) and then Mn-dependent peroxidase (MnP) were reached maximum level(0.5-1.5 U/ml) in glucose-yeast-peptone(GYP) medium. On the other hand, activity of lignin peroxidase(LiP) was not detected in this medium. I selected the No. 42 strain of P. ostreatus produced high levels of Mn-dependent peroxidase and laccase based on the screening method.
Wood of Robinia pseudoacacia and bark of Populus alba$\times$P. glandulosa, Fraxinus rhynchophylla and Ulmus davidiana var. japonica were collected and extracted with acetone-water(7:3, v/v) in glass jar to examine whether its bioactive compounds exist. The concentrated extracts were fractionated with hexane, chloroform, ethylacetate and water, and then freeze-dried for column chromatography and bioactive tests. The isolated compounds were sakuranetin-5-O-$\beta$-D-glucopyranoside from Populus alba $\times$Pl glandulosa, 4--ethyoxy-(+)-leucorobinetinidin frm R. pseudoacacia and fraxetion from F. rhynchophylla and were characterized by $^1H$ and$^{13}C $ NMR and positive FAB-MS. Decay-resistant activity was expressed by weight loss ratio and hyphae growth inhibition in the wood dust agar medium inoculated wood rot fungi. R. pseudoacacia showed best anti-decaying property in both test and its methanol untreated samples, indicating higher activity than methanol treated samples in hyphae grwoth test. In antioxidative test, $\alpha$-tocopherol, one of natural antioxidants, and BHT, one of synthetic antioxidants, were used as references to cmpare with the antioxidant activities of the extacted fractions. Ethylacetate fraction of F. rhynchophylla bark indicated the hightest activity in this test and all fractions of R. pseudiacacia extractives also indicated higher activities compared with the other fractions. In the isolated compounds, aesculetin isolated from F. rhynchophylla bark showed best activity and followed by robonetinidin from R. pseudoacaica.
