  • The various culture conditions of Trichoderma viride(ATCC 3454) and Phanerochaete chrysosporium(ATCC 26921) with glucose-pepton medium, Mandels medium, YMG medium for wood degradable enzyme were examined. Mycellium of the two species grew profusely on glucose-pepton medium. Maximum fungal growth was observed about 10days. But CMCase, Fpase, laccase activity in the culture medium with glucose-pepton was not detected. When grown in fermenter culture using Mandels medium, Trichoderma viride produced CMCase and Fpase. Its CMCase activity was 0.15 lU/ml and Fpase activity was 0.3 IU/ml within about 4-6days. Phanerochaete chrysosporium grown in a YMG medium gave the best enzyme activity when they were grown under stationary culture with an atmosphere of 100% oxygen. Levels of laccase activity of 3.0 mull were achieved in stationary culture under 100% oxygen. The enzyme condensation by ultrafiltration method caused a 2-fold(cellulase) and 6-fold(laccase) as compared to control activity.

목판 교환 방법을 활용한 다색 협힐 제작기법에 관한 연구 (A study of multicolored clamp resist dyeing techniques using a wooden printing-block exchange method)

  • The aim of this study is to define the new expressive techniques for multicolored clamp resist (hyuphill) dyeing, based on empirical verification on relics that are estimated to be dyed by the exchange of more than two wood blocks: a previously undiscovered technique. Clamp resist dyeing (assumed to be made by exchanging wood blocks) have uneven resist printing lines or cloudy gradation. These are reproduced as follows: first, they have uneven contour lines, particularly with the color blue. It is possible to exchange wood blocks separately on patterns with uneven resist printing lines, and it has been verified that the exchange of wood blocks makes these irregular resist printing lines. It has also been verified that exchanging the wood blocks according to the gradation (to emphasize the cubic effect on the patterns) yields clamp resist dyeing with no resist printing lines but with cloudy gradations that have accented borders. This study provides basic information that enables methods of multicolored clamp resist dyeing through wood block exchange to be deduced (something that has not been attempted for a long time). Thus, the revival of the modern Korean dyeing culture based on the conservation and perseverance of the traditional dyeing techniques can be achieved.

목초액의 향균 및 DPPH 라디칼 소거 활성에 관한 연구 (Study of Antimicrobial and DPPH Radical Scavenger Activity of Wood Vinegar)

  • The antimicrobial and antioxidative activities were investigated to confirm the utility of wood vinegar. Antimicrobial activity was performed by paper disc method and liquid culture. The growth inhibition was observed in all microbial species at a dose of as low as 25 $\mu$m of wood vinegar by paper disc method. Also, in liquid culture, S. cerevisiae and P. aeruginosa were more inhibited the growth than others in the concentration of $2\%$ (v/v). For measuring of antioxidative activity, wood vinegar was fractionated with acidic, phenolic, basic and neutral fraction; and their antioxidant activities were measured by the radical scavenging effect on DPPH radical. In four fractions, phenolic fraction showed high antioxidative activity.

The Production of Methyl Trans-cinnamate in the Submerged Cultures of Tricholoma matsutake Mycelia

  • Methyl trans-cinnamate is a significant flavor compound of Tricholoma matsutake. Attempts were made to produce this compound by culturing the mycelium using submerged culture. No methyl trans-cinnamate could be detected when the mycelium was cultured in the basal liquid medium. However, the addition of Pinus desiflora extracts to the medium, methyl trans-cinnamate was largely produced. To find out compounds or fractions inducing methyl trans-cinnamate, dichloromethane fraction obtained from the wood extracts of P. densiflora was subjected to column chromatography. Three sub-fractions were obtained from the $CH_2CI_2$ fraction. Submerged cultured mycelium treated with sub-fraction 1 has the highest content of methyl trans-cinnamate. Maximum methyl trans-cinnamate $(470.2{\mu}g/g)$ was obtained when the first sub-fraction of dichloromethane fraction of the P. densiflora wood extracts was added to the medium. This indicates that wood extracts of P. densiflora contains inducer of the methyl trans-cinnamate production in the T. matsutake submerged culture.

In Vitro Mycorrhization and Morphological Characterization of Xanthoconium affine with Pinus densiflora

  • In this study, we investigated the culture conditions of four ectomycorrhizal fungi, namely, Amanita spissacea NIFoS 2719, Pisolithus arhizus NIFoS 2784, Suillus spraguei NIFoS 2848, and Xanthoconium affine NIFoS 2716, in solid and liquid culture media. In addition, we attempted to induce in vitro mycorrhization of the fungi with Pinus densiflora. Prior to liquid culture, we determined the optimal culture conditions for each species in solid media. The results revealed that all species examined are capable of growth in potato dextrose agar (PDA), malt extract agar (MEA), and modified Melin-Norkran's medium (MMN), although their preferred growth media were different. Liquid culture experiments showed that inorganic nitrogen did not enhance the mycelial growth of all four species. Therefore, we used MMN-based liquid inocula to promote the growth of ectomycorrhizal fungi in our symbiosis culture system. Mycorrhization was observed in Xanthoconium affine NIFoS 2716. Morphological analysis revealed that fungi-inoculated roots of P. densiflora form simple and dichotomous lateral roots with dense mycelia. In addition, inoculation with X. affine NIFoS 2716 promoted root and shoot developments.

Optimization of in Vitro Cultivation of Inonotus Obliquus

  • This study was performed to get the basic information concerned to the optimum culture condition of Inonotus obliquus. Several solid media, PDA, MEA and Czapek-Dox, and three liquid media were adopted for the in vitro cultivation. Some main features of the fungal morphological characteristics under cultivation conditions were observed and described. Preliminary results showed that appearance of the mycelial mat, hyphal size and substrate pigmentation differed according to the media. The PDA medium was the most favorable substrate for the growth on solid culture, followed by MEA and Czapek-Dox media. Concerned to the addition of amino acids, 5 amino acids, such as alanine, alginine, isoleucine, leucine and threonine, enhanced to the mycelial growth. Isoleucine was shown the best fungal growth. An important morphological hyphal structure for the fungus, the setae, was found in abundance and diverse its shape and size. In liquid culture, fresh potato broth was the best growth stimulant of the fungus, followed by Malt extract and potato broth. Addition of yeast extract to the liquid media had improved the biomass, but not laccase production.

Decolorization of Dyes by Selected Wood Degradation Fungus

  • The objectives of this study were to select superior fungus for lignin degradation and to decolor dyes by selected fungus. Ligninolytic fungi were screened and isolated from decayed woods. Ten ligninolytic fungi were selected by ligninolytic enzyme activity on the PDA media containing rhemazol brilliant blue R, guaiacol and gallic acid. Their lignin degradation abilities were tested on the extractive-free wood powder of Quercus acutissima and Pinus densiflora. As a result, 8J-28 was selected as superior fungus for lignin degradation. Also, decolorization abilities of dyes were examined by shaking and static culture. And congo red, crystal violet, poly R-478, methylene blue used to investigate decolorization abilities of dyes. As a result, 8J-28 showed over 90% in decolorization of congo red, crystal violet, poly R-478.

Tyromyces palustris를 이용한 구리의 제거 (Copper Uptake by Tyromyces palustris)

  • In this research, the removal or uptake of heavy metals such as coppers by using oxalic acid metabolism of wood rot fungi, Tyromyces palustris were endeavored. As results, the addition of oxalic acid to copper containing culture did not cause the mycelium growth, but Tyromyces palustris was able to grow in this culture without inhibition. Tyromyces palustris grew with the cicular halo type in copper containing culture, and this type was formed as collectives after examining by microscope, and considered as copper oxalates by analyzing FT-IR comparison experiment with standards. According to this result, Tyromyces palustris has secreted oxalic acid during incubation, this secreted oxalic acid was combined with coppers, and formed copper oxalates by chelating reactions. In other words, the oxalic acid was might be as non-toxifying agent of coppers in medium. By using this copper removal mechanisms, Tyromyces palustris immobilized sawdust was used in bench scale air lift system for removing coppers. The added coppers were almost removed from the system within 72hrs. Therefore, this nonenzymatic wood degradation mechanism may give a possibility for removing coppers from copper containing waste water.

Biocontrol of Blue Stain in Pine Wood with Lyophilized Mycelium of Ophiostoma quercus Albino Strain

  • Mycelium of Ophiostoma quercus albino strain cultured in liquid culture media was harvested, lyophilized, and stored for examining biocontrol efficacy against wood discoloration by staining fungi in the laboratory and field conditions. Dry weight of mycelium grown in brown sugar yeast extract broth(BYB) showed 3.8 times higher than that grown in potato dextrose broth(PDB). The optimum culture period in BYB was 4 weeks. In vitality test of the albino strain, the lyophilized mycelium stored in liquid nitrogen($-196^{\circ}C$) or in a refrigerator($4^{\circ}C$) kept the vitality until 13 months after storage; however, the mycelium stored at room temperature lost the vitality completely after 13 months. The mycelium stored in liquid nitrogen or in a refrigerator protected wood chips from the discoloration by pretreating mycelial suspension on pine wood chips. The mycelium stored at room temperature for 7 months also showed complete protection. These results suggest that the lyophilized mycelium have a biocontrol efficacy only if it keeps the least vitality. In the field conditions, both albino strain and $Woodguard^{(R)}$(commercial chemical protectant) showed significant differences(p=0.05) in discoloration rate as compared to the non-treated control when these were treated on the wood logs of Pinus rigida. The albino strain showed better protection than $Woodguard^{(R)}$. Isolation frequency of blue stain fungi from the chips of wood logs treated with the albino strain was 0% at three months after treatment, while that treated with $Woodguard^{(R)}$ was 76.7%. In another experiment, pre-treatment of mycelial suspension on the cut surface of wood logs also showed significant protection from wood discoloration. Spraying of both albino strain on the cut surface and insecticides on the bark also showed relatively good control effects as compared to insecticide alone on the bark or nontreated control.
