Quercetin Glycosides from Bark of Maple (Acer komarovii Pojark.)

  • The chemical constituents of Acer komarovii Pojark. which belongs to Aceraceae has never been reported. The bark of A. komarovii was extracted with 70% aqueous acetone, and the concentrated extract was successively partitioned with n-hexane, dichloromethane, ethyl acetate and $H_2O$. From the ethyl acetate soluble fraction, four compounds were isolated by the repeated Sephadex LH-20 and RP C-18 column chromatography. From the results of spectroscopic methods including FAB-MS, 1D and 2D NMR, the structures of the compounds were determined as quercetin (1), guaijaverin (2), hirsutrin (3) and hyperin (4). These compounds (1-4) have not been reported in this tree yet.

  • This study was carried out to investigate the constituents of Sophora japonica(Leguminosae) bark. To isolate compounds, bark was extracted with ethanol and then partitioned with hexane, dichloromethane, ethylacetate and butanol successively. After partitioned, DCM fraction was subjected to column chromatography with various solvent system in silica gel and/or Sephadex LH-20. Structures were elucidated by spectroscopic methods including MS, 1H, 13C and 2D-NMR experiments. Four compounds were isolated from the bark and identified as 3',7-dihydroxy-4'-methoxyisoflavone (cyclosin), puerol A, maackiain-3-O-𝛽-D-glucopyranoside (trifolirhizin), and 4', 5, 7-trihydroxyisoflavone (genistein). Among these compounds, cyclosin and trifolirhzin were first isolated from S. japonica.

A curvature method for beam-column with different materials and arbitrary cross-section shapes

  • This paper presents a curvature method for analysis of beam-columns with different materials and arbitrary cross-section shapes and subjected to combined biaxial moments and axial load. Both material and geometric nonlinearities (the p-delta effect in this case) were incorporated. The proposed method considers biaxial curvatures and uniform normal strains of discrete cross-sections of beam-columns as basic unknowns, and seeks for a solution of the column deflection curve that satisfies force equilibrium conditions. A piecewise representation of the beam-column deflection curve is constructed based on the curvatures and angles of rotation of the segmented cross-sections. The resulting bending moments were evaluated based on the deformed column shape and the axial load. The moment curvature relationship and the beam-column deflection calculation are presented in matrix form and the Newton-Raphson method is employed to ensure fast and stable convergence. Comparison with results of analytic solutions and eccentric compression tests of wood beam-columns implies that this method is reliable and effective for beam-columns subjected to eccentric compression load, lateral bracings and complex boundary conditions.

  • 북가주에 있는 삼나무 수종에서 cadinane과 acorane류의 sesquiterpenoids를 chromatography를 사용하여 분리한 후, 화학적인 방법과 분광학적인 방법을 이용하여 분리된 성분의 화학구조를 규명하였다. 그 sesquiterpenoids의 생합성 경로를 정량적인 상관성에 의하여 밝혔다. 또한, 여러 삼나무 수종에 존재하는 sesquiterpenoids의 분류학적 차이에 대하여 논하였다.

Protease Activity from Fruit Body of Sarcodon aspratus (능이자실체의 Protease 활성)

  • This study was performed to investigate the protease activity from fruit body of Sarcodon aspratus and its features. The specific protease activity was increased with the increasing purification steps, 2.62 times by desalting, 17 times by CMC column chromatography, 113.8 times by DEAE-Sephadex A-50 column chromatography, and 728.3 times by Sephadex G-75 column chromatography. Proteases were identified as two different enzymes having different isoelectric points at pH 4.35 (its recovery rate 8%) and pH 4.7 (its recovery rate 3.5%). Those proteases were purified by 3,025 folds and 3,257 folds in terms of specific activity. Two proteases having different isoelectric points had similar enzymatic properties. This protease was estimated to be 43,000 daltons of molecular weights by SDS-PAGE. This protease with optimum pH 4 was almost stable in the pH range of 4~7. Optimal temperature of protease activity was 40 to 50℃, and the protease activity was completely inhibited at 70℃ for 30 min.

The comparative analysis of smoke components delivered from cigarette papers manufactured by flax and wood pulp (궐련지의 섬유종류가 담배 연기성분에 미치는 영향)

  • This study was conducted to evaluate the effect of cigarette papers, flax and wood, on the delivery of mainstream smoke. The main components of cigarette papers were cellulose, hemicellulose, lignin, and pectin. Lignin contents, known as precursor of smoke's phenolic compounds, of the flax and wood cigarette papers were 5.8% and 10.6%, respectively. The pyrolysis products of cigarette papers were similar by the profile of total ion chromatogram. But, the area % of some components, such as 1,3-cyclopentanedione, 3,5- dimethyl cyclopentane-1,2-dione, 2-hydroxy-3-methyl-2-cyclopentenone, dihydro-2(3H)-furanone, 3-methyl-2(5H)-fruanone, and 5-methyl-2-furaldehyde delivered through pyrolysis of the flax cigarette paper were higher than that of wood cigarette paper. Otherwise, the area % of some components, such as 2-methyl-cyclopentene-l-one), 2,3-butanedione, 2-cyclopentene-l-one, and 5-hydroxy-2-methyl-furaldehyde, 2-furaldehyde delivered through pyrolvsis of the wood cigarette paper were higher than that of flax cigarette paper. To identify the difference between two cigarette papers, we used the cigarette column filled with the cut cigarette paper instead of the cut tobacco leaf. The amounts of semi-volatile fraction delivered from flax cigarette paper was more than that of wood cigarette paper. But, by using the cut tobacco, there was no big difference of delivery amount between flax and wood cigarette papers. Also, aroma of TPM by collecting from brening cut tobacco wrapped in flax and wood papers showed a different pattern by the electonic nose system. Although the difference between two cigarette papers by using the cut tobacco was smaller than that of cut cigarette paper, this result indicated that the fax and wood had the different effects on the delivery of smoke components as shown in the sensory test results.

Theoretical Modeling of the Resonant Column Testing with the Viscosity of a Specimen Considered (점성을 고려한 공진주 실험의 이론적 모델링)

  • The resonant column testing determines the shear modulus and material damping factor dependent on the shear strain magnitude, based on the wave-propagation theory. The determination of the dynamic soil properties requires the theoretical formulation of the dynamic behavior of the resonant column testing system. One of the theoretical formulations is the use of the wave equation for the soil specimen in the resonant column testing device. Wood, Richart and Hall derived the wave equation by assuming the linear elastic soil, and didn't take the material damping into consideration. Hardin incorporated the viscoelastic damping of soil in the wave equation, but he had to assume the material damping factor for the determination of the shear modulus. For the better theoretical formulation of the resonant column testing, this study derived a new wave equation to include the viscosity of soil, and proposed an approach for the solution. Also, in this study, the equation of motion for the testing system, which is another approach of the theoretical formulation of the resonant column testing, was also derived. The equation of motion leads to the better understanding of the resonant column testing, which includes the dynamic magnification factor and the phase angle of the response. For the verification of the proposed equation of motion for the resonant column testing, the finite element analysis was performed for the resonant column testing. The comparison of the dynamic magnification factors and the phase angles far the system response were performed.

A Study on the Component Analysis of Sappan Wood Extracts (소목 추출물의 구조분석)

  • Colorants were extracted from the heartwood of sappan lin. with MeOH under reflux, and the concentrate or the powder of dye was prepared by low pressure concentration method using suitable organic solvent. Various components were isolated from sappan wood, and the chemical structure and mechanism of compound having the excellent antibacterial and deodorization properties were analyzed. The results obtained are as follows ; The seventeen components of sappan wood were seperated by HPLC chromatography, and the five components among them were existed more than 6% and the other components were existed lower than 0.6%. The resolving powers of the non-polar solvent and polar solvent systems were evaluated by their ability to resolve the samples. It showed that chloroform-methanol-water(800:150:10) system has the best resolving power. Although the seperation rate is very slow, polyamide C-100 column chromatography gives a clear seperation of sappan wood. On the basis of the spectrometric data such as IR, UV, $GC-Mass,\;^1H-NMR,\;^{13}C-NMR\;and\;^1H-^{ 13}C-NMR$, the chemical structure of compound haying the excellent antibacterial and deodorization properties was established as brazilin containing the functional groups such as two quaternary carbon, one benzyl carbon, methylene contiguous to oxygen and methylene caused by oxygen atom.

The Production of Methyl Trans-cinnamate in the Submerged Cultures of Tricholoma matsutake Mycelia

  • Methyl trans-cinnamate is a significant flavor compound of Tricholoma matsutake. Attempts were made to produce this compound by culturing the mycelium using submerged culture. No methyl trans-cinnamate could be detected when the mycelium was cultured in the basal liquid medium. However, the addition of Pinus desiflora extracts to the medium, methyl trans-cinnamate was largely produced. To find out compounds or fractions inducing methyl trans-cinnamate, dichloromethane fraction obtained from the wood extracts of P. densiflora was subjected to column chromatography. Three sub-fractions were obtained from the $CH_2CI_2$ fraction. Submerged cultured mycelium treated with sub-fraction 1 has the highest content of methyl trans-cinnamate. Maximum methyl trans-cinnamate $(470.2{\mu}g/g)$ was obtained when the first sub-fraction of dichloromethane fraction of the P. densiflora wood extracts was added to the medium. This indicates that wood extracts of P. densiflora contains inducer of the methyl trans-cinnamate production in the T. matsutake submerged culture.

Demethoxylation of Milled Wood Lignin and Lignin Related Compounds by Laccase from White-rot Fungus, Cerrena unicolor

  • Highly purified Cerrena unicolor laccase (benzenediol:oxygen oxidoreductase, EC 1.10.3.2) caused the demethoxylation of milled wood lignin and several lignin related substances. The constitutive form of the enzyme produced extracellularly by C. unicolor fermenter culture was isolated and purified by ion-exchange chromatography on the DEAE-Toyopearl column and by affinity chromatography on a ConA-Sepharose and Syringyl-AH-Sepharose 4B columns. The enzyme was further immobilized on functionalized porous glass (CPG) and keratin coated CPG. The demethylating activity was monitored both by estimation of released methanol and by detection of the level of methoxyl groups (also in some water miscible solvents) after incubation of lignin materials with laccase preparations (free and immobilized). The effects of the incubation time and temperature on the demethoxylating activity of immobilized laccase preparations were also studied.
