Prevalence of antibody against 38 kDa outer membrane protein of Yersinia enterocolitica in swine (국내 사육돼지에서의 Yersinia enterocolitica 38 kDa outer membrane protein에 대한 항체가 분포)

  • Yersinia enterocolitica is an inhabitant in the lower intestinal tract of many domestic and wild animals as well as in the nature. Of the several forms of diseases caused by Y. enterocolitica, an acute enteritis, especially in young children, is the most common form. Infection of the bacteria usually occurs through fecal-oral route by contaminated foods or water, especially mountainspring water. Of the domestic animals, swine has been known as one of the most important carrier in the human infection. Based on the knowledge, prevalence of antibody against Y enterocolitica was investigated with swine sera collected from Korea for the survey of Y enterocolitica infection in swine. As the first step of this survey, we analyzed outer membrane protein (OMP) profiles of the representative strains of Y enterocolitica isolated from the feces of piglets and mountainspring water in Korea. Thirty-eight kDa OMP was identified as the common OMP regardless of origin, serotype, or biotype of Y enterocolitica isolates. Presence of antibody specific for 38 kDa OMP of Y enterocolitica in 1,076 swine sera collected from November 1999 to October 2000 was analysed with ELISA. Antibody titer in sows was significantly higher than that in piglets, growing pigs and finishing pigs (p<0.05). Also, there was seasonal difference in the prevalence of antibody against Y enterocolitica. These results would provide the basic knowledge for controlling the Y enterocolitica infection in human as well as swine.

  • All of the animals and the plants in ecosystem are intimately connected to one another and the changes of forests and surroundings affect directly wild animals. This study was conducted at Hangyeong-myeon Cheongsu-ri located in the western part of Jeju Island belonging to Hangyeong Andeok Gotjawal Zone and Jocheon-eup Seonheul-ri located in the eastern part of Jeju Island belonging to Jocheon Hamdeok Gotjawal Zone. The survey on advent of birds was carried out twice a month from January 2014 to December 2015. We divided habitat environments into three survey sites such as a forest, a shrub forest and a farmland. A total of 65 species and 4,802 individuals were observed during the survey period. In a forest, 36 species and 1,287 individuals were observed while A shrub forest had 40 species and 1,554 individuals. And in a farmland, 41 species and 1,961 individuals were observed. The only 10 species were observed in forest and the only 7 species in shrub forest and the only 10 species in farmland. The species diversity and the evenness of a farmland were the highest, and the species richness was the highest in a shrub forest, and the dominance of a forest was the highest among the three areas. The similarity index between a shurb forest and a farmland was high while that between a forest and a farmland was low. The similarity index related with breeding appeared that a forest and a farmland was different from each other.

  • Nowadays, mankind is suffering from serious pollutions. In proportion to industrial modernization, polltional problems have been increased and arised new one endlessly. Todays, we are cope with a newkind pollution materials threatening the life of mankind with root out. It is Environmental Hormon, that is, Endocrine Disrupting Chemical(EDs). EDs was made by dioxin, PCB, DDT, cadmium, lead, ether and even diethylstillbestrol(DES) that used to contraceptive pill etc. These EDs makes changing from normal human hormon to abnormal one and it kill off or fade out spermatozoa. Then the spermatozoa was infected with EDs makes man imposex, sexual malformations and infertility also. It is the more critical phenomenons that EDs invade into the field of wild-life; birds, invertebrate animals, fishes, mammalia especially. England TV(BBC) journalist Cadury, D. introduced a lot of case studies that many kind of male animals was changed feminization by EDs in her book (The Feminization of Nature;1997). Otherwise, Gibbs, P.E. and Bryan, G. W. reported about dog-whelk snail (a sort of sea shell) was changed gender from male to female by EDs in their study. Briefly speaking, Environmental Hormon (EDs) stem from polluted materials maloperate human and animal informational codes of hormon. It is a significant event to an individual ontogeny and ecological system. To overcome these problem WHO, OECD, UNEP held many work-shops, international seminars and researches but it is not enough yet. On May of 1997, the International Meeting of Environmental Administrators makes a $\ulcorner$Declaration on EDs Problems$\lrcorner$ for solving it fortunately and NHK(japan) reported Super Critical Fluid as a new materials for decrease toxicity of dioxin in recent. Meat Eating Practice in Korea (한국의 육식문화)

  • Koryo Dynasty has greatly effected the meat eating practice in Korea. And by focusing on this period, this paper has in vestigated how this practice influenced and effected our meat eating culture. The 'Orders to Prohibit Butchery' written on Koryo's history books were to encourage stockbreeding rather than to follow the Buddhist policy they followed. By encouraging stockbreeding, they wanted to promote the usage of cattle in farming and thus increasing agriculture industry as a whole. Nonetheless, records show that hunting was permitted to a certain degree. And this allowed the civilians to depend their meat supply from hunting and for the fire field farmers to capture wild animals that harmed their crops. Moreover, through 'Kiwujae' (Kiwujae - a shamans service to pray for rain / ritual (praying) for rain.), we could see that earlier part of Karyo's rituals and ceremonies followed the Buddhist tradition while the latter followed the Shamanism tradition. Perhaps this was the result of allowing 'meat' for the service offerings. As Shamanism could be considered as a religion that allowed 'meat', prevalence of Shamanism was promoting meat-eating at mess(after these rituals and ceremonies that offered food (meat inclusive) to their guardian or god, the civilians would dine together.). In relation, this public eating practice slowed down the progress for storage technique. Therefore, meat-eating was developed through public and mass dining rather than through the form of family or private. On this account, we can safely regard meat-eating practice as a 'public event'. On the other hand, the history of castration is not so long in Korea. And the purpose of such practice was to use the stock for farming rather than to yield high quality meat. It is known that Mongol in Koryo period has greatly influenced meat cooking in Korea. And the exemplary dish is the 'tang' (tang - kind of soup. However less creamy, clearer broth and with more ingredients than soup.). However, the tang we ate in everyday life had the same cooking method as the tang we offered for services. Moreover, since we did not use castrated animals for our offering as the Mongolians, we must not have been greatly influenced by them. But if so, perhaps the influences would have been limited to the nobility.

Variability of laying hen behaviour depending on the breed

  • Objective: For many generations, most species of farm animals have been subjected to intense and strictly targeted selection for improvement of their performance traits. This has led to substantial changes in animal anatomy and physiology, which resulted in considerable differences between the current animal breeds and their wild ancestors. The aim of the study was to determine whether there is breed-specific variability in behaviour as well as differences in emotional reactivity and preferences of laying hens. Methods: The investigations involved 50 Green-legged Partridge, 50 Polbar, and 50 Leghorn hens. All birds were kept in the same conditions, and the behavioural tests were carried out at 30 weeks of age. We used the tonic immobility test and a modified open-field test including such objects as water, commercial feed, feed enriched with cereal grains, finely cut straw, and insect larvae, a sandpit, a mirror, and a shelter imitating a hen nest. Results: The research results demonstrate that the birds of the analysed breeds differ not only in the excitability and emotional reactivity but, importantly, also in the preferences for environment-enriching elements. Ensuring hens' well-being should therefore be based on environmental modifications that will facilitate acquisition of essential elements of chickens' behaviour. The greatest emotional reactivity was found in the Leghorn breed, which may be a result of correlated selection aimed at an increase in chicken productivity. Conclusion: The differences in the behaviour of the birds from the analysed breeds indicate that laying hens cannot be regarded as one group of animals with the same environmental requirements.

Isolation and identification of mammalian orthoreovirus type 3 from a Korean roe deer (Capreolus pygargus)

  • Mammalian reovirus (MRV) causes respiratory and intestinal disease in mammals. Although MRV isolates have been reported to circulate in several animals, there are no reports on Korean MRV isolates from wildlife. We investigated the biological and molecular characteristics of Korean MRV isolates based on the nucleotide sequence of the segment 1 gene. In total, 144 swabs from wild animals were prepared for virus isolation. Based on virus isolation with specific cytopathic effects, indirect fluorescence assays, electron microscopy, and reverse transcription-polymerase chain reaction, only one isolate was confirmed to be MRV from a Korean roe deer (Capreolus pygargus). The isolate exhibited a hemagglutination activity level of 16 units with pig erythrocytes and had a maximum viral titer of 105.7 50% tissue culture infectious dose (TCID50)/mL in Vero cells at 5 days after inoculation. The nucleotide and amino-acid sequences of the partial segment S1 of the MReo2045 isolate were determined and compared with those of other MRV strains. The MReo2045 isolate had nucleotide sequences similar to MRV-3 and was most similar (96.1%) to the T3/Bat/Germany/342/08 strain, which was isolated in Germany in 2008. The MReo2045 isolate will be useful as an antigen for sero-epidemiological studies and developing diagnostic tools.

Genetic Analysis of H7N7 Avian Influenza Virus Isolated From Waterfowl in South Korea in 2016 (2016년 한국 야생조류에서 분리한 H7N7 조류인플루엔자 바이러스 유전자 분석)

  • Type A influenza virus is circulating in wild birds and can infect wide ranges of hosts such as humans, pigs, domestic birds, and other mammals. Many subtypes of avian influenza viruses are circulating in aquatic birds. Most avian influenza viruses found in aquatic birds are low pathogenic avian influenza viruses. Highly pathogenic avian influenza viruses have been found in waterfowls since 2005. It is known that H5 and H7 subtypes of avian influenza viruses can be mutated into highly pathogenic avian influenza viruses in domestic poultry. In this study, we isolated novel reassortant H7N7 avian influenza virus from the fecal materials of migratory birds in the Western part of South Korea in 2016, and analyzed the sequences of all its eight genes. The genetic analysis of our isolate, A/waterfowl/Korea/S017/2016 (H7N7) indicates that it was reassortant avian influenza virus containing genes of both avian influenza viruses of wild birds and domestic ducks. Phylogenetic analysis showed that our isolate belongs to Eurasian lineage of avian influenza virus. Since avian influenza viruses continue to evolve, and H7-subtype avian influenza virus can mutate into the highly pathogenic avian influenza viruses, which cause the great threat to humans and animals, we closely survey the infections in both wild birds, and domestic poultry, and mammals.

Amphibian Biotope Planning to Research on Reproduction of Bombina orientalis (무당개구리(Bombina orientalis) 번식 연구를 위한 양서류 바이오톱(amphibian biotope) 조성)

  • Bombina orientalis, or Korean fire-bellied toad is belonging to Class Amphinia, Order Anura, Family Bombinatoridae, Genus Bombina. Ecological characteristics, such as small size, slow moving and swimming, gentle attitude to other species, under laboratory and/or wild condition lead the toad as suitable experimental animals with easy handling. Seasonal reproductive biology except breeding season (usually from Apr. to late Jun.) are unknown still caused by sampling methods for hibernating specimens have not been established yet. Most species of amphibian, include B. orientalis, are declined around the world related with global warming, therefore, wild caught specimen for any purposes must be limited controled soon. Even though, some pre-studies of biotope for increasing biodiversity have been reported, construction of an artificial biotope to get natural breeding cycling specimen for study of amphibian reproduction is unique modelling and only trial. In an effort to set year-round sampling for study of reproduction of B. orientalis, we suggest protocols for planning, construction manual and SOP(Standard Operation Process) to manage amphibian biotope. During pilot operation of a designed and constructed biotope, the seasonal acclimation during winter hibernation of B. orientalis were successful. It will be helpful for studying the reproductive biology of wild amphibians in temperate region and allow to get reasonable research conditions in amphibian laboratory.

Screening the level of cyanogenic glucosides (dhurrin) in sorghum accessions using HPLC analysis

  • Sorghum (Sorghum bicolor (L.) Moench.) is one of the most important crops for human and animal nutrition. Nonetheless, sorghum has a cyanogenic glucoside compound which can be degraded into hydrogen cyanide, toxic to humans and animals even with tiny amount. In consequence, breeding materials with a low cyanide level has been a top priority in sorghum breeding programs. To fulfill our long-term goal, we are screening sorghum accessions with low cyanide level, which would be an important breeding material for food safety. We collected seeds of various sorghum accessions and analyzed relevant metabolites to find useful breeding materials of sorghum accessions containing low cyanide. Fourteen wild relatives were obtained from the University of Georgia in US, a reference accession BTx623, and three local varieties from National Agrobiodiversity Center of Rural Development Administration in Korea, and one wild species from the Wild Plant Resources Seed Bank of Korea University in Korea. Sorghum plants were grown in plastic greenhouse under natural conditions. After growing, leaf samples were harvested at different developmental stages: seedling phase, vegetative phase (right before flowering), and reproductive phase (ripening). Using collected samples, quantification analysis were performed by an HPLC system for three metabolites (dhurrin, 4-hydroxybenzaldehyde, and 4-hydroxyphenylacetic acid) in sorghum plants. Prior to metabolome analysis, specific experimental condition for HPLC system was set to be able to separate three metabolites simultaneously. Under this condition, these metabolites were quantified in each accession by HPLC system. We observed that the metabolite contents were changed differently by developmental stages and accessions. We clustered these results into five groups as patterns of their contents by developmental stages. Most of accessions showed that 4-hydroxybenzaldehyde content was very high at seedling stage and decreased rapidly at vegetative phase. Interestingly, the patterns of dhurrin content were very different among clusters. However, 4-hydroxyphenylacetic acid content was maintained at low levels by developmental stages in most accessions. The results would demonstrate how dhurrin and alternative degradation pathways are differentiated in each accession.

A Study on the Symbolism of Auspicious Animal Sculptures Installed on Woldae(月臺) Stone Railing at the Geunjeongjeon(勤政殿) of Gyeongbokgung(景福宮) - Based on Twenty-eight Mansions - (경복궁 근정전 월대 석난간에 설치된 서수 조각물의 내용 및 상징적 의미 연구 -28수 관련 내용을 중심으로-)

  • There are stone sculptures diversely shaped in several parts of Gyeongbokgung. Geunjeongjeon is a place where the stone sculptures are intensively placed among the various palace buildings of Gyeongbokgung, and it is more important because it contains the phases of the age of the dynasty and the symbolism of governing philosophy. What can be seen in the structure of auspicious animal sculptures is based on the form of instinctive thought that the heavenly and human order systems are in an organic correspondence with each other, especially with regard to astronomical events such as Samwon Sasin 12jisin 28su. The parts that have been interpreted only in Sasin and 12jisin in the interpretations such as wild dogs, Gyoryong, camels, wolves, and apes were able to clarify the contents by revealing the form of 28su. In particular, as Beopsu Ssangbeopsu at the corner may be related to the guardian deity who governs water, called Yimun, Chimi, and Chimun, it is one of Bibo's ways to protect the palace that was vulnerable to fire. In addition, the shape of the existing 28su placed Dambi, but Ha Woldae at the Geunjeongjeon was assumed to have a camel statue and a planned arrangement of double meaning with the Bibo form of Pungsu Sasinsa, and it is also a feature of the arrangement of Woldae at the Geunjeongjeon. The actual composition of auspicious animals at the Nambogye of Geunjeongjeon was in the order of Haechi, wild dogs, horses and Jujak, and the contents of "Gyeongbokgung Construction Daily Record" were in the order of Haechi, horses, wild dogs, and Jujak. As to different composition layouts, based on the contents of the "Gyeongbok palace Construction Daily Record", the composition of Samjae Cheonjiin (天地人) was interpreted differently from the conventional interpretation of the arrangement of Woldae. All of these forms are associated with defense systems in the four directions and have become animal representations of each direction. The auspicious animal statues placed on the railing of Geunjeongjeon Woldae can be seen as reflected in a single building with the three dimensional personality that includes Pungsu's Bibo personality with the symbolic meaning that reveals the centrality as Jeong Jeon, the nation's best politics let alone the ideological system of the ancient astronomical of the East called Men Heaven Unity.
