Developmental and Morphological Characterization of the Wild Silkmoth, Actias gnoma, in Korea

  • The wild silkmoth, Actias gnoma was firstly collected at Suwon located in the middle part of Korea. The developmental and morphological characteristics of A. gnoma reared under the laboratory conditions were analysed. The egg shape was shorter elliptic. The major and minor diameter of the eggs were 2.04 mm and 1.83 mm, respectively. White mucous material was remained inside the eggs after hatching. Also, the body color of the larvae was completely changed from dark brown to light yellowish-green at the 3rd instar, but it did not occur until pupation. In the feeding test on several plants, the oak tree, Quercus acutissima, was newly identified as a host plant. The final larval instar was mostly 6th, but in some larvae it was 7th. A few of larvae ate their own casts just after ecdysis. The whole larval duration ranged from 45 to 59 days. The single cocoon weight, cocoon shell weight and cocoon shell percentage were 1.65 g,31.3 cg and 18.9%, respectively, The cocoon shape and color were spindle and light brown, respectively. The morphology of the silk gland was greatly different from those of Bombyx mori, Antheraea yamamai and Antheraea peryi: the thickness of the middle and posterior silk glands was almost identical.

닭 정자(精子)의 형태(形態)와 염색성(染色性) 조사(調査) (Studies on the Morphology and Stainability of Chicken Spermatozoa)

  • The present studies were designed to investigate the morphology and stainability of the chicken spermatozoa. Semen samples were collected by abdominal massage from 10 cocks of Arbor, Acres strain (egg breed) and 10 cocks of white Cornish strain (meat breed). The semen samples were diluted with Sarker's solution and were washed. Some of the semen smear slides were stained with seven differential stain methods and was compared with one another by light microscope. In addition to the staining already compared, the length of heads, middle pieces and tails of 400 spermatozoa of two chicken breed was measured with micrometer. The results obtained from these, studies were as follows: 1. Eosin stain appeared to give good results than hematoxylin, pre-treated protease and eosin or hematoxylin stain, pre-treated protease and hematoxylin-eosin stain, carbol-fuchsin, stain and Giemsa 9 technique in differential staining of spermatozoal three portions and pre-treated protease and eosin stain appeared as good staining methods for middle piece of spermatozoa. 2. The average length of chicken spermatozoa was $90.4{\pm}4.0{\mu}m$, and the average length of the head, middle piece and tail of spermatozoa was $13.0{\pm}0.5{\mu}m$, $3.8{\pm}0.2{\mu}m$ and $73.6{\pm}3.8{\mu}m$ lesoectively. 3. The average length of spermatozoa of Arbor Acres strain was $89.2{\pm}5.0{\mu}m$ and the average length of the head, middle piece and tail of spermatozoa was $12.9{\pm}0.5{\mu}m$, $3.8{\pm}0.2{\mu}m$ and $72.5{\pm}4.7{\mu}m$ respectively. The average length of spermatozoa of with Cornish was $91.6{\pm}3.0{\mu}m$ and the average length of the head, middle piece and tail of spermatozoa was $13.1{\pm}0.5{\mu}m$, $3.8{\pm}0.2{\mu}m$ and $74.7{\pm}2.8{\mu}m$ respectively.

Physiological Effects of Diethylstilbestrol Exposure on the Development of the Chicken Oviduct

  • Estrogen has dramatic effects on the development and function of the reproductive tract in mammals. Although diethylstilbestrol (DES) triggers the development of reproductive organs in immature animals, continued exposure to DES induces dysfunction of the female reproductive tract in mice. To investigate the effects of neonatal estrogen exposure on the reproductive tract of female chickens, we implanted DES pellets into the abdominal region of immature female chicks and then examined the effects of DES on the oviducts of both immature chicks and sexually mature chickens (30 weeks old). DES induced mass growth and differentiation of the oviduct in immature chicks. The chick oviduct increased by 2.7- and 29-fold in length and weight, respectively, following primary DES stimulation. In secondary DES stimulation, the length and weight of the chick oviduct increased by 4.5- and 74-fold, respectively. Additionally, DES treatments caused abnormal development of the infundibulum and magnum in hen oviducts. Furthermore, infundibulum abnormality gave rise to unusual ovulation of follicles and resulted in infertility and dysfunction of the magnum, such as less production of egg white proteins. Our results indicate that DES exposure during early developmental stages in chickens has detrimental effects on the development and maintenance of the female reproductive tract after sexual maturation.

Influence of Hyaluronic Acid on the Different Levels of Lysozyme and Peroxidase in the Aspects of Candidacidal Activities

  • Purpose: The purpose of the study was to investigate the influences of hyaluronic acid on the candidacidal activities of lysozyme, the peroxidase system, and the glucose oxidase-mediated peroxidase (GO-PO) system at different concentrations of antimicrobial enzymes. Methods: Hyaluronic acid was used at a final concentration of 0.5 mg/mL. Hen egg-white lysozyme (HEWL) was used at concentrations ranging from 10 to $100{\mu}g/mL$. The peroxidase system included bovine lactoperoxidase (bLPO), potassium thiocyanate (KSCN, 1 mM), and hydrogen peroxide ($100{\mu}M$). The GO-PO system included bLPO, KSCN (1 mM), glucose oxidase (10 units/mL), and glucose ($30{\mu}g/mL$). The final concentration of bLPO in the peroxidase and GO-PO systems ranged from 12.5 to $100{\mu}g/mL$. Candida albicans strains ATCC 10231, 11006, and 18804 were utilized. Candidacidal activities of antimicrobials and the influence of hyaluronic acid on their candidacidal activities were determined based on colony forming units. Results: Candidacidal activities of the peroxidase and GO-PO systems increased with increasing concentrations of bLPO. This tendency was the same in the presence or absence of hyaluronic acid. Candidacidal activity of HEWL was not significantly concentration-dependent. Candidacidal activities of the GO-PO system were higher than those of the corresponding peroxidase system. Candidacidal activity was inhibited in the presence of hyaluronic acid in the following order: HEWL, the peroxidase system, and the GO-PO system. Conclusions: Hyaluronic acid inhibited the candidacidal activities of HEWL, the peroxidase system, and the GO-PO system. The GO-PO system exhibited better candidacidal activity than HEWL and the peroxidase system both in the presence and absence of hyaluronic acid.

銀花甘草湯과 加味方이 抗 알레르기, 解熱, 消炎, 鎭痛效果에 미치는 影響 (An Experimental Studies on the Anti-allergic Reaction, the Antipyretic Action, the Anti-inflammatory and the Analgesic Action of Yenhwagamchotang and Gamiyenhwagamchotang)

  • Yenhwagamchotang has been widely used in treatment of inflammatory disease which is based on Oriental Medical literatures. These studies were attempted experimental effects of Yenhwagamchotang(sample A), Yenhwagamchotang plus Forsythia koreana NAKAI.(sample B), and Yenhwa-gamchotang plus Taraxacum mongolicum HAND- MAZZ(sample C),on the Anti-allergic reaction, the antipyretic action,the anti-inflammatory and the analgesic action,in rats. THe results of the studies were as follow: 1. Vascular permeability responses to intradermal serotonin in rats were showed significant effect at all sample groups. 2. The homologous passive cutaneous anaphylaxis in rats provoked by the IgE-like antibody aganist egg white albumin showed the decreasing effect. 3. The delayed type hypersensitivity responses to picryl chloride in mice were showed significant effect at all sample groups. 4. The delayed type hypersensitivity response to sheep red blood cell in mice were showed significant effect at all sample groups. 5. In anti-pyretic effect by yeast method were showed significant effect at all sample groups. 6. The anti-inflammatory effect by carrageenin method were showed significant effect at all sample groups. 7. The analgesic action by acetic acid method in mice were showed significant effect at all sample groups. According to the above result, Yenhwagamchotang(sample A), Yenhwagamchotang plus Forsythia koreana NAKAI(sample B), AND yenhwagamchotang plus Taraxacum mongolicum HNAN- MAZZ.(sample C ) were concluded to have the anti-allergic reaction, the antipyretic action, the anti-inflammatory, the analgesic action.

라이소자임 활성을 보유한 Lactobacillus rhamnosus 배양물의 병원성 미생물에 대한 항균 효과 (Anti-Bacterial Effect of Lactobacillus rhamnosus Cell-Free Supernatant Possessing Lysozyme Activity Against Pathogenic Bacteria)

  • Recently, there has been a growing demand for natural preservatives because of increased consumer interest in health. In this study, we produced Lactobacillus rhamnosus cell-free supernatant (LCFS) and evaluated and compared its antimicrobial activity with existing natural preservatives against pathogenic microorganisms and in chicken breast meat contaminated with Escherichia coli and Staphylococcus aureus. Lactobacillus rhamnosus cell-free supernatant possessed 30 units of lysozyme activity and contained 18,835 mg/L of lactic acid, 2,051 mg/L of citric acid and 5,060 mg/L of acetic acid. Additionally, LCFS inhibited the growth of fourteen pathogenic bacteria, S. aureus, Bacillus cereus, Listeria monocytogenes, Vibrio parahaemolyticus, Listeria innocua, S. epidermidis, L. ivanovii, E. coli, Pseudomonas aeruginosa, Shigella sonnei, Shi. flexneri, Proteus vulgaris, Pseudomonas fluorescens, and Klebsiella pneumoniae. The antibacterial activity of LCFS was stronger than that of egg white lysozyme (EWL), Durafresh (DF) and grapefruit seed extract (GSE). Additionally, LCFS maintained its antimicrobial activity after heat treatment at $50^{\circ}C{\sim}95^{\circ}C$ and at pH values of 3~9. Moreover, LCFS inhibited the growth of E. coli and S. aureus in chicken breast meat. In conclusion, it is expected that LCFS, which contains both lysozyme and three organic acids, will be useful as a good natural preservative in the food industry.

비근육 단백질의 수화조건이 겔 형성에 미치는 영향 (Effect of Hydration Condition of Non-Muscle Protein on Gelling)

  • [ $10\%$ ]의 단백질농도에서 5, 15, 25, 35$^{\circ}C$에서 각각 48시간까지 수화시킨 첨가단백질의 겔 형성능을 조사하였을 때 대두단백이 혈장단백, 건조난백과 같은 다른 2가지 첨가단백질 사이에는 중요한 차이가 있었으며, 이는 대두단백은 모든 조건에서 겔을 형성하지 못하였고, 수화는 5$^{\circ}C$, 12시간 24시간에서 혈장단백의 젤리강도를 $10\%$ 정도 상승시켰으며, 건조난백의 경우 5, 15, 25$^{\circ}C$에서 12, 24시간에서 $60\%$ 정도 젤리강도를 증가시켰다. 이와 같이 겔 형성능이 상승한 경우는 수화가 단백질의 인력과 척력의 적절한 균형을 제공한 것으로 추정되며, 실제 단백질의 겔 형성능을 이용하는 많은 식품이 있고 또한 가공공정이 다양하지만 적절한 수화공정의 추가는 겔 형성능을 상승시킬 수 있을 것으로 판단되었다.

고압조건에서 가열 처리된 Iysozyme의 변성 (Denaturation of Heat Treated Lysozyme under High Pressure Conditions)

  • 압출성형가공(extrusion cooking)에 의한 단백질의 조직화 메카니즘을 해석하기 위해 난백lysozyme을 고압조건에서 가열처리한 모델계 시료의 불용화 현상과 분자량적인 변화를 조사하였다. $100,\;300,\;600\;kg/cm^2$의 고압조건에서 $70,\;120,\;150^{\circ}C$로 가열온도가 높은 시료일수록 용해성은 감소하였으며 $200^{\circ}C$로 가열한 시료에서는 처리압력이 증가할수록 용해도가 현저히 감소하였다. $150^{\circ}C$$200^{\circ}C$로 가열처리한 시료에서는 dimer 이상의 polymer가 생성되었고 monomer보다 분자량이 작은 band의 생성도 인정되었는데 그 추정 분자량은 약 $6,000{\sim}9,000$의 분포이었으며 이 보다 저분자의 peptide는 존재하지 않았다. $120^{\circ}C$에서 가열한 시료의 고분자 형성은 주로 분자간 disulfide 결합에 의한 것이었고 $150^{\circ}C$ 이상으로 가열한 시료의 고분자 형성에는 disulfide결합 이외의 분자간 결합도 상당수 관여한 것으로 판단된다.

Structure and Function of the Autolysin SagA in the Type IV Secretion System of Brucella abortus

  • A recent genetic study with Brucella abortus revealed the secretion activator gene A (SagA) as an autolysin component creating pores in the peptidoglycan (PGN) layer for the type IV secretion system (T4SS) and peptidoglycan hydrolase inhibitor A (PhiA) as an inhibitor of SagA. In this study, we determined the crystal structures of both SagA and PhiA. Notably, the SagA structure contained a PGN fragment in a space between the N- and C-terminal domains, showing the substrate-dependent hinge motion of the domains. The purified SagA fully hydrolyzed the meso-diaminopimelic acid (DAP)-type PGN, showing a higher activity than hen egg-white lysozyme. The PhiA protein exhibiting tetrameric assembly failed to inhibit SagA activity in our experiments. Our findings provide implications for the molecular basis of the SagA-PhiA system of B. abortus. The development of inhibitors of SagA would further contribute to controlling brucellosis by attenuating the function of T4SS, the major virulence factor of Brucella.

Enzymatic Hydrolysis of Ovotransferrin and the Functional Properties of Its Hydrolysates

  • Bioactive peptides have great potentials as nutraceutical and pharmaceutical agents that can improve human health. The objectives of this research were to produce functional peptides from ovotransferrin, a major egg white protein, using single enzyme treatments, and to analyze the properties of the hydrolysates produced. Lyophilized ovotransferrin was dissolved in distilled water at 20 mg/mL, treated with protease, elastase, papain, trypsin, or α-chymotrypsin at 1% (w/v) level of substrate, and incubated for 0-24 h at the optimal temperature of each enzyme (protease 55℃, papain 37℃, elastase 25℃, trypsin 37℃, α-chymotrypsin 37℃). The hydrolysates were tested for antioxidant, metal-chelating, and antimicrobial activities. Protease, papain, trypsin, and α-chymotrypsin hydrolyzed ovotransferrin relatively well after 3 h of incubation, but it took 24 h with elastase to reach a similar degree of hydrolysis. The hydrolysates obtained after 3 h of incubation with protease, papain, trypsin, α-chymotrypsin, and after 24 h with elastase were selected as the best products to analyze their functional properties. None of the hydrolysates exhibited antioxidant properties in the oil emulsion nor antimicrobial property at 20 mg/mL concentration. However, ovotransferrin with α-chymotrypsin and with elastase had higher Fe3+-chelating activities (1.06±0.88%, 1.25±0.24%) than the native ovotransferrin (0.46±0.60%). Overall, the results indicated that the single-enzyme treatments of ovotransferrin were not effective to produce peptides with antioxidant, antimicrobial, or Fe3+-chelating activity. Further research on the effects of enzyme combinations may be needed.
