This study was done to investigate the effect of vitamin C on hepatic biliary and microsomal function during ischemia and reperfusion. Rats were treated with vitamin C(20, 100, 400, 1600 mg/kg) or with vehicle(saline) and then subjected to 60 min no-flow hepatic ischemia in vivo. Control animals were time-matched sham ischemic animals. After 1 or 5 hr of reperfusion, bile was collected, blood was obtained from the abdominal aorta, and liver microsomes were isolated. In vehicle-treated ischemic rats, serum ALT and AST levels peaked at 5 hr and were significantly attenuated by vitamin C 20 mg/kg and 100 mg/kg treatment. Similarly, hepatic wet weight-to-dry weight ratio was decreased in the vehicle-treated ischemic group. Vitamin C 20 mg/kg and 100 mg/kg treatment minimized the increase in this ratio. Lipid peroxidation was elevated in vehicle-treated ischemic group, but this elevation was also inhibited by vitamin C 20 mg/kg and 100 mg/kg treatment. Bile flow and cholate output, but not bilirubin output, were markedly decreased by ischemia/reperfuzion. Vitamin C 20 mg/kg and 100mg/kg treatment restored the secretion but vitamin C 1600 mg/kg reduced the cholate output. Cytochrome P-450 content was decreased by ischemia/reperfusion and restored by vitamin C 20 mg/kg and 100 mg/kg treatment to the level of sham operated group but decreased by vitamin C 1600 mg/kg. Aminopyrine N-demethylase activity was decreased and aniline p-hydroxylase activity was increased by ischemia/reperfusion. The changes in the activities of aminopyrine were prevented by vitamin C 20 mg/kg and 100 mg/kg treatment, but not by 400 mg/kg and 1600 mg/kg treatment. Our findings suggest that ischemia/reperfusion diminishes hepatic secretory functions as well as microsomal drug metabolizing systems, small doses(20, 100 mg/kg) of vitamin C significantly ameliorates and large doses(400, 1600 mg/kg) of vitamin C aggravated these ischemia/reperfusion-induced changes.
The germination characteristics of Diplachne fusca (L.) P. Beauv. seeds were investigated under different seed storage conditions to find out reliable system for maintaining the seeds with high and uniform germination rate and thus for possible use of the seeds in herbicide screening with a continuous seed supply. When the seeds were stored under wet-low temperature($4^{\circ}C$) condition, the germination rate was 88% after 4-week-storage. The germination rate slightly declined after the storage for longer than 3 months. Dry seeds stored at room temperature exhibited very low germination rate. The wet-low temperature treatment was effective for inducing the germination of the seeds which had been stored under dry-room temperature condition for 4 months. The germination rate was 70% after 2-week- storage under the wet-low temperature condition. The germination rate was much higher under an alternate temperature condition than under a continuous temperature condition. The optimum temperature was 35/$25^{\circ}C$(14/10hrs). The seeds had a capability to germinate under NaCl-treated condition even at a concentration of 1.0%, whereas the germination of Echinochloa crus-galli seeds was completely inhibited by 0.5% NaCl. This result indicates that D. fusca has an advantage over E. crus-galli to survive in reclaimed 1and from the sea.
Dried tobacco leaf debris infected with tobacco mosaic virus (TMV) was subjected to heat treatment (6$0^{\circ}C$~10$0^{\circ}C$) with or without addition of moisture and to room temperature for natural decay to examine the periods of time required for the inactivation of PMV in the inoculum source. Wet conditions (60% moisture content of the debris) for heat treatment were more efficient than dry conditions to inactivate the virus at 7$0^{\circ}C$~10$0^{\circ}C$, and which decrease of temperature, the time needed for the viral inactivation increased greatly. At 6$0^{\circ}C$ and 7$0^{\circ}C$, the temperaturein a compost heap during the actively decomposing period, it takes about 15 days or more for the complete inactivation of the virus. However, considering the decrease of the viral infectivity during the decomposition, a shorter period of time will be required to inactivate TMV in the conditions mentioned above, suggesting that a well decomposed organic manure containing tobacco leaf debris may not have infective TMV and may not provide a potential inoculum source.
Background : Aquaporins (AQPs) may play a role in the pathogenesis of pulmonary inflammation and edema. This study investigated the role ofAQPs in acute lung injury following bleomycin inhalation in rats. Methods : Sprague-Dawley rats were treated via inhalation with 10 U/kg bleomycin hydrochloride dissolved in 5 ml of normal saline. The control rats were treated with 5 ml normal saline. The animals (n = 6-8 rats per group) were sacrificed at 4, 7, and 14 d. The changes in AQP1, AQP4, and AQP5 expression levels over time were analyzed by Western blotting. The nitrate and nitrite concentrations in the bronchoalveolar lavage fluid (BALF) were measured using a modified Griess reaction. ELISA was used to check cytokines. Results : The respiration rates were significantly higher 4 and 7 days after the bleomycin treatment compared with those of the control rats. The tidal volume was lower in rats at 4 days after the bleomycin treatment, and the wet/dry weights of the lung were significantly higher than those of the control group. The nitrite and nitrate concentrations in the BALF from the rats at 4 days after exposure to bleomycin were greater than those from the saline-treated rats. Immunoblotting studies demonstrated that the AQP1 and AQP4 expression levels were lower in the rats at 4 days. However, the AQP4 expression level was higher at 7 days. The AQP5 expression level increased at 4, 7 and 14 days after the bleomycin treatment. Conclusion : This study demonstrates that AQPs are expressed differently in bleomycin-induced pulmonary edema.
The effects of after-treatment condition on the permeability of polysulfone hollow fiber membranes were investigated. It was well known that the permeability of and the rejection of hollow fiber membranes prepared by dry-wet spinning technique was highly affected by spinning conditions such as composition of spinning solution, inner and outer coagulant and spinning height. But these are also governed by after-treatment condition such as drying, hot water treatment and glycerine treatment. Since it was difficult to remove DMAc and NMP in membranes by washing with in cold water and even traces of such solvent affected the reproducibility of the membrane characteristics, the hot water-treatment should be required.
Objectives: To compared the effect of different levels of moisture of root canal on the sealing ability after filling with four different types of sealer. Materials and Methods: Single-rooted teeth (n = 90) instrumented to and apical size of 0.06 / 45 were randomly assigned to 12 experimental groups (n = 7 per group), positive/negative control groups (n = 3 per group). The teeth of the experimental groups (a. DRY; b. PAPER POINT DRY; c. WET) were obturated with sealer (Group 1-3: Sealapex; Group 4-6: AH plus; Group 7-9: Tubuli-seal; Group 10-12: EndoRez) and warm vertical compaction method. After 7 days in $37^{\circ}C$, 100% humidity, the coronal-to-apical microleakage was evaluated quantitatively using a glucose leakage model. The leaked glucose concentration was measured with spectrophotometer at 1, 3, 7, 14, 21, and 30 days. Data were recorded ad mmol/L and statistically analysed with the two-way ANOVA and Duncan test (p = 0.05). Results: Throughout the experimental period Tubuli-seal/WET (Group 9) showed the highest mean cumulative glucose penetration (178.75 mmol/L), whereas AH plus/DRY (Group 4) had the least (20.78 mmol/L). Conclusions: The results of this study demonstrated that the moisture condition of root canals at the time of obturation and the type of sealer that was used had a significant effect on leakage and sealing ability. Thus drying procedure according to sealer types is a critical step and should not be missed in endodontic treatment.

In the production of rice flour, wet milling is a method of milling rice after soaking it in water, and it takes a lot of time and cost from milling to drying. To overcome this problem, the floury type rice was developed for dry milling and it is known to have round starch granules, low content of damaged starch after milling, and a starch structure similar to wheat. Because of its unique properties different from normal rice, it is necessary to research on processing and characteristics of floury-type rice to expand its utility in the food industry. Therefore, this study aimed to prepare the pregelatinized floury type rice (Baromi2) by autoclave and investigate their physicochemical properties. As the heat treatment time increased, the brightness decreased from 83.8 to 76.8, however, both redness and yellowness increased from 0.57 to 4.5 and from 14.58 to 21.13, respectively. Despite of same treatment time, soaking in water (10 min) before autoclaving increased the solubility and swelling power of Baromi2 over 2 times. The peak viscosity of native Baromi2 was over 2000 RVU, on the other hand, there was a significantly decrease to less than 1000 RVU of pregelatinized Baromi2. Heat treatment without immersion caused partial gelatinization of starch, resulting that some starch granules maintaining their integrity. Whereas there were no starch granules in heat treatment with soaking in water due to complete gelatinization. This study would be helpful to the suggestion of using heat-treated floury-type rice as an intermediate material in the food industry in the future.
Threshing operation may be one of the most important processes in the paddy post-production system as far as the grain loss and labor requirement are concerned . head-feeding type threshers commercially available now in Korea originally were developed for threshing dry paddy in the range of 15 to 17 % in wet basis. However, threshing wet-paddy with the grain moisture content above 20 % has been strongly recommended, especially for new high-yielding Indica -type varieties ; (1) to reduce high grain loss incurred due to the handling operations, and (2) to prevent the quantitative and qualitative loss of milled -rice when unthreshed grains are rewetted due to the rainfall. The objective of this study were to investigate the adaptability of both a head-feeding type thresher and a throw-in type thresher to wet-paddy , and to find out the possiblilities of improving the components of these threshers threshing. Four varieties, Suweon 264 and Milyang 24 as Tongil sister line varieties, minehikari and Jinhueng as Japonica-type varieties, were used at the different levels of the moisture content of grains. Both the feed rate and the cylinder speed were varied for each material and each machine. The thresher output quality , composition of tailing return, and separating loss were analyzed from the sampels taken at each treatment. A separate experiment for measurement opf the power requirement of the head-feeding type thresher was also performed. The results are summarized as follows : 1. There was a difference in the thresher output quality between rice varieties. In case of wet-paddy threshing at 550 rpm , grains with branchlet and torn heads for the Suweon 264 were 12 % and 7 % of the total output in weight, respectively, and for the Minehikari 4.5 % and 2 % respectively. In case of dry paddy threshing , those for the Suweon 264 were 8 % and 5% , and for the Minehikari 4% and 1% respectively. However, those for the Milyang 23 , which is highly susceptable to shattering, were much lower with 1 % and 0.5% respectively, regardless of the moisture content of the paddy. Therefore, it is desirable to breed rice varieties of the same physical properties as well as to improve a thresher adaptable to all the varieties. Torn heads, which increased with the moisture content of rall the varieties except the Milyang 23 , decreased as the cylinder speed increased, but grains with branchlet didnt decrease. The damaged kernels increased with the cylinder speed. 3. The thresher output quality was not affected much by the feed rate. But grains with branchlet and torn heads increased slightly with the feed rate for the head-feeding type thresher since higher resistance lowered at the cylinder speed. 4. In order to reduce grains with branchlet and torn heads in wet-paddy threshing , it is desirable to improve the head-feeding type thresher by developing a new type of cylinder which to not give excess impact on kernels or a concave which has differenct sizes of holes at different locations along the cylinder. 5. For the head-feeding type thresher, there was a difference in separating loss between the varieties. At the cylinder speed of 600 rpm the separating losses for the Minehikari and the Suweon 264 were 1.2% and 0.6% respectively. The separating loss of the head-feeding type thresher was not affected by the moisture content of paddy while that of the Mini-aged thresher increased with the moisture content. 6. From the analysis of the tailings return , to appeared that the tailings return mechanism didn't function properly because lots of single grains and rubbishes were unnecessarily returned. 7. Adding a vibrating sieve to the head-feeding type thresher could increase the efficiency of separation. Consequently , the tailing return mechanism would function properly since unnecessary return could be educed greatly. 8. The power required for the head-feeding type thresher was not affected by the moisture content of paddy, but the average power increased linearly with the feed rate. The power also increased with the cylinder speed.
Twenty five 75% Holstein Friesian cross bred bullocks fed rice straw (Oryza sativa) of long form, were fed with the following five treatments. 1. Rice straw, untreated (RS) 2. RS + water (1:1), stored for 24 hours (WRS) 3. RS (100 kg) + urea solution (4 kg urea/100 litre water) and dried (USRS) 4. RS (100 kg) + urea solution (as in 3) stored in wet condition for 24 hours (UWRS) 5. RS (100 kg) + urea solution (as in 3) stored in pit for 21 days (UTRS). Potential digestibility of treatments of RS was evaluated by monitoring (in vitro) Simulating Rumen like Fermentation (SRLF). The results indicated that Dry Matter Intake (DMI), digestibility of nutrients, N utilization were of the order UTRS > UWRS > USRS > WRS and RS (p < 0.05 to p < 0.01). SRLF index was high (255.84) for UTRS and least (145.58) for USRS. It was intermediary (199.66) for UWRS. The acetyl content (AC) of UTRS with higher hemicellulose (HCE) digestibility (80.8%) was low compared to UWRS, USRS, RS and WRS. The acetate content was of the order UTRS < UWRS < USRS < WRS and RS thereby indicating that reduction in acetyl content was an index of positive response of urea-treatment of RS. In addition, the ratio of HCE/AC in faeces of UTRS was 0.87 as against the ratios (2.26-2.48) observed in other treatments recording reduction in AC due to urea-treatment. Among the treatments, USRS only supplemented N while UTRS in addition to utilization N, increased the digestibility of structural carbohydrates. Reduction in treatment time from 21 days to 1 day (UWRS) resulted in improvements similar to those of UTRS.

The increase in carbon stock and sustainability of crop production are the main challenges in agricultural fields relevant to climate change. Methane is the most important greenhouse gas emitted from paddy fields. This study was conducted to investigate the effects of tillage and cultivation methods on methane emissions in rice production in 2014 and 2015. Different combinations of tillage and cultivation were implemented, including conventional tillage-transplanting (T-T), tillage-wet hill seeding (T-W), minimum tillage-dry seeding (MT-D), and no-tillage-dry seeding (NT-D). The amount of methane emitted was the highest in T-T treatment. In MT-D and NT-D treatments, methane emissions were significantly decreased by 77%, compared with that in T-T treatment. Conversely, the soil total carbon (STC) content was higher in MT-D and NT-D plots than in tillage plots. In both years, methane emissions were highly correlated with the dry weight of rice ($R^2=0.62{\sim}0.96$), although the cumulative emissions during the rice growing period was higher in 2014 than in 2015. T-T treatment showed the highest $R^2$ (0.93) among the four treatments. Rice grain yields did not significantly differ with the tillage and cultivation methods used. These results suggest that NT-D practice in rice production could reduce the methane emissions and increase the STC content without loss in grain yield.
