오미자 추출물의 추출특성 및 항산화 효과 (Extraction Characteristics and Antioxidative Activity of Schiznadra chinensis Extracts)

  • 오미자 추출액의 추출 특성과 추출액의 항산화성을 검토하여 오미자의 추출 조건을 결정함으로써 오미자 유효성분의 식품 소재화를 위한 기초자료를 얻고자 하였다. 용매와의 비율을 결정하기 위해 8배, 10배, 12배, 14배의 water를 가하여 추출 수율을 비교한 결과 water와의 비율이 10배일 때 추출 수율이 가장 좋은 것으로 나타났다. 탁도는 Water 추출액에서 가장 높았으며, 추출용매의 ethanol 농도가 증가할수록 낮아졌다. 적정산도는 추출용매의 50% ethanol에서 가장 높았으나, 전체적으로 산도가 매우 낮았다. 유리당 함량은 fructose $1.24{\sim}4.42mg%$, glucose $3.65{\sim}4.71mg%$, sucrose $0.89{\sim}1.69mg%$로 glucose의 함량이 가장 높았으며, 당질의 대부분을 glucose와 fructose인 것으로 나타났다. 총 폴리페놀 함량을 나타낸 것으로 50% ethanol 추출액은 0.98%, 75% ethanol 추출액은 0.89%, water 추출액은 0.58%로 나타나 50% ethanol 추출액에서 가장 높은 함량을 나타내었으며. water에 비해 유기용매 추출시 높은 함량을 보였다. 전자공여능은 water 추출액 60.87%, 50% ethanol 추출액 57.24%, 75% ethanol 추출액 55.61%로 나타나 용매별 전자공여능 효과가 큰 차이는 나타나지 않았으며. water 추출액이 다른 용매에 비해 높은 전자공여 효과를 보여 주었다. SOD 유사환성 측정 결과는 50% ethanol 추출액에서 33.65%로 가장 높은 활성을 나타냈었으며, 50% ethanol 추출액이 다른 두 용매 보다 우수한 것으로 나타나 총 폴리페놀 함량과 전자공여능 측정 결과 와 비슷한 경향을 보여주었다.

  • The microcapsules containing ethanol in a water-soluble polymer shell could be formed by the spray-drying technique. A mixed solution of ethanol, water and a water-soluble polymer is spray-dried at the lowest possible temperature to obtain a powder product in which the water is substantially removed and the ethanol is encapsulated in water-soluble polymer shell because of the hydrophillic property of polymer and permeability difference between ethanol and water. In this study, the effect of sodium lauryl sulfate (SLS) on the ethanol content, ethanol encapsulation efficiency and yield were investigated to maximize the microencapsulation of ethanol.

  • 덱스트린/에탄올/물(1/1/l, w/w/w)의 혼합용액에 라우릴황산나트륨 0.5%(w/w) 및 홍삼 엑기스 20%(w/w)를 첨가한 조성을 가지고 분무건조하여 제조한 분말 홍삼주는 최대 에탄올(31.17$\pm$ 1.33%(w/w)) 및 ginsenoside Rbl(243.0$\pm$ 7.0$\mu\textrm{g}$/g)이 봉입된 미립구로서 용해 및 복용감이 우수한 새로운 홍삼 엑기스의 제형으로서의 가능성이 있다.

물과 에탄을 이성분 혼합용매 계에서 부피 및 온도 변화에 관한 연구 (A Study on the Change of Volume and Temperature in Aqueous Binary Solvent with Ethanol)

  • This paper is to study on the change of volume and temperature of the solution which mixed water with ethanol. And its main purpose is to examine closely how the volume changes, and to find the method to maximize the change of the volume. The summaries for results of the study are; First, we were known that water and ethanol are homogeneously mixed. But two solvents does not mix homogeneously by different specific gravity at early stages. Second, we could see that the volume changed large at homogeneous mixed water with ethanol by stirrer, the change of volume is the largest value when water mixed with ethanol in the ratio of one to one. Third, when water mixed with ethanol in the ratio of one to one, the change of temperature is very large by activated hydrogen bond. We conclude that it is the best result when ratio of water and ethanol is one to one and the solution is well mixed.

마가목 줄기 열수 및 에탄올 추출물의 항산화 활성 (Antioxidant Activities of Hot Water and Ethanol Extracts from the stem of Sorbus commixta Heal)

  • Objectives : In order to investigate the possibility of Sorbus commixta Heal. stem (SC) as a natural material, antioxidant activities of the hot water and ethanol extracts were examined. Methods : The samples of SC were pulverized, and fractions were extracted repeatedly three times from hot water and 70% ethanol at room temperature for 2 hours. The antioxidant activities were analyzed from total polyphenol, flavonoid contents, DPPH, ABTS, hydroxyl radical, $Fe2^+$ chelating, and nitrite scavenging activity. Results : Total polyphenol contents were significantly higher in ethanol extract group ($504.39{\mu}g/m{\ell}$) than in hot water extract group ($364.64{\mu}g/m{\ell}$). Total flavonoid contents were also significantly higher in ethanol extract group ($160.09{\mu}g/m{\ell}$) than in hot water extract group ($124.59{\mu}g/m{\ell}$). DPPH, ABTS, $Fe2^+$ chelating were slightly higher in ethanol extract gorups than in hot water extract groups, and increased in a dose-dependent manner. The hydroxyl radical scavenging activity (18.42~23.61%) of ethanol extract groups were shown to be approximately twice higher than that (7.63~10.37%) of hot water extract groups at $12.5{\sim}50{\mu}g/m{\ell}$ concentration. Nitrite scavenging activities of both ethanol and hot-water extract groups were shown to be higher in a dose dependent manner at the concentration of $12.5{\sim}50{\mu}g/m{\ell}$ at pH 3.0 than at pH 1.2, and ethanol extract groups (86.55~96.64%) had higher activity than the hot water extract groups (42.59~92.63%), which was higher than that of the control group antioxidant BHT (72.96~80.11%). Conclusions : The extracts of SC displayed antioxidant activities which suggested a natural material can be developed to functional material.

Oenanthe javanica extract accelerates ethanol metabolism in ethanol-treated animals

  • The effect of water dropwort (Oenanthe javanica DC) extract in eliminating ethanol was evaluated in New Zealand white rabbit and ICR mice. When a hot-water extract of water dropwort extract and ethanol was injected into New Zealand white rabbit, the plasma ethanol level was rapidly reduced, similar to metadoxine treatment. Specifically, the n-butanol fraction of hot-water extract was the strongest in eliminating plasma alcohol in ICR mice. When ethanol was orally ingested, administration of the hot-water extract eliminated up to 44% of the plasma ethanol in mice while the n-butanol fraction eliminated around 70%. Alcohol removal behaved in a dose-dependent manner in response to 50-200 mg/kg of n-butanol fraction. These data show O. javanica extract is effective in overcoming alcohol intoxication by the accelerating ethanol metabolism.

구기자 추출물의 추출특성 및 항산화 효과 (Extraction Characteristics and Antioxidative Activity of Lycium chinense Extracts)

  • 구기자 추출액의 추출 특성과 추출액의 항산화성을 검토하여 구기자의 추출 조건을 결정함으로써 구기자 유효성분의 식품 소재화를 위한 기초자료를 얻고자 하였다 용매와의 비율을 결정하기 위해 8배, 10배, 12배, 14배의 water를 가하여 추출 수율을 비교한 결과 water와의 비율이 10배일때 추출 수율이 가장 좋은 것으로 나타났다. 추출물의 가용성 고형분은 water 추출액에서 5.0 $^{\circ}$Brix로 가장 높은 함량을 나타내었다. 추출액의 탁도는 50$\%$ ethanol 추출 시 가장 높았고, 75$\%$ ethanol 추출 시 가장 낮았다. 적정산도는 추출용매의 ethanol 함량이 감소할수록 산도가 높아지는 경향을 보였으며, 유리당의 함량은 fructose 4.53$\~$5.22 mg$\%$, glucose2.79$\~$4.40 mg$\%$, sucrose 0.35$\~$l.18 mg$\%$ 로 나타났으며, water 추출액이 다른 추출 용매에 비해 높게 나타났다. 총폴리페놀의 추출 용매별 함량은 50$\%$ ethanol 추출액에서 2.93$\%$로 가장 높은 함량을 나타내었으며, water 추출액이 75$\%$ ethanol 추출액 보다 높은 함량을 나타내었다. 구기자추출액은 50$\%$ ethanol에서 66.97$\%$로 나타나 다른 용매에 비해 높은 전자공여 효과를 보여 주었으며, 폴리페놀 함량과 비슷한 경향을 나타내었다. SOD 유사활성 측정 결과는 50$\%$ ethanol 추출액에서 37.33$\%$로 가장 높은 활성을 나타냈었으며, 총 폴리페놀 함량과 전자공여능 측정 결과와 비슷한 경향으로, 50$\%$ ethanol 추출액이 다른 두 용매 보다 우수한 것으로 나타났다.

Alcohol-induced hepatic fibrosis in pig

  • A number of toxicants have been incriminated as a causing hepatic disease. Among many detrimental injury, alcohol has been noted for hepatitis, fatty liver, fibrosis, and hepatic cirrhosis. The purpose of this study was to develop animal model for hepatic fibrosis in pigs fed ethanol, and to search for a new anti-fibrogenic agent via this model. Twelve male Landrace pigs were divided into 3 groups of 4 animals each. Group 1, 2 and 3 were fed with active ceramic water only, ceramic water + liquid diet containing 15% ethanol and normal tap water + liquid diet containing 15% ethanol for 12 weeks, respectively. At week 12, all pigs were immediately sacrificed for collection each tissue and blood. Serologically, serum ALT and AST levels were significantly reversed in group 2, as compared to group 3. They were normal range in pigs of group 1. Microscopically, macrovesicular lipid droplets and moderate hepatocellular necrosis were evident in the tap water + ethanol fed group 3. However, the active ceramic water treated group 1 showed normal architecture. Moreover, in group 2, mild fatty changes and necrosis were observed in hepatocytes. Collagen fibers were increased in spaces surrounding periportal and interlobular connective tissues in the group 3 of tap water + ethanol, but collagen synthesis and its thickness of fibrotic septa connecting portal tracts were markedly reduced in the group 2 of ceramic water + ethanol. Myofibroblasts were detected mainly in the interlobular connective tissues of pig liver of group 3 treated ethanol and tap water. Few to no myofibroblasts were observed in groups 1 and 2. CYP2E1 was not or rarely detected in group 1 fed ceramic water. However, group 2 showed slightly activation of CYP2E1 in the area of pericentral vein, while CYP2E1 was significantly activated in group 3 fed tap water and ethanol. Based on the above data, we believe that we have developed a unique alcohol induced fibrosis model in pig, which will be useful in developing anti-fibrotic agents and drugs. Furthermore, the active ceramic water used in our study had an inhibitory and may be protective against ethanol induced hepatic toxicity and fibrosis.

주름조개풀(Oplismenus undulatifolius) 추출물의 생리활성 효소 억제 효과 (Inhibitory activities on biological enzymes of extracts from Oplismenus undulatifolius)

  • 주름조개풀에 함유되어 있는 phenolic contents는 water, 70% ethanol을 용매로 하여 추출하였을 때 각각 7.7, 10.1 mg/g의 함량을 나타내었다. 추출물의 phenolic 농도를 $50-200{\mu}g/mL$로 조절하여 항산화 효과를 측정한 결과, DPPH에서는 water, ethanol 추출물 $50{\mu}g/mL$ phenolics 농도에서 각각 78, 82%였고, ABTS radical decolorization는 $100{\mu}g/mL$ phenolics 농도에서 각각 92, 76%의 radical 소거능을 나타내었다. PF는 $200{\mu}g/mL$ phenolics 농도에서 water, ethanol 추출물에서 각각 1.51, 2.08 PF로 측정되었으며, TBARs값은 water, ethanol 추출물 $50{\mu}g/mL$ phenolics 농도에서 각각 84, 99%의 활성을 나타내었다. 기능성 식품 활성을 측정하는 ${\alpha}-Glucosidase$ 저해활성을 측정한 결과 ethanol 추출물 $200{\mu}g/mL$ phenolics 농도에서 44%의 억제력을 나타내었고, ${\alpha}-amylase$ 저해활성은 water 추출물 $50-200{\mu}g/mL$ phenolics 농도에서 37-88%의 효소 억제력을 나타내었다. 미용 식품 활성을 나타내는 tyrosinase 저해활성을 측정한 결과 ethanol 추출물에서 82%의 tyrosinase 저해효과를 나타내었다. 또한 elastase 저해활성을 측정한 결과 water, ethanol 추출물에서 각각 4, 61%로 나타내었고, collagenase 저해활성을 측정한 결과 water, ethanol 추출물 $200{\mu}g/mL$ phenolics 농도에서 각각 39, 67%로 ethanol 추출물에서 높은 억제력을 나타내어 우수한 저해효과를 확인하였다. Astringent 효과를 측정한 결과 water, ethanol 추출물 $200{\mu}g/mL$ phenolic 농도에서 각각 13, 32%의 astringent 효과를 나타내었다. Hyaluronidae 저해활성을 측정한 결과 ethanol 추출물 $200{\mu}g/mL$ phenolic 농도에서 46%의 Hyaluronidae 저해효과를 나타내었다.

  • The luminescence intensity of $Tb^{3+}$ bound to picolinic acid, squaric acid, and 2,3-pyrazinedicarboxylic acid has been studied in water and water-ethanol mixtures. It has been found that the emission intensity of $Tb^{3+}$ in the complexes is enhanced with the increase of ethanol content in water-ethanol mixtures. Several factors affecting on the emission intensity of $Tb^{3+}$ in waterethanol mixtures are discussed.
