Metabolic syndrome, including obesity, glucose intolerance and elevated blood pressure, is related to type 2 diabetes and cardiovascular disease. Previous studies have reported the anti-oxidative, anti-inflammatory and anti-diabetic effects of purple corn extract. We investigated the efficacy of purple corn extract (PC) against high-fat diet (HFD)-induced obesity and glucose intolerance, and examined the underlying mechanisms by analyzing expression of proteins and genes involved in glucose regulation and macrophage infiltration. C57BL/6 mice were fed with normal chow diet (ND), or HFD treated with distilled water (DW, control) or PC, for 10 weeks. Although body weights were similar in the HFD-fed groups, we observed a decrease in the liver and epididymal adipose tissue (EAT) weights, and enhanced glucose tolerance test (GTT) results in the PC group, as compared with DW group. Liver showed increased Akt phosphorylation in the PC-treated mice; however, no changes were observed in the EAT, for all groups. In PC-treated mice, decreased macrophage infiltration was seen in the EAT, with a reduced expression of macrophage marker genes. Finally, proinflammatory cytokine gene expressions were decreased by PC in the EAT, and a modest trend for downregulation was observed in the liver. Hence, we conclude that PC may decrease glucose intolerance by increasing the phosphorylation of Akt and reducing the macrophage infiltration into the EAT.
Anemia, the condition of the diminished concentration of hemoglobin per erythrocyte is common in patients with cancer and is a frequent complication of myelosuppressive chemotherapy. Cham-Dang-Gui (Angelicae Gigantis Radix) has been used in traditional Korean medicine to treat hematologic deficiencies. In this study, Cyclophosphamide (CYP), an alkylating agent that has a broad spectrum of anti-cancer activities, was intraperitoneally injected into the experimental animals to suppress the bone marrow thereafter, causing anemia. The hemopoietic effects of Cham-Dang-Gui were examined using anemic rat model. Rats were divided into five groups: CON (control group), ANS (CYP-injected + normal diet), AND (CYP-injected + normal diet + Cham-Dang-Gui), ALS (CYP-injected + low iron diet), and ALD (CYP-injected + low iron diet + Cham- Dang-Gui) groups. CYP (30 mg/kg) was intraperitoneally injected to rats for 3 days to induce anemic condition. Saline or Cham-Dang-Gui was administrated orally during the entire experimental period. CYP injection decreased body weight gain and food consumption in comparison with CON group. Oral administration of Cham-Dang-Gui extract with normal iron diet significantly prevented the lower body weight gain. The blood level of hemoglobin, iron status (serum iron, transferrin, ferritin and TIBC) and blood level of vitamin B-12 in Cham-Dang-Gui treated groups were significantly higher than those of Cham-Dang-Gui untreated groups regardless of amount of iron in the diet. Taken together, it could be concluded that the Cham-Dang-Gui extract could improve anemic condition induced by CYP injection by improving hematological value, iron status and vitamin B12 status in rats.
The purpose of this study was to evaluate the microleakage of light cured glass ionomer restorative materials in class 5 cavities. In this in vitro study, class 5 cavities were prepared on buccal and lingual surfaces of forty extracted human premolars and molars on cementum margin. These specimen were randomly divided into four groups of 10 each : Group 1 was Fugi II (control), Group 2 was Fugi II LC, Group 3 was Vitremer, and Group 4 was Dyract. Group 2 was also divided once more into 2 groups of 5 each : Group 2-1 was pretreated with dentin conditioner and Group 2-2 was not. All teeth were restored according to the manufacturer's instructions. After 500 thermocycling between $5^{\circ}C$ and $55^{\circ}C$, the 40 teeth were placed in 2 % Methylene blue dye for 24hr, then rinsed with tab water. The specimen were embedded in clear resin, then sectioned buccolingually through the center of restoration with a low speed diamond saw. The dye penetration on each of the specimen were then observed with a stereomicroscope at 20. The results of the study were statistically analyzed using the Student-Newman-Keuls Methods and the Mann-Whitney Rank Sum Test. Tooth restorative interfaces were evaluated using SEM analysis. Results were as follows, 1. Compared to conventional glass ionomer restoratioqs, all light cured glass ionomer restorations were fairly resistant to microleakage (P<0.05). 2. Groups 3 (Vitremer) and Group 4 (Dyract) were found to be the most resistant, Group 2 (Fugi II LC) fairly resistant, and Group 1 (Fugi II) least resistant to microleakage(P<0.05). 3. No significant differences were found between Group 2-1 and Group 2-2 (P>0.08). 4. With the backscattered SEM analysis, the degrees to which tight bonding occurred were also observed in all the groups except for Group 1. Group 4 showed the highest degree of tight bonding than any other materials used in this study.

The purpose of this study was to elucidate the effect of blood-and saliva-contamination during dentin pretreatment procedure on tensile bond strength, and to investigate the effect of contaminant-removing treatments on the recovery of bond strength of dentin bonding agents. Dentin specimens prepared from freshly extracted bovine mandibular anterior teeth were divided into non-contaminated control and contaminated experimental groups. The specimens of the contaminated group were contaminated with saliva or blood after etching or priming procedure, followed by contaminant-removing treatments. All the specimens were bonded with All Bond$^{(R)}$ 2 dentin bonding agent and Bisfil$^{TM}$ composite resin or Scotchbond$^{TM}$ Multipurpose and Z100. After all the bonded specimens were stored in $37^{\circ}C$ distilled water for 24 hours, tensile bond strengths were measured. The contaminated dentin and fractured dentin surfaces were examined under the scanning electron microscope. The results were as follows : Contaminated specimens showed lower bond strength than non-contaminated ones regardless of the kind of contaminant, contamination time and contaminant-removing treatments, except specimens which were acid-etched following saliva contamination after etching in All Bond$^{(R)}$ 2 groups (p<0.05). Blood contaminant resulted in much bond strength decrease than saliva ones (p<0.01), and contamination after priming resulted in much decrease in bond strength than after etching (p<0.01). Re-etching resulted in increase of bond strength in the specimens contaminated with saliva after etching but not in blood contaminated ones. Re-priming resulted in increase of bond strength in the specimens contaminated after priming regardless of the kind of contaminant.
Objectives: It is difficult to achieve adhesion between resin cement and zirconia ceramics using routine surface preparation methods. The aim of this study was to evaluate the effects of $CO_2$ and Er:YAG laser treatment on the bond strength of resin cement to zirconia ceramics. Materials and Methods: In this in-vitro study 45 zirconia disks (6 mm in diameter and 2 mm in thickness) were assigned to 3 groups (n = 15). In control group (CNT) no laser treatment was used. In groups COL and EYL, $CO_2$ and Er:YAG lasers were used for pretreatment of zirconia surface, respectively. Composite resin disks were cemented on zirconia disk using dual-curing resin cement. Shear bond strength tests were performed at a crosshead speed of 0.5 mm/min after 24 hr distilled water storage. Data were analyzed by one-way ANOVA and post hoc Tukey's HSD tests. Results: The means and standard deviations of shear bond strength values in the EYL, COL and CNT groups were $8.65{\pm}1.75$, $12.12{\pm}3.02$, and $5.97{\pm}1.14MPa$, respectively. Data showed that application of $CO_2$ and Er:YAG lasers resulted in a significant higher shear bond strength of resin cement to zirconia ceramics (p < 0.0001). The highest bond strength was recorded in the COL group (p < 0.0001). In the CNT group all the failures were adhesive. However, in the laser groups, 80% of the failures were of the adhesive type. Conclusions: Pretreatment of zirconia ceramic via $CO_2$ and Er:YAG laser improves the bond strength of resin cement to zirconia ceramic, with higher bond strength values in the $CO_2$ laser treated samples.
The purpose of this study was to investigate the effects of green tea ingestion on hepatocarcinogenesis before and after its initiation. Male Sprague-Dawley rats were fed an AIN76A diet with or without green tea. Initiation was induced by a single dose (200 mg/kg) of diethylnitrosamine at week 4 and 0.02% (w/w) 2-acetylaminofluorene was supplied in the diets. The control group had free access to water for 13 weeks (CTR13). Tea infusion was provided from the beginning of the experiment for 13 weeks (PRE13) or from the post-initiation stage until week 13 (POST13). Three other groups (CTR24, PRE24 and POST24) were added to examine the longer-term effects (24 weeks) with the same experimental design. The percentage area of liver sections that were positive for hepatic placental glutathione S-transferase (GST-P), which was used as a marker of preneoplastic lesions, was smaller in PRE13 ($20.2{\pm}5.0%$, $mean{\pm}SD$) and POST13 ($26.0{\pm}4.8%$) than in CTR13 ($33.2{\pm}5.8%$, p<0.05). Over the longer period, the GST-P lesions were significantly smaller for both PRE24 and POST24 ($21.6{\pm}8.5%$ and $22.2{\pm}4.0%$, respectively) than for CTR24 ($28.6{\pm}5.1%$, p<0.05), but there was no significant difference between PRE24 and POST24. The liver content of thiobarbituric acid reactive substances was significantly lower in the tea groups than in the controls (p<0.05). However, no significant differences were observed among groups of GST activity. The results show that tea consumption exhibits a stronger short-term initiation-inhibiting ability in liver carcinogenesis, but over a longer period, the preventive effects of green tea ingestion do not differ in post- and pre-initiation.

The effects of mungbean sprout juice on cadmium-induced hepatotoxicity in rats were investigated. Sprague-Dawley rats weighing about 90g were divided into 4 groups and raised for 6 weeks. ; control group(CON), mungbean sprouts juice-administered group(MSJ), cadmium-administered group(CAD) fed water containing 40 ppm cadmium chloride and mung bean sprouts juice and cadmium-administered group(MCD). The diet was supplied every day for the measurement of feed efficiency ratio(FER) and net weight gain was measured every 3 days. The activities of serum glutamic oxaloactic transaminase(GOT) and glutamic pyruvic transaminase(GPT), superoxide dimutase(SOD), catalase and glutathione peroxidase(GSH-Px) in the liver and the hepatic contents of glutathione were examined. The contents of Cd in liver and kidney of the rats were determined by using ICP(Inductively Coupled Plasma Emission Spectrophotometer). Growth rate and FER were decreased in CAD group, compared with CON group but the changes were not significant in MCD group. The activities of serum GOT and GPT, SOD, catalase and GSH-Px in the liver were increased by Cd administration, but the alterations were decreased by supplementation with mungbean sprouts juice. The level of glutathione decreased in CAD group(26.8$\pm$9.0mg/g liver), whereas it increased in MCD group(36.4$\pm$15.8mg gliver). The content of Cd in the liver and kidney in MCD group(9.57 ppm, 4.88 ppm) was decreased, compared with CAD group(12.81 ppm, 5.46 ppm). This result suggested that mungbean sprout juice has a lowering effect on the accumulation of Cd in the liver and kidney and it is believed that the juice has some protective effects to Cd-induced hepatotoxicity in rats, but the mechanism of these effects was obscure.

Isolation and identification of the major bacteria causing spoilage of buckwheat starch jelly, and the effects of chitosan concentrations (0, 0.5, 1.0, 1.5, 2.0%) on shelf-life and quality of buckwheat starch jelly were investigated. Eight strains were isolated from spoiled buckwheat starch jelly and identified as Serratia liquefaciens and Staphylococus lentus. During storage of buckwheat starch jelly for 6 days at 18$^{\circ}C$, viable counts were lower at higher chitosan concentrations, especially at 1.5% and 2.0%. Water activity was less reduced at higher chitosan concentrations. Color L* value decreased, and a* values increased slightly with storage periods. In sensory evaluation, buckwheat starch jelly containng 1.0% chitosan was evaluated the best, with lower overall acceptability at higher chitosan concentrations. Buckwheat starch jelly containing 1.0% chitosan showed a longer shelf-life by 1~2 days than the control.

The effects of Monascus koji on the fermentation and quality of mul-kimchi (CCK) were investigated. Monascus koji at a final concentration of 1% against total amounts of saline and salted Chinese was added to MK and CCK, respectively, and fermented at 1$0^{\circ}C$. The koji of MK and CCK had a little effect on pH and acidity. Antimicrobial effect of water extracts from Monascus koji showed at the concentration of 3% for L. brevis and 5% for L. acidophilus and L. bulgaricus, respectively. Whereas no effect was founded in the strains of L. plantrum, L. GG and Leuc. mesenteroides. It was possible to control the color of kimchi by using Monascus koji instead of red pepper. Especially, color acceptability of MK added the koji was greatly improved. The content of reducing sugar and the score of sweet taste were slightly increased in the kimchi added the koji, whereas no great difference in the crispness was found. Overall acceptability of CCK was not apparent, but that of MK was greatly enhanced. It is apparent that the inhibitions of HMG-CoA reductase in MK and CCK added 1% Monascus koji were 10~20 and 7~16%, respectively.

This study was carried out to determine the antimutagenic effect of soybean sprouts cultured in water containing germanium by Ames test and SOS chromotest. Germanium significantly inhibited the mutagenicity induced by aflatoxin B$_1$ (AFB$_1$) in Salmonella typhimurium TA100 by Ames test, and 4-nitroquinoline-N-oxide (4-NQO) in SOS chromotest. Juice from germanium treated soybean sprouts (GTS) inhibited 57∼75% mutagenicity induced by AFB$_1$, N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and 4-NQO compared with 20∼48% inhibition rate of control soybean sprouts (germanium non-treated soybean sprouts, GNTS) in the Ames test. Also, methanol extracts from GTS inhibited 65% mutagenicity induced by AFB$_1$ in Salmonella typhimurium TA100, and 51% mutagenicity by 4-NQO in SOS chromotest. Therefore, it suggests that GTS has strong potential antimutagenic effect.
