용매와 추출조건이 계피추출액의 항산화성에 미치는 영향 (Effect of Solvents and Some Extraction Conditions on Antioxidant Activity in Cinnamon Extracts)

  • 용매의 종류와 추출온도, 시간, 용매첨가량, 추출횟수 등의 추출조건이 계피추출액의 이화학적 특성에 미치는 영향을 조사하였다. 수소공여능으로 나타낸 항산화활성도는 12가지 용매중 물과 70% 에탄올 추출액에서 높게 나타나 물과 70% 에탄올을 다음의 추출조건을 조사하기 위한 적정용매로 선정하였다. 추출온도를 $20{\sim}100^{\circ}C$로, 추출시간을 $2{\sim}10$시간으로 하여 추출액을 조제했을 때 항산화활성도와 총 phenol 함량은 $80^{\circ}C$와 8시간 추출액에서 높게 나타났고 285nm와 490nm에서의 흡광도는 온도가 높을수록 증가하였다. 용매 첨가비에 있어서는 계피중량의 20배 이상 첨가시에 큰 변화가 없었으며 추출횟수는 3회 이상의 추출액에서 항산화활성도 및 기타 특성이 현저히 감소하였다.

생산조인(生酸棗仁)과 초산조인(炒酸棗仁)의 추출방법에 따른 효능비교연구 (Study on the Comparison of Effects by Extraction Methods of Roast and Raw Semen Zizyphi Spinosae)

  • To observe the difference and change of the efficacy and ingredient appearing according to whether semen zizyphi spinosae has been roasted and its extraction method, locomotor activity, anticonvulsant activity, sleeping effect of each hot water and UMPM low temperature extracts of roast and raw semen zizyphi spinosae were measured through animal tests. Roast semen zizyphi spinosae showed superior pharmacological efficacy than raw semen zizyphi spinosae. Also, hot water extraction method showed superior pharmacological efficacy than low temperature extraction method(UMPM) that uses ultrasonic waves and microwaves. In short, it was shown that herbal medicine's extraction method needs to maximize medicinal effects by trying optimum extraction methods according to efficacy by considering characteristics of each herbal medicine. Together, there's also the need for in-depth studies on water-soluble ingredients of semen zizyphi spinosae that may be extracted through full hot water extraction method.

Change in plasma cortisol and glucose levels of Oncorhynchus keta according to water temperature

  • Chum salmon (Oncorhynchus keta) is a species of anadromous salmonid inhabiting coastal rivers in the North Pacific and the Arctic in the Bering and is the most widely distributed among Pacific salmon species. It is an important fish species in Korea as the salmon releasing project is being actively carried out. This study was conducted to investigate changes in the physiological activity of O. keta according to water temperature. Three experiments were conducted according to the water temperature and period, and the plasma concentrations of cortisol and glucose were analyzed from the blood samples of the experimental groups. Experiment I is a short-term water temperature experiment, in which water temperature stimulation was given for 4 hours at water temperatures of 12℃, 16℃ (control), 20℃, and 24℃, and a recovery period was given for 4 hours. Experiment II is an experiment in which water temperature stimulation was given for 24 hours, 48 hours, and 72 hours at a high temperature of 24℃, and a recovery period was given for 12 hours, respectively. Experiment III is a long-term water temperature experiment, in which the water temperature was 12℃, 16℃ (control), 20℃, and 24℃ for 8 weeks. As a result of the experiment, in Experiment I, there was no significant difference in the survival rate between the experimental groups, but the concentration of cortisol and glucose in the plasma according to the set water temperature showed a significant difference. In Experiment II, there was no significant trend according to the maintenance time of the high-temperature state, but as the temperature increased, the plasma cortisol and glucose levels significantly increased compared to the control group. In Experiment III, all of the experimental group C (24℃) died in the 1st week, and there was no significant difference in the plasma glucose at the 1st and 8th weeks among the remaining experimental groups.

Alcohol Oxidase와 Alcohol Dehydrogenase를 이용한 기상에서의 Ethanol의 산화반응 (Oxidation of Ethanol in the Gas Phase with Alcohol Oxidase and Alcohol Dehydrogenase)

  • Alcohol oxidase와 alcohol dehydrogenase를 DEAE-Cellulose와 ePG에 고정화시켜 에탄올을 산 화시키는 반응을 기상에서 수행하였다. 이 기상반응 의 반응활성은 반응온도와 효소의 hydration 정도에 큰 영향을 받았다. 온도에 의한 효소활성의 변화는 액상에셔의 경우와 비슷한 경향을 보였다. 즉 기상 반응에서도 $35^{\circ}C$에서 $40^{\circ}C$ 사이에서 최고의 활성을 나타내였다. 효소의 hydration 정도는 효소의 활성 에 매우 큰 영향을 끼치는 인자였는데 water activity가 0.8 정도까지는 water activIty가 증가함 에 따라 효소의 활성이 급격히 증가하다가 그 이상 에서는 다소 감소하는 경향을 보였다. 쇼소의 얀정 성도 water activity에 의해서 크게 영향을 받았는데 water actIvity가 0.5 정도까지는 water activity가 증가함에 따라 안정성이 비교적 급격히 감소하였고 그 이상의 water activity에서는 효소의 안정성이 water activity의 영향을 크게 받지 않고 어느 정도 엘정하게 유지되었다. 담체의 흡착성능이 기상반응에 마치는 영향을 알아보가 위하여 셔로 흡 착성능이 다른 DEAE-Cellulose와 CPG에 고정화 된 효소를 사용하여 반응을 비교해 본 결과 담체의 흡착성능은 반응에 별 영향을 주지 못했다. Alcoh이 O oxidase의 기상반응 속도상수(Km=0.24mM)는 액상에서의 값(Km=11.22mM)과 order of magni tude가 약 2 정도의 차이를 보여주었는데 이는 기질 인 에탄올의 기 액 평형상태에셔의 각상간의 농도의 order of magnitude 차와 비슷하였다. 같은 산화효 소류이나 조효소를 펠요로 하고 반응속도식이 많이 다른 alcohol dehydrogenase 효소에도, 기상반응 속도상수와 사용기질의 기 액평형 데이타와의 이와 같은 연관성이 성립하였다. 이같은 연관성이 산화환 원효소류가 아닌 다른 효소류에도 적용될지의 여부는 앞으로의 연구로 밝혀야 할 것이다.

Regional differences in winter activity of hibernating greater horseshoe bats (Rhinolophus ferrumequinum) from Korea

  • Background: Hibernating bats exhibit ubiquitous winter activity in temperate zones, but there is considerable between- and within-species variety in their intensity and purpose. Bats may fly during winter for sustenance or travel to other hibernacula. This study compared inter-regional variation in the winter activity of the greater horseshoe bat (Rhinolophus ferrumequinum). We predicted that weather and hibernacula-environmental conditions would influence winter activity patterns. Results: Winter activity patterns differed between regions. In the Anseong area, we confirmed movement inside the hibernaculum, but in Hampyeong, we observed movement both inside and between hibernacula. The two regions differ by $4^{\circ}C$ in average winter temperatures. Anseong experiences 22 days during which average daily temperatures exceeded $5^{\circ}C$, whereas Hampyeong experienced 50 such days. During the hibernating period, bat body weight decreased by approximately 17-20% in both regions. Conclusions: Ambient temperatures and winter-roost environments appear to be behind regional differences in hibernating bat activity. As winter temperatures in Korea do not favor insect activity, feeding probability is low for bats. However, bats may need to access water. At Anseong, underground water flows inside the hibernaculum when the reservoir outside is frozen. At Hampyeong, the hibernaculum does not contain a water source, but the reservoir outside does not freeze during winter. In conclusion, water-source location is the most likely explanation for regional variation in the winter activity of hibernating bats.

In Vitro Evaluation of Antioxidant Activity of Lycium barbarum Hot Water Extract and Optimization of Production Using Response Surface Methodology

  • This study is concerned with the optimization of the manufacturing process of a hot water extract containing antioxidant activity from Lycium barbarum, traditionally known to have various physiological activities. For the establishment of the optimization process, the central composite design of response surface methodology(RSM) was used. Thirteen extraction processes were performed by encoding the independent variables, extraction temperature (65.9℃-94.1℃) and extraction time (2.59 hr-5.41 hr). As a result of the experiment, the optimal manufacturing conditions for the extract were 340.0 mg/100 g of GAE at an extraction temperature of 94.1℃ and an extraction time of 5 hr. The maximum yield of flavonoids was 22.44 mg/100 g of HES at an extraction temperature of 94.1℃ and an extraction time of 4 hr. The conditions for producing the extract with the maximum antioxidant capacity (DPPH 92.12%) were 90℃ and 4.5 hr extraction time. Therefore, the optimal manufacturing process conditions for extracts containing total phenol content, flavonoid content, and DPPH radical scavenging activity, which are dependent variables, were extraction temperature of 90-95℃ and extraction time of 4 hr, which were not significantly different from the actual values. Therefore, Lycium barbarum extract rich in total phenol and flavonoid content related to antioxidant function is expected to be used as a functional food and cosmetic material.

질화세균의 암모니아 산화능과 이에 미치는 환경요인의 영향 (Ammonia oxidation activity of nitrifying bacteria and effects of some environmental factors)

  • Ammonia oxidation activity of polluted water samples in Jinhae Bay and isolated strain from the seawater was investigated, and effects of environmental factors such as temperature, salinity, substrate concentration to the ammonia oxidation were also investigated. The ammonia oxidation activities of sediments, 0.01-0.04mg eq. $NO_2-N/l/h$, were exceptionally higher than that of sea water, $0.5{\sim}1{\mu}g$ eq. $NO_2-N/l/h$. the activities of muddy sediments at station 4 and 2 were 0.03~0.04mg eq. $NO_2-N/l/h$ and that of sandy sediment at station 3 was 0.002mg eq. $NO_2-N/l/h$. In the case of sea water, the activity of polluted area, station 1, was 2 times higher than that of offshore, station 4. The isolated strain reached log phase after 30days culturs and its oxidation activity was $2{\sim}3{\mu}g$ eq. $NO_2-N/day$. The maximum oxidation of ammonia by IA 13 strain occured at 30mg/l oxidation increased with the salinity rising up to 100% seawater concentraion. And temperature for maximum oxidation of ammonia was $35^{\circ}C$. the oxidation increased with the salinity rising up to 100% seawater concentration.

저장조건과 수분함량에 따른 버어리종 잎담배 수분활성도 및 이화학적 특성 변화 (Changes of Water Activity and Physico-chemical Properties of Burley Tobacco as Affected by Storage and Moisture Contents)

  • The study was carried out to investigate water activity, physicochemical properties and color degree of burley tobacco stored in shelf with different moisture content for 56 days. The study was consist of two experimental designs, that is, moisture content in burley tobacco was kept to 13 to 15 %, 17 to 19 %, and 21 to 23 % under ambient temperature of $20^{\circ}C$ with relative humidity of 24 %, and another group of in burley tobacco was only kept in ambient temperature of $3^{\circ}C$ with relative humidity of 50 % for 56 days with same water content of former treatment. Normal leaves were kept for 56 days under 21 to 23% of moisture content with increased water activity and TSNA(tobacco-specific nitrosamine content. It was considered that favorable leaves conditions are due to favorable microbial growth resulting in increased TSNA content. The total nitrogen content has not changed in two experimental groups for 56 days, otherwise the nitrate form nitrogen and total alkaloid contents were only increased by 21 to 23 % of water content. Degree of pH change was slightly decreased in both storage and water treatment. It was more decreased in 21 to 23% of moisture content than that of other moisture content. In stored leaf tobacco, higher moisture content caused lower brightness, yellow degree, and carbonization quotient.

비음처리에 따른 붓순나무의 광합성, 엽록소 함량 및 엽 특성 (Photosynthesis, Chlorophyll Contents and Leaf Characteristics of Illicium anisatum under Different Shading Treatments)

  • Illicium anisatum was bred under four different light intensity. Those condition were full sunlight(PPFD $1600{\mu} mol\;m^{-2}s^{-1}$), 30% treatment(PPFD $400{\mu} mol\;m^{-2}s^{-1}$), 50% treatment(PPFD $250{\mu} mol\;m^{-2}s^{-1}$) and 70% treatment(PPFD $100{\mu} mol\;m^{-2}s^{-1}$), respectively. Chlorophyll a and b were increased according to decrease of light intensity. Thirty percent and 50% treatment had not significant different in chlorophyll a and b. Thirty percent treatment was shown the best photosynthetic activity through invested photosynthetic rate, intercellular $CO_2$ concentration and water use efficiency. Photosynthetic activity trend of 50% treatment was similar to 30% treatment. Seventy percent treatment was shown the best photosynthetic activity at low light intensity but that was decreased to lower value than 30% and 50% treatment under high intensity. Control, bred full sunlight, was shown the worst photosynthetic activity at measured all light intensity. That result could imply that was caused by photo-inhibition because of long term exposed of shade tolerant plant at high light intensity. Leaf characteristics had not significant different in leaf length, width and area but leaf dry weight had similar trend to photosynthetic activity.

생물활성탄 공정에서 Geosmin과 MIB의 제거 특성 : 생물분해와 흡착 (Removal Characteristics of Geosmin and MIB in BAC Process : Biodegradation and Adsorption)

  • 본 연구에서는 생물활성탄 공정에서 geosmin과 MIB의 제거시 생물분해와 흡착기작의 역할을 BAC와 biofilter를 이용하여 평가하였다. 정상상태에 도달한 BAC 공정에서 부착 박테리아들의 활성이 저하된 저수온기($9^{\circ}C$)에도 EBCT 30분의 조건에서는 geosmin과 MIB가 완전 제거되었다. 유입수의 수온이 $26^{\circ}C$일 때가 $9^{\circ}C$일 때보다 상층부분에서 부착 박테리아의 생체량과 활성도가 높았고, 여층의 하부로 갈수록 부착 박테리아의 생체량과 활성도의 감소율이 높게 나타났다. 이는 수온이 높을 때 상층부분에서 BOM 제거율이 높아 하층에 공급할 수 있는 BOM 양이 감소하기 때문이다. 부착 박테리아의 활성이 억제된 BAC 공정의 geosmin과 MIB의 제거율은 활성화된 BAC 공정에서의 제거율과 비교하여 큰 차이를 나타내지 않아 BAC 공정에서 geosmin과 MIB의 제거에 흡착기작의 기여도가 높게 나타났다. BAC 공정에서 geosmin과 MIB의 제거시 부착 박테리아에 의한 생물분해 기작과 활성탄 표면이 가지는 흡착 기작이 경쟁적으로 작용하였다.
