Our aim was to evaluate the quality of pine seeds after long-term storage and to analyze the correlation between germination and physiological characteristics. Therefore, we investigated the germination and physiological characteristics of seeds stored for different periods and storage temperatures. Pine seeds lost little viability and vigor after 12 years of storage at 4℃; it is even possible to prolong the storage period at -18℃. The deterioration process operating through long-term storage was characterized by a loss of seed vigor before a loss in seed viability. Electrical conductivity of the seed leachate was significantly higher in seeds that completely lost their viability, and was also found to be significantly correlated with the germination percentage, T50, mean germination time, and germination speed. Among the minerals leaked to the leachate, K had the highest concentration, followed by Na, Ca, Cu, Mg, and Fe, while Mn and Zn were not detected. Concentrations of K, Ca, Cu, Mg, and Fe in the leachate differed significantly by treatment, and in the cases of K, Ca, Cu, Na, and Mg, we observed large differences in the leachate between the seeds that were collected in 2003 and those collected in other years.. Germination percentage was significantly negatively correlated to the concentrations of Ca, Cu, K, Mg, and Na. The coefficient of uniformity of germination was significantly negatively correlated to Cu concentration. The other germination characteristics were not significantly correlated with the mineral concentration. Therefore, the mineral concentrations of pine seed leachate were not reliable indicators of seed deterioration when the differences in quality between seed lots were small. However, electrical conductivity could be an indicator of seed viability and vigor of pine seeds under long-term storage.
The viability loss or death of weed seeds buried in soil can be induced by infrared irradiation which has good penetration in moist soil. By using this principle of pre-emergence soil-treatment, the study was carried out to obtain basic information needed to develop the effective weed control method for the production of less polluted agricultural products. An apparatus for irradiating infrared was constructed by using ceramic material with high emissivity. The LPG was used as fuel for producing infrared by heating ceramic material. The soil heated in this study was loam with four levels of moisture contents (0.6, 5.7, 10.7, 15.1 % wb). The temperature distribution was measured at various soil depths when soil with different moisture content was irradiated with infrared for three different times (30, 60, 90 sec). The soil depths with duration time of minimum 3 minutes over $80^{\circ}C$, temperature inducing viability loss of weed seeds, were investigated. When the moisture content of soil was 0.6 and 5.7 % wb, the soil depths which can induce viability loss of weed seeds was greatly increased with increasing irradiation time. However, any depths of soil tested in this study was not reached to the temperature of $80^{\circ}C$ when 30 seconds of irradiation time was applied on soil with moisture content of 10.7 or 15.1 % wb. Generally, the soil depth needed for viability loss of weed seeds was decreased with increasing moisture content of soil. Also, longer irradiation time was required to induce viability loss of weed seeds with increasing moisture content of soil.
The purpose of this research was to study the effect of freeze drying on the microorganisms in kefir. Influences of lyoprotectants and rehydrated media (water at $4^{\circ}C$, $25^{\circ}C$; 10% reconstituted milk at $4^{\circ}C$, $25^{\circ}C$) on the viability of lactic acid bacteria and yeasts in freeze-dried kefir were investigated. Kefir was made from cow milk which was inoculated with 5% kefir grains, and incubated at $20^{\circ}C$ for 20 h. Lyoprotectants (galactose, lactose, maltose, sucrose and trehalose) were added independently before dehydration of kefir by freeze drying. Results indicated significant loss in viability of microorganisms in kefir after freeze-drying. Addition of 10% galactose or 10% sucrose as lyoprotectants significantly increased the survival rates of both lactic acid bacteria and yeasts (p<0.05). The $4^{\circ}C$ rehydration temperature showed the best viabilities for yeasts, however, viability was not significantly affected by rehydration media (p>0.05).
In this study, we investigated the antioxidant activity of hesperidin and hesperetin, which are the active compounds from Citrus junos, in the cellular system. Under cellular model of oxidative damage using LLC-$PK_1$ renal epithelial cell, the oxidative damage induced by 2,2'-azobis (2-amidinopropane) dihydrochloride (AAPH) led to the loss of cell viability, while treatment of hesperidin and hesperetin increased significantly the cell viability as dose-dependent manner. In addition, NO-induced cellular oxidative damage by sodium nitroprusside were significantly recovered by the treatment of hesperidin and hesperetin, showing the increase of cell viability. But hesperidin and hesperetin showed no significant protective effect on $O_2{^-}$-induced cellular oxidative damage. The present study indicates that hesperidin and hesperetin protect against free radical, especially AAPH-induced peroxyl radical. In particular, hesperetin has stronger protective effect against oxidative stress than hesperidin.
The present work evaluated the effects of alcoholic extracts of salvia (Salvia officinalis), thyme (Thymus vulgaris), and 2 pure compounds (thymol and menthol) on the viability of Echinococcus granulosus protoscolices in vitro. Four different concentrations of each extract (2,500, 1,500, 1,000, and 500 ${\mu}g$/ml) and 3 different concentrations each of thymol and menthol (50, 10, and 1 ${\mu}g$/ml) were used. Concentration of 2500 ${\mu}g$/ml of both extracts showed a significant protoscolicidal activity on the 6th day. Complete loss of viability of protoscolices occurred with 500 ${\mu}g$/ml concentration of both extracts at day 6 and day 7 post-treatment (PT), respectively. Pure compounds, i.e., menthol and thymol, showed potent effects with 50 ${\mu}g$/ml concentration at day 2 and day 5 PT, respectively. These effects were compared with those of albendazole sulfoxide (800 ${\mu}g$/ml), a commonly used treatment drug for hydatidosis. Krebs-Ringer solution and the hydatid cystic fluid at a ratio of 4:1 was a good preservative solution which kept the protoscolices viable for 15 days.

The effect of microencapsulation on previously isolated Lactobacillus delbrueckii 94/L4 as starter culture for yogurt, and Lactobacillus casei 97/L3 as a probiotic candidate was investigated. Preliminary results showed that L. delbrueckii 94/L4 exhibited tolerance to bile, unlike L. casei 97/L3. Freeze drying significantly (p < 0.05) reduced the viability of both isolates by log 0.71-2.70. Although microencapsulation preserved the viability of L. casei 97/L3 cells exposed to simulated gastrointestinal tract conditions for 120 min, it did not impart significant (p < 0.05) protection against loss of viability during the first 30 min of exposure. Conversely, microencapsulated L. delbrueckii 94/L4 with the addition of Streptococcus thermophilus 24/S1 as starter culture was successfully incorporated into milk to form yogurt, yielding a significantly (p < 0.05) improved product quality.

In order to promote the efficiency of strawberry breeding programs, pollen viability of strawberry, 'Seolhyang' was investigated using the fluorochromatic reaction test and seed set under controlled environment. Pistil receptivity was also assessed by hand pollination. Four varieties including 'Maehyang' were used for the test of pistil receptivity with 'Seolhyang' as a pollen parent. Pollen viability remained high for several days under dry conditions as below 33% relative humidity while the greatest loss of viability occurred at 76% relative humidity. The viability was rapidly decreased at high humidity and almost all grains were unviable in 7 days after storage. Pollen viability does not appear to be drastically reduced if the relative humidity is low. Therefore, humidity is more important factor than temperature for the pollen viability in Fragaria${\times}$ananassa. The rate of seed set by hand pollination lasted higher than the average of 77.2% from 2 to 8 days after emasculation when the daily average temperature was around $15^{\circ}C$ in plastic house. It began to decline gradually from 10 days and had decreased dramatically after 12 days except several cultivars. Based on the daily mean accumulated temperature, it is recommended to have an artificial pollination between the range of $45{\sim}140^{\circ}C$ after the emasculation to increase the rate of seed set in strawberry.
This study aims to investigate the biological activities related to hair loss of short anti-oxidant peptides (DK peptides) $5{\alpha}$-reductase inhibition and anti-oxidation. The series of DK peptides were generated amphipathic helical properties using leucines, lysines and tryptophan residues. Cell viability and free radical scavenging activities were performed using 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl-tetrazolium bromide (MTT) and 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assay, respectively. The DK peptides were investigated for inhibitory activity against $5{\alpha}$-reductase. Antioxidant activities were determined by means of, 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assays. All peptides could inhibit $5{\alpha}$-reductase in lipopolysaccharide-stimulated macrophage. In conclusion, DK peptides was suggested as the most attractive ingredients for improving hair loss, because of the high inhibitory against $5{\alpha}$-reductase inhibition.
Objectives: The root of Platycodon grandiflorum (PG) has been known to possess a range of pharmacological activities including anti-cancer, anti-inflammatory, and anti-oxidant effects. The present study was designed to investigate whether or not PG-induced cell death was connected with autophagy and apoptosis in NCI-H460 human lung cancer cells. Methods: Effects on the cell viability and apoptotic activity were quantified using MTT assays and flow cytometry analysis, respectively. Protein activation was measured by immunoblotting. Autophagy was measured by LC3 immunofluorescence and immunoblotting. ROS production and loss of mitochondria membrane potential (MMP) were checked with flow cytometry analysis. Results: Following exposure to PG, NCI-H460 cell proliferation decreased simultaneously inducing autophagic vacuoles and up-regulation of microtubule-associated protein 1 light chain 3 and beclin-1 protein expressions. Interestingly, pre-treated with autophagy inhibitors, 3-methyladenin or bafilomycin A1 further triggered reduction of cell viability. PG treatment also induced apoptosis that was related modulation of Bcl-2 family proteins, death receptors and activation of caspases. In addition, PG stimulation clearly enhanced loss of MMP and reactive oxygen species (ROS) generation. Conclusions: Our results suggest that PG elicited both autophagy and apoptosis by increasing loss of MMP and ROS production. PG induced-autophagy may play a cell protective role.
Rat renal proximal tubule suspension was prepared from adult male Sprague Dawley rat (250-300g) by mechanical (non-enzymatical) method and evaluated as a pontential model for mechanistic studies and early screening of nephrotoxicity, using anionic antibiotics (cephaloridine). Cephaloridine (CPL) produced an increase in LDH release into media. This release results from decrease a proximal tubule cell viability and subsequently increase the permeability of cell viability and subsequently increase the permeability of cell membrane. Since loss of intracellular potassium and ATP into media is the sign of disruption of cell membrane, especially basolateral membrane (BLM), CPL induced proximal tubule cell compromise also appear be associated with BLM, maybe $Na^+-K^+$ ATPase. Also seen was significant depression in brush border membrane (BBM) ALP activity and no significantly increase in BBM GGT activities. The inhibition of typical anion, PAH accumulation (especially, CPL 5 mM) and cation, TEA (especially, 4hours incubation) were seen dose dependently. This is because of CPL accumulation in renal proximal tubule and increase of cytotoxicity.
