In order to gain insight into the molecular changes at the protein level in plants exposed to low temperature for a long period of time (vernalization), proteome analyses of vernalization-treated Arabidopsis thaliana have been carried out by two-dimensional gel electrophoresis followed by matrix-assisted laser desorption/ ionization time-of-flight mass spectrometry. Fourteen proteins including ATP binding/GTP binding/translation elongation factor and glycine-rich RNA-binding protein 7 (GRP7) showed differential expression in vernalization-treated Arabidopsis thaliana. GRP7 showed the most dramatic increase in expression suggesting its involvement in response to vernalization treatment.

To investigate genes related to vernalization and cold- resistance in barley (Hordeum vulgare L. cv. Nagaoka), differentially expressed genes were identified from cold-resistant barley leaves with suppression subtractive hybridization (SSH) and Northern blot analyses. The nucleotide and the deduced amino acid sequences of the putative gene products were compared. The bvrn-7 showed high homology(84%) with gene related to vernalization, and the bvrn-3, bvrn-12, bvrn-28, bvrn-29 and bvrn-36 related to cold-resistant genes had high identity of 88~98% with low temperature-induced genes. The results indicate that the 6 genes were closely related to vernalization and cold-resistance during low temperature treatment.

A traditional wheat breeding program needs more than 12-13 years to develop a new cultivar. In recent years, 'Speed breeding (SB)' system, which uses extremely extended photoperiod (22 h), enabled up to 4-6 generations of spring wheat per year. However, since almost 70% of wheat cultivars are winter type, and over 95% of total cultivation area is for winter wheat in Korea, optimized vernalization treatment was essential for improving the SB system. Several vernalization temperatures and durations were tested with various genotypes, and the 4 weeks of 8-10 ℃ vernalization treatment was the most effective to develop 4 generations per year, for both spring and winter type wheat cultivars. This 'Speed vernalization (SV)' system followed by SB, allowed developing a new F6 recombinant inbred lines (RILs) within 2 years. Among the 184 RILs, which derived from a cross between Jokoyung and Joongmo2008, two outstanding lines were selected for yield trial test, and then named Milyang52 and Milyang53. Compared to the traditional wheat breeding program, over 60% of the time was saved to develop these two lines. Marker-assisted selection and backcross were also combined with the SV system. YW3215-2B-1 (Jokoyung*3/Gamet), which has similar agronomic traits with Jokyoung and the same Glu-B1 allele with Garnet, was developed within 2.5 years. Thus, the SV system combined with molecular breeding technology would help breeders to make a new cultivar with less time and high efficiency.
Expression of 43 flowering-related genes was examined in two inbred lines of Chinese cabbage, Chiifu and Kenshin, under different photoperiod, vernalization and flower development stages. The floral genes cloned by RT-PCR with degenerated primers showed high homology with Arabidopsis counterparts. Genes in two inbred lines, TOC, CRY1, CO, RGAL and GAl, were highly expressed under all tested conditions. However, expression of three genes was regulated by particular experimental conditions. The expression of LHY gene was predominant in Chiifu under the short-day conditions, whereas the expression of RGAL gene was influenced by vernalization in both inbred lines. Besides, the expression of NAP gene was induced by vernalization only in Chiifu. Most of the flower identity-related genes were expressed during flower development. The transcript level for several genes was not detected in this experiment.
The effects of vernalization and darkness on flower initiation were reviewed by using naked barley varieties. The dark effects were pronounced on flower induction even at 25$^{\circ}C$. There were no differences in flowering responses among varieties. The effects of vernalization and darkness combined with 25$^{\circ}C$were 100% and 60% respectively which showed a significant difference. When the alternation of darkness with light was taken as first and last the flowering rate was 92 percent. When it was intercalated by light treatments the flowering rate was 91 per cent. Thus the effects of vernalization and darkness seems to differ.

This study was carried out to determine the effects of vernalization, temperature, and soil water deficit (SD) on mesophyll cells, growth, and yield of Chinese cabbage (Brassica campestris L). The palisade parenchyma and spongy tissues of Chinese cabbage were observed under full irrigation and two weeks of SD treatment. These cells were severely collapsed by four weeks SD treatment. The SD treatment had the greatest influence on the growth of Chinese cabbage among the tested treatment factors (vernalization, temperature, and SD), growth significantly decreased by severe drought treatment (four weeks SD treatment). In addition, the relative growth rate, unit leaf rate, leaf area ratio, specific leaf area, and leaf weight ratio were significantly affected by SD treatment; however, other individual factors and their combined treatments did not influence the analyzed growth parameters. The yield under vernalization after high temperature and full irrigation treatments was 3,056 kg/10 a, which was the greatest among all the tested treatments, while four-week SD treatment significantly reduced the yield. Head formation of Chinese cabbage was not altered under SD treatment, and vernalization treatments did not induce bolting. Our results indicated that collapsing mesophyll cells and reduced growth and yield were induced by SD treatment. Thus we suggest that optimal irrigation system should be install to avoid or overcome crippling drought conditions in the open field.

Flowering is one of the most important development traits related to the production of Brassica rapa crops. After planting, a sudden low temperature triggers premature flowering, which leads to a reduction in the yield and quality of harvested production. Therefore, understanding the mechanism of flowering control is important in the agricultural productivity for preventing Brassica rapa crops. Vernalization is generally known as the main factor of flowering in the Brassica plant. However, in the subspecies of Brassica rapa, some accession such as Yellow sarson and Komatsuna display the flowering phenotype without vernalization. Circadian genes, which diurnally regulate plant physiology, have a role for photoperiodic flowering but are related to the regulation of the vernalizarion mechanism. In this report, the 22 B. rapa accession were divided into two groups, vernalization and non-vernalization, and the sequenced circadian gene, BrPRR1s. Among them, the BrPRR1b gene was found to have deletion regions, which could classify the two groups. The PCR primer was designed to amplify a short band of 422bp in the vernalization type and a long band of 451bp in the non-vernalization type. This primer set was applied to distinguish the flowering types in the 43 B. rapa accession and 4 Brassica genus crop, Broccoli, cabbage, mustard, and rape. The PCR analysis results and flowering time information of each crop demonstrated that the primer set can be used as marker to discern the flowering type in Brassica crops. This marker system can be applied to the B. rapa breeding when selecting the flowering character of new progenies or introducing varieties at an early stage. In addition, these results displayed that the circadian clock genes can be a good strategy for the flowering control of B. rapa crops.
Seven barley varieties were tested under three different vernalization durations for observing the effects of vernalization duration and different growth habit on shoot apex development and heading date. The final leaf number per main stem in 3 and 6 weeks vernalized seedlings did not vary among varieties, but ranged 7 to 14 leaves in non-vernalized seedlings. The winter types had more leaves than the spring types. Days for each leaf emergence in non-vernalization were retarded 1. 3 to 1. 5 days in comparison with 3 or 6 weeks vernalized seedlings. In general, the leaf emergence speed of spring types was faster than that of winter types. The VI stage whose double ridge formed, did not vary in 6 weeks vernalization, but spring (Gangbori & Dongbori 2) and facultative or winter types showed two conspicuous difference patterns. The differences of days to X stage were great among different vernalization duration and varieties; the stage of spring types was reached faster than that of winter types. The early varieties within the same growth habit were reached to X stage faster, and the time of flag leaf emergence showed the similar tendency to the differentiation of X stage. The time of the first rapid stem internode elongation became late as for incompletely vernalized seedlings. The time within the same vernalization duration became later in winter types than in spring types, and even within the same growth habit, the time of early varieties became faster than that of late varieties. The growth habit in especially non-vernalized seedlings had highly significant correlation coefficients with the times of leaf development speed, leaf number per main stem, the first rapid stem internode and young spike elongations, X stage, and flag leaf emergence. However, the relationship between growth habit and time of heading in the field was not close.
