  • Compensation of temperature is very important to make high precision load cell. In this study, we developed a new type load cell. The structure of the load cell has four strain gauges on single side of the load cell beam. Also a new temperature compensation method was proposed and these characteristics were better than previous one. This study will offer application to other type of load cell and another sensors.

The Study on Rough Colony Type Mutant of Pseudomoms mori(Boyer et Lambert) Stevens, caused Mulberry Bacterial Blight: Pathogenicity and General Characteristics (뽕나무 세균성위축병균 Pseudomonas mori (Boyer et Lambert) STEVENS의 Rough Colony Type Mutant에 관한 연구 ; 병원성 및 일반적 성질)

  • The study has been carried out to compare the pathogenicity, physiological characteristics and genetic reliability between rough colony type strain and smooth colony type strains of Pseudomonas mori (Boyer et Lambert) Stevens which were isolated from diseased plant parts in 5 different areas throughout country. The results are summarized as follow. 1. The rough colony type strain showed more agressive reactions to tested host plant varieties than smooth colony type strains though there was no differences in the appearence of lesion types caused by both strains. 2. Both colony types were differentiated morphologically in that the rough colony type strain was having more than 200r long filamentous body without flagella where as the smooth colony type strains have short rods with one or several polar flagella. 3. The colony of smooth type strains was circular, entire, smooth and opaque, while the rough type strain shelved endulated, irregular margin, rough and wrinkled colony on nutrient agar media. 4. There were no differences between both colony types in the physiological and serological test. 5. Both of smooth and rough colony type strains showed genetic reliability through more than 100 succeeding cultures on the media, and were stable to various chemicals such as 1 to 3 percent of NaCl, 5 kinds of organic acid and 4 kinds of antibiotics.

Recent results on the analysis of viscoelastic constitutive equations

  • Recent results obtained for the port-pom model and the constitutive equations with time-strain separability are examined. The time-strain separability in viscoelastic systems Is not a rule derived from fundamental principles but merely a hypothesis based on experimental phenomena, stress relaxation at long times. The violation of separability in the short-time response just after a step strain is also well understood (Archer, 1999). In constitutive modeling, time-strain separability has been extensively employed because of its theoretical simplicity and practical convenience. Here we present a simple analysis that verifies this hypothesis inevitably incurs mathematical inconsistency in the viewpoint of stability. Employing an asymptotic analysis, we show that both differential and integral constitutive equations based on time-strain separability are either Hadamard-type unstable or dissipative unstable. The conclusion drawn in this study is shown to be applicable to the Doi-Edwards model (with independent alignment approximation). Hence, the Hadamardtype instability of the Doi-Edwards model results from the time-strain separability in its formulation, and its remedy may lie in the transition mechanism from Rouse to reptational relaxation supposed by Doi and Edwards. Recently in order to describe the complex rheological behavior of polymer melts with long side branches like low density polyethylene, new constitutive equations called the port-pom equations have been derived in the integral/differential form and also in the simplifled differential type by McLeish and carson on the basis of the reptation dynamics with simplifled branch structure taken into account. In this study mathematical stability analysis under short and high frequency wave disturbances has been performed for these constitutive equations. It is proved that the differential model is globally Hadamard stable, and the integral model seems stable, as long as the orientation tensor remains positive definite or the smooth strain history in the flow is previously given. However cautious attention has to be paid when one employs the simplified version of the constitutive equations without arm withdrawal, since neglecting the arm withdrawal immediately yields Hadamard instability. In the flow regime of creep shear flow where the applied constant shear stress exceeds the maximum achievable value in the steady flow curves, the constitutive equations exhibit severe instability that the solution possesses strong discontinuity at the moment of change of chain dynamics mechanisms.

  • The serological typing of staphylococcus hyicus subsp. hyicus isolated from pigs with exudative epidermitis and healthy pigs was attempted by slide agglutination technique with absorbed sera prepared from antisera against swine strains of the organism. Of 204 Staph. hyicus subsp. hyicus isolated, 202 (99.0%) were classified into 6 serotypes tentatively designated as A, B, C, D, E and F. The organisms were found to possess common antigens and a strain specific antigen. In 26 epizootics of exudative epidermitis, type A was found in 4(15.4%), type B in 12(46.2%), type C in 9(34.6%) and type E in 1(3.8%). All of the piglets of each affected litter were infected by serologically identical strain. In 178 strains isolated from the skin of healthy pigs, type A was found in 14.1%, type B in 23.0%, type C in 30.3%, type D in 1.7%, type E in 7.9% and type F in 21.9%, and 2 strains were untypable. There was a difference in the occurrence of serotypes of Staph. hyicus subsp. hyicus among the farms examined.

Site-specific Disruption of Glyoxylate Bypass and Its Effect in Lysine-producing Corynebacterium lactofermentum Strain

  • The role of glyoxylate bypass in a lysine-producing Corynebacterium lactofermentum strain was analyzed. Unlike the wild type, the strain expressed enzymes of glyoxylate bypass during growth in the fermentation broth containing glucose as the carbon source. To evaluate the importance of glyoxylate bypass in the strain, we disrupted chromosomal aceA by using a cloned fragment of the gene. Site-specific disruption of aceA which codes for the isocitrate lyase, the first enzyme of the bypass, was confirmed by Southern blot analysis. The aceA mutant strain completely lost isocitrate lyase activity and ability to grow in a minimal medium containing acetate as the sole carbon source. The mutant strain was similar to its parental strain in growth characteristics and produced comparable amounts of lysine in shake flasks containing glucose as the carbon source. The amount of oxaloacetate accumulated in the fermentation medium was similar for both strains, suggesting that expression of glyoxylate bypass does not necessarily lead to the increase in intracellular oxaloacetate. These data clearly demonstrate that glyoxylate bypass does not function as one of the routes of carbon supply for lysine production in the strain. It appears that the leakiness of the glyoxylate bypass in the strain might be the result of a secondary mutation which arose during previous strain development by random mutagenesis.

Interaction between the Rice Pathogens, Fusarium graminearum and Burkholderia glumae

  • Species belonging to the genus Fusarium are widely distributed and cause diseases in many plants. Isolation of fungal strains from air or cereals is necessary for disease forecasting, disease diagnosis, and population genetics [1]. Previously we showed that Fusarium species are resistant to toxoflavin produced by the bacterial rice pathogen Burkholderia glumae while other fungal genera are sensitive to the toxin, resulting in the development of a selective medium for Fusarium species using toxoflavin [2]. In this study, we have tried to elucidate the resistant mechanism of F. graminearum against toxoflavin and interaction between the two pathogens in nature. To test whether B. glumae affects the development of F. graminearum, the wild-type F. graminearum strains were incubated with either the bacterial strain or supernatant of the bacterial culture. Both conditions increased the conidial production five times more than when the fungus was incubated alone. While co-incubation resulted in dramatic increase of conidial production, conidia germination delayed by either the bacterial strain or supernatant. These results suggest that certain factors produced by B. glumae induce conidial production and delay conidial germination in F. graminearum. To identify genes related to toxoflavin resistance in F. graminearum, we screened the transcriptional factor mutant library previously generated in F. graminearum [3] and identified one mutant that is sensitive to toxoflavin. We analyzed transcriptomes of the wild-type strain and the mutant strain under either absence or presence of toxoflavin through RNAseq. Expression level of total genes of 13,820 was measured by reads per kilobase per million mapped reads (RPKM). Under the criteria with more than two-fold changes, 1,440 genes were upregulated and 1,267 genes were down-regulated in wild-type strain than mutant strain in response to toxoflavin treatment. A comparison of gene expression profiling between the wild type and mutant through gene ontology analysis showed that genes related to metabolic process and oxidation-reduction process were highly enriched in the mutant strain. The data analyses will focus on elucidating the resistance mechanism of F. graminearum against toxoflavin and the interaction between the two pathogens in rice. Further evolutionary history will be traced through figuring out the gene function in populations and in other filamentous fungi.

  • This paper presents the Inkjet-printed strain gauge using PEDOT:PSS. The strain gauge (width 0.6 mm, length 20 mm, thickness $0.3{\mu}m$) was printed on the PET film using PEDOT:PSS ink. The resistance variation of the fabricated strain gauge was measured by the digital multi-meter with the displacement range of -4 to 10 mm. As the measured result, resistance variation (${\Delta}R/R_0$) has approximately 0.75%, linearity of 99.87%. The fabricated strain gauge is expected to the various applications such as tape type pressure sensor, PMS(pressure mapping sensor), wearable devices.

  • Strain gages were widely used transducers. Essentially a strain gage was an electric element to which an appropriate type was attached. Strain was sensed by gages and provided electrical output proportional to applied forced. This paper describes the recent development of a thick film strain gage ceramic pressure sensors. The thick film resistors as strain gage in the Wheatstone bridge were fabricated with a novel mixture of ruthenium. The thick-film technology of resistors were printed on the ceramic diaphragm back side by screen printing and cured at $850^{\circ}C$. The mechanical measurements were performed with the computer simulation results(ANSYS 5.1). The output sensitivity was 1.2mV/V, of which max. nonlinearity was less than 0.29%, hysteresis was less than 0.38%FS.

Strain Analysis in GFRP Cross-Ply Laminates Using TR-EFPI Optical Fiber Sensor (광섬유 TR-EFPI 센서를 이용한 GFRP 직교 적층판의 변형률 해석)

  • Longitudinal strains({$varepsilon}_x$) of the core and skin layers in glass fiber reinforced plastic(GFRP) cross-ply composite laminates have been studied using the embedded optical fiber sensor of totally-reflected extrinsic Fabry-Perot interferometer(TR-EFPI). Foil-type strain gauges bonded on both the upper and lower surfaces were used for the measurement of the surface strains. Both TR-EFPI sensor and strain gauge bonded on the specimen surface showed excellent agreement within -0.0086 ~ +0.0302% strain. It was shown that values of {$varepsilon}_x$ in the interior of the surface layer and the core layer measured by embedded TR-EFPI sensor was significantly higher than that of the specimen surface measured by strain gauges. The experimental results were ascertained with finite element analysis. Embedded TR-EFPI optical fiber sensor could measure accurately the internal strains which were different from the surface.

  • The dynamic softening behavior of type 430 ferritic stainless steel could be characterized by the hot torsion test in the temperature range of 900-110$0^{\circ}C$ and the strain rate range of 0.05-5/sec. It is found that the continuous dynamic recrystallization (CDRX) was a major dynamic softening mechanism. The effects of process variables strain ($\varepsilon$) stain rate($\varepsilon$)and temperature (T) on CDRX could be individually established from the analysis of flow stress curves and microstructure. The effect of CDRX individually established from the analysis of flow stress curves and microstructure. The effect of CDRX increased with increasing strain rate and decreasing temperature in continuous deformation. The multipass deformation processes were performed with 10 pass deformations. The CDRX effect occurred in multipass deformatioon. The grain refinement could be achieved from multipass deformation The grain refinement increased with increasing strain rate and decreasing temperature. Also the CDRX in multipass deformation was affected by interpass time and pass strain. The total strain was to be found key parameter to occur CDRX.
