Transcriptional Alteration of p53 Related Processes As a Key Factor for Skeletal Muscle Characteristics in Sus scrofa

  • The pig could be a useful model to characterize molecular aspects determining several delicate phenotypes because they have been bred for those characteristics. The Korean native pig (KNP) is a regional breed in Korea that was characterized by relatively high intramuscular fat content and reddish meat color compared to other western breeds such as Yorkshire (YS). YS grew faster and contained more lean muscle than KNP. We compared the KNP to Yorksire to find molecular clues determining muscle characteristics. The comparison of skeletal gene expression profiles between these two breeds showed molecular differences in muscle. We found 82 differentially expressed genes (DEGs) defined by fold change (more than 1.5 fold difference) and statistical significance (within 5% of false discovery rate). Functional analyses of these DEGs indicated up-regulation of most genes involved in cell cycle arrest, down-regulation of most genes involved in cellular differentiation and its inhibition, down-regulation of most genes encoding component of muscular-structural system, and up-regulation of most genes involved in diverse metabolism in KNP. Especially, DEGs in above-mentioned categories included a large number of genes encoding proteins directly or indirectly involved in p53 pathway. Our results indicated a possible role of p53 to determine muscle characteristics between these two breeds.

Presenting an advanced component-based method to investigate flexural behavior and optimize the end-plate connection cost

  • A very widely used analytical method (mathematical model), mentioned in Eurocode 3, to examine the connections' bending behavior is the component-based method that has certain weak points shown in the plastic behavior part of the moment-rotation curves. In the component method available in Eurocode 3, for simplicity, the effect of strain hardening is omitted, and the bending behavior of the connection is modeled with the help of a two-line diagram. To make the component method more efficient and reliable, this research proposed its advanced version, wherein the plastic part of the diagram was developed beyond the guidelines of the mentioned Regulation, implemented to connect the end plate, and verified with the moment-rotation curves found from the laboratory model and the finite element method in ABAQUS. The findings indicated that the advanced component method (the method developed in this research) could predict the plastic part of the moment-rotation curve as well as the conventional component-based method in Eurocode 3. The comparison between the laboratory model and the outputs of the conventional and advanced component methods, as well as the outputs of the finite elements approach using ABAQUS, revealed a different percentage in the ultimate moment for bolt-extended end-plate connections. Specifically, the difference percentages were -31.56%, 2.46%, and 9.84%, respectively. Another aim of this research was to determine the optimal dimensions of the end plate joint to reduce costs without letting the mechanical constraints related to the bending moment and the resulting initial stiffness, are not compromised as well as the safety and integrity of the connection. In this research, the thickness and dimensions of the end plate and the location and diameter of the bolts were the design variables, which were optimized using Particle Swarm Optimization (PSO), Snake Optimization (SO), and Teaching Learning-Based Optimization (TLBO) to minimization the connection cost of the end plate connection. According to the results, the TLBO method yielded better solutions than others, reducing the connection costs from 43.97 to 17.45€ (60.3%), which shows the method's proper efficiency.

  • 많은 세균들은 환경적 스트레스에 대항하기 위해 세균 생존에 유용한 특정 유전자들의 전사를 유도하는 대체시그마 인자 RpoS를 활용한다. 세포 내 RpoS 단백질의 농도는 주로 ClpXP 단백질 분해효소의 조절을 통해 결정된다. RpoS를 ClpXP로 전달하기 위해서는 adaptor 단백질 RssB가 반드시 필요하다. Two-component-type response regulator RssB는 RpoS와 지속적으로 상호작용을 하지만, 다양한 환경변화에 의해 RssB-RpoS 상호작용이 억제되어 세균에서 RpoS 양을 증가시킨다. 본 총설에서는 최근까지 연구 된 RssB-RpoS 상호작용에 관여하는 RssB의 anti-adaptor 단백질 IraD, IraM, IraP 등의 조절인자들과 RssB의 N-terminal 수용체 도메인의 인산화에 대해 설명하고 요약하였다. 이러한 RssB의 정교한 활성을 통한 RpoS 분해조절 과정은 외부환경 스트레스로부터 보다 효율적으로 세균을 보호할 수 있다.

Regulation of autonomic functions following two high frequency yogic breathing techniques

  • Yoga is an ancient Indian system of life, encompassing various practices including practices for self-discipline and also for regulating the health states of the individual, being practiced for thousands of years. The present study aims at understanding the effect of two high frequency breathing practices over autonomic nervous system. Forty healthy male volunteers of age $21{\pm}2$ years with $9{\pm}3$ months of Yoga practice experience were recruited. The two high frequency Yoga breathing practices, kapalabhati (KB) and bhastrika (BH) were given as interventions randomly on either of the two days to minimise laboratory bias. They were assessed before and immediately after the interventions for heart rate, respiratory rate, heart rate variability (HRV), blood pressure and peripheral oxygen saturation. There was a significant increase in heart rate (p<0.01; p<0.001), systolic blood pressure (p<0.01; p<0.001), NN50 (p<0.01; p<0.001) component of HRV for both KB and BH groups respectively. There was a significant reduction in respiratory rate in both the groups (p<0.001, and p<0.05, BH and KB respectively) immediately following intervention. A significant increase in LF component of HRV and reduction in Diastolic blood pressure and high frequency (HF)component following KB was also observed (p<0.05, for all comparisons). The Mean peripheral oxygen saturation remained unaltered in both the groups (p>0.05).The results suggest that high frequency yoga breathing practices induce physiological arousal immediately as evidenced by increased blood pressure and heart rate. The sympathetic arousal was more following KB session as evidenced by an increased diastolic blood pressure, LF power and a decrease in HF power of HRV as compared to the BH session.

LIN-23, an E3 Ubiquitin Ligase Component, Is Required for the Repression of CDC-25.2 Activity during Intestinal Development in Caenorhabditis elegans

  • Caenorhabditis elegans (C. elegans) utilizes two different cell-cycle modes, binucleations during the L1 larval stage and endoreduplications at four larval moltings, for its postembryonic intestinal development. Previous genetic studies indicated that CDC-25.2 is specifically required for binucleations at the L1 larval stage and is repressed before endoreduplications. Furthermore, LIN-23, the C. elegans ${\beta}$-TrCP ortholog, appears to function as a repressor of CDC-25.2 to prevent excess intestinal divisions. We previously reported that intestinal hyperplasia in lin-23(e1883) mutants was effectively suppressed by the RNAi depletion of cdc-25.2. Nevertheless, LIN-23 targeting CDC-25.2 for ubiquitination as a component of E3 ubiquitin ligase has not yet been tested. In this study, LIN-23 is shown to be the major E3 ubiquitin ligase component, recognizing CDC-25.2 to repress their activities for proper transition of cell-cycle modes during the C. elegans postembryonic intestinal development. In addition, for the first time that LIN-23 physically interacts with both CDC-25.1 and CDC-25.2 and facilitates ubiquitination for timely regulation of their activities during the intestinal development.

Genome-wide Screening to Identify Responsive Regulators Involved in the Virulence of Xanthomonas oryzae pv. oryzae

  • Two-component systems (TCSs) are critical to the pathogenesis of Xanthomonas oryzae pv. oryzae (Xoo). We mutated 55 of 62 genes annotated as responsive regulators (RRs) of TCSs in the genome of Xoo strain PXO99A and identified 9 genes involved in Xoo virulence. Four (rpfG, hrpG, stoS, and detR) of the 9 genes were previously reported as key regulators of Xoo virulence and the other 5 have not been characterized. Lesion lengths on rice leaves inoculated with the mutants were shorter than those of the wild type and were significantly restored with gene complementation. The population density of the 5 mutants in planta was smaller than that of PXO99A at 14 days after inoculation, but the growth curves of the mutants in rich medium were similar to those of the wild type. These newly reported RR genes will facilitate studies on the function of TCSs and of the integrated regulation of TCSs for Xoo pathogenesis.

Effect of Mutations of Five Conserved Histidine Residues in the Catalytic Subunit of the cbb3 Cytochrome c Oxidase on its Function

  • The cbb3 cytochrome c oxidase has the dual function as a terminal oxidase and oxygen sensor in the photosynthetic bacterium, Rhodobacter sphaeroides. The cbb3 oxidase forms a signal transduction pathway together with the PrrBA two-component system that controls photosynthesis gene expression in response to changes in oxygen tension in the environment. Under aerobic conditions the cbb3 oxidase generates an inhibitory signal, which shifts the equilibrium of PrrB kinase/phosphatase activities towards the phosphatase mode. Photosynthesis genes are thereby turned off under aerobic conditions. The catalytic subunit (CcoN) of the R. sphaeroides cbb3 oxidase contains five histidine residues (H2l4, B233, H303, H320, and H444) that are conserved in all CcoN subunits of the cbb3 oxidase, but not in the catalytic subunits of other members of copper-heme superfamily oxidases. H214A mutation of CcoN affected neither catalytic activity nor sensory (signaling) function of the cbb3 oxidase, whereas H320A mutation led to almost complete loss of both catalytic activity and sensory function of the cbb3 oxidase. H233V and H444A mutations brought about the partial loss of catalytic activity and sensory function of the cbb3 oxidase. Interestingly, the H303A mutant form of the cbb3 oxidase retains the catalytic function as a cytochrome c oxidase as compared to the wild-type oxidase, while it is defective in signaling function as an oxygen sensor. H303 appears to be implicated in either signal sensing or generation of the inhibitory signal to the PrrBA two-component system.

Seismic response of steel braced frames equipped with shape memory alloy-based hybrid devices

  • This paper highlights the role of innovative vibration control system based on two promising properties in a parallel configuration. Hybrid device consists of two main components; recentering wires of shape memory alloy (SMA) and steel pipe section as an energy dissipater element. This approach concentrates damage in the steel pipe and prevents the main structural members from yielding. By regulation of the main adjustable design parameter, an optimum performance of the device is obtained. The effectiveness of the device in passive control of structures is evaluated through nonlinear time history analyses of a five-story steel frame with and without the hybrid device. Comparing the results proves that the hybrid device has a considerable potential to mitigate the residual drift ratio, peak absolute acceleration and peak interstory drift of the structure.

Phosphorylation-Dependent Septin Interaction of Bni5 is Important for Cytokinesis

  • In budding yeast, septin plays as a scaffold to recruits protein components and regulates crucial cellular events including bud site selection, bud morphogenesis, Cdc28 activation pathway, and cytokinesis. Phosphorylation of Bni5 isolated as a suppressor for septin defect is essential to Swe1-dependent regulation of bud morphogenesis and mitotic entry. The mechanism by which Bni5 regulates normal septin function is not completely understood. Here, we provide evidence that Bni5 phosphorylation is important for interaction with septin component Cdc11 and for timely delocalization from septin filament at late mitosis. Phosphorylation-deficient bni5-4A was synthetically lethal with $hof1{\Delta}$. bni5-4A cells had defective structure of septin ring and connected cell morphology, indicative of defects in cytokinesis. Two-hybrid analysis revealed that bni5-4A has a defect in direct interaction with Cdc11 and Cdc12. GFP-tagged bni5-4A was normally localized at mother-bud neck of budded cells before middle of mitosis. In contrast, at large-budded telophase cells, bni5-4A-GFP was defective in localization and disappeared from the neck approximately 2 min earlier than that of wild type, as evidenced by time-lapse analysis. Therefore, earlier delocalization of bni5-4A from septin filament is consistent with phosphorylation-dependent interaction with the septin component. These results suggest that timely de localization of Bni5 by phosphorylation is important for septin function and regulation of cytokinesis.

Rhodobacter sphaeroides에서의 광합성유전자(puf, puc, puhA, bchC, bchE, bchF와 bchI)의 발현조절 (Regulation of Photosynthesis Genes (puf, puc, puhA, bchC, bchE, bchF, and bchI) in Rhodobacter sphaeroides)

  • 본 연구에서는 lacZ transcriptional fusion plasmid를 이용하여 광합성 세균인 Rhodobacter sphaeroides에서의 7가지 광합성유전자 (puf, puc, puhA, bchC, bchE, bchF, bchI) 발현의 경향과 조절을 조사하였다. R. sphaeroides에서 puhA와 bchI를 제외한 모든 광합성유전자들이 호기적 조건과 비교했을 때 혐기적 조건에서 더욱 강하게 발현되었다. puhA 유전자는 bchFNBHLM-RSP0290과 operon을 형성하며, bchI 유전자는 crtA와 operon을 이루는 것으로 나타났다. 광합성 조건에서 자란 R. sphaeroides의 puf, puc, bchCXYZ operon의 발현은 빛의 세기에 비례하는 반면, bchFNBHLM(RSP0290 puhA) operon의 발현은 빛의 세기에 반비례 하였다. bchEJG의 발현은 $10\;W/m^2$의 빛이 조사된 광합성 조건에서 제일 낮았으며, $100\;W/m^2$의 빛의 광합성 조건에서 가장 높았다. R. sphaeroides의 산소인지와 빛 인지에 관련된 세 가지 주요 조절기작에 의한 광합성유전자 조절은 다음과 같다. puf와 bchC는 PpsR repressor와 PrrBA two-component system에 의해 조절된다. 그리고 puc operon은 PpsR, FnrL, PrrBA system에 의해 조절된다. bchE의 발현은 FnrL과 PrrBA system에 의해 조절되는 반면, bchF는 오로지 PpsR에 의해서만 조절된다. PpsR repressor는 강한 세기의 빛 조건에서 bchf 발현억제의 원인이 되며, FnrL은 그 자체가 산소를 인지하는 기능 이외에도 세포질의 산화/환원 상태의 인지에 관련될 것으로 보인다.
