Evaluation of Agronomic Characteristics, Nutritional Contents, and Insect Response of the Transgenic Potato Resistant to Glufosinate Ammonium (제초제 저항성 형질전환 감자의 농업적 특성, 영양 성분 및 해충 반응성 평가)

  • The agronomic characteristics, nutritional contents, and insect response of the potato clones transformed with a glufosinate ammonium resistance gene were evaluated. Among the 4 transgenic potato clones, the Bar 3 clone was selected as a promising one for commercialization. The Bar 3 clone showed similar tuber yield capacity but higher herbicide resistance as compared with the non-transgenic potato cv. Dejima. The herbicide resistance of the Bar 3 clone was more than 5 times higher when tested with the herbicide concentration recommended by the producer. The major agronomic characteristics of the Bar 3 clone were not different from those of the non-transgenic Dejima. The annual variation in yields and agronomic characteristics showed similar tendency for 2 years from the third to fourth generation after transformation. The tubers of the Bar 3 clone also showed low occurrence in common scab and physiological disorders such as cracking and secondary growth. But the reasons for such results are yet to be studied. Also, it was considered that the Bar 3 clone have a potential of reducing not only common scab occurrence but also soil erosion during potato cultivation in field. The nutritional contents (mineral compound, vitamin C and amino acid) and response to Spodoptera exigua of the transgenic potato clones were not significantly different.

Efects of Biodegradable Mulching Films Containing Rice Powder on Sweetpotato Growth (쌀 분말이 함유된 생분해성 멀칭필름이 고구마 생육에 미치는 영향)

  • In this study, two types of biodegradable film prototypes were produced using plastic resin containing rice powder. The application of these biodegradable films in sweetpotato (Ipomoea batatas L. Lam) fields and their impacts of plant growth, yield, and the soil environment were assessed, in comparison with Polyethylene (PE) film. The light transmittance of the biodegradable film containing 30% of 350 mesh rice powder (BF30-350RP) was 0.8%, which was lower than the 2.0% light transmittance of the biodegradable film containing 40% of 500 mesh rice powder (BF40-500RP) and 2.7% light transmittance of PE film. Surface temperature measurements on clear day indicated that the PE film exhibited the lowest temperature, with the minimal difference observed between BF40-500RP and BF30-350RP. Assessment of the damage ratio resulting from agricultural work revealed a ranking of 0.4% for the PE film, 3.3% for BF500-400RP, and 5.3% for BF350-30RP. Visible decomposition of BF40-500RP and BF30-350RP commenced after 40 and 30 days of outdoor exposure, reaching 62.3% and 70.4% decomposition at 90 days post-exposure, respectively. The decomposition of biodegradable films applied to sweetpotato fields progressed more slowly in BF40-500RP than in BF30-350RP. The BF40-500RP film on the surface of the ridges was decomposed by 5%, 30%, 55%, and 90% after 30, 60, 90, and 120 days after planting sweetpotato cuttings, respectively. Both types of biodegradable films at the ridge and furrow borders were completely decomposed after 75 days of sweetpotato planting. In a field where the surface was sealed by mulching without growing sweetpotatoes, the soil moisture and its deviation were lower in the order of PE film, BF40-500RP, and BF30-350RP, but the differences were not significant. The soil temperature was higher for PE film mulching than for the biodegradable films containing rice powder, but the differences were small. Two months after sweetpotato planting, the daily average soil moisture decreased by 2.5%point for BF30-350RP mulching, 1.5%point for BF40-500RP mulching, and 1.1%point for PE film mulching over seven days. Soil temperature was similar for both biodegradable film mulches, but increased steadily for the PE film mulch, reaching a daily average of 0.1℃ higher than for the biodegradable films. Sweetpotato vine growth and tuber yield were similar for all the mulching films tested.

Manipulation of Cassava Cultivation and Utilization to Improve Protein to Energy Biomass for Livestock Feeding in the Tropics

  • Cassava (Manihot esculenta, Crantz), an annual tropical tuber crop, was nutritionally evaluated as a foliage for ruminants, especially dairy cattle. Cultivation of cassava biomass to produce hay is based on a first harvest of the foliage at three months after planting, followed every two months thereafter until one year. Inter-cropping of leguminous fodder as food-feed between rows of cassava, such as Leucaena leucocephala or cowpea (Vigna unculata), enriches soil fertility and provides additional fodder. Cassava hay contained 20 to 25% crude protein in the dry matter with good profile of amino acids. Feeding trials with cattle revealed high levels of DM intake (3.2% of BW) and high DM digestibility (71%). The hay contains tannin-protein complexes which could act as rumen by - pass protein for digestion in the small intestine. As cassava hay contains condensed tannins, it could have subsequent impact on changing rumen ecology particularly changing rumen microbes population. Therefore, supplementation with cassava hay at 1-2 kg/hd/d to dairy cattle could markedly reduce concentrate requirements, and increase milk yield and composition. Moreover, cassava hay supplementation in dairy cattle could increase milk thiocyanate which could possibly enhance milk quality and milk storage, especially in small holder-dairy farming. Condensed tannins contained in cassava hay have also been shown to potentially reduce gastrointestinal nematodes in ruminants and therefore could act as an anthelmintic agent. Cassava hay is therefore an excellent multi-nutrient source for animals, especially for dairy cattle during the long dry season, and has the potential to increase the productivity and profitability of sustainable livestock production systems in the tropics.

Effects of Solarization and Green Manual Crops for Promotion of Fall Patato Grawth (가을감자 生産性向上을 위한 綠肥作物栽培와 太陽熱 土壤消毒의 效果)

  • This study was conducted to effect promotion of fall potato growth in the place which green manual crops had been previously cultivated, crushed and tilled and polyethylene transparent vinyl had been mulched from Aug. 5 to Aug. 29 in 1997. The maximum temperature of below-ground at the depth of 10cm was $42^{\circ}C$ during the mulching period, and the pH and the organic compound content of soil was very increased by the cultivation of green manual crops and the mulching of polyethylene transparent vinyl. The total yields of green manual crops which had been previously cultivated were as the following orders; the hybrid of sudan grass, soybean, alfalfa, otherwise, fresh weight of stems and the tuber yield of fall potato were significantly affected in the plot of soybean + fall potato after the soil solarization with the mulching. As these results, it was possible to improve crops growth and to sterilize soil by the mulching treatment of polyethylene vinyl after cultivation of green manual crops during the high temperature of the summer season.

Screening of Herbal Plant extracts Showing Antimicrobial Activity against Some Food Spoilage and Pathogenic Microorganisms (일부 식품 부패성 및 병원성 미생물에 대해 항균활성을 나타내는 생약자원의 검색)

  • This study was carried out to screen herbs among herbal plants showing antimicrobial activity against some food spoilage and pathogenic microorganisms. One hundred fifteen kinds of herbal plants were extracted by 70% ethanol, and then they have been screened for antimicroorganisms. Six herbal plants such as Salviae radix, Dryopteris rhizoma, Terminaliae fructus, Araliae radix, Psoraleae fructus and Schisandrae fructus showed strong antimicrobial activities against Bacillus subtilis. Antimicrobial activities were showed in Anemarrhena radix and Dryopteris rhizoma on Candida albicans, and in Anemarrhenae radix, Dryopteris rhizoma and Polygalae radix on Schizosaccharomyces sp. It was revealed that eight herbal plants such as Dryopteris rhizoma, Salviae radix, Sappan ligunum, Sinomeniae radix, Schisandrae fructus, Rhui fructus, Sophorae radix and Inulae radix also showed antimicrobial activities on Streptococcus mutans. In addition, Anemarrhena radix, Curcuma tuber, Inulae radix, Polygonum radix, Sanguisorbae radix, Scutellariae radix and Terminaliae fructus and showed antimicrobial activities on Trichophyton mentagrophytes. Four kinds of herbal plants such as Dropteris rhizoma, Salviae radix, Terminaliae fructus and Scutellariae radix which showed broad antimicrobial spectrums were mixed by 1 : 1 ratio with the other herbal paints showing relatively strong microbial activities such as Terminaliae fructus, Sinomeniae radix and Scutellariae radix etc. The extracts of mixed herbal paints showed higher antimicrobial activities than those of single herbal plant.

  • The experiment was conducted to evaluate weeding effect and to inhibit tuberization of Eleocharis kuroguwai authority by single or systematic applications of several herbicides. E. kuroguwai was controlled above 90 percentage by pretilachlor and butachlor+bensulfuron application at 5-10 DAT(Days after transplanting) followed by bentazon at 50 DAT and weeded 88 percentage by mefenacet+bensulfuron application at 15 DAT. Tuberization inhibition rate of E. kuroguwai was higher at the application of butachlor+bensulfuron at 5 DAT followed by bentazon at 50 DAT than any other applications. The weight variation of E. kuroguwai tubers formed at 15-20cm soil stratum was larger than that at 10-15cm. Tuber of E. kuroguwai was hardly formed at top soil stratum (0-5cm) and deep soil (15-20cm) by the systematic application of bentazon at 50 DAT. Grain yield of rice from systematic application with bentazon at 50 DAT and mefenacet+bensulfuron at 10-15 DAT was similar to that of hand weeded plot.

Effect of Seedling Quality on the Seedling Raising Period of Stem Cutting and Yield Characteristics of 'Solara' Potatoes in Aeroponics Cultivation (감자 'Solara' 경삽묘의 육묘기간에 따른 묘소질 및 수경재배에서의 수량 특성)

  • This study was conducted to dentify the optimum plantlet type of 'Solara' potatoes (Solanum tuberosum L.) for growth in an aeroponics system. Plantlets of 'Solara' were transplanted on March 16, 2015 in a greenhouse, and growth and yield characteristics were investigated at 70 and 78 days after transplanting, respectively. Stem length was shorter in plantlet of 15-day-old stem cuttings and acclimatization of culture, and the stem length of plantlets of stem cuttings tended to increase with increasing stem cutting age. The fresh weight of plants was the highest in the plantlets of 40-day-old stem cuttings and the lowest in non-rooted stem cuttings and acclimatization of culture. The highest number of first stolons was obtained in 35-day-old stem cuttings. The number of second stolons was the highest in plantlets of 35-day-old stem cuttings, acclimatization of culture, and 30- day-old stem cuttings. The total number of tubers was higher in plantlets of 35-day-old stem cuttings and acclimatization of culture, and the number of tubers above 3 g was the highest in plantlets of 35-day-old stem cuttings. The weight of tubers above 3 g was the heaviest in plantlets of 35-day-old stem cuttings(1,947 g per 10 plants), followed by plantlets of 30-day-old stem cuttings. These results indicate that plantlets of 30 to 35-day-old stem cuttings could be the best for production of 'Solara' potato tubers in an aeroponics system.

Studies on the Mass Propagation of Pinellia ternata (Thunb.) Breit in Vitro (조직배양에 의한 반하〔Pinellia ternata(Thunb.) Briet〕의 대량번식에 관한 연구)

  • In order to find out the best media, explants and environmental conditions for induction of calluses and organogeneses of Pinellia ternata (Thunb.) Breit in vitro, various parts of adult have been cultured on Murashige & Skoog's medium containing various levels of 2,4-dichlorophenoxy acetic acid(2,4-D) and kinetin. The results obtained were as follows: Calluses were induced from the surface of apical meristem and leaf tissue. Formation and growth of calluses in petiole ex plants were best on the MS medium complemented with 2,4-D 2.0 mg/l and kinetin 0.2mg/l. But callus formation in stem ex plants of the nearest tuber was not induced at all kinds of media. Plantlets occured at all treatment except absence of growth regulator. Their numbers, size, leaf and fresh weight were promoted by 2,4-D 2.0mg/l and kinetin 0.2mg/l. Root growth was increased on the medium containing higher 2,4-D concentrations. Size and fresh weight of callus were increased at 25$^{\circ}C$ compared with 10, 20 and 30$^{\circ}C$, respectively. Optimal pH value was at 6.0 for growth of callus. Morphological aberrations were observed in plantlets, especially in regenerated leaves. The separation of the broad leaved plantlets and albino were observed in some cultures. Growth of plantlets after transplantation was best in pots with the sterilized vermiculte. But abnormal variants withered up.

Growth and Biomass Productivity of Seedlings from Seeds in Jerusalem Artichoke (Helianthus tuberosus L.) (돼지감자 실생묘의 생장특성과 바이오매스 생산성)

  • In order to examine the possibility of seed propagation of jerusalem artichoke (Helianthus tuberosus L.) which has been propagated vegatatively so far, it was investigated the growth characteristics of seedlings and the biomass productivity by seed propagation in jerusalem artichoke. The results indicated that the seedlings from seeds exhibited the physiological dwarfing as a rosette habit. The physiological dwarfing as a rosette habit in jerusalem artichoke seedlings from seeds was a kind of appearance affected by photoperiod. This abnormal habit in seedlings from JA6 seeds during the growing period was persisted in short photoperiod of 10 hrs but was disappeared in long photoperiod of 14 hrs. In the condition of long photoperiod, seedlings from seeds grew normally and the average tuber yields obtained from seed propagation was finally 3.51 ton/10a, which can be the satisfying level to the use of seed propagation despite its complexity such as the needs of specific seedling time because it gives a high biomass productivity. This potential utilization of seed propagation in jerusalem artichoke can be basically applied to the collection or preservation of germ plasm and the improvement of jerusalem artichoke.

  • A simulation modeling for predicting the harvesting date with high potato solids consists of development of mathematical models. The mathematical model on potato growth and its development should be obtained by using agricultural elements which analyze relations of solar radiation quantity, temperature, photon quantity, carbon dioxide exchange rate, water stress and loss, relative humidity, light intensity, and wind etc. But more reliable way to predict harvesting date against climatic change employs in vivo energy consumption for growth and induction shape in a slight environmental adaptation. Therefore, to calculate in vivo energy loss, we take a concept of estimate of the amount of basal metabolism in each tuber on the basis of $Wm={\int}^m_tf(x)dt$ and $Tp=\frac{Tm{\cdot}Wm^{Tp}}{Wm^{Tm}}$. In the validation experiments, results of measuring solid accumulation of potato harvested at simulated date agreed fairly well with the actual measured values in each regional field during the growth period of 2005-2009. The calculation method could be used to predict an appropriate harvesting date for a production of high potato solids according to weather conditions.
