Seasonal variations of triterpene glycosides contents in the leaf of Centella asiatica (L.) Urban (병풀 잎에서 triterpene glycosides의 시기별 함량변화)

  • The seasonal variation of two triterpene glycosides, madecassoside and asisaticoside, in the aerial parts of Centella asiatica from May through October was investigated. In leaves, the highest level of triterpene glycosides content was about 108.1 mg/g in September. In petiols, the content was as low as 19.02 mg/g. According to the experimental results obtained, we found that Centella asiatica leaf can accumulate the triterpene glycosides and September is the optimal season for the collection.

Serum Cholesterol Lowering Effect of Triterpene Acetate Obtained from Lactuca indica (왕고들빼기로부터 얻은 Triterpene Acetate의 혈청 콜레스테롤 저하효과)

  • The diets with three types of triterpenoid constituents, which were isolated from Lactuca indica, provoked significant changes of serum lipoprotein-cholesterol meta bolisms of hypercholesterolemic rats induced by high-cholesterol diet, together with the reduction of atherogenic index. Especially, triterpene acetates which have triterpene moieties such as ${\beta}-amyrin$, ${\alpha}-amyrin$, lupeol, pseudotaraxasterol, taraxasterol and germanicol showed a considerable hypocholesterolemic activity. The rat given orally with triterpene acetates did not exhibit a significantly higher value of atherogenic index than that of normal rats.

  • Chromatographic separation of Laduca raddeana extract afforded a mixture of fatty acyl triterpene, triterpene acetates and primary long chain alcohol. The kind of triterpene moieties in these two triter­penoids was six, i.e., ${\beta}-amyrin,\;{\alpha}-amyrin$, lupeol, pseudotaraxasterol, taraxasterol and germanicol on the basis of chemical and spectroscopic methods. The acyl moieties in the corresponding acyl mixture were characterized as acetates, myristate, palmitate, stearate and arachidate. And a mixture of primary long chain alcohol were composed of teracosanol and hexacosanol.

  • A new cytotoxic triterpene hydroperoxide (3) was isolated from the methylene chloride extract of the aerial part of Youngia japonica together with four known triterpenes (1, 2, 4 and 5). Their structures were identified by means of physico-chemical and spectral data to be $\beta$-amyrin acetate (1), taraxasterol (2), 2l$\alpha$-hydroperoxy-taraxasterol (3), oleanolic acid (4) and ursolic acid (5). Compounds 3 and 5 showed moderate cytotoxicity against five tumor cell lines.

  • Along with five known triterpene glycosides, a new triterpene glucosyl ester, named crataegioside, was isolated from the roots of Rubus crataesifolius Bunge. The structure was established as ilexosapogenin A 28-O-$\beta$-D-glucopyranosyl ester by chemical and spectroscopic methods.

  • A triterpene glycoside was first isolated from berries of Rubus coreanus Miquel in the family of Rosaceae, which have been used as a tonic in chinese drug. The compound was identified as $28-{\beta}-D-glycopyranosyl$ ester of $2{\alpha},3{\beta},19{\alpha},23-tetrahydroxyurs-12-en-28-oic$ acid by IR, MS, $^{1}H-NMR$, $^{13}C-NMR$ spectra.

  • A new oleanane-triterpene, 3${\beta}$-acetoxy-11${\alpha}$-benzoyloxy-13${\beta}$-hydroxyolean-1 2-one (1), was isolated along with a known quinone-methide triterpene, pristimerin (2), from the root bark of Siphonodon celastrinneus Griff., a Thai medicinal plant of the family Celastraceae. Their structures were determined based on spectroscopic analysis.

  • Phytochemical analysis on rice bran and its pitch was performed by the tool of TLC, UV- and IR-spetroscopy, so that it was found that they contained ${\gamma}-oryzanol$ and free sterol. GC-MS analysis of free sterol revealed that it was composed of ${\beta}-sitosterol$, campesterol and stigmasterol. Successive phytochemical analysis of ${\gamma}-oryzanol$ revealed that it was composed of ferulic acid ester of triterpene and sterol, respectively. Triterpene moieties of ${\gamma}-oryzanol$ were identified as follows: cycloartanol, cycloartenol, 24-methylenecycloartanol and unknown triterpene; And sterol moieties were found to be identical with free sterols. In addition, characteristic absorption band in UV spectrum (220-340 nm) was exclusively due to ${\gamma}-oryzanol$. Thus, it was suggested that rice brans of nearly all species of Oryza sativa can be quantitatively analyzed by UV absorption spectrometry, even when water soluble pigments was contained in the rice bran.

Analysis of Triterpene Glycoside Levels and Antioxidant Activity in the Different Shoot Tissues of Centella asiatica (L.) Urban (병풀 지상부 조직에서 시기별 triterpene glycoside 함량 및 항산화활성 분석)

  • Centella asiatica is one of the local herbs that is claimed to possess various physiological effects. C. asiatica also accumulates large amounts of pentacyclic triterpenoid saponins known as centelloids. These terpenoids usually include asiatic acid, asiaticoside, madecassoside, and madecassic acids. In the present study, to understand the changes of triterpene glycoside levels in the different shoot tissues of C. asiatica during seasonal cultivation, we investigated the High-Performance Liquid Chromatography (HPLC) analysis via different extraction methods, such as water, 20% ethanol and methanol extracts. Significant increases were observed in the levels of madecassoside and asiaticoside in the leaf extracts by methanol compare with extracts using water or 20% ethanol. Additionally, we also analyzed the various antioxidant activity in the different shoot tissues of C. asiatica using different extracts, such as leaves, petioles and both materials. Among these petiole extracts showed high 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTs) scavenging activity in all extracts, whereas 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity exhibited high activity levels in the leaf part using methanol and ethanol extracts. Levels of total phenolics and flavonoid also showed the highest levels in the leaf tissues using all extracts such as methanol, ethanol and water. Our results indicated that the increased levels of triterpene glycoside and antioxidant activity in the leaf parts of the C. asiatica were indicating that useful metabolites were mainly maintained through seasonal cultivation such as madecassoside, asiaticoside.
