  • The purpose of this study was to investigate the impact DM duration on cardiovascular disease risk factors in Korean Type 2 diabetes mellitus patients. The subject were 250 (male: 134, female: 118) Korean Type 2 DM patients recruited from a general hospital's DM clinic. Anthropometric and hematological variables and dietary intake were assessed. The subject's mean age was 60.2 years and duration of diabetes was 9.5 years. As far as DM duration was concerned, male subjects with long duration (${\geq}$ 5 years and longer) showed significantly lower weight, fat free mass, skeletal muscle mass, BMI (p < 0.05) and higher 2hrs-postprandial plasma glucose (p < 0.05) and concentration of Hemoglobin A1c (p < 0.01) compared to those with less 5years of duration. In female subjects, same trend was found. Female subjects with long duration (${\geq}$ 5 years and longer) showed significantly higher fat free mass (p < 0.05) and skeletal muscle mass (p < 0.05), Hemoglobin A1c (p < 0.01), HDL-cholesterol (p < 0.01) and triglyceride (p < 0.05), dietary intakes of protein (p < 0.05) and fat (p < 0.05) compared to those with less 5years of duration. In conclusion, Type II DM patients of long duration had higher blood lipid concentrations and higher carbohydrates intakes. Subjects followed diet prescription strictly showed lower blood lipid concentration, suggesting the effectiveness of diet to manage Type 2 DM patients.

  • BACKGROUND/OBJECTIVES: Heavy alcohol consumption causes the development of alcoholic liver disease (ALD), a neglected but important public health problem. Many studies have pointed out that probiotics could improve gut health, which is also considered to be a cause of ALD. Therefore, this study screened the probiotics, Lactobacillus casei GKC1 (GKC1), L. fermentum GKF3 (GKF3), Bifidobacterium lactis GKK2 (GKK2), L. rhamnosus GKLC1 (GKLC1), L. paracasei GKS6 (GKS6), and L. plantarum GKM3 (GKM3), for their potential benefits in alleviating ALD for applications to disease prevention. SUBJECTS/METHODS: C57BL/6N mice were divided into 8 groups (n = 6 in each): normal control, positive control (alcohol-diet fed), and treatments of feeding probiotics GKC1, GKF3, GKK2, GKLC1, GKS6, and GKM3 under an oral dose 0.82 g/kg B.W. per day by oral gavage. The experiment was conducted for 8 weeks, and the concentrations of alanine aminotransferase (ALT), aspartate aminotransferase, triglyceride (TG), and total cholesterol (TC) in mice were measured. The glutathione (GSH), catalase (CAT), and histology were analyzed after sacrifice. RESULTS: The results showed a decrease in the serum ALT, liver TG, and liver TC levels in the GKS6, GKM3, and GKLC1 groups compared to the positive control. In addition, the decreasing GSH and CAT levels were inhibited in the GKS6 and GKM3 groups. The histopathological results showed that all probiotics could reduce the accumulation of liver fat. Furthermore, there was a significant difference in GKLC1 with lower stomach damage compared to the alcohol-fed mice without any addition of probiotics. CONCLUSIONS: GKLC1, GKS6, and GKM3 can be used as supplements for alleviating the development of ALD.

  • The objective of this study was to examine diet-induced changes in the expression of UCP2 mRNA in the liver and UCP3 mRNA in the skeletal muscle of mice fed a high-sucrose or high-fat diet. Male ICR mice, aged 4 weeks, were divided into three dietary groups and fed control (N) or modified AIN-76 high-sucrose (US) or high-fat (HF) diets for 12 weeks. The serum total cholesterol (TC) and LDL-cholesterol concentrations of the HF group were significantly higher than those of the N and HS groups. The hepatic TC and triglyceride contents of the HS and HF groups were also significantly higher than those of the N group. The HS diet group had higher serum leptin and insulin levels compared to those of the HF group. Hepatic UCP2 mRNA expression was significantly higher in the HS group than in the N group, but the level in the HF group did not differ from that of the N group. Muscular UCP3 mRNA level was significantly higher in the HF group and especially in the HS group than in N the group. We observed that two gene (UCP2, 3) levels exhibited a similar tendency. These results suggest that UCPs mRNA levels and energy expenditure may be altered or controlled by various dietary patterns. Further research is needed to elucidate the effects of diet on the regulation of many obesity-related genes.

  • The purpose of this study was to assess the effects of Cordyceps militaris extract supplementation on the improvement of blood glucose, lipid compositions and hepatic functional enzyme activities in the serum of streptozotocin(STZ, 50 mg/kg BW, IP injection)-induced diabetic rats fed the experimental diets for 5 weeks. The concentrations of blood glucose and lipid compositions(total cholesterol, ${\beta}$-lipoprotein, free cholesterol, cholesteryl ester, triglyceride and phospholipid) and atherosclerotic index in serum were significantly lower in the DM+CM group than in the DM group, whereas the ratio of HDL-cholesterol concentration to total cholesterol in the DM+CM group were higher than in the DM group. The activities of hepatic functional enzymes(aminotransferase, LDH and ALP) in serum were lower in the Cordyceps militaris extract administration group(group DM+CM) than in the hyperglycemic DM group. The results shown above suggested that Cordyceps militaris extract supplementation effectively improves blood glucose and atherosclerosis in the serum of diabetic rats.

  • In this study, we investigated the dose-dependent effects of mixtures of Atractylodes macrocephala (AM) and Amomum villosum (AV) water extracts in a ratio of 3:1 on high fat diet (HFD)-induced obesity model. Oral administration of various concentrations with mixtures of AM and AV extracts in a ratio of 3:1 for 6 weeks inhibited HFD-induced increases of body, liver and epididymal fat weights in a dose-dependent fashions. Those effects may be mediated by decreased expressions of lipogenesis-related genes such as acetyl coA carboxylase (ACC) and fatty acid synthase (FAS) in liver. Also, increase of insulin and decrease of adiponectin in serum by HFD supply were inhibited by three different dosages of mixtures of AM and AV extracts in a ratio of 3:1. HFD supply induced increases of serum total cholesterol, triglyceride and LDL cholesterol. However, hyperlipidemia was significantly decreased in dose-dependent manners by treatment with mixtures of AM and AV extracts. Based on the results of the present study, hypolipidemic and anti-obesity effects by mixtures of AM and AV extracts were found in HFD-induced obesity model. Further clinical investigation is needed to develop anti-obesity therapeutic or preventive agents by using mixtures of AM and AV extracts.

  • This study was performed to evaluate the biological activity and protective effect of Cassia tora ethanol extracts against D-galactosamine induced hepatotoxicity in rats. Male Sprague-Dawley rats were grouped into normal group, D-galactosamine treated group(control), D-galactosamine plus 0.25% Cassia tora extracts treated group and D-galactosamine plus 0.5% Cassia tora extracts treated group. Normal and control group were fed control diet and Cassia tora extracts treated groups were fed experimental diets containing 0.25% or 0.5% Cassia tora ethanol extracts for 5 weeks. Body weight gain and liver weight of rats were not significantly different between groups. Cholesterol and triglyceride concentrations in serum and liver were significantly lower in rats treated only with D-galactosamine compared to normal group, and improved in Cassia tora extracts supplemented rats. D-galactosamine treatment significantly increased serum aspartate transaminase, alanine transaminase, gamma glutamyl transferase and alkaline phosphatase, however, the activities of aspartate transaminase and alanine transaminase were significantly decreased in Cassia tora extracts supplemented rats when compared with D-galactosamine treated control group. Cassia tora extracts significantly suppressed the D-galactosamine-induced elevation of liver thiobarbituric acid reactive substances(TBARS) contents. Superoxide dismutase activity was decreased by D-galactosamine treatment, however by the supplementation of Cassia tora ethanol extracts, significantly increased in dose-dependent manner. Glutathione peroxidase activity in rats fed diets containing Cassia tora extracts was decreased compared to control. Based on these results, we concluded that Cassia tora ethanol extracts may prevents the D-galactosamine-induced hepatotoxicity probably via an antioxidant mechanism.

  • This study is performed to estimate the prevalence of metabolic syndrome among male workers and to identify the relationships with many related factors including anthropometry, hematological index, serum lipid level, dietary-related behaviors and health-related behaviors. According to the age groups, the 20s are significantly higher in normal and risk groups than in the metabolic syndrome (MS) group, the 30s are significantly higher in MS group than the other groups. The levels of AST and ${\gamma}$-GTP both show significant differences in the order of MS group ($30.3{\pm}8.8U/l$, $91.1{\pm}40.2U/l$) > risk group ($25.7{\pm}8.1U/l$, $41.8{\pm}20.2U/l$) > normal group ($22.8{\pm}6.0U/l$, $26.6{\pm}10.7U/l$). For the frequency of breakfast consumption, the response of 'Every day' is significantly higher in MS group than normal and risk groups, but the response of 'Not at all' is significantly higher in normal group than MS and risk groups. The drinking amount is positively correlated with ${\gamma}$-GTP in normal group, and it is negatively correlated with the hematocrit level, but it is positively correlated with the systolic blood pressure in MS group. AST is positively correlated with glucose concentrations of the MS group. The ALT is positively correlated with waist circumferences and systolic blood pressure in the risk group. The results of this study show that breakfast frequency, education level, drinking amount, drinking frequency, exercise frequency, AST, ALT and ${\gamma}$-GTP levels are all important risk factors of MS. Therefore, it is very important to maintain a healthy life style for the prevention of MS incidence.

  • Two experiments were conducted to evaluate the use of various levels of full-fat sunflower seeds (FFSS) on broiler performance and carcass characteristics. In the first experiment, FFSS was included in a basal diet at 70, 140, and 210 g/kg and the $AME_n$ values of the experimental diets were determined. The linear regression equation of $AME_n$ values on rate of inclusion was calculated. Extrapolation value for the $AME_n$ of FFSS at 100% inclusion was 14.22 MJ/kg. In the second experiment, diets containing various levels (0, 70, 140, and 210 g/kg) of FFSS were given to broilers (Ross strain) from 0 to 49 d. At 28 days of age, blood parameters and digestive enzyme activities were determined and carcass parameters were evaluated at 49 days of age. Weight gain, feed intake and feed conversion ratio (FCR) were improved (p<0.05) when broilers were fed various levels of FFSS in the starter and finisher diets. Breast, thigh, gastrointestinal tract and gizzard weight percentages were not affected by dietary treatments; however, liver weight percentage was decreased significantly (p<0.05) and weight of abdominal fat decreased but this effect was not significant. The activities of digestive enzyme (protease and ${\alpha}$-amylase) were not influenced by the treatments. Activity of alkaline phosphatase, concentrations of calcium, phosphorus, glucose, triglyceride, protein, high density lipoprotein (HDL) and low density lipoprotein (LDL) were not affected by incorporation of FFSS in the broiler diet. Although concentration of HDL increased and LDL decreased, these effects were not significant. The results of this study indicate that FFSS can be used at up to 21% in broiler diets without adverse effects on performance or other parameters of chickens.
