Development of Early Maturing Rice Stripe Virus Disease-Resistant 'Haedamssal' through Marker-Assisted Selection (MAS를 이용한 줄무늬잎마름병 저항성 조생종 벼 '해담쌀' 개발)

  • 'Haedamssal' is an early maturing and rice stripe virus disease-resistant cultivar adaptable for early-transplanting cultivation that was developed by the rice breeding team of the Department of Southern Crop, NICS, RDA, in 2014. This cultivar was derived from the cross YR25869 (YR21247-B-B-B-49-1/Sasanishiki BL4//Koshihikari) and YR25868 (Unkwang//YR21247-B-B-B-49-1/Sasanishiki BL4) made in the 2005/2006 winter season and was advanced to the F5 generation by a bulk breeding method using rapid generation advance. To incorporate rice stripe virus resistance, marker-assisted selection on the RSV gene was conducted in 3-way and 6-way cross F1 generation using the tightly linked marker RM6897. From testing in the replicated yield trial in 2011, a promising line YR26258-B-B-B-33-3 was selected and it was designated as 'Milyang276'. A local adaptability test of 'Milyang276' was performed at three locations from 2012 to 2014 and it was named as 'Haedamssal', which was a good eating quality variety. The culm length was 67 cm in yield trials, which was 4 cm shorter than 'Jopyeong'. The number of spikelets per panicle was lower than 'Jopyeong', whereas the number of tillers per hill was higher. This variety was resistant to RSV disease, bacterial blight, and leaf blast disease. The milled rice yield of 'Haedamssal' was 5.48 MT per ha at the early transplanting in the local adaptability test. 'Haedamssal' is well adapted to early transplanting cultivation in the southern plain area (Registration No. 6811).

  • Objective : Many recent reports have shown that the mature mammalian brain harbors multipotent stem cells, rendering the brain capable of generating new neurons and glia throughout life. Harvested stem cells from an adult rat are transplanted in order to evaluate the cell survival and differentiation. Methods : Using a percoll gradient with a high speed centrifugation method, we isolate neural stem/progenitor cells were isolated from the subventricular zone[SVZ] of a syngeneic adult Fisher 344 rats brain. For 14days expansion, the cultured cells comprised of a heterogeneous population with the majority of cells expressing nestin and/or GFAP. After expanding the SVZ cells in the presence of basic fibroblast growth factor-2, and transplanting then into the hippocampus of normal rats, the survival and differentiation of those cells were examined. For transplantation, the cultured cells were labeled with BrdU two days prior to use. In order to test their survival, the cells were transplanted into the dorsal hippocampus of normal adult Fisher 344 rats. Results : The preliminary data showed that at 7days after transplantation, BrdU+ transplanted cells were observed around the injection deposition sites. Immuno-fluorescent microscopy revealed that the cells co-expressed BrdU+ and neuronal marker ${\beta}$-tubulin III. Conclusion : The data demonstrate that the in vitro expanded SVZ cells can survive in a heterotypic environment and develop a neuronal phenotype in the neurogenic region. However more research will be needed to examine the longer survival time points and quantifying the differentiation in the transplanted cells in an injured brain environment.

Number of sampling leaves for reflectance measurement of Chinese cabbage and kale

  • Objective of this study was to investigate effects of pre-processing method and number of sampling leaves on stability of the reflectance measurement for Chinese cabbage and kale leaves. Chinese cabbage and kale were transplanted and cultivated in a plant factory. Leaf samples of the kale and cabbage were collected at 4 weeks after transplanting of the seedlings. Spectra data were collected with an UV/VIS/NIR spectrometer in the wavelength region from 190 to 1130 nm. All leaves (mature and young leaves) were measured on 9 and 12 points in the blade part in the upper area for kale and cabbage leaves, respectively. To reduce the spectral noise, the raw spectral data were preprocessed by different methods: i) moving average, ii) Savitzky-Golay filter, iii) local regression using weighted linear least squares and a $1^{st}$ degree polynomial model (lowess), iv) local regression using weighted linear least squares and a $2^{nd}$ degree polynomial model (loess), v) a robust version of 'lowess', vi) a robust version of 'loess', with 7, 11, 15 smoothing points. Effects of number of sampling leaves were investigated by reflectance difference (RD) and cross-correlation (CC) methods. Results indicated that the contribution of the spectral data collected at 4 sampling leaves were good for both of the crops for reflectance measurement that does not change stability of measurement much. Furthermore, moving average method with 11 smoothing points was believed to provide reliable pre-processed data for further analysis.

Estimation of Rice-Planted Area using Landsat TM Imagery in Dangjin-gun area (Landsat TM 화상을 이용한 당진군 일원의 논면적 추정)

  • For estimating paddy field area with Landsat TM images, two dates, May 31, 1991 (transplanting stage) and August 19, 1991 (heading stage) were selected by the data analysis of digital numbers considering rice cropping calendar. Four different estimating methods (1) rule-based classification method, (2) supervised classification(maximum likelihood), (3) unsupervised classification (ISODATA, No. of class:15), (4) unsupervised classification (ISODATA, No. of class:20) were examined. Paddy field area was estimated to 7291.19 ha by non-classification method. In comparison with topographical map (1:25,000), accuracy far paddy field area was 92%. A new image stacked by 10 layers, Landsat TM band 3,4,5, RVI, and wetness in May 31,1991 and August 19,1991 was made to estimate paddy field area by both supervised and unsupervised classification method. Paddy field was classified to 9100.98 ha by supervised classification. Error matrix showed 97.2% overall accuracy far training samples. Accuracy compared with topographical map was 95%. Unsupervised classifications by ISODATA using principal axis. Paddy field area by two different classification number of criteria were 6663.60 ha and 5704.56 ha and accuracy compared with topographical map was 87% and 82%. Irrespective of the estimating methods, paddy fields were discriminated very well by using two-date Landsat TM images in May 31,1991 (transplanting stage) and August 19,1991 (heading stage). Among estimation methods, rule-based classification method was the easiest to analyze and fast to process.

Transition of Rice Culture Practices during Chosun Dynasty through Old References. Ⅵ Fertilizing Practices (주요 고농서를 통한 조선시대의 도작기술 전개 과정 연구 Ⅵ. 시비기술)

  • Different from 'Seyokwiryang' (歲易爲良 : fallowing as the best method) which was an agricultural technique of ancient China, fertilization of rice fields was already practiced in the end of Koryo age in Korea. 'Bunjongbeob'(糞種法 : fertilizer applicating method on seed) or 'Bunkwabeob' (糞科法 : fertilizer applicating method on each plant) was practiced before green manure of bean crops and 'Bunjeonbeob'(糞田法 : fertilizer practicing method on fields) were done. In the 15th century 'Dojeonbunjeonbeob'(稻田糞田法 : fertilizer applicating method on paddy) in 'Jikseol' was divided the materials to be used into the soil brought from another place, trees, grasses, and the manure. Also, it discribed the fertilization between first plowing and the second, and proper fertilization for particular soil conditions. In case of transplanting techniques, the fertilization practices were specified into nursery and rice fields, and restoration of organic matter was systemized by plowing for cultivation in the reclaimed areas. In the 17th century, through 'Jikseolbo'($\ulcorner$直說補$\lrcorner$), the habitual practice of Kyungsang province was systemized and 'Bunyangsool'(糞壤術 : technique of fertilization) of 'Jodoangkicheo' (早稻秧基處 : rearing fields of early-ripening rice) was completed. Specific things was the manufacturing and utilizing techniques of 'Bunhoe'(糞灰 : mixture of manure and ash), 'Yohoe'(尿灰 : mixture of urine and ash), and additional fertilizers. In the 18 to 19th century, the materials of fertilization were greatly enlarged to recover the waste lands and to support the reinforcement of soil fertility for increasing the system of two cropping a year. Also, 'Jeobunbeob'(貯糞法 : method of manure storage) and additional fertilization were emphasized, and use of wagons for it was emphasized to improve the theory of fertilization and working efficiency. As mentioned above, limitation of fertilizing materials was conquered by 'Dojeonbunyang'(稻田糞壤 : techniques of practicing fertilizers in paddy) and the system of additional fertilization was established. The fertilization methods were improved with 'Jeobunbeob' due to the theory and recognition necessary for high rates of fertilizers.

Gibberellin Application Method and Concentration Affect to Growth, Runner, and Daughter Plant Production in 'Maehyang' Strawberry during Nursery Period (육묘기 '매향' 딸기의 생육, 런너 및 자묘 생산에 미치는 지베렐린 처리방법 및 농도의 영향)

  • This study was aimed to evaluate the effect of application method and concentration of gibberellin $A_3$ ($GA_3$) on the growth, runner production, and seedling quality of strawberry plants (Fragaria ${\times}$ ananassa Duch. cv. Maehyang) during nursery period. The mother plants of strawberry were transplanted in pot ($64{\times}27{\times}18cm$) filled with commercial growing medium on March 20, 2018. $GA_3$ concentration was applied as 0, 50, 100 or $200mg{\cdot}L^{-1}$ with spray or drench to 45 mL per plant at 4 weeks after transplanting, respectively. Nutrient solution was supplied with the EC $1.5dS{\cdot}m^{-1}$ after the transplanting and supplied 350 mL per pot twice a day (15 min per one time) after rooting. The growth characteristics of mother plants of strawberry were measured at 7 weeks after treatment, and growth characteristics of daughter plants of strawberry were measured at 10 weeks after treatment. Runner length and diameter of mother plant was the longest or thickest in the spray with $200mg{\cdot}L^{-1}$ than the other treatments, respectively. Soil-plant analysis development (SPAD) value of mother plant was the lowest in spray with $200mg{\cdot}L^{-1}$. However, leaf length, leaf width, and crown diameter showed no significant differences in all treatment among application method and concentration of $GA_3$. As the concentration of $GA_3$ increased, physiological disorder like stretchiness of crown occurred more. The physiological disorder was the most occurred in spray treatment with $200mg{\cdot}L^{-1}$, but drench treatment occurred less than spray treatment. The number of runners and daughter plants increased with increasing concentration of $GA_3$ regardless of application methods. In the growth characteristics of the daughter plants, leaf length and leaf width of first daughter plant, plant height, crown diameter, leaf area and SPAD value of second daughter plant, and plant height of third daughter plant were the significantly greatest in drench with $100mg{\cdot}L^{-1}$ treatment. This results indicate that growth and runner production of mother plants and growth of daughter plants of strawberry were the best achieved by drench application in the $100mg{\cdot}L^{-1}$ $GA_3$.

Sowing Method in Plug Tray for Production of Plug Seedlings of Rehmannia glutinosa (Gaertn.) Libosch. ex Steud. (지황의 공정묘 생산을 위한 플러그 트레이 파종 방법)

  • Conventionally, the seeds of Rehmannia glutinosa (Gaertn.) Libosch. ex Steud. have been directly sown at the field without using the plug seedling method. Plug seedlings have the advantage of promoting germination and convenient transplanting. However, there is little information about propagation of R. glutinosa using the plug seedling method. This study was conducted to investigate the optimal seed rhizome length, diameter, and sowing direction of R. glutinosa for establishing the plug seedling method. Seed rhizome length and diameter were separated by 1, 2, 3 cm and 0.3-0.5, 0.6-1.0, 1.1-1.5 cm, respectively. And seed rhizomes were sown in vertical and horizontal directions. The survival rate in 1 cm length of seed rhizomes was lower than in other treatments. The leaf length, leaf width, number of leaves, SPAD, leaf area, and fresh and dry weights of shoot and roots were the greatest in 3 cm length of seed rhizomes. As the seed rhizome diameter decreased, the growth characteristics of R. glutinosa tended to increase. When R. glutinosa was sown horizontal direction, the leaf length, leaf width, number of leaves, and leaf area were significantly higher than in the vertical direction. In conclusion, when sowing seed rhizome in a plug tray, using a length of 3 cm, a diameter of less than 1 cm, and sowing in a horizontal direction is considered an appropriate sowing method for R. glutinosa.

Studies on Direct Sowing-Dry Paddy Rice Culture in the Middle Part of Korea (중부지방에 있어서의 수도건답직파재배 기술체계확립에 관한 시험연구)

  • Experiments on rice concerning it s varieties, fertilization, seedling dates and herbicides have been conducted to determine the most desirable method of direct sowing cultivation on dry paddy field land in the middle part of Korea. The results obtained at the Office of Rural Development of Choongnam Province are as follows:. 1. Sixteen different derivatives from the main varieties of low land rice were cultivated on a dry paddy field by the direct sowing method; at the same time, a few varieties were tried adopting the common transplanting cultivation method. The yield and yield factors from these two groups were examined to give the following results: a) Compared to the common transplanting cultivation, the direct sowing method showed remarkably increased number of panicles while the number of flowers per panicle was shown to be significantly decreased. The maturing ratio was detected to be lowered. The yield horn them differed according to the different varieties : good yield was obtained from Hokwang while Norin #25 proved poor when compared with the common transplanting cultivation method. b) Among sixteen varieties tested, Sunsou, Norin #25, Jaigou, Hokwang, Palkueng and Gosi showed comparatively high yields, their yield being more than 325 kilograms per 10 Are, but Nampoong, Paldal, Nongkwang, Norin #29, Eunbangju #101 and Shiro gane showed less yield, their yield being less than 271 kilograms per 10 Are, the relations between the yield and the yield factors can be summarized as follows; Number of varieties and their rice yield. 1) The varieties which were great in the, number of panicles and high in yield=Jaigoun, Hokwang Palkueng and Gosi. 2) The varieties which were low in the number of panicles and high in yield=Sounsou and Norin #25. 3) The varieties which were great in the number of panicles and poor in yield=Eunbangju #101 and Sirogane. 4) The varieties which were poor in the number of panicles and poor in yield: Nampung, Paldal and Norin #29. Number of flowers per panicle and yield. 1) The varieties which were great in the number of flowers per panicle and high in yield: Sounsou, Norin #25 and Gosi. 2) The varieties which were poor in the number of flowers per panicle and high in yield ; Jaigoun, Hokwang and Palkueng. 3) The varieties which were great in the number of flowers per panicle and poor in yield: Paldal and Nampung. 4) The varieties which were poor in the number of flowers per panicle and poor in yield: Norin #29. Eunbangju #101 and Sirogane. Maturing ratio and yield. 1) The varieties which were high in the maturing ratio and high in yield: Jaigoun, Sounsou, Norin #25 and Palkueng. 2) The varieties which were low in the maturing ratio and high in yield: Hokwang and Gosi. 3) The varieties which were early maturing rat io and low in yield: Hokwang and Gosi. 4) The varieties which were late maturing ratio and poor in yield: Eunbangju #101, Nampungand Sirogane 1, 000 grain weight and yield. 1) The varieties which were heavy in 1, 000 grains weight and high in yield=Norin #25 and Hokwang. 2) The varieties which were light in 1, 000 grains weight and high in yield=Sounsou and Jaigoun. 3) The varieties which were heavy in 1, 000 grains weight and poor in yield=Nongkwang and Eunbanju. 4) The varieties which were light in 1, 000 grains weight and poor in yield=Norin #29 and Sirogane. 2. The experiment on fertilization showed that the most desirable amount to be given per 10 Are was 10 kilograms of Nitrogen, 5 kilograms of phosphate and 6 kilograms of potassium; and when the Nitrogen given exceeded 8 kilograms, its effect was better when given in amsll consecutive (split) amounts, while the maturing ratio and the number of the flowers per panicle increased when Nitrogen was given in large amount during the later stage of growth of rice. 3. The experiment on the date and amount of seedling showed that the tested variety, Sunsou gave the best results when planted on the days between 25 April and 10 May. Eight liters per 10 Are were preferable if planted early and 12 liters per 10 Are if planted late. The reason why the later planting gave a lower yield was that the number of flowers per panicle was fewer. 4. The experiment on the irrigation for rice with direct sowing cultivation immersed in water showed that it was the most satisfactory when irrigated on 25th June, 55 days after its seedling, its plot giving the best yield. The plots 10th June and 15th July showed just as good results. However, irrigated later, than 15th July it showed lower yields. 5. Compared to the yield of the plot controlled by the common method, the yield from the plots treated with chemical herbicide such as LOROX, TOK, PCP, SWEP, Mo-338 on dry condition soil seemed poorer, but significant difference was not found statistically. On the other hand in the case where chemical herbicides such as TOK, Mo-338, Stam F-34 or ORDRAM were used after irrigation, the yield from the ORDRAM and TOK treated plots did not show significant differences compared to the common hand weed controling method, but those treated with chemicals other than the above showed a lower yield.

Effect of the Whole-layer Application of Slow-release Fertilizer on Growth and Yield of Rice (완효성비료의 전층시비가 벼 생육과 수량에 미치는 영향)

  • This experiment was carried out to find out the optimum application method of slow-release fertilizer under different soil textures, clay loam(Deogpyeong series), sandy loam (Gangseo series) in the southern region(Milyang, Kyeongnam) of Korea. The fertilizers used were Chosun slow-release fertilizer(silicate latex coated fertilizer: N-P$_2$O$_{5}$-K$_2$O=18-12-13) and Meister 15 (thermoplastic resin coated fertilizer : N-P$_2$O$_{5}$-K$_2$O=14-14-14). The two whole basal application methods such as side band placement at transplanting time of rice plant(cv.Donghaebyeo) and incorporation with soil as basal were tested. The released amount of ammonium nitrogen from the soils for 4 days submerged was 95ppm in sandy loam and 60ppm in clay loam. The greenish degree of rice leaf was higher at the whole plow layer placement method than the others. The nitrogen efficiencies of the fertilizers were Meister 15 > convention > Chosun in order and between the application methods were similiar. The growth status was better at the whole plow layer placement application method of Meister 15 regardless of soils and seedling ages. The ripening ratio was increased at slow-release fertilizer application, and between application methods, whole plow layer placement was lower because of lodging damage. The yield in clay loam soil showed a significant difference between the ferilizers, but there were slight differences between the application methods and fertilizers in sandy loam soil. As a result, the slow-release fertilizer, Meister 15, applied at the whole layer showed a good rice growth and seemed a plausible fertilizing method.

Acclimation of Ectomycorrhizal Pine (Pinus densiflora) Seedlings Inoculated with Tricholoma matsutake by the Treatment of PDMP and Tween Solutions (송이균 접종으로 외생균근을 형성한 소나무 묘목의 PDMP 및 Tween 용액 처리에 의한 순화)

  • Trials to find a method for overcoming the depletion of Tricholoma matsutake mycorrhiza from the artificially inoculated pine seedling (Pinus densiflora) when it is transplanted in situ were carried out. The culturing jars containing mycorrhizal seedlings were punched at the bottom to make the jar like a pot with keeping the soil structure of the original medium in vitro, and 8 kinds of irrigation solution were applied. Irrigation of distilled water caused disappearance of T. matsutake mycorrhizae, but biweekly irrigation of 2.5% PDMP (24 g potato dextrose broth, 3 g malt extract, 1 g peptone) solution or 0.5% Tween 80 solution until 3 months of acclimation could keep mycorrhizal association stronger. The percentage of mycorrhizal roots out of total length of roots was increased during the acclimation, and the acclimation rate reached up to 70%. Thus, we recommend that the mycorrhizal pine seedlings inoculated with T. matsutake would be acclimated before transplanting into the field.
